The seasonal variations of nitrifying bacterial population sampled from 3 sites in Moon-Chon reservoir were analyzed by in situ hybridization with fluorescently labeled rRNA-targeted oligonucleotide probes from August 2000 until July 2001. In addition, physico-chemical parameters such as temperature, pH, chi-a and DOC were measured to determine correlations between those factors and the size of nitrifying bacterial populations. Total bacterial numbers varied in the range of $0.8{\sim}1.5{\times}10^6\;cells/ml$ independent of sites and had the maximal values in March at all 3 stations. The ratio of eubacteria to total bacteria ranged from 44.9% to 79.5%, and the ratio of each nitrifying bacteria to eubacterial numbers reached only $1.0{\sim}7.4%$. The variations of ammonia-oxidizing bacteria ranged from $1.1{\times}10^4$ to $3.0{\times}10^4\;cells/ml$ without noticeable peak values whereas those of nitrite-oxidizing bacteria varied in $1.3{\sim}5.7{\times}10^4\;cells/ml$ with the increasing tendency in winter regardless of the sites. Moreover it was observed that the numbers of nitrite-oxidizing bacteria were higher than those of ammonia-oxidizing bacteria. Total bacterial numbers correlated with water temperature (r = 0.355, p<0.05) and DOC (r = 0.58G, p<0.01) positively whereas nitrite-oxidizing bacteria correlated with temperature (r = -0.416, p<0.05) and pH (r = -0.568, p = 0.001) negatively. In addition, DOC represented good correlations with eubacterial numbers (r = 0.448, p<0.01). These results indicate that temperature, DOC and pH might be one of the main factors affecting variations of bacterial populations in the aquatic ecosystem. It was also suggested that FISH method is a useful tool for detection of slow growing nitrifying bacteria.
For the purpose of development of bioremediation technology for soil contaminated by chlorinated phenols, this study was focused on the isolation and characterization of bacteria capable of degrading chlorinated phenols, the establishment of analytical methods for chlorinated phenols, and the investigation of the contaminated sites. One site near the Incheon Industrial Complex was identified as a pentachlorophenol (PCP)-contaminated spot. The soil brought from the PCP-contaminated site contained 10-100$mu\textrm{g}$/g wet soil of PCP. Many bacterial strains capable of growing on a minimal medium containing PCP were isolated from 15 soil samples collected throughout the land, and among them, 10 active isolates were finally selected for the further studies on the biodegradability and for the use in in situ bioremediation of contaminated soil. These isolates showed species-specific pattern in PCP-decrease and cell growth in a minimal medium containing 500-1,000mg/ιPCP. Strain Bul degraded 90% of PCP at 216 hrs after incubation. Expecially, strain Bu34 was capable of degrading 4,000mg/ι PCP and was identified as Pseudomonas putida Bu34. It is seemed that the isolated active bacteria could be effectively used for the bioremediation of PCP-contaminated sites.

The Study mainly focused on bioremediation of 16 types PAHs and heavy metals in contaminated marine sediments at filed scale study using slow release biostimulant ball (BSB) was investigated. In our experiment, filed scale study ($1m{\times}1m$) was performed and the effect of BSB on PAHs and heavy metals were analysed. BSB size and distance were determined and optimum size and distance were 3cm and 5.5cm respectively. BSB containing nutrients of acetate, nitrate and sulphate which can enhance the activity of microorganism to increase degrading capacity of PAHs and enhance the heavy metals stabilization also to decrease bioavailability. PAHs containing 16 types of 2, 3, 4, 5 and 6 rings compound PAHs were found and to degrade upto 100% in 2, 3 rings, upto 90 to 94% in 4 and 5 rings and 6 ring compound was degrade up to 70%. For heavy metals stabilization percentage was increased using BSB sediment and exchangeable portion was decreased and residual portion was increased in all analysed heavy metals. BSB enhance the PAHs degradation and stabilization of heavy metals percentages. BSB is a promising method for remediation of PAHs and heavy metals in contaminated marine sediments.
