The Effect of Exercise Therapy on Serum Lipid Level and Antioxidants of obese College Female Students (운동요법이 비만여대생의 혈청지질 및 항산화계에 미치는 영향)

  • The purpose of this study is to analyze the effects of exercise therapy on serum lipid levels and antioxidants of obese and normal college female students. The subject group composed of ten normal-weight students(below 30% body fat ratio) and ten obese students(above 30% body fat ratio). After a pilot test, the subjects were given an eight-weeks exercise program. Before and after the exercise program, the subjects were given test for serum lipid and antioxidants were analyzed. The SAS program was used in the data analysis. The statistical measurements employed here were T-test, Wilcoxon signed rank test, and Wilcoxon rank sum test. The results of this research are as follows. 1) The effects of exercise therapy on serum lipid levels : Before the exercise therapy, the levels of Total-cholesterol, Triglyceride and LDL-cholesterol of the obese group were highier than those of the normal-weight group. However, the HDL-cholesterol levels were highier in the normal-weight group than in the obese group, but these differences were not significant. With the exercise therapy, the levels of Total-cholesterol increased gradually. The HDL-cholesterol increased gradually, the LDL-cholesterol level decreased in both groups, However, the Triglyceride level decreased in the obese group and increased in normal group, but the difference was not significant. 2) The effects of the exercise therapy on serum antioxidants ; Before exercise therapy, the serum FR and GSSG levels of the obese group were significantly highier than those of the normal-weight group(p=0.00, p=0.04). The serum GSH level of the normal-weight group was highier than that of the obese group, and the serum MDA level of the obese group was highier than that of the normal-weight group. Again these differences were not significant. With exercise therapy, serum FR level was reduced and serum GSSG level significantly increased in both group(obese group p=0.01, normal-weight group p=0.01), The serum GSH level of the obese group significantly increased (p=0.01), and serum MDA level significantly increased in the obese group(p=0.01), but the difference in the normal-weight group was not significant. These results show that regular exercise therapy reduces serum FR levels and activation of antioxidant systems, and suppress oxidative stress. These effects were slightly highier in the obese group than in the normal-weight group. The regular exercise therapy decreased the serum Triglyceride levels more in the obese group than in the normal-weight group. However the improvement of the serum lipid profile may require a longer exercise period than eight weeks. The results show that the exercise therapy was overall more effective in the obese group than the normal-weight group.

Chlorogenic Acid Enhances Glucose Metabolism and Antioxidant System in High-fat Diet and Streptozotocin-induced Diabetic Mice (고지방식이와 스트렙토조토신으로 유도한 당뇨마우스에서 Chlorogenic Acid의 혈당강하 및 항산화 효과)

  • This study investigated dose-response effects of chlorogenic acid (CA) on glucose metabolism and the antioxidant system in streptozotocin (STZ)-induced diabetic mice with a high-fat diet (HFD). Male ICR mice were fed with a HFD (37% calories from fat) for 4 weeks prior to intraperitoneal injection with STZ (100 mg/kg body weight). Diabetic mice were supplemented with two doses of CA (0.02% and 0.05%, wt/wt) for 6 weeks. Both doses of CA significantly improved fasting blood glucose level, glucose tolerance and insulin tolerance without any changes in plasma insulin and C-peptide levels. Plasma leptin concentration was significantly higher in the CA-supplemented groups than in the diabetic control group. Both doses of CA significantly increased hepatic glucokinase activity and decreased glucose-6-phosphatase activity compared to the diabetic control group. The ratio of glucokinase/glucose-6-phosphatase was dose-independently higher in CA-supplemented mice than in diabetic control mice. CA supplementation dose-independently elevated superoxide dismutase and catalase activities, whereas it lowered lipid peroxide levels compared to the diabetic control mice in the liver and erythrocyte. These results suggest that low-dose CA may be used as a hypoglycemic agent in a high-fat diet and STZ-induced diabetic mice.

Antioxidant properties and oxidative stability of celery seeds ethanol extract using in vitro assays and oil-in-water emulsion (샐러리 종자 에탄올 추출물의 산화방지 활성 및 수중유적형 유화계에서의 산화안정성)

  • This study was conducted to examine the antioxidant activity of 80% ethanol extract of celery seeds and to verify the effectiveness of extracts as a natural antioxidant to improve the stability of oil-in-water emulsions. The radical scavenging activity of 80% ethanol extract of celery seeds was significantly increased at 0.125, 0.25, and 0.5 mg/mL (p<0.05). Additionally, the total phenolic content and FRAP value were equal to $8.2{\pm}2.3mol$ tannic acid equivalent/g extract and $195.0{\pm}12.6mol$ ascorbic acid equivalent/g extract, respectively. The headspace oxygen content was significantly higher in the group treated with 80% ethanol extract of celery seeds than in the control group (p<0.05). The amounts of lipid hydroperoxide and conjugated diene were significantly reduced compared to the control group (p<0.05). The results showed that the extract of celery seeds had excellent antioxidant ability and it could be used as a natural antioxidant owing to the increased oxidative stability of the emulsified product.

