Study on the Screening and Development of Antibiotics in the Mushrooms -The Screening of Bacterial Antibiotics in Basidiomycetes (I)- (버섯 중 항균물질의 검색 및 개발에 관한 연구 -그람양성균에 대한 항균물질의 검색 (1보)-)

  • To investigate the antibacterial activities of 53 species of mushrooms for Gram-positive bacteria, bioactive substances from mushrooms were extracted with petroleum ether, 80% ethanol, and distilled water. The 14 extracts of 12 species of mushrooms, largely with 80% ethanol, showed antibacterial activities against 6 species of Gram-positive bacteria selectively. Especially, the extracts of Thylopilus PS-5, Pulveroboletus PS-7, and Russula PS-9 showed various antibacterial activities against several bacteria. Minimal inhibitory concentration (MIC) of the 80% ethanol extracts from Pulveroboletus PS-7 and Boletus PS-8 was $300\;{\mu}g/ml$ against Bacillus subtilis, and that of the 80% ethanol extracts from Trichaptium PS-3 was $500\;{\mu}g/ml$ against Micrococcus luteus. MIC of other extracts with antibacterial activities was above $1000\;{\mu}g/ml$.

Characterization of Antimicrobial Substance Produced by Lactobacillus paraplantarum KNUC25 Isolated from Kimchi (김치로부터 분리된 Lactobacillus paraplantarum KNUC25가 만드는 항균 물질의 특성)

  • The KNUC25 strain isolated from over-fermented whole Chinese cabbage kimchi was examined for its physiological characteristics using API 50 CHL system assay and identified as Lactobacillus paraplantarum by analysis of whole-cell protein SDS-PAGE pattern assay and similarity of 16S rDNA sequence. L. paraplantarum KNUC25 had a broad antimicrobial activity spectrum from Gram positive to Gram negative bacteria. Scanning electron micrograph analysis showed that KNUC25 might attack to cell surface of indicator cells and destruction can lead to inhibition of the cell growth. The antimicrobial substance of the KNUC25 strain was stable to various degrading enzymes and at high temperature and not a plasmid-born matter. Resistance to proteolytic enzymes showed that an antimicrobial activity of KNUC25 might not be caused by proteinous substance. Maximum production of antimicrobial substance was the exponential growth phase at $30^{\circ}C$.

Identification of Antifungal Activity Subatnces on Seedborn Disease from Garlic and Taxus Extracts (마늘, 주목의 추출물로부터 종자전염성병에 대한 항균활성물질 동정)

  • Antifungal substances were isolated and identified from garlic and taxus extracts to develop safe and broad fungicide. The inhibitory effect of seedborn disease of sesame, pepper, radish, chinese cabbage by formulation of antifungal substances was investigated. The antifungal substance isolated through column chromatography from garlic and taxus extracts was confirmed by GC-MS as allicin($C_6H_{10}OS_2$) and taxol($C_{47}H_{51}NO_{14}$) and the quantified content from each extracts by HPLC analysis was 0.62%, 0.29%, respectively. The formulation composed of garlic and taxus extracts controlled effectively the seedborn fungi tested in this study at 10X dilution, but at 100X dilution the inhibitory effect decreased. Phytotoxicity of these formulations did not recognized.

Characterization of the Bacteriocin Produced by a Leuconostoc mesenteroides Strain Inhibiting the Growth of Lactobacillus sakei (Lactobacillus sakei 생육저해활성 보유 Leuconostoc mesenteroides가 생산하는 Bacteriocin의 특성)

  • Lactobacillus sakei is known to be the most populous lactic acid bacteria in over-ripened kimchi. Twenty three strains of Leuconostoc species inhibiting the growth of Lb. sakei were isolated from kimchi and amongst these the Leuconostoc mesenteroides strain CK0122 exhibited the highest antagonistic activity against Lb. sakei. The culture supernatant of the strain CK0122 was fractionated by a molecular weight cutter and lyophilized. The fraction with a molecular weight of less than 3,000 Da showed antagonistic activity against Lb. sakei. The antibacterial activity of the active fraction was sensitive to proteinase K treatment, confirming its proteinaceous nature (bacteriocin). The crude bacteriocin was active in the pH range of 4 to 7 and extremely stable after 15 min of heat treatment at $121^{\circ}C$. The crude bacteriocin inhibited the growth of Bacillus cereus, Listeria monocytogenes, Staphylococcus aureus, Alcaligenes xylosoxydans, Flavobacterium sp., and Salmonella typhimurium.

  • This study was conducted to develop environment-friendly agricultural products with anti-microbial activity against Acidovorax avenae subsp. citrulli as a pathogen of bacterial fruit blotch in cucurbit. Schlechtendalia chinensis was extracted by MeOH and solvent fraction. The hexane fraction, which showed highest value of anti-microbial activity, was analyzed by GC-MS. Each mass spectra, corresponding to each peak of chromatogram, was compared to MS database of Wiley library. As a result, myristic acid, palmitic acid and 3-n-pentadecylphenol were identified as maine compounds showing antimicrobial activity against A. avenae subsp. citrulli. Bioassay using commercial myristic acid, palmitic acid and 3-n-pentadecylphenol to test for the anti-microbial activity conformed the anti-microbial activity of potential active compounds, and myristic acid and 3-n-pentadecylphenol showed strong activity. In conclusion, myristic acid and 3-n-pentadecylphenol identified from S. chinensis were anti-microbial chemicals.

