Development of Natural Antimicrobial Agents for Preserving Greenhouse Fresh Produce (시설원예산물의 선도유지를 위한 항균소재의 개발)

  • To investigate the function and effectiveness of natural antimicrobial agent(NAA), as freshness preserving material, extracted from medicinal herbs, their antimicrobial activities were tested against Bacillus cereus, Corynebacterium xerosis, Peudomonas syringae and Enterobacter aerogenes causing the postharvest decay of greenhouse produce. The ethanol extracts of Polygonum cuspidatum Sieb. et Zucc and Syzyrium aromaticum (L.) Merr. et Perry showed remarkable inhibitory effect on the growth of above-mentioned reference microorganisms when treated with mon than 500ppm. The antimicrobial activities of NAA were very stable in the wide spectrum of temperature (40$^{\circ}C$-180$^{\circ}C$) and pH(4-10).

  • In this study, we propose an antibacterial cosmetic case using UV wavelengths. UVC have excellent antibacterial strength in terms of economics and convenience. The purpose of experiments was to prove it and to select a suitable permeability material. In experiments, UVA, UVC machanics and the competitor product were used to measure and compare the antibacterial strength of antibacterial cosmetic case using UV wavelength. S. aureus, a gram-positive strain, was used subject to the experiment, and the antibacterial strength measured and compared the number of colonies using the plate count technique. As a result, both UVA and UVC showed antibacterial activity, and UVC showed the most powerful antibacterial activity. In Conclusion, the Quartz glass was the most suitable of the permeable materials, but the more thick the examined quartz material became the less antibacterial strength.

  • In order to development of new denture cleaners using natural antimicrobial agents, we have investigated various species of medicinal plants for the antimicrobial activity against Streptococcus matans and Candida albicans. Citrus extract showed significant antifungal activities in a dose-dependent manner against Candida albicans. Candidasis is also a common problem affecting older adults using full denture. We also developed liquid type denture cleaner formulation using citrus extracts.

Functional Finishing of Cotton Fabrics by Treatment with Chitosan (키토산 처리에 의한 면직물의 기능화가공)

  • Cotton fabric was treated with chitosan by pad-dry(-cure) method to impart antimicrobial properties. Four chitosans of different degree of deacetylation (DAC: 65~95%) with similar molecular weight(MW: ca. 50, 000) and one chitosan oligomer(MW 1, 800, DAC 86%) were used. In order to improve the durability to laundering of antimicrobial activity for the fabrics treated with chitosan oligomer, crosslinker or binder was included in the finishing formulation. Antimicrobial activity against Staphylococcus aureus and Proteus vulgaris was evaluated by the Shake Flask Method. The treated fabrics were laundered up to 20 times according to AATCC Test Method 60-1986 or JIS 0217-104 and antimicrobial activity of the laundered fabrics was evaluated. The antimicrobial activity was increased with the increase of concentration and degree of deacetylation of chitosan. And the cured fabrics showed better durability to laundering than the not-cured fabrics according to AATCC Test Method 60-1986. Crosslinker and binder decreased antimicrobial according of the fabrics treated with chitosan oligomer and were not effective to improve the durability to laundering according to JIS 0217-104.   • Antimicrobal effects of Akebia quinata DECAISNE were studied. Their Stems and Fruits were extracted with ethanol and water. The ethanol extracts, boiling ethanol extracts, boiling water extracts were not showed the inhibitory effects. But water extracts of stems and fruits were showed inhibitory effects on the 2species.(Bacillus subtills KCTC 1021, Bacillus cereus KCTC 1012)

  • 참멍게의 체액세포로부터 산추출 후, 조 추출물에서 천연항균소재를 개발하기 위해 먼저 멍게 조 추출액을 직접 Tricine-SDS PAGE를 통하여 주요 펩타이드들의 분자량의 범위를 살펴본 결과 6kDa 이하의 분자량의 펩타이드들이 다량 존재함을 알수 있었다. 펩타이드들의 size별 항균활성을 알아보기 위해 여러 사이즈의(100, 50, 30, 10 kDa)의 한외여과만으로 여과하여 그 여과액들의 specific 활성을 알아본 결과 여과막의 cut-off size에 상관없이 거의 인정한 specific activity를 가짐을 알 수 있었다. 멍게 조 추출액의 여러 미생물에 대한 항균 스펙트럼을 알아보기 위해 E.coli, P. aeruginosa, S. typhi, V. parahaemolyticus, L. monocytogenes, B. sutillus, S. aureus, S. mutans 균주들을 $10^5 CFU/ml$로 부터 4log 감소시키는 농도를 측정한 결과 각각 200, 50, 60, 10, 25, 30, 100, 100ppm 농도였으며, 대표적 상용화 항균 펩타이드인 Nisin과의 항균활성 비교 결과 비슷하거나 월등히 뛰어난 결과를 보여주었다. 또한 추출액의 열안정성을 측정하기 위해 $100^{\circ}C$에서 10분간 가열한 후 원액과의 항균력의 차이를 Radial diffusion assay로 알아본 결과 항균력의 차이가 거의 없음을 알 수 있었다.

