Antimicrobial Activity of Solvent Fraction from Cornus officianalis (산수유의 용매분획별 항균활성)

  • To develop natural food preservatives, ethanol and water extracts were prepared from the cornus (Cornus of officianalis) and antimicrobial activities were examined against 10 microoganisms which were food borne pathogens and/or food poisoning microoganisms, food-related bacteria and yeasts. Ethanol extract exhibited antimicrobial activity for the microoganisms tested, except lactic acid bacteria and yeast. Especially, minimum inhibitory concentrations (MIC) of the ethanol extracts were determined as 0.25 mg/mL against bacteria and 2 mg/mL against target lactic bacteria and yeasts. Antimicrobial activity of the ethanol extracts were not destroyed by the heating at 121$^{\circ}C$ for 15 min and not affected by pH. The ethanol extract of cornus exhibiting high antimicrobial activities were fractionated in the other of diethylether and butanol fractions to test antimicrobial activity The antimicrobial activity adjust bacteria test was highest in the ethanol fraction.

Antifungal Activity or Coptis japonica Root-stem extract and Identification of Antifungal Substances (황련추출액의 항균활성과 항균성물질의 동정)

  • Crude extract of Coptis japonica root-stem was evaluated for antifungal activity against Phytophthora capsici, Fusarium oxysporum, Colletotrichum dematium, Colletotrichum truncatum, Botrytis cinerea, Botryosphaeria dothidea and Alternaria porri, and antifungal active compound from the extract was identified. In addition, the usefulness of the extract for some plants disease control was investigated. Crude extract of C. japonica root-stem exhibited antifungal activity against P. capsici, F. oxysporum, C. dematium, B. cinerea, B. dothidea and A. porri. Antifungal activity of the substance isolated from C. japonica root-stem was similar to a standard chemical berberine-Cl. Red-pepper fruit rot, sesame stem rot and welsh-onion alternaria leaf spot were effectively controlled by the crude extract of C. japonica root-stem. Phytotoxicity was not observed in the red-pepper and welsh-onion leaves and red-pepper and strawberry fruits with exogenous foliage application of the crude extract. Seeds germination and radicle growth of red-pepper and sesame were inhibited by the crude extract of C. japonica root-stem. 4.24g of yellowish compound per 100g of C. japonica root-stem was obtained. The compound was identified as berberine-Cl by HPLC.

  • To develop natural food preservatives, ethanol and water extracts were prepared from the coriander (Coriandrum sativum L.) and antimicrobial activities were examined against 10 microorganisms which were food borne pathogens and/or food poisoning microorganisms and food-related bacteria and yeasts. Isolation of Antimicrobial Substance from Schizandra chinensis Baillon and Antimicrobial Effect (오미자로부터 항균활성 물질의 분리 및 항균효과)

  • In order to isolate antimicrobial substances from Schizandra chinensis, the dried fruits were extracted with the methanol and the extract showed a strong antimicrobial activity. Also, the methanol exract was further fractionated with hexane, dichloromethane, ethylacetate and buthanol. The ethyl acetate-soluble fraction showed the strongest antimicrobial activity. These fraction were further separated by using various chromatographic methods including thin layer chromatography, silicagel open column chromatography and prep. HPLC. A major component S-EA-5-T1 and S-EA-5-T3 from the ethyl acetate fraction, which showed a strong antimicrobial activity was identified by Mass and NMR spectrometry. Two compounds were isolated and identified as trimethylcitrate and the essential oil of Schizandra chinensis and was estimated as gomisin C, respectively. The growth of S. typhimurium was also inhibited about 1.65 to 2.86 log cycle in minced pork by the addition 1% of Schizandra chinensis extract for 12 days at $4^{\circ}C$. These results suggested that these compounds have a strong potential as a natual food preservatives.

  • To obtain antimicrobial compounds against fish pathogenic bacteria from natural products, 80% methanolic extracts from 14 species of medicinal plant were screened for antimicrobial activity against fish pathogenic bacteria, Edwardsiella tarda and Vibrio anguillarum. Among them, Glycyrrhiza glabra, Rhus vemiciflua and Sanguisorba officinalis were effective for growth inhibition of Gram-negative bacteria, both E. tarda YSF and V. anguillarum YSR. Through the activity-guided isolation for R. verniciflua extract that exhibited the highest antimicrobial activity among three extracts, one antimicrobial compound (1) was isolated and identified as methyl-3,4,5-trihydroxybenzoate, or methyl gallate. This compound significantly inhibited the growth of tested strains of both E. tarda and V. anguillarum exhibiting MIC of 1 mg/ml for each strain.

Antimicrobial Activity of the Aerial Part of Artemisia capillaris Extracts on the Food-Borne Pathogens (인진호 추출물의 식중독 세균들에 대한 생육억제 효과)

  • The solvent extracts of the aerial part of Artemisia capillaris extracted by using several solvents with different polarities were prepared and their antimicrobial activities were examined. The antimicrobial activities and cell growth inhibitions were investigated to each strain with the different concentrations of the aerial part of Artemisia capillaris Acetone extract showed the highest antimicrobial activity. Minimum inhibitory concentrations (MIC) for each strain were appeared to 20 mg/mL at Staphylococcus aureus and Bacillus subtilis, 40 mg/mL at Vibrio parahaemolyticus, and 80 mg/mL at Salmonella tyhimurium. The cell growth inhibitions were shown on Staphylococcus aureus, Bacillus subtilis, Vibrio parahaemolyticus, and Salmonella typhimurium for 48 hours. The acetone extract was further fractionated sequentially with hexane, chloroform, ethyl acetate, and butanol for purifying crude acetone extract. The solvent fraction of hexane, chloroform, ethyl acetate, and butanol showed the different antimicrobial activity, respectively.

