Effect of Dietary Fatty Acids on Fatty Acid Composition of Platelet Phospholipids, Thromboxane B2 Formation, and Platelet Aggregation in Men (식이 지방산이 혈소판 인지질의 지방산 조성, 혈장 Thromboxane B2의 농도 및 혈소판 응집에 미치는 영향)

  • The degree of platelet aggregation, thromboxane B2(TXB2)formation and fatty acid composition of platelet phospholipids(PL) were investigated in 24 healthy male subjects who for five weeks consumed either corn oil(CO) rich in linoleic acid(LA), perilla oil (PO) rich in $\alpha$-linoleic acid($\alpha$-LAN), or canola oil(CNO) rich in oleic acid(OA) as a major fat source. Total fat intake was 30% of total calories and prescribed oil intake of each dietary group was 50% of the total fat intake. In the CO group, significantly decreased contents of polyunsaturated fatty acids(PUFA), n-6 PUFA, n-3 PUFA and eicosapentanoic acid(EPA) were observed, and significantly increased contents of OA and saturated fatty acids(SFA) were observed in platelet PL after 3 weeks and 5 weeks of dietary treatment. In the PO group, contents of OA and docosahexanoic acid(DHA) were increased, and the ratio of n-6/n-3 was decreased significantly in platelet PL after dietary treatment. The CNO group showed significatnlty decreased contents of PUFA, P/S ratio, n-6 PUFA, LA,(EPA+DHA)/arachidonic acid(AA), and significantly increased SFA contents after 3 weeks of the oil-based diet. The dietary-induced effects on fatty acid composition of platelet PL were observed mostly after 3 weeks of the oil-based diet. The dietary-induced effects on fatty acid composition of platelet PL were observed mostly after 3 weeks. Plasma TXB2 levels were increased after 3 and 5 weeks of dietary treatment. However, only the CO and CNO groups showed significantly increased plasma TXB2 levles after 3 and 5 weeks of dietary treatment. However, only the CO and CNO groups showed significantly increased plasma TXB2 levels after 5 weeks of experimental diets, when compared with initial values. Degree of platelet aggregation increased only in the CO group after dietary treatment. As a result, at week 5 the degree of platelet aggregation of the CO group was significantly higher than those of the PO and CNO groups. Among the three oil-based diets, the PO-based diet seems to have beneficial effects on atherosclerosis by influencing plasma TXB2 levels and the degree of platelet aggregation, while the CO-based diet showed the most adverse effects. Our results imply that plasma TXB2 levels might be affected by dietary fatty acid composition.

Anti-microbial and Anti-thrombosis Activities of Lees of Sweet Potato Soju (고구마 소주 주박의 항균 및 항혈전 활성)

  • Sweet potato soju (SPS), a form of traditional distilled alcoholic liquor in Korea, is manufactured by the distillation of fermented broth under normal pressure, thus providing it for a uniquely smooth taste infused with the flavor of sweet potato. After distillation, the lees of SPS is produced as by-product and discarded. In this study, the ethanol and hot water extracts of lees of SPS, and their subsequent organic solvent fractions using hexane, ethylacetate (EA), butanol, and water residue were prepared in an effort at the efficient re-use of the lees of SPS. The ethanol extraction yield was 1.36-fold higher than that of the hot water extraction, and the EA fraction revealed the highest total polyphenol content among the solvent fractions. The various extracts and solvent fractions did not demonstrate hemolytic activity at up to 0.5 mg/ml concentrations against human red blood cells. In the bioactivity assay, only the EA fraction displayed a broad spectrum of anti-microbial activity against different pathogenic and food spoilage bacteria, and demonstrated significant anti-coagulation activity by inhibitions of thrombin, prothrombin and blood coagulation factors. Furthermore, only the EA fraction from the hot water extract of the lees of SPS showed anti-platelet aggregation activity, which is comparable to aspirin (a commercially available drug). Our results suggest that the EA fraction of the hot water extract prepared from the lees of SPS has a high potential as a novel resource for anti-microbial and anti-thrombosis agents.

