Molecular Cloning and Expression of the Novel Attacin-Like Antibacterial Protein Gene Isolated from the Bombyx mori (집누에로부터 새로운 attactin 유산 항세균성 펩타이드 유전자의 분리 및 발현)

  • Hyalophora cecropia attacin-like antibacterial gene was isolated from Bombyx mori induced with nonpathogenic bacteria. It was expressed in Spodopfera frugiperda 9 (Sf9 cells using baculovirus expression vector system (BEVS), and examined its antibacterial activity. With a cDNA library constructed from fifthinstar B. mori injected with Escherichia coli(4 X IOhcellsllarva), differential screening was performed using naive and induced mRNA probes. BmInc6 clone was screened by partial nucleotide sequence and GenBank database analysis. A complete nucleotide sequence of Bmlnc6 cDNA was determined (GenBank, AF005384). Its insert size was 852 bp and had open reading frame that started translation at position 35 and stopped at 679. And its putative polyadenylational signal existed at 812 bp. The number of amino acid deduced from Bmlnc6 cDNA was 214 and hydropathy analysis showed that this peptide was hydrophilic. This peptide deduced by BmInc6 was named nuecin. When the nuecin gene was expressed in Sf9 cells using BEVS, about 950 bp of the transcripts was detected. In addition, SDS-PAGE analysis showed that the molecular weights of intracellular expressed protein and the mature protein secreted to culture media were approximately 23 and 20 kDa, respectively. The antibacterial activity of nuecin against E. coli and Bacillus subtilis was significantly high, demonstrating that nuecin had a wider antibacterial spectrum with gram negative and positive bacteria than attacin.

  • In the bioactivity test of nine sesquiterpene lactons isolated from Hemisteptia lyrata, Chrysanthemum zawadskii and Chrysanthemum boreale, we investigated their antimicrobial activities against ten bacteria and six phytopathogenic fungi. Hemistepsin A and hemistepsin B isolated from H. lyrata and tulipinolide isolated from C. boreale exhibited strong antibacterial activity. In the antifungal test, hemistepsin B, angeloylcumambrin B, tigloylcumambrin B and costunolide showed broad activity. Both antibacterial and antifungal experiments exhibited quite clear specificity against microbial strains.

  • The antibacterial protein gene, nuecin was expressed in Sf9 cells using baculovirus expression vector system (BEVS). The antibactetial activity of mature nuecin against Pectobacterium carotovorum subsp. carotovorum, Ralstonia solanacearum and Pseudomonas tolaasii was significantly high, demonstrating that nuecin had a wider antibacterial spectrum on gram negative and positive bacteria. The result appears to be superior to other antibacterial peptide, attacin. The nuecin was purified by SP-sepharose and Mono Q HR ion-exchange chromatography, and then by Superdex 200 HR 10/30 column. The purified nuecin is quite stable at 80$\^{C}$ and 100$\^{C}$ for several hours of incubation and in a wide pH range (pH 2-12).

  • The essential oil from Chamaecyparis obtusa was investigated for biological activities in anti-oxidative, anti-inflammation and antibacterial method, respectively. The Growth inhibitory effect of C. obtusa oil on the bacteria was evaluated with MIC (minimum inhibitory concentration), $IC_{50}\;(50\%$ inhibitory concentration), and paper disc method. Two kinds of gram positive strains and two kinds of gram negative strains were used in this study. Gram positive strains were B. subtilis and S. aureus. and Gram negative strains were E. coli and P. aeruginosa. Gram positive strains showed much more intensive effect than gram negative strains. Anti-oxidative effect was investigated with DPPH (1,1-diphenyl-2-picrylhidrazyl) in methanol based and $IC_{50}\;was\;0.78\%.$ Our results suggest that the essential oil from Chamaecyparis obtusa has effects on anti-bacterial, anti- oxidative and anti-inflammation in in vitro and in uiuo. Then this material could be expect synergic effect with other candidated extracts and oils.

