Antibacterial Activity of Continentalic Acid from Aralia continentalis (독활(獨活)(Aralia continentalis)추출물 Continentalic Acid의 항균활성 연구)

  • 독활(Aralia continentalis)로부터 항균활성 물질을 찾아내기 위한 일환으로 항균실험과 분리실험을 병행하여 실시하였다. 항균활성물질 분리는 독활을 클로로포롬으로 추출하여 실리카겔($SiO_2$)과 분취용액체크로마토그래피(prep-HPLC)법으로 2종의 화합물을 분리하여 핵자기공명(NMR) 등 분광학적인 기법으로 이용하여 구조동정을 하였다. 이때 2종의 화합물은 (-)-pimara-8(1),15-diene-19-oic acid와 (24E)-stagmasta-5,22-dien-$3{\beta}-ol$임이 확인되었다. 2종의 화합물에 대한 메티실린 내성 황색포도상구균(MRSA) 및 메터실린 감응 황색포도상구균(MSSA)의 표준균주와 임상분리균주(MRSA)에서의 최소억제농도(MIC)가 $8-16{\mu}g/mL$로 나타났다. 그러므로 본 연구의 결과로부터 화합물(-)pimara-8(1),15-diene-19-oic acid은 항생제 내성균에 대한 치료제로서 개발 가능성을 확인할 수 있었다.

Antibacterial and Mechanical Properties of Glass Ionomer Cements Containing Functionalized Multi-wall Carbon Nanotube(MWCNT) (기능화 다중벽 탄소나노튜브를 첨가한 글라스아이오노머 시멘트의 항균효과 및 기계적 성질)

  • The purpose of this study was to investigate the antibacterial effect of functional multiwall carbon nanotube (MWCNT) on the conventional glass ionomer cement(GIC). The MWCNT was incorporated into a commercial powder at 0.125, 0.25, 0.5 and 1.0 wt%. Specimens for vickers hardness(VHN) disks(Ø$10mm{\times}2mm$) each were prepared. Indention were made using a load of 200 N and 20 s dwell on four specimens for each distilled water storage time 1d, 7d, and 14days. The antibacterial effects using agar diffusion test with S. mutans and S. aureus. The diameters of the inhibition zones produced around the materials were measured. The results revealed that all the vickers hardness values were increase significantly with incubation time(p<0.05). Regarding the antibacterial effect for S. mutans, all the tested groups showed a slightly higher value not significantly with the control group(p<0.05). However S. aureus statistical analysis indicated a significant difference for antibacterial agents between control and MWCNT containing 0.25 wt%(p<0.05). These results of this study provide that the conventional GIC with containing MWCNT show good antibacterial effect against and favorable mechanical properties. Further this study on the efficient functionalization of multiwall carbon nanotube will be needed.

Extraction Process Optimization of Poncirus trifoliata and Prunus mume for Antibacterial Activity against Vibrio parahaemolyticus (장염비브리오 항균활성을 위한 탱자와 매실의 추출조건 최적화)

  • The present investigation was carried out particularly to optimize the extraction process of Poncirus trifoliata and Prunus mume to develop a functional foodstuff having antibacterial activity against Vibrio parahaemolyticus. The extracts were prepared from the fruits under various conditions, and then optimum extraction conditions were decided in case maximal antibacterial activity was obtained. In extraction process by using hot water, the best antibacterial activity of Poncirus trifoliata was achieved at $80{\sim}100^{\circ}C$ for 30 min, while that of Prunus mume was achieved at $100^{\circ}C$ for 30 min, as showing 14.8 and 16.6 mm of inhibition diameter, respectively. The extraction process with addition of NaOH increased the activity of Prunus mume, but addition of $K_2CO_3$ and $NaHCO_3$ drastically decreased the activity. In the processes at high temperature up to $120^{\circ}C$ by using pressure extractor and with various concentrations of NaOH, the maximum activity was observed in the extract of Prunus mume at $120^{\circ}C$ for 30 min with 0.05 N NaOH. The fermented ethanol extract of the fruits showed less activity than those of the extracts with water and NaOH solution.