Antioxidative and Inhibitory Activities on Tyrosinase of Hippophae rhamnoides Leaf Extracts (비타민나무 잎 추출물의 항산화 및 타이로시네이즈 저해활성)

  • In this study, the antioxidative and inhibitory effects on tyrosinase and elastase of Hippophae rhamnoides (H. rhamnoides) leaf extracts were investigated. The ethyl acetate fraction of H. rhamnoides extracts showed more effective free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$ = 4.68 ${\mu}g$/mL). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the aglycone fraction in the luminol-dependent $Fe^{3+}$-EDTA/$H_2O_2$ system was 0.19 ${\mu}g$/mL. The aglycone fraction exhibited more prominent cellular protective effects (${\tau}_{50}$, 133.3 min at 10 ${\mu}g$/mL) in the $^1O_2$-induced photohemolysis of human erythrocytes. The inhibitory effect ($IC_{50}$) of the aglycone fraction on tyrosinase was 54.86 ${\mu}g$/mL, and more effective than arbutin known as whitening agent. These results indicate that fractions of Hippophae rhamnoides extract can be used as antioxidants in biological system, particulaly skin exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes against ROS.

Evaluation of Antioxidant Potential of Extract/Fractions of Equisetum arvense (I) (쇠뜨기 추출물의 항산화 작용에 관한 연구(I))

  • In this study, the antioxidative effects of Equisetum arvense extracts were investigated. The free radical (1,1 diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$) of extract/fractions of Equisetum arvense was in the order: 50 % ethanol extract ($182.04{\mu}g/mL$) < ethylacetate fraction ($54.50{\mu}g/mL$) < deglycosylated flavonoid aglycone fraction ($14.13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of some Equisetum arvense extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated using the luminol- dependent chemiluminescence assay. The order of ROS scavenging activity was deglycosylated flavonoid aglycone fraction ($OSC_{50}$, $3.54{\mu}g/mL$) < 50 % ethanol extract ($0.80{\mu}g/mL$) < ethylacetate fraction ($0.006{\mu}g/mL$). Ethylacetate fraction showed the most prominent scavenging activity. The protective effects of extract/fractions of Equisetum arvense on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethanol extract (50%) suppressed photohemolysis in a concentration dependent manner, particularly deglycosylated aglycone extract exhibited the most prominent celluar protective effect ($\tau_{50}$, 161.10 min at $10{\mu}g/mL$). These results indicate that extract/fractions of Equisetum arvense can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS.

Effect of Inclusion of Chitosan-Oligosaccahariede in Drinking Water on the Blood Component Profile, Immunity and Antioxidative Enzyme in Broiler Chickens (키토산 올리고당의 음수내 급여가 육계의 혈액성상, 면역력 및 항산화 효소계에 미치는 영향)

  • The study was conducted to the effect of inclusion of chitosan-oligosaccharide in drinking water on the blood component profile, immunity and antioxidative enzyme of broiler chickens. A total of 28,000 broiler chickens (Arbor Acre) at 1 days of age were fed the commercial diet until 35 days of age, the treatment divided into two treatments, normal control group and chitosan-oligosaccharide in drinking water group. In concentration of glucose, treatment was significantly decreased than control (P<0.05), but not statistically different on the Triglyceride, Total cholesterol, HDL-cholesterol and LDL-cholesterol in the Blood component profile of broiler chicks. The concentration of glutamic oxalacetic transaminase (GOT) was statistically different but the concentration of glutamic pyruvate transaminase (GPT) was significantly decreased in treatment fed chitosan-oligosaccharide than control (P<0.05). Immunoglobullins in the blood, concentration of IgG was not significantly different among control and treatment but concentration of IgM was significantly increased in fed chitosan-oligosaccharide than control (P<0.05). Antioxidant and super oxide dismutase (SOD) was not different among control and treatment and concentration of catalase and glutathione peroxidase were significantly increased in fed chitosan-oligosaccharide than control (P<0.05).

Inhibitory Effects of Ginger and Garlic Extracts on the DNA Damage (마늘 및 생강추출물의 DNA 손상억제작용)

  • The inhibition mechanism of DNA damage by lipid peroxidation was studied through the reaction systems of plasmid pBR322 DNA, linoleic acid and the ethanol extracts obtained from ginger and garlic. The DNA damage was greatly inhibited by the addition of ginger and garlic extracts, and their scavenging effects of active oxygens were also great. It is considered that the inhibitory effects of these extracts on the DNA damage are mainly due to their scavenging effects of active oxygen radicals.

