The Regulation Mechanisms of Kinesin Motor Proteins (Kinesin 모터 단백질의 조절 기전)

  • Proper intracellular transport is essential for normal cell function. Intracellular transport is mediated by microtubule-dependent molecular motor proteins, as well as kinesin and cytoplasmic dynein, which move their cargo along long, microtubule tracks in cells. Kinesins are ATP-dependent plus-end-directed motor proteins in the intracellular transport of organelles, vesicles, RNA complexes, and protein complexes. The mislocalization of these different types of cargo has been linked to cell dysfunction and degeneration. The cargo transport of kinesins can be described by the following steps: binding to the appropriate cargo and/or adaptor proteins, activation of the kinesin's motility and movement along the microtubule, and the release of the cargo at the correct destination. Recently, several studies have revealed the mechanisms for the regulation of kinesin motor activity, including cargo loading and unloading. Intracellular cargo transport is also modulated by adaptor proteins, which link the kinesins to their cargo. The regulatory proteins, which include protein kinases and phosphatases, regulate kinesin motor activity directly through the phosphorylation or dephosphorylation of kinesins and indirectly through the modification of adaptor proteins, such as c-Jun NH-terminal kinase-interacting proteins, or of the microtubule network. These findings lay the groundwork for understanding how kinesins are differentially engaged in intracellular cargo transport. In addition, understanding the regulatory mechanisms of each kinesin is an area of key interest within cell biology and neurophysiology. In this study, we reviewed kinesins' regulation proteins and discuss how their regulation affects cargo recognition and transport.

Phase Behavior and Structural Analyses of the THF + H2 Binary Clathrate Hydrate (THF + H2 이성분계 크러스레이트 하이드레이트의 상거동 및 구조 분석)

  • In this study, we investigated that hydrate formation and phase behavior of the $THF+H_2$ binary clathrate hydrates. In order to confirm the binary clathrate hydrate formation we employed the Raman and NMR spectroscopies that are known to be quite powerful tools, particularly for examining the cage occupancy pattern of guest molecules. In addition, we traced the P-T profiles from clathrate hydrate formation and dissociation process, which made it possible for the resulting phase equilibrium boundary to be clearly established. In the binary clathrate hydrate matrix we further identified that the relatively large THF molecules can only occupy the large $5^{12}6^4$ cavities, while the small $H_2$ molecules are entrapped in the empty $5^{12}$ cavities in structure-II, making the hydrate to be stable above 273 K even at relatively low pressure condition. Considering that pure $H_2$ hydrate can be produced at the extreme pressures higher that 1,000 bar, we can conclude that the water-soluble second guest inclusion induces $H_2$ storage and transportation to be readily achievable under much milder conditions.

  • Electrochemical ammonia synthesis from water and nitrogen using a Pt/GDC/Pt cell was experimentally investigated. Electrochemical analysis and ammonia synthesis in the moisture-saturated nitrogen environment were performed under the operating temperature range $400{\sim}600^{\circ}C$ and the applied potential range OCV (Open Circuit Voltage)-1.2V. Even though the ammonia synthesis rate was augmented with the increase in the operating temperature (i.e. increase in the applied current) under the constant potential, the faradaic efficiency was decreased because of the limitation of dissociative chemisorption of nitrogen on the Pt electrode. The maximum synthesis rate of ammonia was $3.67{\times}10^{-11}mols^{-1}cm^{-2}$ with 0.1% faradaic efficiency at $600^{\circ}C$.

Fabrication of Nano $Y_{2}O_{3}-CeO_{2}$ Sintered Body Using Dispersion Stability (분산 안정성을 이용한 나노 $Y_{2}O_{3}-CeO_{2}$ 소결체의 제조)

  • The dispersion stability of nano $Y_2O_3-CeO_2$ system was investigated using colloid surface chemistry. Green body of $Y_2O_3$ doped $CeO_2$ was prepared by slip casting in and aqueous system. The dispersion stability of suspension between powders and organic additive was accomplished through electrokinetic behavior of suspension, which was done by ESA apparatus. The dynamic mobility of particles was enhanced when the anionic dispersant of the amount of 1wt% was added. The dissolution of $Y^{3+}$ ion in suspension occurred in the acidic region so that pH value in slurries did not move to below 7.0. In the $CeO_2-Y_2O_3$ system, optimal preparation of suspension was made after adding the anionic dispersant as the amount of 1wt% and pH value of 11.0, and then slip-cast and sintered at 1400$^{\circ}$C, 2 hrs. It appeared relative density of >98% and homogeneous distribution of Y element in depth direction as well as in the microstructure of surface.