Anti-platelet Aggregation Effect of Cheongpyesagan-tang In Vitro (청폐사간탕(淸肺瀉肝湯)의 혈소판 응집억제 작용에 대한 in vitro 연구)

  • Objective : The study was designed to test the anti-platelet effect of the extract Cheongpyesagan-tang and compare it with aspirin in vitro. Methods : The extract from Cheongpyesagan-tang was made by the pharmacy department of Kyung Hee Oriental Medical Hospital. The extract was investigated for inhibition against the collagen induced aggregation of human platelet suspensions on aggregometry. Aspirin and aspirin-Cheongpyesagan-tang were investigated together. Results : 1. In collagen induced human platelet aggregation test, the extract from Cheongpyesagan-tang significantly inhibited in concentration 30mg/ml (p<0.05), 40mg/ml, 50mg/ml (p<0.001) and the effect depended on concentration over 20mg/ml. 2. Aspirin and aspirin-Cheongpyesagan-tang inhibited collagen induced human platelet aggregation significantly (p<0.001). Aspirin-extract of Cheongpyesagan-tang inhibition rate was higher than aspirin only (p<0.05). Conclusions : The extract of Cheongpyesagan-tang has anti-platelet aggregation and synergic effect with aspirin on human platelet in vitro.

Anti-oxidation and Anti-thrombosis Activities of Different Parts of Salicornia herbacea L (함초의 부위별 항산화 및 항혈전 활성)

  • This study was designed to develop the functional beverage using Salicornia herbacea L (SH). To evaluate the anti-oxidation and anti-thrombosis activities of SH, the ethanol extracts of leaf (LF), stem (ST) and seed (SD) of SH were prepared. Components analysis showed the total polyphenol of LF > ST > SD, and the total flavonoid of LF > SE > ST, respectively. Amounts of total sugar and reducing sugar of LF and ST were similar, but the SD had only 1/10 of sugar contents of LF. Anti-oxidation activities of each extract determined by calculated RC50 showed LF > SD > ST for DPPH anion, nitrite and reducing power, and LF = SD > ST for ABTS cation scavenging activity, respectively. In anti-coagulation assay, SD showed strong inhibition activity against thrombin and blood coagulation factors. All the extracts had no platelet aggregation activities against human platelet and no hemolysis against human RBC. Our results suggest that the SD from SH has a great potential as a new anti-coagulation agent.

  • Gender differences in the effects of gonadectomy and high cholesterol diet on body weight and body cholesterol were investigated by using Sprague Dawley rats. Body weight, plasma and liver levels of cholesterol and triglyceride and platelet aggregation were examined in ovariectomized(OVX) or orchidectomized(ODX) rats with their intacts after feeding diet with or without 0.5% cholesterol. Body weight was significantly increased(p < 0.01) in OVX rats and significantly decreased(p < 0.01) in ODX rats compared to their respective intact rats, and cholesterol diet significantly(p < 0.05) decreased body weight in gonadectomized rats. Liver lobes from rats fed cholesterol diet were opaque and larger than those from rats find control dict, resulting in a significant increase(p < 0.01) in LW/BW ratio. Plasma and liver levels of total cholesterol were significantly increased (p < 0.01) in female rats regardless ovariectomy when find 0.5% cholesterol diet, but those levels in male rats were increased only when they were orchidectomized(p < 0.0l). Plasma HDL-cholesterol was significantly decreased(p < 0.05) in both sexes when find cholesterol diet. HDL-cholesterol were higher in female than male rats regardless treatments(p < 0.05). Liver triglyceride was significantly increased(p < 0.05) in both sexes when find cholesterol dict. Plasma level of triglyceride was not different among groups except significant decrease(p < 0.05) in cholesterol find ODX rats. Maximum platelet aggregation in female rats was significantly lower(p < 0.05) than male, but ovariectomy and cholesterol diet caused an increase te the level of male rats. Microscopic examination showed cholesterol diet caused a lipid accumulation in liver. Results indicate that intact female rats have higher response to hypercholestcrolemic diet than intact male rats and orchidectomy causes male rats more responsive to hypercholesterolemic diet. However, ovariectomy causes an increase female food efficiency ratio to the level of male rats, significantly increasing body weight.