Antimicrobial Activity of Aqueous Ethanol Extracts of Perilla frutescens var. acuta Leaf (차조기의 에탄올과 물 혼합 추출물의 항균활성)

  • This study was conducted to examine the antimicrobial activity of Perilla frutescens var. acuta leaf fractions extracted with a mixture of ethanol and water. The Ca and Mg contents of the leaf were 595.75 mg% and 467.0 mg%, respectively, and they were the highest among all of the test minerals. The extract yield increased w e content of water in e extraction solvent. Antimicrobial activity against Staphylococcus aureus, Bacillus subtilis and Pseudomonas aeruginosa was found in the 50,70 and 95% ethanol extracts. Of the various fractions extracted from the mixture of ethanol and water, the ethyl acetate fraction showed antibacterial activity against all microorganisms tested in this experiment, and the antibacterial activity of ethyl acetate fraction from the water extract was the strongest. The phenol and flavonoid content in the ethyl acetate fraction showed no correlation with the concentration of ethanol in the extract solvent; however, their contents were higher in the 30% ethanol and water extraction which the antimicrobial activity of the extract was the strongest.

Antimicrobial Activity of Extracted by Supercritical Fluid from Origanum vulgare, Cinnamomum cassia, Chamaecyparis obtusa and Scutellariae baicalensis (오레가노, 육계, 편백 및 황금의 초임계 유체 추출물의 항균 활성)

  • The variety of functional plants has an attention for new natural food preservation and natural antiseptic development. The extracts from functional plants with various methods (ethanol extraction, hot water extraction and supercritical fluid extraction) tested antimicrobial activity against 10 strains including the pathogenic and food poisoning bacteria, the yeast and fungi. The antimicrobial activities of supercritical fluid extracts were shown higher than ethanol extract and hot water extract when tested with disc-diffusion method and minimum inhibitory concentration (MIC). Antimicrobial activity of supercritical fluid extract was two times higher than ethanol extract in Cinnaonomum cassia. In addition, the supercritical fluid extractions of Chamaecyparis obtuas and the C. cassia showed the higher yield than Origanum vulgare and Scutellariae baicalensis. The supercritical fluid extract of C. cassia showed an antimicrobial activity against all strains tested. The supercritical fluid extract of S. baicalensis showed strong antimicrobial activity on Listeria monocytogenes. Supercritical fluid extraction of O. vulgare and C. obtuas showed strong antimicrobial activity on Salmonella typhimuriium. In MIC test, C. obtuas was shown the best natural material for the preparation of natural antimicrobial agent by supercritical fluid extraction. In conclusion, these results suggest that supercritical fluid extraction technique was effective to obtain functional ingredient with higher antimicrobial activity in the development of new antimicrobial reagent from natural materials.

Functional Finishing of Cotton Fabrics by Treatment with Chitosan (키토산 처리에 의한 면직물의 기능화가공)

  • Cotton fabric was treated with chitosan by pad-dry(-cure) method to impart antimicrobial properties. Four chitosans of different degree of deacetylation (DAC: 65~95%) with similar molecular weight(MW: ca. 50, 000) and one chitosan oligomer(MW 1, 800, DAC 86%) were used. In order to improve the durability to laundering of antimicrobial activity for the fabrics treated with chitosan oligomer, crosslinker or binder was included in the finishing formulation. Antimicrobial activity against Staphylococcus aureus and Proteus vulgaris was evaluated by the Shake Flask Method. The treated fabrics were laundered up to 20 times according to AATCC Test Method 60-1986 or JIS 0217-104 and antimicrobial activity of the laundered fabrics was evaluated. The antimicrobial activity was increased with the increase of concentration and degree of deacetylation of chitosan. And the cured fabrics showed better durability to laundering than the not-cured fabrics according to AATCC Test Method 60-1986. Crosslinker and binder decreased antimicrobial according of the fabrics treated with chitosan oligomer and were not effective to improve the durability to laundering according to JIS 0217-104. Isolation and Identification of Antimicrobial Compound from Dandelion(Taraxacum platycarpum D.) (민들레로부터 항균성 화합물의 분리 및 동정)

  • Antimicrobial activity of dandelion(Taraxacum platycarpum D.) was investigated. Methanol extract of dried dandelion was fractionated to hexane, chloroform, ethylacetate, butanol and aqueous fraction. Ethylacetate fraction among these fractions showed the highest inhibitory effect on the microorganisms such as B. subtilis, L. monocytogenes, S. aureus, E. coli and V. parahaemolyticus at $500\mu\textrm{g}/disc$. Ethylacetate fraction was further fractionated into 13 fractions by silica gel column and thin layer chromatography(TLC). The results showed that ethylacetate fractions No. 4, 5 and 6 had high antimicrobial activity. These were mixed again, re separated and five fractions were obtained. Among them, No. 2 fraction had the highest inhibitory effect on the microorganisms, which was then separated into six fractions. In the 3rd fractionation, No. 3 fraction was identified as benzoic acid by HPLC, $^{1}H-NMR$ and GC MS.