The Antioxidative and Antimicrobial Effects of Internal Organs of Aplysia kurodai Fractions (군소내장 분획물의 항산화 및 항균효과)

  • This study was carried out to investigate the antioxidative and antimicrobial activities of internal organs of Aplysia kurodai (AK). The internal organs of AK were extracted with methanol (AKM), which was then further fractionated into four subfractions by using solvent partition method, affording hexane (AKMH), methanol (AKMM), butanol (AKMB), and aqueous (AKMA) soluble fractions. The antioxidative activity of fractions from AK was investigated by measuring the scavenging activities of AK against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species (ROS). Among the various solvent fractions, AKMB showed a marked scavenging effect against DPPH radical, peroxynitrite ($ONOO^-$) and reactive oxygen species (ROS). The antimicrobial activity was increased in proportion to its concentration by the paper disk method. Among the various solvent fractions, AKMM fractions of AK showed the strongest antimicrobial activities. The methanol extracts exhibited antimicrobial activity against all organisms tested, while the hexane extracts showed antimicrobial activity only against Proteus vulgaris. The results suggest that the AK may be suitable for development as a food preservative and alternative antioxidant.

Isolation and Characterization of Bio-active Materials from Prickly Lettuce (Lactuca serriola) (가시상추 유래 생리활성물질의 분리 및 특성 규명)

  • The bio-active materials were isolated from prickly lettuce (Lactuca serriola) by using several extraction solvents. The contents of general compounds, vitamins, polyphenols and flavonoids of prickly lettuce were analysed. In addition, nitrite scavenging activity, anti-oxidative activity and anti-microbial activity of methanol extracts were measured. The ethylacetate (EtOAc) fraction of methanol extracts of prickly lettuce showed the best on the nitrite scavenging activity, anti-oxidant activity and anti-microbial activity. And also, the contents of polyphenols and flavonoids were the highest among all fractions. The subfraction 2 (EtOAc:MeOH=3:1) of EtOAc fraction showed the strongest anti-oxidative activity among 5 subfractions. The subfraction 2 was identified as ${\rho}$-coumaric acid and caffeic acid by GC-MS. The anti-microbial activity of each solvent fraction from prickly lettuce was measured. The EtOAc fraction showed the strongest anti-microbial activity. Overall, these results may provide the basic data to understand the biological activities of bio-active materials derived from prickly lettuce.

Antimicrobial Activity of the Coriolus versicolor Liquid Culture Extracts Against Antibiotic Resistant Bacteria and Purification of Active Substance (구름버섯 균사체 배양 추출물의 복합내성 세균에 대한 항균활성 및 활성물질의 정제)

  • The liquid culture extract of Coriolus versicolor was prepared by directly boiling the whole culture broth 7 days after incubation in 12% citrus extract medium. After removal of mycelial debris through filtration, this extract was further extracted with equal volume of ethyl acetate (1 : 1, v/v). The ethyl acetate extracts showed significant antibacterial activities against Stapylococcus aureus CCARM3230 and Psudomonas aeruginosa CCARM2171, which are resistant to several antibiotics. The most active fraction was eluted from a silica gel column with a mixture of dichloromethane and methanol (9 : 1, v/v) and the purity of this active substance was confirmed by HPLC analysis. The results suggest that the purified active substance could be a good source for the development of a new antimicrobial agent, especially for the treatment of antibiotic resistant bacteria.

Antimicrobial activity of Dryopteris crassirhizoma against Streptococcus sobrinus (Streptococcus sobrinus에 대한 Dryopteris crassirhizoma의 항균활성)

  • As part of our screening of anticariogenic agents from medicinal plants, the ethanol extracts of 10 herbs widely used in the fork medicine were tested for the antimicrobial activity against a cariogenic bacterium Streptococcus sobrinus. The ethanol extracts of Dryopteris crassirhizoma showed antimicrobial activities against S. sobrinus. These results suggested that the extracts of Dryopteris crassirhizoma could be the potential source of antimicrobial agent against S. sobrinus.

  • Ninty three species of domestic weeds were collected and screened for antimicrobial, antitumor, antioxidant and herbicidal activities. Among them, few showed antifungal activities. Cuscuta japonica showed inhibitory activity against Alternaria mali, Ambrosia artemisiifolia and Geranium sibiricum against Phytophthora capsici, Aster yomema and Aster pilosus against Phytophthora parasitica. Ambrosia artemisiifolia, Artemisia princeps, Artemisia capillaris, Ludwigia prostrata, Chrysanthemum zawadskii, Bidens frondosa, and Geranium sibiricum showed broad antibacterial activities. Carex chordorhiza, Artemisia capillaris, Persicaria nodosa, Senecio koreanus, Pariicum bisulcatum, Geranium sibiricum showed antiblebbing activity on human chronic leukemia K562 cell, among them, Persicaria nodosa was the strongist. Angelica decursiva, Equisetum arvense, Cimicifuga heracleifolia, Persicaria nodosa, Geranium sibiricum, Oenothera odorata, Cyperus sanguinolentus showed antioxidant activities. Ludwigia prostrata and Peucedanum terebinthaceum showed strong herbicidal activities.