  • 본 연구는 접속사를 통한 텍스트 통합 과정이 논리적 추론 종류에 따라 다른 정보처리 과정 혹은 다른 종류의 단원적 구조(modular structure in language processing)에 의해 처리되는지를 조사하기 위해 실시되었다. 또한, 접속사를 통한 추론 과정이 실어증의 증상 종류에 따라 다른 종류의 언어정보처리 손상이 있는지를 평가하기 위해 실시되었다. 실험에 참가한 환자는 이해성 실어증환자(Wernicke aphasic), 전반성 실어증 환자(Global aphasic), 표현성 실어증 환자(Broca aphasic) 등이었다. en 종류의 과제를 이용하였다. 한 과제는 앞 뒤 문장을 논리적 관계성을 표현하는 접속사를 채워 넣는 과제였고 다른 과제는 접속사가 포함된 텍스트가 옳은지를 판단하는 정오 판단 과제였다. 실험재료 문장에 사용된 접속사는 추가적인 정보를 제공하는 '그리고'와 대등 관계를 나타내는 '그러나' 및 인과 관계를 표현하는 '그래서' 였다. 이 세 종류의 접속사는 각기 다른 논리적 관계성을 나타낸다. 실험 결과는 실어증 환자가 전반적으로 채워 넣기 과제에서 보다는 정오 판단 과제에서 더 많은 실수를 보였으며, 표현성 실어증 환자보다는 이해성 실어증 환자가 더 많은 오류를 보였다. 또한, 세 종류의 접속사 중에 '그리고'가 표함된 텍스트에서 더 많은 실수를 보였다. 이 연구에서 나타난 흥미 있는 결과는 표현성 실어증 환자는 '그러나' 접속사가 포함된 텍스트에서의 수행이 '그래서'가 포함된 경우에서보다 좋은 반면에 전반성 실어증 환자는 '그래서'를 포함하는 텍스트에서의 수행이 '그러나'를 포함하는 텍스트에서의 수행이 더 우수해서 이중해리(double dissociation)가 나타난다는 사실이다. 이 결과는 선후 문장이 어떤 종류의 논리적 관계성을 지니는가에 따라 다른 종류의 정보처리가 진행된다는 것을 암시하는 결과이다.>$\textrm{cm}^2$.。C로 비교적 양호한 초전박막의 전기적 특성을 나타내었다.(Mg+Fe)비를 갖고 전자에 비해 Al이 풍부한 환경에서 생성되었으며, 따라서 활석과 연관되지 않은 녹니석은 생성시 광체와 인접한 화강아질 편마암에 의해 주로영향을 받았을 것으로 생각된다. 녹니석의 이러한 2가지 화학조성상의 경향은 녹니석과 공존하는 운모류나 각섬석류들의 화학분석결과와도 잘 일치한다. 이러한 결과는 이 지역의 활석 광상이 초염기성암 기원의 사문암이 열수변질작용을 받아 생성되었음을 명확하게 지시하며, 따라서 활석 광석내에 존재하는 녹니석은 활석의 근원 광물로서 녹니석편암 및 녹니석 편마암 매의 녹니석이 활석화되고 남은 잔존광물이 아니라, 주변암에 의해 성분상의 영향을 받은 열수와 사문암과의 변질교대작용에 의한 활석화과정 중에 주로 생성된 것으로 추정된다. 이러한 결과는 연구지역의 활석광상이 초염기성암의 사문암화 작용과 활석화 작용의 두 가지 변질작용에 의해 형성되어졌음을 알려준다.농도 증가 없이 폐 조직에 약 50배 정도의 고농도 cisplatin을 투여할 수 있었으며, 또한 분리 폐 관류 시 cisplatin에 의한 직접적 폐 독성은 발견되지 않았다이 낮았으나 통계학적 의의는 없었다[10.0%(4/40) : 8.2%(20/244), p>0.05]. 결론: 비디오흉강경술에서 재발을 낮추기 위해 수술시 폐야 전체를 관찰하여 존재하는 폐기포를 놓치지 않는 것이 중요하며, 폐기포를 확인하지 못한 경우와 이차성 자연기흉에 대해서는 흉막유착술에 더 세심한 주의가 필요하다는 것을 확인하였다. 비디오흉강경수술은 통증이 적고, 입원기간이 짧고, 사회로의 복귀가 빠르며, 고위험군에 적용할 수 있고, 무엇보다도 미용상의 이점이 크다는 면에서 자연기흉에 대해 유용한 치료방법임에는 틀림이 없으나 개흉술에 비해 재발율이 높고 비용이 비싸다는 문제가 제기되고 있는 만큼

