Age-related Changes of DNA Damage and Antioxidative Capacity in Rats (노화과정에 따른 흰쥐의 DNA손상과 항산화능의 변화)

  • This study was performed to assess age-related changes in DNA damage and antioxidative capacity in 4, 8, 12, 16, 20, and 24 months old Sprague-Dawley male rats. The following were measured the degree of oxidative DNA damage as indicated by levels of 8-hydroxy-2'-deoxyguanosine (80HdG) in the kidney ; the peroxidized lipid concentrations in the plasma and the liver, as indicated by the levels of thiobarbituric acid reactive substances (TBARS); and the levels of antioxidant enzyme activities in the erythrocytes and the liver. Both body weight (BW) and epididymal fat pad (EFP) weight per BW increased with age until 16 months, then decreased slightly from 20 to 24 months. However, the weights of the liver, kidney and spleen per BW decreased with age. Concentrations of 8-OHdG in the kidney increased with age, only slightly front 4 to 16 months, and then markedly from 16 to 24 months. TBARS concentrations in the plasma and liver were shown to increase with age, being lowest in the 4 month-old group and highest in the 24 month-old group. Superoxide dismutase (SOD) activity in the erythrocytes increased with age Catalase activity in the erythrocytes increased from 4 to 16 months, then decreased from 20 to 24 months. Glutathione peroxidase (GSH-Px) activity in the erythrocytes showed no age-related change. Liver SOD activity decreased with age, particularly from 16 to 20 months, but catalase and GSH-Px activities in the liver showed no significant changes. These results showed that during the normal aging of SD rats, DNA damage in the kidney and TBARS concentrations in the plasma and liver increased with age, particularly after 16 months, and the imbalance of antioxidative enzyme activities in the erythrocytes accelerated with age.

  • This study was performed to investigate the effect of kimchi intake on antiaging characteristics in liver of senescence-accelerated mouse (SAM) in terms of free radical production and anti-oxidative enzyme activities. Two hundred twenty SAM were divided into four groups and fed kimchi diet for 12 months. Experimental groups were kimchi free AIN-76 diet (control) group, Korean cabbage kimchi diet (KCK) group, mustard leaf added (30%) Korean cabbage kimchi diet (MKCK) group, and mustard leaf kimchi diet (MLK) group. Amount of freez-dried kimchi added to the diet was 5% that is equivalent to 50 g of fresh kimchi. Concentrations of total free radical, OH radical, $H_2O$$_2$in the liver significantly increased as aged (p<0.05). But those free radical concentrations from kimchi diet groups were lower than those of control (p<0.05). Among kimchi groups, MKCK and MLK groues showed greater inhibiting effect than KCK.   • High-tech medical equipment has increased the utilization of radiation in the medical field. As a result, research on radiation protection using natural materials has become an important social issue. Selenium is a natural substance that is highly expressed in prostate known that an essential role in prostate cells. Selenium was orally administered to Rat and irradiated with 10 Gy of radiation. Then, the prostate tissue w as used as a target organ for 1 day, 7 days and 21 days to investigate the radiation protection effect of selenium through changes of blood components, Superoxide Dismutase and histological changes. As a result, there was a significant protective effect of hematopoietic immune system(hemoglobin concentration, neutrophil, platelet) in the group irradiated with selenium(p<0.05). the observation of tissue changes selenium is an effective component to increase Superoxide Dismutase activity, and it was confirmed that it has an effect of inhibiting the expression of hypertrophy of prostate by irradiation. Therefore, it is considered that selenium can be utilized as a radioprotective agent by inducing prevention of prostate-related diseases.

Varietal Difference in Antioxidative Activity of Ethanolic Extracts from Colored Rice Bran (유색미 에탄올 추출물의 품종간 항산화 활성 변이)

  • Interrelation between the antioxidative activities of 70% ethanol extracts from bran fraction of 23 kinds of colored rice and chuchung as a control were examined. Antioxidative activities were evaluated by assaying reducing power, electron-donation ability to DPPH free radical, scavenging activity of hydroxy radical $({\cdot}OH)$ generated through Fenton reaction and inhibitory activity on lipid peroxidation using linoleic acid autoxidation system, respectively. Among 24 varieties of colored rice LK 1-3-6-12-1-1 had the strongest reducing power followed by Elwee, DZ 78, Jumlalocal-1 and SC-45 in decreasing order. The electron-donating ability to DPPH radical was higher in order of HP 883-1-1-1-B-1-1, HP 833-1-3-1-1-1, LK 2-7-12-1-1 and DZ 78. The hydroxy radical scavenging activity was higher in order of DK-1, IR 1544-38-2-2-1-2-2, SC-5 and SC-45 but LK 2-7-12-1-1 had oxidative effect. In the liaoleic acid autoxidation model system, RGS No 336, LK 1B-2-1-1, LK 1B-4-12-1-1, LK 1A-2-12-1-1, LK 2-7-12-1-1 and HP 883-1-1-1-B-1-1 exhibited strong antioxidative activities but Elwee, Jumlalocal-l and SC-45 showed to have oxidative effects. The rice variety of highest pigment content was Elwee and the next were RGS-No 336, IR 1544-38-2-2-1-2-2 and SC-5 with the order of higher content. The reducing power was correlated with the quantity of the pigment in the ethanolic extract of rice bran and SC-5 showed relatively high antioxidative activity in every results of antioxidative activity tests.

