Antioxidant and Anti-aging Activities of Ethanol Extracts from Defatted Perilla frutescens (들깨박 추출물의 항산화 및 항노화 효과)

  • Purpose: This study was conducted to investigate the antioxidative, collagenase and elastase activities of defatted perilla ethanol extracts. In addition, the possibility of its use as a functional cosmetic material has been studied. Methods: In order to investigate the antioxidant function of the perilla extract, electron spin resonance (ESR) was measured with a spectrometer by analyzing the scavenging ability of diphenyl-β-picrylhydrazyl (DPPH) free radicals. Results: The antioxidant activity of DPPH free radical scavenging activity of the perilla extract was about 68% at 85 ㎍/ml, 85% at 20 ㎍/ml, 90% at 40-100 ㎍/ml, and showed antioxidant ability. The inhibitory activity of collagenase was increased to 4.1% at 20 mg/ml, 12% at 40 mg/ml and 26.7% at 100 mg/ml. The inhibitory activity increased in proportion to the concentration. The inhibitory activity of elastase was increased to 3.8% at 20 mg/ml, 15% at 40 mg/ml, 28% at 80 mg/ml and 35.7% at 100 mg/ml. Conclusions: These results suggest that the ethanol extract of perilla may inhibit the antioxidant activity and the activity of collagenase and elastase to improve the skin aging and wrinkles.

Studies on Quality Changes and Antioxidant Activity During the Fermentation of the Salt Fermented Whangseoke (황석어(Collichthys nireatus Jordan et starks) 젓갈의 숙성과정 중 품질변화와 항산화작용에 관한 연구)

  • The effect of storage temperature on the quality and antioxidative activity Whangseoke sauce was studied over a period of 240 days. Fermented Whangseoke with $25\%$ salt were stored at $25^{\circ}C$. The quality change and antioxidant activity of Whangseoke in linoleic acid emulsion was evaluated with various parameters including acids values, peroxide values, TBA values, reducing sugar, brown color intensity, electron donating ability and reducing power at various time intervals for 240 days of storage. In general, it was observed, in all sample, that peroxide values, brown color intensity, electron donating ability and reducing power gradually increased, while reducing sugar decreased during storage at $25^{\circ}C$. The antioxidative activities of fermented Whangseoke were determined on tile linoleic acid emulsion system. The results showed that Whangseoke had antioxidant activity. These results suggest that antioxidant activity of Whangseoke seemed to influence by Maillard reaction products during the storage periods.

Effects of Fatty Acids and Vitamin E Supplementation on Antioxidant Systems in the Liver and Serum of the Second Generation Rat (지방산 및 비타민 E 보충 식이가 제2세대 흰쥐 간조직과 혈청의 항산화체계에 미치는 영향)

  • Effects of dietary fatty acids and vitamin E on antioxidant system were studied in rat liver and serum. Sources of dietary fat (10 wt%) were safflower oil (SO) poor in $\omega$3 fatty acid and mixed oil (MO) with computer-adjusted fatty acid ratios (AA/DHA=1.4, $\omega$6/$\omega$3=6.3, P/M/S=1.0/l.5/1) with (ME) and without (MO) vitamin E (500 mg/kg diet). Rats were fed the three kinds of diet from 3~4 wks prior to the conception. At the age of 3 and 9 wks of the second generation rat, antioxidant vitamins and glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) activities were measured in the liver and serum. The concentrations of $\beta$-carotene were lower in ME than in MO and SO in the liver at the age of 3 wks. It seemed that vitamin E has an inhibitory action on the uptake of $\beta$-carotene or acts as a preferred antioxidant to $\beta$-carotene. The concentrations of lycopene were lower in SO than in MO in the liver at the age of 3 wks. The concentrations of cryptoxanthin showed no significant changes within groups. The activities of GSH-Px tended to increase in ME compared to MO and the ratios of SOD/GSH-Px tended to decrease in ME compared to MO in the liver at the age of 3 weeks. The activities of antioxidant enzyme at the age of 3 weeks and 9 weeks were similar. This suggested that the activity level of antioxidant enzymes reached to the adult level at the age of 3 weeks which is the end point of lactation period.