Screening of the Antibacterial Effects by Ethanol Extracts from Natural Plant in Jeju against Propionibacterium acnes (Propionibacterium acnes에 대한 제주 자생식물 에탄올 추출물의 항균효과 검색)

  • Propionibacterium acnes, a major pathogens bacterium of acne, is one of the microflora associated with the inflammation. In this study, the antibacterial effects of 70% ethanol extracts from the 61 natural plants in Jeju against P. acnes were investigated using the disc diffusion method. The 45 natural plant extracts showed antibacterial effects and the 16 natural plants extracts didn't show antibacterial effects. The Phyllanthus urinaria L. (stem and leaves) extract showed thr highest antimicrobial activity with $18.96{\pm}0.69mm$ clear zone of the growth of P. acnes, followed by Angelica gigas Nakai (roots), Gynostemma pentaphyllum (Thunb.) Makino, Angelica gigas Nakai (stem and leaves), Isodon inflexus (Thunb.) Kudo, camellia sinensis L. (flowers), Rosmarinus officinalis, etc. These results suggest that the natural plant in Jeju can be used as functional cosmetic biomaterials or antimicrobial agents against P. acnes.

Characteristics of Resin on Antimicrobial Properties of Calibration Devices. (교정장치의 항균에 따른 레진상의 특성)

  • Polymethyl methacrylate (PMMA) is concerned with promoting oral infection due to its low antibacterial activity. To overcome this, the nanoparticles of Ag-MSN, nGO, and CNP were mixed with MMA liquid in a weight ratio of 0, 0.25, 0.5, 1.0, 2.0% compared to resin powder using Orthocryl from Dentarum, a calibration resin, and then instructed by the manufacturer. Accordingly, a specimen for calibration was prepared by mixing PMMA: MMA (1.2: 1) ratio, and physical properties of the calibration resin, antifungal experiments, and statistical analysis were performed. As a result of antibacterial experiments, the antibacterial properties of Ag-MSN increased. In nGO, the antibacterial adhesive effect increased hydrophilicity, not a change in surface roughness. The higher the CNP concentration, the higher the antibacterial activity. This suggests its potential usefulness as an antibacterial dental material for orthodontic devices and temporary restorations.

Antimicrobial Activity and Characteristics of Protamine Extracted from Tuna Spermary (참치백자로부터 추출한 Protamine 의 특성 및 항균활성)

  • The purpose of this study was to investigate the antimicrobial effects and characteristics of protamine extracted from spermary of tuna. The result of amino acids analysis showed that the contents of arginine were 46%. It was 10% lower when compared to standard protamine (SP: Asama kasei LTD, Japan). Also, there were significant difference in the contents of proline adn glycine. The average molecular weights of main protein in TP were 13,400 Da, whereas those in SP were 11,300 Da and 2,600 Da. To increase antimicrobial activities of TP, pepsin or trypsin was treated at $37^{\circ}C$. After TP was hydrolyzed with pepsin (pepsin hydrolyzed protamine: PHP) for 4hrs, the average molecular weights of the main protein were 11,300 Da and 3,900 Da, and the antimicrobial activities were significantly increased compared to TP. After TP was hydrolyzed with trypsin (trypsin hydrolyzed protamine: THP) for more than 10 min, the average molecular weights of the main protein were below 2,500 Da. PHP had higher antimicrobial effects on some gram positive bacteria (Staphylococcus aureus, Lactobacillus helveticus, Lactobacillus plantarum) than SP. However, THP had no antimicrobial activities. When TP was hydrolyzed with pepsin (PHP), its antimicrobial activities increased in the same level with those of SP, and this increase might be resulted rather from the changes of molecular weights of the main protein than from the contents of arginine in protamine.