Hypocholesterolemic Effect of Tangerine Pulp, Sea Tangle or Prickly Pear Cactus on Lipid Level, Intestinal Cholesterol Absorption, Platelet Aggregation and Liver Tissue in Hypercholesterolemic Rats (감귤박, 다시마, 손바닥 선이장 분말을 함유한 식이의 급여가 고콜레스테롤혈증 흰쥐의 체내 지질수준과 장내 콜레스테롤 흡수, 혈소판 응집성 및 간 조직에 미치는 영향)

  • This study done to investigate the hypocholesterolemic effect of ow dietary fiber in a high cholesterol diet in aspect of cholesterol absorption and excretion using Sprague Dawley rats. After feeding diets containing 0.5% cholesterol and 5% tangerine pulp, sea tangle or prickly pear cactus for four weeks, we measured the levels of plasma and liver cholesterol and triglyceride, fecal neutral sterols and fecal radioactivity after ingestion 14(sup)C-cholesterol. We also examined platelet aggregation and histological change in liver tissues in association of hypercholesterolemia. The liver to body weight ratio was significantly(p<0.01) lower in rats fed prickly pear cactus than in other groups. The levels of plasma total cholesterol, LDL-cholesterol and triglyceride were decreased significantly(p<0.01) in the prickly pear cactus group compared with the control, while there was no difference in the liver levels of total cholesterol and triglyceride among groups. Fecal corprostanol and cholesterol were significantly(p<0.01) higher groups of control and prickly pear cactus compared to the other two groups. Radioisotope excretion after ingestion of 14(sup)C-cholesterol was higher in the control group than in tangerine pulp group or sea tangle group, with the highest in prickly pear group. Radioisotope excretion was the highest during the 2nd and 3rd days in all groups. Hematocrit and platelet aggregation were decreased in all fiber groups compared to the control, but not statistically different. Microscopic examination showed that cholesterol diet cause a fat accumulation in the liver and prickly pear cactus decreased the fat accumulation. Result indicates that prickly pear cactus has hypocholesterolemic effect by decreasing absorption and increasing excretion of cholesterol, thereby protective effect on fatty liver. Control group fed diet containing high cholesterol and low fiber seems to have a self control system in cholesterol absorption and excretion preventing hypercholesterolemia.   • SK-1080 is one of the newly developed orally active nonpeptide angiotensinII $AT_1-receptor$ antagonist that selectively acts at $AT_1$ receptor with high affinity. The cardiac effect on ischemia/reperfusion injury of SK-1080 was compared with those of losartan, a prototype of this class, in isolated rat hearts. Isolated perfused rat heart was pretreated with drug for 10 min and then subjected to global ischemia for 30 min followed by reperfusion with- or without drug for 30 min. The possible additive effect of SK-1080 on the platelet aggregation and coagulation in human blood was also studied. We investigated whether SK-1080 effects the platelet aggregation induced by ADP, a platelet agonist partially dependent on $thromboxaneA_2$. The clotting times in the prothrombin time (PT) and activated partial thromboplastin time (APTT) were also examined in human plasma in vitro as coagulation screening test. SK-1080 improved reperfusion function (LVDP, left ventricular developed pressure; PRP, rate-pressure product) in a dose-dependent manner. SK-1080 reduced ADP-induced platelet aggregation compared with vehicle but less than losartan, and did not affect clotting times.

Inhibitory Effects of Ginsenoslde $Rg_3$ on Platelet Aggregation and its Mechanism of Action (Ginsenoside $Rg_3$의 혈소판 응집 억제 효과 및 그 작용기전에 관한 연구)

  • The effects of ginsenosides purified from red ginseng on platelet aggregation were investigated. Preincubation of washed platelets from rats with either ginsenoside Rg3, ginsenosides non-polar fraction (G-NPF), ginsenoside Rg1(Rg1) or ginsenosides polar fraction(G-PF) reduced the plytelet aggrelation induced by collagen in a dose-dependent manner, whereas ginsenoside Rg2 failed to inhibit the aggregation. Their IC50 values of Rg3, G-NPF, Rgl, and G-PF were 8.7$\pm$1.0, 150.3$\pm$0.1, 369.9$\pm$ 1.0, 606.211.3 $\mu\textrm{g}$/ml, respectively. Aggrelation induced by thrombin was also inhibited by Rg3 and G-NPF with IC50 being 5.2$\pm$ 1.1 and 66.5$\pm$0.8 $\mu\textrm{g}$/ml, respectively. The alterations of Intracellular Ca2+ concentration in platelets were monitored using fura-2 as a fluorescent Ca2+ indicator. Both Ca2+ release from internal stores and Ca2+ influx into cytosol were suppressed by Rg3. Rg3 also inhibited granular release of ATP and TXA2 formation induced by thrombin in a dose-dependent manner in the washed platelets. Rg3 also inhibited Aggregation and ATP release from human platelets induced by collagen to a similar extent as were observed in rat platelets. In conclusion, Rg3 is a Potent anti-aggregating component in ginsenosides and may exert its anti-aggrega1ing activity by decreasing TXAa formation and granular secretion in platelets, most likely by inhibiting Ca2+ influx and Ca2+ mobilization from intracellular stores. Thus ginseng may contribute to the prevention and treatment of thrombosis.