Cellular Protective Effects and Antioxidative Activity of Resveratrol (레스베라트롤의 세포보호 및 항산화 효과)

  • In this study, the cellular protective effect of resveratrol on oxidative damage and its antioxidative activity were investigated. The free radical-scavenging activity ($FSC_{50}$) of resveratrol was measured to be $103{\mu}M$. The reactive oxygen species-scavenging activity ($OSC_{50}$) of resveratrol on the ROS generated in a $Fe^{3+}-EDTA/H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. Resveratrol displayed $0.042{\mu}M$ ROS scavenging activity, which is 9.6-fold higher than that of L-ascorbic acid ($0.405{\mu}M$) and had a more prominent cellular protective effect than (+)-${\alpha}$-tocopherol. When HaCaT cells were exposed to $800mJ/cm^2$ of UVB or treated with $30{\mu}M$ rose bengal, resveratrol protected the cells against oxidative stress in a concentration-dependent manner; however, it was unable to protect the cells when the damage was induced by 10 mM $H_2O_2$. These results indicate that resveratrol could be employed to improve and prevent the skin aging through its antioxidative and cellular protective activities.

Antioxidative and Antiaging Effects of Persicaria hydropiper L. Extracts (여뀌 추출물의 항산화 및 항노화에 관한 연구)

  • In this study, we investigated the antioxidative activity and inhibitory effects on elastase and tyrosinase of Persicaria hydropiper L. extracts. The free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activities ($FSC_{50}$) of ethyl acetate fractions of Persicaria hydropiper L. was $5.23\;{\mu}g/mL$. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some Persicaria hydropiper L. extracts on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system were investigated using the luminol - dependent chemiluminescence assay. The ROS scavenging activities ($OSC_{50}$) of ethyl acetate fractions of Persicaria hydropiper L. was $0.40\;{\mu}g/mL$. The protective effects of extract / fractions of Persicaria hydropiper L. on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The Persicaria hydropiper L. extracts suppressed photohemolysis in a concentration dependent manner ($1\;{\sim}\;10\;{\mu}g/mL$). Inhibitory effects ($IC_{50}$) on tyrosinase of aglycone fraction of Persicaria hydropiper L. extracts was $8.90\;{\mu}g/mL$. Inhibitory effects ($IC_{50}$) on elastase of aglycone fraction of Persicaria hydropiper L. extracts was $2.37\;{\mu}g/mL$. These results indicate that extract / fractions of Persicaria hydropiper L. can function as antioxidants in biological systems, particularly skin exposed to UV radiation by anti-oxidative activity and protect cellular membranes against ROS. Persicaria hydropiper L. extract / fractions could be used as a new cosmeceutical for whitening and anti-wrinkle products.

Antioxidative Activities of Whole Plant Extracts of Solanum nigrum L. (까마중(Solanum nigrum L.) 전초 추출물의 항산화 활성)

  • In this study, the antioxidative effects of 50% ethanol extract, ethylacetate fraction and aglycone fraction obtained from dried whole plant of Solanum nigrum L. were investigated. The free radical scavenging activities ($FSC_{50}$) were $215.46{\mu}g/mL$, $42.43{\mu}g/mL$ and $52.28{\mu}g/mL$, respectively. Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) in $Fe^{3+}-EDTA/H_2O_2$ system were $25.25{\mu}g/mL$, $7.05{\mu}g/mL$ and $6.25{\mu}g/mL$, respectively. 50% ethanol extract and aglycone fraction showed the cellular protective effect against $^1O_2$ induced cellular damage of rabbit erythrocytes at $5{\sim}25{\mu}g/mL$, but not at high concentrations. These results indicated that S. nigrum extract/fractions could be used as an antioxidative agent. However, it could induce cellular damage at high concentrations. In conclusion, a special caution is required to use S. nigrum extracts as a cosmetic ingredient.