Methane, which is emitted from natural and anthropogenic sources, is a representative greenhouse gas for global warming. Methanotrophs are widespread in the environment and play an important role in the biological oxidation of methane via methane monooxygenases (MMOs), key enzymes for methane oxidation with broad substrate specificity. Methanotrophs have attracted attention as multifunctional bacteria with promising applications in biological methane mitigation technology and environmental bioremediation. In this review, we have summarized current knowledge regarding the biodiversity of methanotrophs, catalytic properties of MMOs, and high-cell density cultivation technology. In addition, we have reviewed the recent advances in biological methane mitigation technologies using methanotrophs in field-scale systems as well as in lab-scale bioreactors. We have also surveyed information on the dynamics of the methanotrophic community in biological systems and discussed the various challenges pertaining to methanotroph-related biotechnological innovation, such as identification of suitable methanotrophic strains with better and/or novel metabolic activity, development of high-cell density mass cultivation technology, and the microbial consortium (methanotrophs and non-methanotrophs consortium) design and control technology.
This study presents measures to enhance the efficiency of Successive Alkalinity Producing Systems(SAPS), a natural biological purification method that prevents environmental pollution arising from the release of Acid Mine Drainage(AMD) from abandoned mines into rivers and groundwater. The treatment of AMD using SAPS is based on biological processing technology that mostly involves sulfate reducing bacteria(SRB). It has been proven effective in real-world applications, and has been employed in various projects on the purification of AMD. However, seasonal decrease in temperature leads to a deterioration in the efficiency of the process because sulfate-reducing activity is almost non-existent during cold winters and early spring even if SRB is able to survive. Against this backdrop, this study presents measures to enhance the activity of the SRB of the organic layer by integrating light emitting diode(LED)s in SAPS and to maintain the active temperature using LEDs in cold winters. Given that mine drainage facilities are located in areas where power cannot be easily supplied, solar cell modules are proposed as the main power source for LEDs. By conducting further research based on the present study, it will be possible to enhance the efficiency of AMD treatment under extreme cold weather using solar energy and LEDs, which will serve as an environmentally-friendly solution in line with the era of green growth.
Soil contamination in Korea has been accelerated every year. Because of their persistence and cumulative tendency in the environment, soil contaminants have potential long-term environmental and health concerns and it is estimated to cost enormous expense for clean-up. Korea government has legislated the law on conservation of soil environment in mid 1990s, and managed and treated hazardous wastes in contaminated sites as a remediation policy since then. Soil remediation technologies are classified into in-situ/ex-situ or biological/physico-chemical/thermal processes according to applied places or treatment methods, respectively. In Korea, clean-up of polluted sites has been mostly carried out at military areas, railroad-related sites and small-scale oil spilt sites. For these cases, in-situ remediation technologies such as soil vapor extraction (SVE) and bioventing were mainly used. In recent days, an environmental-friendly soil remediation emerged as a new concept - for example, a new soil remediation process using nanotechnology or molecular biological study and an integrated process which can overcome the limitation of individual process. To have better applicability of remediation technologies, comprehensive understandings about the pollutants and soil characteristics and the suitable techniques are required to be investigated. Above all, development of environmental technologies based on the sustainability accompanied by public attention can improve soil environment in Korea.

Studies on the preservation and propagation of tropical and subtropical plants are required, but the studies are insufficient in Korea. Since 2013, the Tropical Plant Resources Research Center has been collaborating on field research and publication to preserve plant diversity in Myanmar protected areas and Sagging areas in cooperation with Ministry of Natural Resources and Environment Conservation and the Myanmar Forest Research Institute. This study is to provide information on cooperative projects. As a result of the joint research between the Korea National Arboretum and Myanmar, in December 2018, the Korea National Arboretum Tropical Plant Resources Research Center is on display for conservation and education purposes with the 65 Myanmar useful materials and specimens of 326 kinds of plants and specimens that Myanmar Forest Research Institute donated. This donation is the first case of Myanmar's export of plant organisms to a foreign country, and as for the Korea National Arboretum, it is the second official case of introducing a mass plant from foreign governments. The study will also accelerate preservation biodiversity and propagation methods of tropical and subtropical plants as well as research on biodiversity conservation and industrial use.
