This paper deals with the antioxidative activities of some spices on the autoxidation of refined sardine oil. The various spices powders(120mesh) were added into the refined sardine oil at the level of $0.1\%$ (w/w), and then incubated at $37^{\circ}C$ Among spices tested, herb spices showed higher antioxidative activities than spicy and seed spices. Especially, the antioxidative activities of herb spices on peroxidation of refined sardine oil were most effective in rosemary and sage. Furthermore, the available antioxidative compounds of rosemary and sage were fractionated into petroleum ether-soluble and -insolubles. Petroleum ether-soluble fractions(PESF) obtained from rosemary and sage on the autoxidation of refined sardine oil had a great antioxidative activities. The yields of PESF obtained from rosemary and sage were $10.3\%\;and\;12.6\%$, respectively. The PESF of rosemary and sage showed higher antioxidative effects than butylated hydroxytoluene(BHT), and indicated predominant metal ion-scavenging effect in PESF-refined sardine oil systems.
A Corynebacterium diphtheriae iron-repressible gene dirA, that was homologous to TSA of Saccharomyces cerevisiae and AhpC subunit of Salmonella typhimurium alkyl hydroperoxide reductase, was amplified with PCR and expressed in E. coli. The DirA purified from the transformed E. coli crude extracts prevented the inactivation of enzyme caused by metal-catalyzed oxidation (MCO) system containing thiols but not by ascorbate/Fe$^{3+}$/$O_2$ MCO system. The DirA concentration, which inhibited the inactivation of glutamine synthetase by 50% (IC$_{50}$) against MCO system, was 0.12 mg/ml. The multimeric forms of DirA were converted to the monomeric form in SDS-PAGE under the thioredoxin system comprised of NADPH, Saccharomyces cerevisiae thioredoxin reductase, and thioredoxin. Also, DirA showed thioredoxin dependent peroxidase activity. All of these results were consistent with the characteristics of a thiol specific antioxidant (TSA) protein having two conserved cysteine residues.

In this study, to evaluate the anti-obesity effects of mulberry leaf tea and its fermented product by Monascus pilosus, we investigated body and organ weight, blood and liver biomarkers in mice fed 1% tea infusions instead of water for 8 weeks. Mice were divided into three groups such as a normal control (NC), unfermented mulberry leaf tea infusion (UMI) and fermented mulberry leaf tea infusion (FMI). Although it is not significant, tea infusion groups showed reduction of body weight gains compared with NC group. Moreover, contents of LDL-cholesterol and lipid peroxide (LPO), altherogenic index, and xanthin oxidase (XO) activity were significantly decreased, and glutathione S-transferase (GST) activity was significantly elevated. The results from this study suggested that UMI and FMI may have an anti-obesity activity, upregulate antioxidant enzymes and reduce levels of oxidants related to liver damage.
The antioxidative activity of antioxidative substances produced from several bacterial strains isolated from fermented foods were tested by $DPPH\;({\alpha},{\alpha}'-diphenyl-{\beta}-picrylhydrazyl)$ free radical scavenging activity. One of the strains showing the highest antioxidative activity was identified as Bacillus sp. based on the morphological, biochemical, physiological characteristics, and 16S rRNA sequence, and named FF-7. The most optimal medium condition for the production of antioxidative substance from Bacillus sp. FF-7 was 2% galactose as carbon source and l% tryptone as nitrogen source. The antioxidative substance produced from FF-7 in these cultural medium was also tested by in vitro experimental models, the peruxidation of linoleic acid and the peroxidation of rat tissues microsomes by using thiobarbituric acid (TBA) for assay of free malondialdehyde production. The antioxidative activity against lipid peroxidation of rat tissues microsomes was shown in the following order; brain 97.50% > heart 79.95% > kidney 77.84% > spleen 77.47% > testis 69.96% > liver 62.45%. The antioxidative substance produced from FF-7 on linoleic acid peroxidation by IBA method was effectively inhibited during four days, and 0.05% BHT (butylated hydroxytoluene) used comparative control was also effectively inhibited. Results showed that the highest antioxidative activity by DPPH method of antioxidative substance produced from Bacillus sp. FF-7 was obtained by supplementing 2% galactose as carton source and l% tryptone as nitrogen source in cultured medium, this substance effectively inhibited the formation of TBARS in brain microsome in vit개 system and in linoleic acid peroxidation.

홍화(Carthamus tinctorius L.)의 씨. 순 및 꽃잎의 수용성 및 메탄올 추출물에 대하여 in vitro 실험계에서 항산화 활성을 비교 검토하였다. 성장기 흰쥐의 뇌 micro-some을 이용한 생테막 지질 과산화 억제정도는 꽃잎 수용성 추출물(97.19%)>순 메탄올 추출물(81.38%)>꽃잎 메탄올 추출물(64.99%)>씨 메탄올 추출물(64.95%) 순으로 나타났다. Limoleic acid 산화 실험계에서는 홍화씨 메탄올 추출물과 홍화씨 수용성 추출물에서 비교적 높은 항산화 활성을 보였다. DPPH 에 의한 수소공여능은 특히 순의 0.1% 메탄올 추출물에서 매우 높은 수소공여작용이 나타나 대조구인 BHT와 거의 비슷한 수준의 항산화 활성을 보였다. 이들 추출물의 폴리페놀 화합물 함량은 홍화 꽃잎 수용성 및 MeOH 추출물은 12.70% 및 8.05% 홍화씨 수용성 및 메탄올 추출물은 6.96% 및 12.34%, 홍화순 수용성 메탄올 추출물 및 메탄올 추출물이 8.75% 및 5.10%로 나타났다.