Effect of Dietary Xanthophylls Supplementation on Pigmentation and Antioxidant Properties in the Egg Yolks (Xanthophylls의 급여가 계란 노른자의 착색 및 항산화성에 미치는 영향)

  • This study was carried out to investigate the effects of dietary xanthophylls supplementation on pigmentation and antioxidant properties in the egg yolk. ISA Brown laying hens aged 50 weeks were fed five kinds of xanthophyll combination such as control(neither natural nor artificial xanthophylls in feed), T1(Commercial diet containing natural xanthopylls in grain + Lutein 10ppm + Capsantin 10ppm), T2(T1 + Capsanthin 65ppm), T3(T1 + Canthaxanthin 65ppm), T4(T1 + Capsanthin 10ppm + Canthaxanthin 65ppm), and T5(T1 + Capsanthin 65ppm + Canthaxanthin 10ppm). The pH values of all egg yolks were not significantly different during storage or feeding periods. CIE L$^{*}$(lightness) values of T2 ~ T5 egg yolks were lower than those of control and T1. Conversely, the CIE a$^{*}$(redness) value of T2 ~ T5 egg yolks showed significantly higher(P<0.05). Egg yolk from chicks fed xanthophylls increased CIE b$^{*}$ values. The CIE b$^{*}$(yellowness) values of T2 ~ T5 egg yolks were higher than control and T1 during storage at 37$^{\circ}C$ for 48 hours respectively. In the antioxidation experiment, dietary xanthophylls supplementation affected lipid antioxidation of egg yolk during storage. The TBARS(O.D.) of egg yolks from chicks fed xanthophylls were lower than that of control during incubation at 37$^{\circ}C$ for 10 hours. In conclusion, dietary xanthophylls supplementation influence to color difference and retardation of lipid oxidation in egg yolk.

Protective Effect of Dietary Buchu (Allium tuberosum Rottler) on Oxidative Stress and Lipofuscin Formation in Streptozotocin-Induced Diabetic Rats (Streptozotocin-유발 당뇨쥐에서 부추식이의 산화적 스트레스 및 Lipofuscin 생성 억제 효과)

  • Diabetes mellitus has been known to be a state of increased oxidative stress. Free radical formation and lipid peroxidation are accelerated in this metabolic disorder. Buchu (Allium tuberosum Rottler) contains lots of antioxidative nutrients such as chlorophyll, vitamin C, $\beta$-carotene, phenolic compounds and sulfur compounds. To investigate the protective effects of buchu, 10% lyophilized buchu diet was fed to streptozotocin (STZ)-induced diabetic rats for 14 weeks and lipid peroxidation, protein oxidation, contents of reactive oxygen species, activities of antioxidative enzymes and contents of accumulated lipofuscin were measured as indicators of oxidative stress. Hepatic MDA and carbonyl contents tended to decrease in 10% buchu diet group compared with control group. Dietary buchu significantly suppressed lipid and protein oxidation in the skin of rats (p<0.05). Contents of hepatic hydroxyl radicals, which exert the highest toxicity among the reactive oxygen species, were significantly decreased in rats fed 10% buchu diet (P<0.05). Activities of antioxidative enzyme, such as superoxide dismutase, catalase, and glutathione peroxidase, tended to increase in liver and skin of rats fed 10% buchu diet, while hepatic catalase activity was significantly increased in buchu group compared with control group. Buchu supplementation significantly inhibited the accumulation of lipofuscin, an end-product of lipid peroxidation reactions induced by reactive oxygen radicals, in eye tissues compared with control diet (p<0.001). In conclusion, buchu supplementation diminished the oxidative stress, so dietary buchu could help to attenuate diabetes complications.

Comparative Evaluation of Biological Activities and Active Compounds of Some Invasive Alien Plants (주요 생태계 교란 외래식물의 생리활성 비교 평가 및 유효성분)

  • To evaluate functional biomaterials of 5 invasive alien plants, total polyphenol and flavonoid contents, antioxidant activity, anti-inflammatory activity, and antibacterial effect were measured. The total polyphenol and flavonoid contents of the extracts were in the order of Rumex acetosella L. > Hypochaeris radicata L. ≥ Lactuca scariola L. > Humulus japonicus Siebold & Zucc. ≥ Solanum viarum Dunal. The DPPH and ABTS radical scavenging activities of the extract were the highest in R. acetosella and correlated well with the total polyphenol contents. In RAW 264.7 cells stimulated with lipopolysaccharide (LPS), nitric oxide (NO) and prostaglandin E2 (PGE2) production inhibitory effect of the extracts (100 ㎍ SE/mL) were 20~60% and 10~70%, respectively, showing the highest inhibitory effect in R. acetocella. The extracts of R. acetosella, H. japonicus and S. viarum showed antibacterial activity against food poisoning-causing microorganisms such as Bacillus subtilis, Escherichia coli, Vibrio parahaemolyticus and Vibrio vulnificus. Furthermore, the H. japonicus extract was found to have effective antibacterial activity against oral microorganisms such as Enterococcus faecalis, Lacticaseibacillus casei, Rothia dentocariosa, Staphylococcus epidermidis and Streptococcus mutans, and its major active ingredients were predicted to be pentadecylic acid, palmitic acid and clionasterol. These results suggest that alien plants have potential as biomaterials with antioxidant, anti-inflammation and antibacterial effects.
