Extraction Yield and Anti-Yeast Activity of Extract from Green Tea Seeds by Pretreatment and Extraction Conditions (녹차씨 전처리와 추출 조건에 따른 녹차씨 추출물의 추출 수율 및 항효모 활성)

  • Green tea (Camellia sinensis) seed extract (GTSE) was prepared under various pretreatment conditions and used to investigate its extraction yield and anti-yeast activity. Anti-yeast activity of GTSE from seeds with or without the coats was the same, whereas the extraction yield was slightly higher in extract from seeds without the coat. Anti-yeast activity of GTSE from seeds with different water contents or particle sizes was the same, whereas the extraction yield was highest in extract from seeds with 7.3% moisture or a smaller particle size. Anti-yeast activity of defatted green tea seed extract (DGTSE) was the same as that of GTSE. Extraction yield was higher in DGTSE from defatted seeds by the oil press machine compared to hexane extraction. Defatted green tea seed (DGTS), a by-product from the oil extraction process, is a good natural source of anti-yeast preservative. The extraction yield and anti-yeast activity of DGTSE were investigated using various extraction solvents, temperatures, and times. The results show that water was an economic extraction solvent, and anti-yeast activity of DGTSE was unstable at $90^{\circ}C$. These results suggest that water, extraction temperature of $50^{\circ}C$, and extraction time of 4 h were the most efficient for extraction of anti-yeast compounds from DGTS.

Optimization of Enzymatic Hydrolysis for the Production of Antitoxic Bovine Hepatic Extract (항독성 소간추출물의 생산을 위한 효소분해의 최적화)

  • Bovine hepatic extract is recognized as possessing detoxifying activity against various liver diseases. In orderto develop a process for its mass production, various enzymatic hydrolysis conditions were tested, and bovine hepatic extracts were prepared. These extracts were then examined for composition, microorganism levels, and vitamin $B_{12}$ content. Among the enzymes tested, papain was selected based on yields for dry residue and amino nitrogen. The other enzymes tested included bromelain, ficin, pancreatin, and protease NP. The optimal hydrolysis conditions were established at 65$^{\circ}C$ for 24 hr, with an addition of 1%(w/w) papain to the beef liver. The prepared spray-dried bovine hepatic extract showed an 11% recovery yield on a raw beef liver basis, with 95% dry residue and 11.8% total nitrogen content. Microorganisms were not detected in the dried extract, and its vitamin $B_{12}$ content was 4.1 ${\mu}$g/g. In summary, the conditions established in this study could be applied for the high yield mass production of bovine hepatic extract.

항이뇨작용기전 검색(Naproxen을 중심으로)

  • Naproxen을 개의 정맥내 투여하였을때 뇨량의 감소와 더불어 삼투질제거율(Cosm), 뇨중 $Na^{+}$$K^{+}$의 배설율( $E_{Na}$ , $E_{k}$ )의 감소와 신세뇨관에서의 $Na^{+}$$K^{+}$의 재흡수율( $R_{Na}$ , $R_{k}$ )과 K/Na비의 증대를 나타내었으며 신혈류량(RPF)이 감소하였다. 한쪽 신동맥내에 naproxen을 투여하였을때 투여신(실험신)에서의 이뇨작용과 대조신에서의 항이뇨작용을 나타내었다. 경동맥에 naproxen을 투여하였을때도 뚜렷한 항이뇨작용을 나타내었다. 이와같은 두경우에서의 항이뇨 작용시의 신기능변화는 정맥내 naproxen의 경우와 같은 양상을 나타내었다. 경동맥내의 naproxen의 항이뇨작용은 신신경 제거에 의하여 영향을 받지 않았으나 정맥내로의 arachidonic acid나 indomethacin의 전처리에 의하여서는 나타나지 않았다. 경동맥내의 naproxen은 정맥내의 spironolactone의 이뇨작용을, 정맥내의 spironolactone은 경동맥내의 naproxen의 항이뇨작용을 억제하였다. 이상의 결과로 보아 naproxen은 중추성 항이뇨작용과 $Na^{+}$저류를 나타내며 이는 prostaglasdin의 합성억제와 aldosterone양 작용에 의하는 것으로 사료된다.