  • 최근에 고령화 사회에 접어들면서 건강하게 활동하며 수명을 다하는 것이 중요한 연구과제가 되고 있다. 이 때문에 영양, 운동, 스트레스, 환경 등과 관련하여 암, 당뇨병, 고혈압, 비만, 동맥경화 등 생활습관병의 극복이 사회문제가 되고 있는 실정이다. 지금까지 고려인삼의 대표적인 동물실험 및 임상학적 연구결과를 종합하여 보면 성인의 난치성 만성질환에 대해서 치료제보다는 질병을 예방하고 회복촉진에 보다 큰 효과 있음을 확인할 수 있다. 이는 인삼이 식품과 의약품의 중간에 위치한 식품의약품(Nutraceutical)의 용도로 활용할 수 있음을 시사한다. 현재까지 인삼제품에 표시되는 기능성의 내용은 약 3,000여편 이상의 약리효능에 관련된 논문이 발표되었다 하더라도 "피로회복, 면역력 증진, 혈소판 응집 억제를 통한 혈액흐름에 도움이 됩니다"이며 이외의 기능성 표시는 현재로서 원칙적으로 불가한 것으로 되어 있다. 그러나 시험물질의 규격화 및 임상시험의 검토기준을 만족시키는 과학적 자료제출에 의한 개별 평가 후 표시내용의 확대가 가능하다. 고려인삼의 다양한 성분 중 ginsenoside는 주요 약효성분으로 인정을 받고 있으며 현재 품질관리의 지표성분으로 활용되고 있다. 이러한 개별 ginsenoside의 약리작용을 보면 서로 유사 또는 상반된 효과를 보여주는 성분들이 공존하고 있어 인삼의 다양한 약리효과와 일맥상통하는 것으로 보고되고 있다. 아울러, ginsenoside는 인삼의 부위 별로 현격한 차이를 보여주기 때문에 소재의 규격화는 매우 중요하다고 판단된다. 따라서, 인삼제품 개발의 가장 중요한 조건은 의약품에 가까운 기능성식품으로 세계적인 제품을 만들기 위해서는 표준화가 선행되어야 한다. 이와 같은 조건을 잘 충족시키는 제품이 스위스 "파마톤의 진사나(백삼추출물, G115)" 제품으로 생각할 수 있다. 따라서, 고려인삼의 수출시장 확대 및 세계적인 제품을 위해서는 진사나에 필적할 수 있는 새로운 홍삼의 표준화된 추출물을 도출하는 것이 무엇보다 급선무라 생각한다. 아울러 고려인삼의 특장점 발굴을 위한 표준화 된 추출분획물, 활성성분 및 효능과 연계한 새로운 고부가가치 창출제품(식품/의약품)개발에 주력해야 할 것이다.

Preventive Effects of Santalum album L. Extracts on Oxidation, Platelet Aggregation and Thrombosis (백단향추출물의 항산화, 항혈소판 응집 및 혈전 용해능에 관한 연구)

  • Objectives : To evaluate the in vitro scavenging activity, inhibitory effect of LDL oxidation of pro-oxidant reactive species, anti-platelet aggregative effects and anti-thrombosis effects in response to treatment with SA using various screening methods including biological and non-biological oxidants. Methods : The antioxidant activity concerning extract from SA was studied with in vitro methods by measuring the antioxidant activity by TEAC, measuring the scavenging effects on reactive oxygen species (ROS) [superoxide anion, hydroxyl radical] and on reactive nitrogen species (RNS) [nitric oxide and peroxynitrite] as well as measuring the inhibitory effect on $Cu^{2+}$-induced human LDL oxidation and the inhibitory effect on thrombin-induced platelet aggregation and thrombosis. Results : SA extracts were found to have a potent scavenging activity regarding oxidative stress as well as an inhibitory effect towards LDL oxidation, platelet aggregation, and thrombosis. Conclusions : The SA extracts have anti-oxidative and anti-atherosclerotic effects in vitro system, which can be used for developing pharmaceutical drugs against oxidative stress and atherosclerosis.