Effects of Vitamin E Supplementation on Lipid Persoxides and Renal Antioxidative Enzyme Activities in KK Mice (비타민 E 보강식이가 KK 마우스에서 지질과산화물과 신장 항산화계 효소 활성도에 미치는 영향)

  • We investigated the effects of vitamin E supplementation on lipid peroxidation and on the activities of antioxidative enzymes in kidney of KK mice of various age and duration of diabetes. Weaned KK mice were fed high fat a diet containing 20% corn oil(wt/wt), and were sacrificed at 4, 6, and 9 months of age. The high vitamin E diet consisted of the high fat diet supplemented with an excessive amount of dl-$\alpha$-tocopheryl acetate (2080 IU/kg diet) . In the diabetic groups, we found an increase in lipofuscin and decrease in antioxidative enzyme activities with aging. Treatment with vitamin E did not modify the level of fasting blood glucose. However, a significant decrease in lipofuscin and increase in antioxidative enzyme acitvities were observed in diabetic mice.

  • Effects of siho(Bupleuri Radix) extracts on lipid composition and antioxidant system were investigated in rat fed high fat diet. body weight gain, concentration of plasma triglyceride, total cholesterol, LDL-cholesterol showed a tendency to decrease in siho(Bupleuri Radix) extracts groups. However, the concentration of HDL-cholesterol showed no significantly different among treatments. Plasma and liver TBARS concentration showed a low values in siho(Bupleuri Radix) extracts groups. Activities of SOD, GSH-Px and CAT showed a tendency to increase in siho(Bupleuri Radix) extracts groups.

Plasma Antioxidant Status and Platelet Antionxidative Enzyme Activities in Patients of Ischemic Heart Disease (허혈성심질환에서의 항산화영양소 상태와 혈소판 항산화계효소 활성에 관한 연구)

  • To investigate antioxidant status and platelet antioxidative enzyme activity in patients with ischemic heart disease, 36 male patients admitted to Kyungpook National University Hospital from June to December 1994 were compared to 36 healthy male control subjects. The percentages of heavy smoking and nonexercise were significantly higher in the patient group compared to the control, but the drinking status was not significantly different between groups. Food habit and food frequency scores were significantly lower in patients than in control subjects. Plasma retinol levels tended to be lower in the patient group, and plasma $\alpha$-tocopherol and $\beta$-carotene levels were not different between groups. There was no difference in the level of plasma thiobarbituric acid reactive substances(TBARS) and in the activities of platelet glutathione peroxidase and catalase. Our results indicate that oxidative stress, which is reflected by the plasma levels of antioxidants and TBARS, did not increase in the patients with ischemic heart disease, and the long-term effects due to smoking, poor food habit and other life styles could possibly contribute to the onset of the disease.

Effect of Dietary Supplementation of Ground Grape Seed on Growth Performance and Antioxidant Status in the Intestine and Liver in Broiler Chickens (천연 항산화제로서 포도씨 분말 첨가가 육용계의 성장 및 항산화 작용에 미치는 영향)

  • A total of twenty, 2-wk-old male broiler chickens were allotted into control diet(CON) or a diet supplemented with 1% ground grape seed(GGS). They had free access to feed and water for 3 wk. Growth performance and antioxidant markers in plasma, intestine and liver were determined. Dietary addition of 1% GGS did not affect weight gain, feed intake, feed conversion and organ weight in 35 day-old broiler chickens significantly. There was no difference in plasma levels of glucose, triglyceride, cholesterol, AST, ALT and LDH activity. However, total antioxidant status(TAS) in blood increased(P<0.05) in chickens fed the diet supplemented with 1% GGS compared to those fed the control diet. In addition, the specific activity of intestinal superoxide dismutase(SOD) increased(P<0.05) in birds fed the diet supplemented with GGS. However, the activities of intestinal gluthathione peroxidase(GSHPx) and gluthathione -S- transferase(GST) and hepatic SOD, GSHPx and GST were not affected by the dietary GGS. The levels of reduced glutathione and lipid peroxidation in the small intestine and liver were not different between the two groups. In conclusion, dietary supplementation of 1% GGS did not result in a negative effect on growth performance. In addition, some antioxidant indicators including blood TAS and intestinal SOD were markedly elevated in response to dietary GGS. Therefore, dietary addition of 1% GGS may be beneficial to improve antioxidant capacity in broiler chicken.