Evaluation of the Natural Antimicrobials against Yeasts in Functional Beverages to Control Quality loss (효모에 의한 기능성 음료 변질 제어를 위한 천연항균물질 항균력 평가)

  • We investigated eight active natural antimicrobials for preservation of functional beverages that are usually degraded by yeasts rather than by bacteria due to a high sugar content and a low pH. Five strains of yeasts (S. cerevisiae, Z. bailii, P. membranaefaciens, C. albicans, and P Anomala) were tested with eight natural antimicrobial agents ($\varepsilon$-polylysine, yucca extract, vitamin $B_1$ derivative, scutellaria baicalensis extract, chitooligosaccharid, allyl isothiocyanate, sucrose-fatty acid ester, and oligosaccharide). The lowest minimal inhibitory concentrations (MIC) were 10 ppm for oligosaccharide and sucrose-fatty acid ester against S. cerevisiae and Z. bailii, 10 ppm for allyl isothiocynate against P. membranaefaciens and C. albican, and 10 ppm for allyl isothiocynate and oligosaccharide against P. anomala. No growth were observed for five kinds of yeasts in functional beverages containing sodium benzonate at concentration of 0.015% or higher. The resistance of S. cerevisiae, Z. bailii, and P. Anomala against natural antimicrobial agents was lower than those of P. membranaefaciens and C. albican. Allyl isothiocyanate, oligosaccharide, and sucrose-fatty acid ester showed the highest antimicrobial activities among the eight tested antimicrobials. These results can be applied to develop new natural antimicrobial agents to improve microbial quality of functional beverages.

Purification and Characterization of Natural Antifungal Protein from Astragal Seeds (Astragalus membranaceus L.). (황기 종자의 천연 항진균성 단백질의 분리정제 및 특성검정)

  • Deterioration of food is in general caused by the presence of microorganisms and chemical compounds of food itself. There exists antimicrobial compound in the food, however, addition of food antiseptics, additives, or physico-chemical processing is a common practice. The safety of artificial chemical antiseptics became a serious public concern, therefore, new natural antiseptic compounds are in need to be developed. We have isolated a new natural antifungal protein (KBS-B2) from Astragal seed through ammonium sulfate precipitation and column chromatography using FPLC Mono-S and Superose 12HR. The purified protein inhibited growth of Candida albicans, and spore germination of food spoiling fungi such as Aspergillus ochraceus, Penicillium expensum, P. digitatum and Botrytis cineria. Antifungal effect of the KBS-B2 protein could be directly assayed by bioautography overlaying the fungal spores on the electrophoresed acrylamide gel. The comparison of N-terminal amino acid sequences of the KBS-B2 with known antifungal protein revealed that had 50% homology to thaumatin and zeamatin like proteins.

  • For the development of antibiotics from Korean mushrooms, the biological activities of extracts from 98 species of mushrooms and from 25 mushrooms were tested against 9 different Gram-negative bacteria and 8 fungi, respectively. Fruiting bodies of each mushrooms were extracted with petroleum ether (P), 80% ethanol (E), and distilled water (H) in that order. P, E, or H extracts from 20 mushroom samples exhibited the antibacterial activity against 8 different Gram-negative bacteria containing Klebsiella pneumoniae, selectively. Among the mushroom extracts with antibiotic activity, E extracts of Boletus umbriniporus, Armillariella tabescens, Rhodophyllus sinuatus, and Suillus luteus showed various antibiotic activities against several bacteria. E extracts of Abortiporus biennis, Phellinus gilvus, and Polyporus dispansus are highly active against Salmonella typhi and their minimum inhibitory concentration (MIC) was all $10\;{\mu}g/ml$. E extract of Armillariella tabescens showed the antifungal activity against Trichopyton mentagrophytes, and its MIC was $300\;{\mu}g/ml$.
