Study on Applicability of Pinus koraiensis Siebold et Zucc Leaf Extract as a Cosmetic Ingredient (잣나무(Pinus koraiensis Siebold et Zucc)잎 추출물의 화장품 소재로서의 활용 가능성에 관한 연구)

  • This study was performed to verify various effects of 70% ethanol extract and distilled water extract of pine leaf such as antioxidant, anti-inflammatory, skin-lightening effects analysis in cells. The result of antioxidant experiment showed that polyphenol content increased concentration dependently in polyphenol and flavonoid effects. The total content of polyphenols and flavonoids was much higher in ethanol extract than water extract. In all the cells of B16F10, RAW264.7 cytotoxicity at each concentration level didn't appear. The result of measuring NO production inhibition showed that it inhibited NO production led to LPS effectively, so anti-inflammatory activity was confirmed. The result of measuring melanin biosynthesis inhibition showed that there was considerably reduction effect, but it performed western blot, and as a result of MITF, Tyrosinase protein revelation, the inhibition of MITF, Tyrosinase revelation was confirmed concentration dependently, Therefore, in this study pine leaf extract was expected to be used as a cosmetic ingredient.

Free radical scavenging, anti-inflammatory and melannin synthesis inhibitory activities of Gloeostereum incarnatum (느릅나무버섯 자실체의 메탄올과 열수추출물의 항산화, 항염증 및 멜라닌 생성저해 활성)

  • Gloeostereum incarnatum is an edible and medicinal mushroom belongs to Family Cyphellaceae of Polyporales, Basidiomycota. The purpose of this study was to investigate the free radical scavenging, anti-inflammatory, and melanin synthesis inhibitory activities of fruiting bodies of Gloeostereum incarnatum. In the free radical scavenging activities, the mushroom extracts showed good 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging and chelating activity on the ferrous ions compared with the positive control, BHT. The mushroom extract suppressed nitric oxide (NO) production in LPS-induced RAW 264.7 macrophage cells in dose dependant manners. Significant reduction of paw edema of rats were observed at 2~6 h after administration with 50 mg/kg of the methanol and hot-water extracts, which were comparable with treatment of 5 mg/kg of indomethacin, the positive control. The melanin synthesis of Melanoma B16/F10 cells treated with $100{\mu}g/mL$ of the methanol and hot water extracts decreased melanin concentration to 50% and 45% compared with the control, arbutin. Therefore, the experimental results showed that methanol and hot-water extracts of Gloeostereum incarnatum fruiting bodies might be used for good sources of anti-inflammatory, free radical scavenging, and skin whitening agents for human health.

  • Trifoliate orange components consist of several kinds, such as monoterpenes, limonoids, flavonoids, and coumarins. Coumarin derivatives were shown to possess valuable pharmacological properties such as anti-inflammatory and dietary effect. Among them, 7-geranyloxycoumarin 6 is a promising chemopreventive agent againist skin, tongue, oesophaqus and colon carcinogenesis in rodents. Seven new coumarin derivatives structurally related to 7-geranyloxycoumarin were synthesised in good yields by $Cs_2CO_3$/acetonitrile condition. We investigated the effect of anti-inflammatory activity on interleukin-6 for synthesised geranyloxycoumarin derivatives. 6-Geranyoxycoumarin 9 (68.9% / $1{\mu}M$ ; 72.6% / $10{\mu}M$) of the anti-inflammatory activity is far higher than 7-Geranyloxycoumarin 5 (40.1% / $1{\mu}M$ ; 61.1% / $10{\mu}M$) and their other derivatives.

  • Sericin is a protein extracted from the cocoon and has many health benefits. This study was conducted to evaluate the anti-wrinkle and anti-inflammatory activity of sericin derived from cocoon as a cosmetic material. The antioxidant activity of sericin was measured by DPPH and ABTS assays. In addition, cell viability was confirmed in Raw 264.7 cells, which are macrophages, and the anti-inflammatory effect of sericin was investigated using the inflammatory response induced using lipopolysaccharide (LPS). It shows that sericin has antioxidant activity and can be an excellent material for anti-aging and anti-inflammatory cosmetics.