  • 화합물 9-methoxy-6-oxo-3,4,4a,5-tetrahydro-3-hydroxy-2,2-dimethyl-naphtho[1,2〕pyran(1)의 전구물질인 7-methoxy-4-oxo-2-prenyl-1-tetralol을 다음과 같은 방법으로 합성하였다. 7-Methoxy-$\alpha$-tetralone에 LDA를 처리한 후, prenyl bromide를 반응시켜 7-methoxy-2-prenyl-1-tetralone을 제조했다 (수율 66%). 7-Methyl-2-prenyl-1-tetralone을 L-seleclride로 환원하여 cis-2-prenyl-1-tetralol을98% 수율로 얻었다. cis-2-Prcnyl-1-tetralol을pyridine 존재하에서 acetic anhydride로서acetylation한 다음 PDC로 산화시켜 4-acetoxy-3-prenyl-1-tetralone을 합성하였다 (수율 45%). cis-4-Acetoxy-3-prenyl-1-tetralone을 가수분해하여 cir-7-methoxy-4-oxo-2-prenyl-1-tenalol을 합성하였다. 6,7-Dimethoxy-4-oxo-2-prenyl-1-letralol도 동일한 방법으로 합성하였다.

Purification and Chemical Properties of Anti-complementary Polysaccharide from Capsici Fructus (고추의 항보체 다당의 정제와 특성)

  • Water-soluble crude polysaccharide (CAP-0) obtained from the Capsici Fructus(the fruits of Capsium annuum L.) showed a potent anti-complementary activity. The anti-complementary activity did not change by pronase digestion of CAP-0, but decreased by the periodate oxidation. CAP-0 was fractionated into four polysaccharide fractions, CAP-1,2,3 and 4, by the addition of cetyltrimethylammonium bromide. CAP-1, showing the highest anti-complementary activity, was refractioned by anion-exchange chromatography to give three major fractions(CAP-1-III, IV and V). CAP-1-III was shown to have low anti-complementary activity, but CAP-1-IV and V had high activity. CAP-1-III and IV were purified on Sephadex G-100 to give each two fractions(CAP-1-IIIa and IIIb, CAP-1-IVa and IVb), respectively. From the results of gel filtration and electrophoresis, these four fractions and CAP-1-V were found to be homogeneous polysaccharides. High molecular polysaccharies(M.W. CAP-1-IIIa 70,000, IVa 195,000, V 140,000) showed relatively higher anti-complementary activity than low molecular polysaccharides(CAP-1-IIIb and IVb).

Production of Anti-dementia Acetylcholinesterase Inhibitor from Pleurotus ostreatus (Heuktari) and Inhibitory Effect on PC12 Neuron Apoptosis (흑타리버섯으로부터 항치매성 Acetylcholinesterase 저해물질의 생산 및 PC12 신경세포사 저해 효과)

  • To develop a new antidementia acetycholinesterase (AChE) inhibitor from edible mushrooms, the inhibitory effects on AChE of water and ethanol extracts from various edible mushrooms were measured. Among the tested compounds, 70% ethanol extracts from Tremella fuciformis showed the highest AChE inhibitory activity, at 25.3% (IC50: 9.9 mg). Water extracts from the fruiting body of Pleurotus ostreatus (Heuktari) showed AChE inhibitory activity of 20.2% (IC50: 12.4 mg). However, the yield (40.8%) from Pleurotus ostreatus (Heuktari) was higher than that from Tremella fuciformis (5.0%). Therefore, we selected Pleurotus ostreatus (Heuktari) as the most promising candidate for a mushroom containing anti-dementia AChE inhibitors. The AChE inhibitor from Pleurotus ostreatus (Heuktari) was optimally extracted when its fruiting body was treated with water for 6 h at 30℃. The anti-dementia effects of the partially purified AChE inhibitor from Pleurotus ostreatus (Heuktari) were observed in PC12 nerve cells.