  • Antimicrobial and antioxidative activities on heartwood extractives of domestic species were investigated to develop a natural fungicide or preservative. Six flavanones including pinostrobin, eriodictyol, naringenin, pinocembrin, taxifolin and verecundin were isolated from Prunus sargentii which has been selected due to its high antimicrobial and antioxidative activities among the tested species. According to the results of antifungal test, pinocembrin was evaluated as the highest antifungal compound among the test compounds, which showed 80% of hyphal growth inhibition rate. Antifungal activity of pinocembrin was similar to hinokitiol(${\beta}$-thujaplicin), strong antimicrobial compound isolated from Thujopsis dolabrata. Naringenin followed pinocembrin in its antifungal activity. However, verecundin did not show any antifungal activity. No compound was effective in antibacterial activities. As a result of the measurement of free radical scavenging activity, antioxidative activities of taxifolin and eriodictyol were 2 times that of ${\alpha}$-tocopherol, and antioxidative index of these compounds were even superior to that of ${\alpha}$-tocopherol. In this regard, it could inferred that high antifungal and antioxidative activities of extractives of P. sargentii were derived from pinocembrin, taxifolin and eriodictyol, respectively.

Antioxidant, antibacterial, antifungal, and anti-inflammatory effects of 15 tree essential oils (수목 방향유 15종의 산화방지, 항세균, 항진균 및 항염증 효과)

  • The current study investigated the beneficial effects of 15 essential oils isolated from tree branches, leaves, and flowers. Among these oils, clove bud and Illicium anisatum oils showed the most potent anti-oxidant effects on 1,1-diphenyl-2-picrylhydrazyl and 2,2'azinbis-(3-ethyl-benzothiazoline-6-sulfonic acid) radical scavenging activities. Next, we evaluated the antibacterial effects of 15 essential oils on Staphylococcus aureus, Listeria monocytogenes, Escherichia coli O157:H7, Salmonella typhimurium, and Streptococcus mutans. Clove bud significantly decreased growth of 5 bacterial strains. In addition, clove bud, Magnolia kobus, Picea abies and Chamaecyparis obtuse significantly reduced growth of the fungi, Aspergillus fumigatus, Aspergillus ochraceus, Candida albicans and Trichophyton rubrum. Additionally, clove bud also remarkably reduced the expression of cyclooxygenase-2 and inducible NO synthase in lipopolysaccharide-activated RAW264.7 cells. These results indicate that essential oils isolated from trees, which exhibit antioxidant, antibacterial, antifungal and anti-inflammatory properties, may be potentially useful in the development of cosmetic ingredients.

Evaluation of Antimicrobial, Antithrombin, and Antioxidant Activity of Aerial Bulbils of Dioscorea batatas Decne (영여자의 항균, 항혈전 및 항산화 활성 평가)

  • The annual productions of yam and its aerial bulbils are estimated to 5,000 and 2,500 ton, respectively. But the majority of bulbils had been discarded without specific use. In this study, methanol extract and its organic solvent fractions were prepared from bulbils of Dioscorea batatas Decne, and their antimicrobial, antithrombin, and antioxidant activities were evaluated, respectively. The methanol extract contained 58.98% of water-soluble materials as like yam's extract. But the bulbils's extract contained 12-folds of total polyphenol and 3.4-folds of total flavonoids compared than yam's extract, respectively. For antimicrobial activity the hexane and ethyl acetate fraction showed strong antibacterial activity at $500{\mu}g/disc$ concentration against gram-negative and gram-positive bacteria. Antifungal activity was not observed in any fractions. Strong antithrombin activity was found in the hexane fractions. At 4.8 mg/mL concentration thrombin time (TT) was over 300 sec, which is 4-folds extended than the TT of yam. In a while, the ethyl acetate fraction showed strong DPPH scavenging activity ($IC_{50}$ of $38.1{\mu}g/mL$), SOD-like activity and reducing power, which are comparable to vitamin C or BHT. Our results suggest that the bulbils of yam as yam tuber have useful bio-activities, such as antibacterial, antithrombosis, and antioxidant activity.

