Isolation of Polyene Antifungal Antibiotics Against Gummy Stem Light Caused by Didymella bryoniae (Streptomyces sp. 유래 Polyene 계 항만고병 항생물질의 분리)

  • Antifungal agents, flavofungin and fungichromin were isolated from the fermentation culture broth of a Streptomyces sp. SKM338. Biological evaluation of these antibiotics indicated that the compounds possesses broad spectrum antifungal activity against various pathogens. Especially, these compounds inhibited throughly growth of Didymella bryoniae, caused Gummy stem blight of melons, occurs in the southeastern Korea. Inhibition of this pathogen may be prevented from directly reducing both pre- and post-harvest yields.

An Antifungal Subatance, 2,4-Diacetylphloroglucinol Produced from Antagonistic Bacterium Pseudo-monas fluorescens 2112 Against Phytophthora capsici (Phytophthora capsici를 길항하는 Pseudononas fluorescens 2112가 생산하는 항진균 항생물질 2,4-diacetylphloroglucinol)

  • An antifungal substance was purified from culture broth of Pseudomonas flulorescens 2112 that showed a broad-spectrum antagonistic activity against various phytopathogenic fungi including capsici. The substance was identified as 2,4-diacetylphloro-glucinol basd on NMR analysis. The 2,4-diacetylphloroglcinol showed antibiotic activity in broad acidic range from pH 1.0 to pH 9.0. About 83% of initial activity was remained after incubation for 30min ar $60^{\circ}C$, however, the activity was dropped up to 50% after 30 min incubation in $80^{\circ}C$. When the nucleotides of P. capsici treated with 2,4-diacetylphloroglucinol were labeled with[$^{3}$ H]-Adenin, the newly synthesized and radioactive-labeled RNA was significantly reduced than those of untreated P. capsici. indicating that the 2,4-diacetylphloroglucinol inhibits RNA synthesis.

  • An antifungal antibiotic-producing bacterium was isolated from soil and identified as Bacillus sp. CJ-1. The culture supernatant was found to have a strong and stable antifungal activity against Colletotrichum gloeosporioides. Culture conditions for the maximum antifungal activity were examined. Glucose and yeast extract were selected as the best carbon and nitrogen sources. The optimum C/N ratio was 3. The optimum temperature and initial pH were determined as $35^{\circ}C$ and 6.0, respectively. Under these conditions, the production for the antibiotic was maximized at 72 hr at $35^{\circ}C$ after cultivation. Microscopic observation showed that the culture supernatant of Bacillus sp. CJ-1 had a strong inhibitory activity on the mycelial growth of the test strain at above $12.5\;{\mu}L\;mL^{-1}$ of concentration.

  • Streptomyces sp. No. 297 previously isolated from a soil sample collected in Mt. Soyo of Kyeongi Province, which produced strong antifungal substances t-cinnamamide and another unknown compound, was identified as Streptomyces griseorubiginosus var. soyoensis. The results of examinations in morphologial, physiological and cultural characteristics of the strain are presented.

  • It were reported that antifungal mechanism of Enterobacter cloacae is a volatile ammonia that produced by the strain in soil, and the production of ammonia is related to the bacterial urease activity. A powerful bacterium SH14 against soil-borne pathogen Fusarium solani, which cause root rot of many important crops, was selected from a ginseng pathogen suppressive soil. The strain SH14 was identified as Bacillus subtilis by cultural, biochemical, morphological method, and $API^{circledR}$ test. From several in vitro tests, the antifungal substance that is produced from B. subtilis SH14 was revealed as heat-stable and low-molecular weight antibiotic substance. In order to construct the multifunctional biocontrol agent, the urease gene of Bacillus pasteurii which can produce pathogenes-suppressive ammonia transferred into antifungal bacterium. First, a partial BamH I digestion fragment of plasmid pBU11 containing the alkalophilic B. pasteurii l1859 urease gene was inserted into the BamH I site of pEB203 and expressed in Escherichia coli JM109. The recombinant plasmid was designated as pGU366. The plasmid pGU366 containing urease gene was introduced into the B. subtilis SH14 with PEG-induced protoplast transformation (PIP) method. The urease gene was very stably expressed in the transformant of B. subtilis SH14. Also, the optimal conditions for transformation were established and the highest transformation frequency was obtained by treatment of lysozyme for 90 min, and then addition of 1.5 ${mu}g$/ml DNA and 40% PEG4000. From the in vitro antifungal test against F. solani, antifungal activity of B. subtilis SH14(pGu366) containing urease gene was much higher than that of the host strain. Genetical development of B. subtilis SH14 by transfer of urease gene can be responsible for enhanced biocontrol efficacy with its antibiotic action.