In this study, oregano was extracted by supercritical extraction and hydrothermal extraction method. In vitro experiments such as antimicrobial and antioxidant activity test were performed. As a result of the disc diffusion method, only the supercritical extracts formed a clear zone. The MIC for S. aureus was found only in the supercritical fluid extracts and it was 1000 ㎍/mL. The hydrothermal extract's MIC is 125 ㎍/mL for C. acnes. Through biofilm inhibition assay, we found that the supercritical fluid oregano extracts inhibit the biofilm of S. aureus by more than 70% even at low concentrations of 125 ㎍/mL. On the other hand, the antioxidant ability of the hydrothermal extract was better than that of the supercritical fluid extracts. Furthermore, we tried to make a skincare ingredient for atopic dermatitis by utilizing the S. aureus biofilm inhibitory ability of oregano supercritical fluid extracts. Liposome was used to overcome the low solubility of the oregano supercritical fluid extracts and increase stability.
Licorice, Glycyrrhizae radix, is one of the oldest and most frequently used botanicals in the oriental medicine. Our previous study showed that dehydrolyasperin C (DGC) isolated from licorice had antioxidant activity and induced phase 2 detoxifying enzymes in mouse hepatoma cells. Therefore, this study was conducted to investigate the effect of exposure time to DGC on quinone reductase (QR), one of the anticarcinogenic biomarkers, and antioxidant potential of plasma using animal model. ICR mice were divided into 7 groups, in which mice in each group were injected with DGC (5 mg/kg b.w.) for 0, 2, 4, 6, 8, 12, 24 hours respectively. Following the treatment the organs including liver, kidney, lung, stomach, large intestine, small and large intestines were collected and subjected to QR activity assay, western blotting, and FRAP assay. Exposure to DGC caused a significant induction of QR activity in stomach and large intestine of mice. Ferric reducing activity of plasma, a typical biomarker for antioxidative potentialshowed that DGC improved antioxidant potential in mice. However, no significant effect of DGC was observed in the other organs.
Zinc is an essential trace element for human and plays an important biological role in antioxidant properties. We have been reported that zinc-enriched S. cerevisiae FF-10 contained 392 mg% in the YM basal and 3,193 mg% in the YM optimal medium. Antioxidative activity of FF-10 was tested in vitro models by DPPH (${\alpha},{\alpha}'$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging activity and lipid peroxidation using linoleic acid (LA) and rat liver homogenate. DPPH radical scavenging activity was higher in the cell-free extract of FF-10 cultured in the YM optimal medium (YMOM) than that in the YM basal medium (YMBM). The inhibition activity of lipid peroxidation using rat liver homogenate was shown in the following order: BHT > YMOM > YMBM and these values were dose dependently. The lipid peroxidation of the control mixture by ferric thiocyanate and TBA methods using LA was increased rapidly as typical peroxidation curve of LA from one day and the antioxidation activity of the cell free extracts by cultivating FF-10 in the YMOM were higher than that of the YMBM. Result of this study indicate that the cell-free extracts containing a high intercellular zinc of S. cerevisiae FF-10 cultured in YMOM showed strong antioxidation capacities in DPPH radical scavenging activity and lipid peroxidation using LA and rat liver homogenate.
활성산소 및 그의 반응산물들에 의해 노화가 진행되고, 특히 혈관의 노화인 동맥경화가 활성산소 및 지질과산화에 의해 유발된다면, 생체 내에서 끊임없이 생성되고 있고 경우에 따라 필수 불가결한 활성산소를 어떻게 잘 처리하는 가가 중요하다. 즉, 활성산소 발생정도에 대한 항산화능의 유도가 미치지 못한다면, 체내의 활성산소 및 그 생산물에 의해 노화, 동맥경화가 진행될 것이다. 활성산소의 생성계와 제거계의 balance가 잘 유지된다면 정상적인 노화로 자연사에 이르게 될 것이고, 이 balance가 깨어져 과도한 활성산소의 축적에 대해 제거계가 따르지 못할 경우에는 활성산소와 그 생산물에 의해 동맥경화와 비정상적인 노화가 진행될 것이다.
식품에 항 산화제로 첨가시키는 BHT와 BHA의 독성에 대해 고찰하였다. BHT와 BHA는 실험동물에서 간, 폐, 신장, 순환계, 생식계, 등에 여러 영향을 주며 이러한 영향은 동물의 종류와 성에 따라 상당한 차이를 나타내고 있다. 대부분 실험에 사용된 BHT와 BHA의 양이 인체가 매일 식품으로부터 흡수할 수 있는 양보다는 훨씬 많은 양을 사용하였기 때문에 동물실험의 결과로 인체에 유독성을 직접 판단하기는 어렵고 이들의 안전성에 대해서는 보다 체계적인 연구가 앞으로 요구된다. 미량의 BHT와 BHA를 장기간 복용하였을 때 생기는 부작용에 대한 연구가 필요하며 또한 이들 항 산화제는 약물대사에 관여하는 효소의 합성내지 활성도를 증가시키는 것으로 알려져 있기 때문에 체내에서 다른 약물의 대사 및 독성에 어떤 상호 작용을 가져오는 지에 대한 면밀한 연구가 요구되고 있다. 더불어 본 논문은 현시점에서 이들 항산화제의 식품에 첨가여부를 논쟁하고자 하는 의도가 전혀 없음을 명백히 하는 바이다.