유황오리 추출물로부터 항종양활성 성분의 분리 및 정제

  • 본 연구는 유황오리로부터 항종양 효과를 나타내는 물질을 용매추출과 각종 chromatography를 사용하여 분리 및 정제하였다. HPLC를 사용하여 정제한 항종양활성 성분의 수율은 유황오리 1Kg당 10mg이었다. HPLC를 이용하여 정제한 항종양활성 물질을 Clonogenic assay로 측정한 결과 $200{\mu}g/m{\ell}$의 농도에서 HEp-2, KB 및 SW-156에서 각각 26%, 28%및 30%의 생존율을 나타내어 항종양 효과가 나타났다. MTT assay에서는 $100{\mu}g/m{\ell}$의 농도는 Farrow, HEp-2 및 KB에 대해서 각각 56%, 58% 및 56%의 세포증식 억제 효과를 나타내었다. bovine normal kidney cell line인 MDBK는 $500{\mu}g/m{\ell}$, $200{\mu}g/m{\ell}$, $100{\mu}g/m{\ell}$에서 각각 48%, 34%, 28%로 증식억제 효과가 나타나 정상세포에 대한 세포독성은 강하지 않은 것으로 판단되어진다.

막분리공정을 이용항 폐수처리 및 회수

  • 폐수처리 분야에 대한 막분리기술의 적용은 산업 전분야에서 그 성능을 입증받고 널리 활용되고 있는 단계이다. 막분리 기술은 단순히 폐수를 처리한다는 개념이 아니라 폐수중에 함유된 유효물질을 회수하여 재사용한다는 점과 폐수중의 물을 높은 수율로 회수하여 재사용한다는 측면, 폐수 오염원의 부하를 크게 줄여준다는 측면에서 많은 잇점을 가지고 있다. 이러한 특징은 각 분야의 제조공정을 "Closed-loop system화" 및 "Zero Discharge" 개념으로 운전함으로써 환경오염의 방지와 더불어 자원의 재활용에 의한 원가절감 효과를 가져올 수 있게 하였다.절감 효과를 가져올 수 있게 하였다.

Antimicrobial, Antihypertensive and Anticancer Activities of Garlic Extracts (마늘 추출물의 항균, 항고혈압 및 항암활성)

  • Antibacterial, antifungal, anticancer, and ACE inhibitory activities of methanol, ethyl acetate, chloroform, and hexane extracts of garlic were investigated. Methanol extract showed highest yield of 7.9-8.8 brix. Ethyl acetate and chloroform extracts showed strong antibacterial activities as determined by inhibition zone (8-16mm) against B. subtilis and P. aeruginosa, and good antifungal activities as determined by inhibition zone (8-30mm) against A. niger, M. miehei, and T. reesei. Wando, Banyans, Chinese, and Seosan garlic methanol extracts showed 65.6, 60.4, 70.1, and 55.3% ACE inhibitory activities, 42.3, 53.8, 50.4, and 54.5% anticancer activities against SNU-1, and 11.3, 20.2, 36.6, and 30.5% anticancer activities against HeLa, respectively.

  • Antibacterial, antifungal, and Angiotensin-I-converting enzyme (ACE) inhibitory activities of buckwheat (Fagopyrum esculentum and F. tataricum) hydrolyzed by Viscozyme L and Alcalase 2.4 L were investigated. The Alcalase 2.4L-hydrolyzed buckwheat showed highest yield of 22.10-24.65%. F. esculentum hydrolysate treated with Viscozyme L from Salmonella typhimurium (clear zone: 3-4.7 mm) and Listeria monocytogenes (clear zone: 4-7.2 mm) showed highest antimicrobial activity among enzymes used. F. esculentum hydrolysate treated with Trichoderma reesei showed strongest antifungal activity among enzymes used (clear zone: 3.7-12 mm). Alcalase 2.4L-hydrolyzed F. esculentum and F. tataricum showed strong ACE inhibitory activities (61.19% and 94.48%, respectively).