  • It has been found that both of the iridium (Ⅰ) complexes, Ir$(ClO_4$)(AN)(CO)$(Ph_3P)_2$(AN = $CH_2$CHCN, $Ph_3P = (C_6H_5)_3$P) and [Ir(AN)(CO)$(Ph_3P)_2]ClO_4$, react with $Cl^-$ to give IrCl(AN)(CO)$(Ph_3P)_2$, and [Ir(AN)(CO)$(Ph_3P)_2]ClO_4$ dissociates AN to yield Ir$(ClO_4)(CO)(Ph_3P)_2$ in the absence of excess AN added, and Ir$(ClO_4)(CO)(Ph_3P)_2$ reacts with $Cl^-$ to produce IrCl(CO)$(Ph_3P)_2$. It is suggested that the catalytic polymerization of AN with Ir$(ClO_4)(AN)(CO)(Ph_3P)_2$ proceeds through the formation of [(CO)(Ph_3P)_2$Ir(-CH=CHCN)(H)($CH_2$=CHCN)]Cl$O_4$ followed by the formation of iridium(alkyl)(alkenyl) type complex which undergoes a reductive elimination to produce the polymer of acrylonitrile.

Cytologic Aspects of Oral Squamous Cell Carcinoma in a Captive African Hedgehog (Altelerix albiventris) (고슴도치의 구강 편평상피암종의 세포학적 고찰 1증례)

  • A 3-year-old intact female hedgehog (Atelerix albiventris) was presented for evaluation of mandibular swelling. Fine needle aspiration of swollen mandibular was performed and smears were stained with Romanowsky type stain for cytological evaluation. Smears were highly cellular with predominance of variably shaped keratinized or non keratinized squamous cells with low numbers of spindloid to abnormally elongated cells. Cytologic impression was squamous cell carcinoma. The mass was surgically removed for histological examination. Microscopically tortuous and anastomosing delicate to broad pegs and nests of neoplastic squamous epithelial cells were supported by a moderate collagenous and spindloid fibroblast stroma. Tumor cells had moderate anisocytosis and mild anisokaryosis and range from moderately to well keratinized, with areas of intratumoral acantholysis accompanied by mixed stromal lymphoplasmacytic, neutrophilic inflammation. The mitotic index is 2-3 per high-power field. Tumor cells were expanding the subcutis subjacent to the layer of skeletal muscle and incorporating the osseous tissue fragments. The final diagnosis was squamous cell carcinoma. The patient survived three months after surgery without any further medical treatments.

Functional Expression of Soluble Streptavidin in Escherichia coli (수용성 streptavidin의 Escherichia coli 에서 기능적 발현)

  • Streptavidin, a protein produced by Streptomyces avidinii, strongly binds up to four molecules of vitamin H, d-biotin exhibiting the dissociation constant of about 10−15 M. This strong binding affinity has been applied for detection and characterization of numerous biological molecules suggesting expression and purification of functional streptavidin should be very useful for the application of this streptavidin-biotin interaction. To express a soluble streptavidin in Escherichia coli, We synthesized streptavidin genes and cloned into pET-22b plasmid, which uses T7 RNA polymerase/T7 promoter expression systems containing pelB leader for secretion into periplasmic space and six polyhistidine tags at C-terminus for purification of expressed proteins. Although streptavidin is toxic to Escherichia coli due to strong biotin binding property, streptavidin was expressed very sufficiently in a range of 10-20 mg/ml. In SDS-PAGE, the size of purified protein was shown as 17 kDa in denatured condition (boiling) and 68 kDa in native condition (without boiling) suggesting tetramerization of monomeric subunit by non-covalent association. Further analysis by size-exclusion chromatography supported streptavidin’s tetrameric structure as well. In addition, soluble streptavidin detected biotinylated proteins in westernblot indicating its functional activity to biotin. Taken these results together, it concluded that our simple expression system was able to show high yield, homotetrameric formation and biotin binding activity analogous to natural streptavidin.

Effect of Cellular Phospholipase A2 Inhibition on Enhancement of Bt Insecticidal Activity (세포성 인지질분해효소 활성 억제에 따른 비티 살충력 증가 효과)

  • Some bacterial metabolites of Xenorhabdus nematophila (Xn) inhibit phospholipase $A_2$ ($PLA_2$) activity to shutdown eicosanoid biosynthesis in target insects. However, little has been known about the target insect $PLA_2$ of these bacterial metabolites. Eight bacterial metabolites identified in Xn culture broth exhibited significant insecticidal activities against larvae of both lepidopteran species of Plutella xylostella and Spodoptera exigua. Moreover, these bacterial metabolites significantly enhanced insecticidal activities of Bacillus thuringiensis (Bt). To determine target $PLA_2$, we cloned and over-expressed cellular $PLA_2$ ($SecPLA_2$) of S. exigua. Purified $SecPLA_2$ catalyzed phospholipids derived from the fat body and released several polyunsaturated fatty acids. Most Xn metabolites significantly inhibited $SecPLA_2$ activity, but were different in their inhibitory activities. There was a positive correlation between the inhibition of $SecPLA_2$ and the enhancement of Bt insecticidal activity. These results indicate that $SecPLA_2$ is a molecular target inhibited by Xn metabolite.