Antimicrobial and Antioxidant activity of the Fruit of Prunus avium L (체리(Prunus avium L.)의 항균 및 항산화 활성)

  • The fruits of sweet cherry are highly appreciated by the consumer due to their precocity and quality, such as their sweetness, color and sourness. In this study, the hot-water extract and its sequential organic solvent fractions were prepared from domestic Napoleon sweet cherry (Prunus avium L.) to investigate antimicrobial and antioxidant activity. The hot-water extract contained about 40% sugars, and the solvent fraction yields for hexane, ethylacetate (EA), butanol, and water residue were 0.01%, 3.45%, 16.30%, and 80.24%, respectively. Contents of total polyphenol and total flavonoid of the fractions were 1.24~5.24%, and 0~3.76%, respectively. Among the fractions, EA fraction showed the highest total polyphenol and total flavonoid concentrations. Evaluation of antimicrobial activity of the extract and fractions revealed that EA fraction and butanol fraction contained strong antibacterial activity against Listeria monocytogenes, Staphylococcus aureus, and Salmonella typhimurium with minimal inhibitory concentration (MIC) of 0.5~1.0 mg/mL. But the extract and fractions tested were not active to Pseudomonas aeruginosa. In a while, only hexane fraction showed anti-Candida activity with 0.5~1.0 mg/mL of MIC. The fraction showed strong activity against different multi-antibiotics resistant strains such as C. albicans CCARM 14020. Antioxidative activity assay showed that EA fraction has a strong DPPH scavenging activity and a reducing power. The $IC_{50}s$ of vitamin E and EA fraction were 15.5 and $195.5\;{\mu}g/mL$, respectively. Our results suggest that the fruit of P. avium L. has high potentials with anti-Candida and antioxidative activity.

Characteristics of Yeongcheon Garlic and Usability of Processed Product Development (영천 마늘의 특성 및 가공제품 개발의 활용성)

  • 본 연구는 경북 제1의 마늘 산지인 영천지역의 마늘과 홍마늘의 특성과 생리활성을 평가하고, 이를 통해 액상스틱 가공제품을 개발하는 것이 그 주요한 목표이다. 영천지역 마늘은 난지형 마늘로 20일, 40일 숙성시킨 홍마늘의 경우 숙성이 진행됨에 따라 당/산 비율이 마늘보다 높아 관능적으로 우수하였다. 뿐만아니라 40일 숙성 홍마늘에서 가장 높은 총 폴리페놀 및 총 플라보노이드 함량을 나타내었으며, 항산화능 역시 가장 우수하였다. 마늘보다 홍마늘의 기능성이 우수하지만 지역 마늘 소비 촉진을 위해 우선적으로 일반마늘을 이용한 가공제품 개발 여구를 진행하였고 부원료로는 지역 생산량이 많은 사과, 포도를 활용하였다. 사과 농축액 첨가 제품과 포도 농축액 첨가 제품에서의 일반 성분분석 결과 2종 모두 우수한 관능성을 나타내어 제품화에 적합하였다. 항세균 및 항진균 활성이 나타나지 않았지만, 항당뇨 활성은 기대하기 어려웠다. 하지만 급성 독성을 나타내지 않은 본 제품은 항산화능에서 모두 우수한 결과를 보여 노화억제, 면역력 증강의 효과가 기대되므로 가공제품으로서의 이용성이 높다고 판단된다.

Antibacterial Activity of Caesalpinia sappan and Coptis chinensis Extracts against Bacillus cereus and Vibrio parahaemolyticus (소목(蘇木)과 황련(黃連) 추출물(抽出物)의 Bacillus cereus와 Vibrio parahaemolyticus 에 대한 항세균활성(抗細菌活性))

  • Objectives : This experimental study was performed to investigate the antibacterial effect of Caesalpinia sappan and Coptis chinensis extract against B. cereus and V. parahaemolyticus. Methods : Methanol extracts of C. sappan and C. chinensis was tested against B. cereus and V. parahaemolyticus by paper disc method. Results : The growth of B. cereus and V. parahaemolyticus was inhibited by C. sappan and C. chinensis extract among 6 kinds of medicinal plant extracts. The extract of C. sappan and C. chinensis extract inhibited the growth of V. parahaemolyticus and B. cereus, respectively. The growth of B. cereus and V. parahaemolyticus had a tendency to increase depend on the concentration of the extract. EtOEt and EtOAc fractions and EtOEt and BuOH fractions of the C. sappan extract had a high antibacterial activity against B. cereus and V. parahaemolyticus, respectively. And, BuOH and $H_2O$ fractions of the C. chinensis extract showed antibacterial activity against B. cereus highly. Conclusions : C. sappan and C. chinensis extract efficiently inhibited the growth of B. cereus and V. parahaemolyticus.