Comparison of Effect of Water and Ethanolic Extract from Roots and Leaves of Allium hookeri (물과 주정을 이용한 삼채 뿌리와 잎 추출물의 항산화 활성 및 항염증 효과 비교)

  • The aim of this study was to evaluate the antioxidant activities and anti-inflammatory effects of water and ethanolic extracts from Allium hookeri roots and leaves. Antioxidant activities of Allium hookeri extracts were determined based on various radical scavenging activities using an ESR spectrophotometer, ferric reducing antioxidant power, 2,2'-azinobis-(3-ethybenzothiazoline-6-sulfonic acid) radical scavenging activity, and oxygen radical absorbance capacity assays. In addition, we investigated anti-inflammatory effects of Allium hookeri extract on lipopolysaccharide (LPS)-induced inflammation in RAW264.7 cells. We also explored the effects of extract from Allium hookeri root on suppression of pro-inflammatory mediators such as nitric oxide (NO) and pro-inflammatory cytokines such as IL-6 and TNF-${\alpha}$ against LPS-induced activation of RAW264.7 cells. Our results demonstrated superior antioxidant activity for leaf extract of Allium hookeri compared to extract from root of Allium hookeri. On the other hand, root extract of Allium hookeri showed better anti-inflammatory activity compared to leaf extract. Our study suggests that Allium hookeri extract exhibits strong antioxidant activity and anti-inflammatory effects and can be developed as a potential therapeutic candidate for diseases involving oxidative stress and inflammation.

  • The anti-inflammatory and analgesic actions of Cedrela sinensis, Oenanthe javanica and ARtemisia princeps var. orientalis were carried out. The methanolic extract of Artemisia princeps var. orientalis showed the inhibitory effect on carrageenin-induced edema as well as vascular permeability in mice. And we also found the alagesic activity in these plants.

Study on the Anti-bacterium, Antioxidant and Anti-inflammatory Effect of Calystegia pubescens Lindl. Extracts (메꽃(Calystegia pubescens Lindl.) 추출물의 항균, 항산화 및 항염증 효과에 관한 연구)

  • Calystegia pubescens Lindl. (C. pubesens) is a native Korean herb that has been traditionally used for its diuretic, fatigue, and blood sugar-lowering effects. In previous studies, C. pubesens has shown antioxidant and whitening effects, but research on its antibacterial and anti-inflammatory properties is limited, and its potential as functional materials is lacking. In this study, we investigated the antibacterial, antioxidant, and anti-inflammatory effects of leaf (CPL) and stem (CPS) extracts of C. pubesens extracted with 70% ethanol. When the anti-bacterial effect was confirmed, CPL showed superior anti-bacterial effect than CPS for C. acne, S. aureus, E. coli and P. aeruginosa. Additionally, CPL exhibited significantly higher antioxidant activity compared to CPS, as demonstrated by DPPH and ABTS+ radical scavenging assays. Therefore, CPL showed superior anti-bacterial and antioxidant effects compared to CPS. Based on these results, further investigation was conducted to confirm the anti-inflammatory effect of CPL using RAW264.7 macrophages. Nitric oxide (NO) produced by lipopolysaccharide (LPS) stimulation was significantly reduced by CPL treatment in a dose-dependent manner. Additionally, the protein and mRNA expression of iNOS was reduced by CPL in LPS-indueced RAW264.7 cells, which was consistent with NO production. In conclusion, this study confirmed that CPL has superior anti-bacterial and antioxidant effects compared to CPS, and demonstrated anti-inflammatory effect of CPL via inhibiting iNOS expression and NO production induced by LPS. Based on the result, we suggest the potential of CPL as a valuable functional materials.