  • A water soluble antifungal antibiotic, Solumycin, was separated from the culture broth of Streptomyces sp. LAM-593, isolated from soil, by butanol extraction, alumina-, 1st and 2nd Sephadex LH-20 column chromatography. The substance was pale yellow crystal which gave a single spot at Rf value 0.24 with ethanol-ammonia water-water (8:1:1), 0.46 with butanol-ethanol-water (5:1:4), 0.84 with 50% methanol on silica gel TLC. It was dissolved well in water, methanol and acidic aq. butanol but not in ethanol, acetone, ethyl acetate, chloroform. acetic acid etc., and gave positive Fehling and Molish reaction. The UV spectrum in methanol showed absorption at 342, 361, 380, and 404 nm. The antibiotic was active against fungi such as Candide, Cryptococcus, Saccharomyces, Trichophyton and Trichosporon, but not to bacteria such as Bacillus, Escherichia and Staphylococcus.

  • Out of 826 isolates of Streptomyces isolated from different soils, their distribution and antibiotic productivity were investigated. Distribution of the organism in the soil was affected by the soil conditions and plants. The highest isolation frequency was occurred from Quercus forest, Robinia forest and grass field, while soils from orchards and cultivating fields showed low density of Streptomyces. More than 49% of the isolates showed antibacterial activity against Bacillus subtilis, Erwinia carotovora subsp. carotovora and Xantomonas campestris pv. oryzae and about 40% of the isolates showed antiyeasty activity against Saccharomyces cerevisiae but only a few isolates showed antibiotic activity against E. coli and Pseudomonas solanacearum. Forty isolates of the Streptomyces showed strong antifungal activity against Pyricularia oryzae. Rate of isolation of Streptomyces was the highest on starch agar among the eight media tested. Antibiotic productivity of the isolates was the highest on potato sucrose agar medium among the 5 media tested.

  • Antifungal activities of pomegranate were investigated. Seventy percent ethanol extracts of seeds, peels, and whole fruits of pomegranate showed similar antifungal activities against Candida albicans in liquid media, whereas extract of whole fruit showed relatively high antifungal activity in solid media. When 70% ethanol extracts were fractionated sequentially with chloroform and ethyl acetate, ethyl acetate fraction exhibited the highest anti-fungal activities against C. albicans. Ethyl acetate fractions of whole fruits and peel portions showed at least 36% and 25% growth against C. albicans, Candida glabrata, Candida tropicalis, and Candida lusitaniae in liquid media, respectively. These results indicate that pomegranate contains antifungal compounds soluble with organic solvents.

  • A multifunctional antagonistic bacterium, producing both antifungal antibiotic and chitinase that could be used as biocontrol agents against fungal plant pathogens was isolated from the plant-disease suppressive soil. The isolate was identified as Promicromonospora sp. KH-28 from various morphological, biochemical and physiological tests. The antifungal antibiotic produced by Promicromonospora sp. KH-28 was soluble in n-butanol, methanol, toluene, n-hexane, ethanol but insoluble in H2O, acetone, chloroform, ethylacetate and ethylether. It inhibited the growth of fungal plant pathogens of Fusarium solani, F. oxysporum, Alternaria mali and Phytophthora capsici. The antagonistic Promicromonospora sp. KH-28 produced optimally the antifungal antibiotic when it was cultivated at pH 7, 3$0^{\circ}C$ for 5 days.

항진균성 항생물질을 생산하는 streptoverticillium sp. NA-4803의 분리 및 동정

  • The aim of the present research program was to develop a strain of actinomycetes producing antifungal substance. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among over 440 isolates, a strain (NA-4803) producing antifungal substance against Trichophyton spp. Nannizzia otae and Pyricularia oryzae was selected. The strain NA-4803 was identified as strain similar to Streptoverticillium blastmyceticum with respect to morphological and physiological characteristics, lecithinase and lipolytic activity, degradation of organic compounds, resistance to antibiotics and utilization of carbon and nitrogen sources. But it showed some differences such as positive reaction of nitrate reduction, negative reaction of L-tyrosine degradation, resistance to cephaloridine, and utilization of I -rhamnose and inulin. The strain NA-4803 was named as Streptoverticillium sp. NA-4803.