Antioxidative Activities and Contents of Polyphenolic Compound of Medicinal Herb Extracts (한약재 추출물의 폴리페놀 화합물과 항산화 활성)

  • The antioxidative activity was measured on the substances of water and ethanol soluble extract from Astragalus membranaceus Bunge, Chrysanthemum morifolium Ramat, Lycium chinensis Miller, Glycyrrhiza uralensis Fischer, Angelica gigas Nakai, Zizyphus jojoba Miller, Paeonia lactiflora Pallas, Cnidium officinale Makino by four different in vitro experimental models of DPPH (a,a'-diphenyl-$\beta$-picrylhydrazyl) method, superoxide dismutase like activity, thiocyanate method, and TBARS (thiobarbituric acid reactive substances) method. The Lycium chinensis Miller contained the highest amount of polyphenolic compounds. The electron donating ability of water extract from Glycyrrhiza uralensis Fischer and ethanol extract from Chrysanthemum were higher than those of the others. The superoxide dismutase-like activity of water extract from Astragalus membranaceus Bunge was the highest among those of all the others. The water extract from Zizyphus jujuba Miller showed the highest antioxidative activity determined by TBARS method. Compared to the control, the inducing period associated with the oxidation degree was delayed up to 8 days in both the water extract from Chrysanthemum, Lycium chinensis Miller, Glycyrrhiza uralensis Fischer, and Paeonia lactiflora Pallas and the in ethanol extract from Chrysanthemum and Glycyrrhiz uralensis Fischer. These results support that water and ethanol extracts from 8 kinds of medicinal herbs contain antioxidative compounds.

Antioxidative Activity of Barley Polyphenol Extract (BPE) Separated from Pearling By-products (도정부산물로부터 분리한 보리 폴리페놀 추출물의 항산화 효과)

  • Barley bran and germ fractions were collected during pearling process. Barley polyphenol extract (BPE) was separated from the pearling fractions, and its antioxidative activity was determined through linoleic acid model system and lipid autoxidation model using corn oil as a substrate. At 0.02% addition level, thiobarbituric acid (TBA) value of BPE from bran fraction III appeared to be similar to that of ${\alpha}-tocopherol$, and bran fraction I and germ extract exhibited similar antioxidative activities to that of BHT. Peroxide value was measured to estimate antioxidative activity of BPE upon lipid autoxidation. As BPE concentration increased, higher antioxidative activity was observed compared to ${\alpha}-tocopherol$ and BHT until 10-15 day storage, whereas no effect was shown after 20-day storage. Relative antioxidant effectiveness (RAE) of $0.02{\sim}0.1%$ BPE from bran fraction I and germ were $128{\sim}135$ and $126{\sim}133$, respectively, and appeared to be higher than that of BHT (126), suggesting that these BPE fractions could be used as natural antioxidants.

Study on Antioxidant and Staminal Activities of Kejihongsamatang (계지홍삼탕의 항산화 및 지구력 효과에 대한 연구)

  • KJHST (Kejihongsarntang) is a modified oriental prescription that consists of five herbs such as Ginseng Radix rubra Koreana, Atractylodis Rhizoma, Zingiberis Rhiaoma, Cinnamomi Ramulus and Glycyrrhizae Radix. For the evaluation of antioxidant and staminal activities of KJHST (Kejihongsarntang), the study was done in comparison of Ginseng Radix rubra (GR). For the antioxidant study, KJHST inhibited hemolysis of erythrocyte and decolored the DPPH(2,2-diphenyl-1-picrylhydrazyl) free radical in a dose depenrlent manner more effectively than GR alone in vitro. KJHST and GR significantly suppressed the time course (1 hr∼6 hr)-level of MDA (malondialdehyde) following AAPH (2,2'-azo-bis-(2-amidino-propane) dihydrochloride) treatment in vivo as compared with control data with no statistical difference. From the evaluation of stamina by swimming test GR and KJHST significantly increased the swimming time in a time and dose dependent manner as compared with control data, while GR was more effective than KJHST in 2 weeks after treatment, though KJHST was more effective than GR at low dose (25 m/kg) 4 weeks after treatment. From the results it can be concluded GR and KJHST had antioxidant and staminal activities.