Hypoglycemic and antioxidant effects of jaceosidin in streptozotocin-induced diabetic mice (Jaceosidin이 streptozotocin으로 유도된 당뇨 쥐의 혈당강하 및 항산화능에 미치는 영향)

  • Purpose: In this study, we investigated the effects of jaceosidin on blood glucose regulation in type 1 diabetic mice. Methods: C57BL/6 mice were divided into four groups; normal control (Normal), diabetes control (D-Control), diabetes low-jaceosidin (D-0.005%), and diabetes high-jaceosidin (D-0.02%). Type 1 diabetes was induced by streptozotocin and mice were then fed a diet containing jaceosidin for eight weeks. Fasting blood glucose, oral glucose tolerance test, insulin tolerance test, lipid peroxidation, and antioxidant enzyme activities were assessed. Results: Jaceosidin supplementation for eight weeks had no effect on body weight, organ weight, and blood lipid profiles. However, jaceosidin supplementation significantly lowered fasting blood glucose level and reduced insulin resistance. We also found that jaceosidin supplementation increased antioxidant capacity by enhancement of catalase and GSH-px activities. Conclusion: These results suggest that jaceosidin could be a therapeutic candidate to ameliorate hyperglycemia through increase of antioxidant enzyme activity.

Protective Effect of Soybean Sauce and Melanoidin on Lipid Oxidation in Rats Fed High PUFA Oils (고도불포화지방산 함량이 높은 유지를 섭취시킨 흰쥐에서 양조간장과 멜라노이딘의 지질산화 억제효과)

  • Soybean sauce fermented with soybean and wheat, has been a major condiment of Korean diets from centuries ago. Melanoidin, a brown pigment generally found in various food systems, is a final product produced in amino-carbonyl reaction during soybean sauce processing. Antioxidative activities of soybean sauce and melanoidin were investigated in vitro system using linoleic acid emulsion. Soybean sauce and glucose-lysine model melanoidin showed the stronger antioxidative effect than control by ferric thiocyanate and conjugated diene assays. In addition, DPPH radical scavenging effect of soybean sauce was higher than melanoidin, which was ascribed to soluble peptide and low molecular protein existing in soybean sauce. To ascertain antioxidative effect of dietary soybean sauce and melanoidin in vivo, the male Wister rats were fed 10% soybean sauce or 10% glucose-lysine model melanoidin with corn oil or fish oil for 5 weeks. Fatty acid compositions in liver and plasma were influenced by oil source. Therefore, EPA and DHA contents of fish oil group were higher than those of corn oil group. When the inhibitory effect of soybean sauce and melanoidin on lipid peroxidation using TBARS methods was measured, fish oil group (FC) showed higher malondialdehyde (MDA) content than corn oil group (CC). However, supplementation of soybean sauce and melanoidin to fish oil group attenuated MDA formation. In the levels of phosphatidyl choline hydroperoxide (PCOOH) in liver and plasma by CL (chemiluminescence)-HPLC method, PCOOH in FC group was significantly higher than that of CC group both in liver and plasma. Supplementation of soybean sauce to fish oil groups significantly inhibited the formation of PCOOH in plasma and liver, while melanoidin suppressed hepatic PCOOH formation. Based on these results, it can be suggested that soybean sauce possesses stronger antioxidative potential than melanoidin.

Blanching Effect on Quality Characteristics and Antioxidant Activities in Yacon Soaked in Doenjang Sauce (데침에 따른 야콘 된장절임의 항산화 활성 및 품질 변화)

  • The objective of this study was to investigate the quality characteristics of blanched and non-blanched yacon (Smallanthus sonchifolius) soaked in Doenjang sauce. The yacon was treated by blanching ($100^{\circ}C$, 4 min), then soaked in Doenjang sauce at $20^{\circ}C$. The pH and total acidity of blanched yacon were not changed significantly after 42 days of soaking. But, the pH and total acidity of non-blanched yacon were rapidly increased and decreased, respectively. The salinity of blanched yacon was higher than that of non-blanched yacon until 35 days. Also, the $Brix^{\circ}$ of blanched yacon was higher than that of non-blanched yacon from 7 days to 35 days. Textural properties of both non-blanched and blanched yacon showed that hardness increased until 21 days but decreased afterwards. The total polyphenol contents of non-blanched yacon decreased after 14 days, but blanched yacon remained constant. The DPPH radical scavenging activity of both blanched and non-blanched yacon decreased gradually as aging continued in the Doenjang sauce. There were no significant differences in sensory characteristics between blanched and non-blanched yacon soaked in Doenjang sauce.