  • 토양에서 분리한 방선균류(actinomycetes) GT103 균주의 배양여액 및 균체추출물은 담배의 주요 병원균인 담배 줄기속마름병균(Erwinia carotovora subsp. carotovora), 담배 탄저병균(Colletotrichum tabacum), 담배역병균(Phytophthora nicotianae var. nicotianae)등에 강한 항균활성을 보였다. 항균활성물질은 용매 추출, silica gel chromatography, HPLC 등을 실시하여 분리.정제하였으며 UV, IR, FAB-MS, \ulcornerH-NMR, \ulcornerC-NMR 분석결과 actinomycin group의 펩타이드계 항생 물질인 actinomycin C\ulcorner로 동정되었다. 이 활성물질의 담배 탄저병에 대한 방제효과는 50$\mu\textrm{g}$/ml 농도에서 100%, 10$\mu\textrm{g}$/ml 농도에서 90%의 방제효과를 보였다.

Purification and Characterization of an Antimicrobial Substance from Bacillus subtilis HH28 Antagonistic to Bacillus cereus (Bacillus cereus를 억제하는 Bacillus subtilis HH28의 항균물질 정제와 특성규명)

  • A bacterium producing antimicrobial substance was isolated from cheonggukjang. The bacterium was identified as a strain of Bacillus subtilis by 16S rDNA sequencing and designated as Bacillus subtilis HH28. The antimicrobial substance produced from Bacillus subtilis HH28 was purified by 0-80% ammonium sulfate precipitation, DEAE-sepharose FF column chromatography, and Sephacryl S-200 HR gel chromatography. The molecular weight of the purified antimicrobial substance was estimated to be approximately 3,500 Da using Tricine sodium dodecyl sulfate-polyacrylamide gel electrophoresis and direct detection analysis. Antimicrobial substance from B. subtilis HH28 not only inhibited B. cereus, but also Listeria monocytogenes and Vibrio parahaemolyticus. The purified antimicrobial substance was stable at $40-80^{\circ}C$, and between pH 2 and 8. Antimicrobial activity of the purified substance was completely destroyed by treatment of protease, proteinase K, and pronase E, indicating that it is proteinaceous.

Antifungal Activities of Equisetin, Zearalenone, and 8'-Hydroxyaearalenone Isolated from Fusarium Species against Plant Pathogenic Fungi. (Fusarium속 균주로부터 분리한 Equisetin, Zearalenone 및 8'-Hydroxyzearalenone의 식물병원곰팡이에 대한 항균활성)

  • Antifungal substances were isolated from solid cultures of Fusarium equiseti FO-68 obtained from arrowhead and Fusarium sp. FO-510 obtained from egg plant, and then their antifungal activities were investigated against plant pathogenic fungi in vitro and in vivo. An antifungal substance was purifed from rice solid cultures of F. equiseti FO-68 and identified as equisetin. In addition, two antibiotic substances were isolated from solid cultures of Fusarium sp. FO-510 and their chemical structures were determined to be zearalenone and 8'-hydroxyzearalenone. in vitro, equisetin and zearalenone inhibited mycelial growth of most of the plant pathogenic fungi tested, whereas 8'-hydroxyzearalenone hardly inhibited fungal growth. In vitro, equisetin effectively controlled the development of tomato gray mold and tomato late blight. Zearalenone exhibited in vivo antifungal activity against rice blast, rice sheath blight, tomato gray mold, and tomato late blight. However, 8'-hydroxyzearale-none did not control the development of plant diseases except tomato gray mold. This is the first report on the antifungal activities of equisetin, zearalenone, and 8'-hydroxyzearalenone.

Search for Plant-originated Antibacterial Compounds Against Pathogen (Acidovorax avenae subsp. citrulli) of Watermelon Bacterial Fruit Blotch (수박 과실썩음병 병원균(Acidovorax avenae subsp. citrulli)에 대한 식물유래 항균 활성물질 탐색)

  • 133 Species of medicinal plants were used for the development of natural agrichemicals with anti-microbial activity against Acidovorax avenae subsp. citrulli, a pathogen of bacterial fruit blotch in watermelon. The MeOH-extracts of these medicinal plants were examined for anti-microbial activity by bioassay. The MeOH-extract of Citrus unshiu Markovich had the strongest antibacterial activity against Acidovorax avenae subsp. citrulli. To identify anti-microbial compounds from Citrus unshiu Markovich, solvent-fractionation was used. The fraction of hexane, which showing the highest value of anti-microbial activity, was analyzed by GC-MS. Each mass spectra, corresponding to each peak of chromatogram, was compared to mass database of Wiley library. As a result, d-Limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, terpineol, palmitic acid, 9,12-octadecadienoic acid, Linolenic acid, and stigmasterol were identified. Among them, d-Limonene, ${\gamma}$-terpinene, ${\beta}$-linalool, and terpineol confirmed to be shown the anti-microbial activity by bioassay. Especially, d-Limonene and ${\gamma}$-terpinene found to have strong activity. In conclusion, we thought d-limonene and ${\gamma}$-terpinene from Citrus unshiu Markovich. Latin, had anti-microbial activity against Acidovorax avenae subsp. citrulli and could be candidates for the control agents for the control of bacterial fruit blotch in watermelon.