Media (bio-stone), aquatic macrophytes (Oenanthe javanica) and herbivorous cladoceran (Daphnia similoides) have been used in artificial floating island (AFI) systems for water pollution control. Efficiency in chl-a concentration controlling of AFI was tested using different combinations of each device: G-BD-mixture bio-stone and Daphnia similoides, G-OB-mixture Oenanthe javanica and bio-stone, G-BOD-mixture bio-stone, Oenanthe javanica and Daphnia similoides, and the out-put water quality improvement was compared with G-C-control (no device was applied). We analyzed removal efficiency of chl-a concentration and nutrient concentrations in the artificially eutrophic water in the laboratory experimental facility. The results showed average removal rates of Chlorophyll a, TN and TP for different four groups: 69.24%, 16.61%, -0.61%; 68.39%, 14.11%, 10.52%; 78.30%, 6.69%, 25.09%; 35.42%, -3.47%, -25.18%, respectively. The results have suggested that the mixture of media, plants and zooplankton is the most efficient combination for Chlorophyll a control, while the mixture of macrophytes and bio-stone have better efficiency nutrient control.
As we are trying to in-situ treat (purify or immobilize) heavy metals or radionuclides in groundwater, one of the geochemical factors to be necessarily considered is the value of oxidation/reduction potential (ORP) of the groundwater. A biogeochemical impact on the characteristic ORP change of groundwater taken from the KAERI underground was observed as a function of time by adding electron-donor (lactate), electron-acceptor (sulfate), and indigenous bacteria in a laboratory condition. There was a slight increase of Eh (slow oxidation) of the pure groundwater with time under a $N_2$-filled glove-box. However, most of groundwaters that contained lactate, sulfate or bacteria showed Eh decrease (reduction) characteristics. In particular, when 'Baculatum', a local indigenous sulfate-reducing bacterium, was injected into the KAERI groundwater, it turned to become a highly-reduced one having a decreased Eh to around -500 mV. Although the sulfate-reducing bacterium thus has much greater ability to reduce groundwater than other metal-reducing bacteria, it surely necessitated some dissolved ferrous-sulfate and finally generated sulfide minerals (e.g., mackinawite), which made a prediction for subsequent reactions difficult. As a result, the ORP of groundwater was largely affected even by a slight injection of nutrient without bacteria, indicating that oxidation state, solubility and sorption characteristics of dissolved contaminants, which are affected by the ORP, could be changed and controlled through in-situ biostimulation method.
In this study, a comprehensive exploration and evaluation of washland candidate locations by means of field monitoring as well as spatial analysis in six major river system (Han, Nakdong, Nam, Geum, Youngsan, and Seomjin Rivers). Washland(in other words, river detention basin) is an artificial wetland system which is connected to streams or rivers likely to riverine wetlands. Major purpose of washland creation is to control floodings, water supply and purification, providence of eco-cultural space to human and natural populations. Characteristics and functions of riverine wetlands can be expected as well, thus it is believed to be an efficient multi-purpose water body that is artificially created, in terms of hydrology and ecology. Geographical information and field monitoring results for the washland candidate locations were evaluated in 2009, with respect to optimal location exploration, ecosystem connectivity and educational-cultural circumstances. A total of $269\;km^2$ washland candidate locations were found from spatial analysis (main channel of Rivers South Han, 71.5; Nakdong 54.1; Nam, 2.3; Geum, 79.0; Youngsan 46.4; Seomjin 15.7), and they tended to be distributed in mid- to lower part of the rivers to which tributaries are confluent. Field monitoring at 106 sites revealed that some sites located in the Rivers Nam and Geum is appropriate for restoration or artificial creation as riverine wetlands. Several sites in the Nakdong and Seomjin Rivers were close to riverine wetlands (e.g., Upo), habitats of endangered species (e.g., otters), or adjacent to educational facility (e.g., museums) or cultural heritages (e.g., temples). Those sites can be utilized in hydrological, ecological, educational, and cultural ways when evidence of detailed hydrological evaluation is provided. In conclusion, determination of washland locations in the major river basins has to consider habitat expansion as well as hydrological function (i.e. flood control) basically, and further utility (e.g. educational function) will increase the values of washland establishment.
