Construction and Characterization of Vector Expressing Low Level of Translation Factor eIF5B (단백질합성인자 eIF5B의 저 발현 효모벡터의 제조 및 특성)

  • eIF5B is a translation initiation factor that delivers Met-$tRNA^{Met}$ to AUG start codon and subsequently joins the small and large ribosomes. In order to study the function of eIF5B encoded by FUN12, we constructed FUN12 which lacked 5' end of its sequence. We found that this construct lacking almost all of its promoter in pRS plasmid partially complemented slow growth phenotype of fun12 deletion strain. Interestingly, this construct expressed N-terminally truncated eIF5B and its expression level was about 5% of that of wild type eIF5B. Low amount of the eIF5B expressed additionally in fun12 deletion strain played a direct role as a limiting factor for its growth. This limiting factor eIF5B in those strains also modulates activities of overall translation in vitro.

Bee Venom Suppresses Lipopolysaccharide-stimulated Expression of Cyclooxygenase-2 and Inducible Nitric Oxide Synthase in Mouse BV2 Microglial Cells (봉독약침액이 BV2 세포에서 LPS로 유발된 염증반응에 미치는 영향)

  • 목적 : 본 연구는 봉독 약침액이 BV2 microglial cell에서 LPS로 유발된 염증반응에 대한 억제효과를 관찰하고자 하였다. 방법 : 봉독 약침액의 항염증작용을 관찰하기 위하여 BV2 microglial cell에 봉독약침액을 1시간전에 농도별$(0.1,\;1,\;100\;{\mu}g/ml)$로 전처치한 후 LPS $(5\;{\mu}g/ml)$로 24시간 동안 처리하여 RT-PCR, western blot, $PGE_2$, assay, NO synthesis assay등의 방법으로 관찰하였다. 결과 : LPS 염증유발에 의해서 BV2 microglial cell에서 COX-2 및 NOS 발현이 증가하였고, 이 러한 증가는 prostaglandin E2 및 NO 합성을 증가시켰다. 이에 반하여 봉독약침액으로 전처치한 군에서는 COX-2 및 NOS 발현을 억제시켜 결과적으로 prostaglandin 합성 및 NO 합성을 억제시킴을 확인할 수 있었다. 또한 LPS 염증유발에 의해서 활성화된 NF-kB의 발현을 억제 시켰다. 결론 : 봉독약침 액은 LPS 염증유발에 의해서 증가된 prostaglandin E2 및 NO 합성을 억제시킴으로써 여러 가지 염종질환의 치료에 유효한 효과가 있을 것으로 사려 된다.

A Study on the Analysis of Jeju Island Precipitation Patterns using the Convolution Neural Network (합성곱신경망을 이용한 제주도 강수패턴 분석 연구)

  • Since Jeju is the absolute weight of agriculture and tourism, the analysis of precipitation is more important than other regions. Currently, some numerical models are used for analysis of precipitation of Jeju Island using observation data from meteorological satellites. However, since precipitation changes are more diverse than other regions, it is difficult to obtain satisfactory results using the existing numerical models. In this paper, we propose a Jeju precipitation pattern analysis method using the texture analysis method based on Convolution Neural Network (CNN). The proposed method converts the water vapor image and the temperature information of the area of ​​Jeju Island from the weather satellite into texture images. Then converted images are fed into the CNN to analyse the precipitation patterns of Jeju Island. We implement the proposed method and show the effectiveness of the proposed method through experiments.

Deformation History of Precambrian Metamorphic Rocks in the Yeongyang-Uljin Area, Korea (영양-울진 지역 선캠브리아기 변성암류의 변형작용사)

  • Precambrian metamorphic rocks of Yeongyang-Uljin area, which is located in the eastern part of Sobaegsan Massif, Korea, are composed of Pyeonghae, Giseong, Wonnam Formations and Hada leuco granite gneisses. These show a zonal distribution of WNW-ESE trend, and are intruded by Mesozoic igneous rocks and are unconformably overlain by Mesozoic sedimentary rocks. This study clarifies the deformation history of Precambrian metamorphic rocks after the formation of gneissosity or schistosity on the basis of the geometric and kinematic features and the forming sequence of multi-deformed rock structures, and suggests that the geological structures of this area experienced at least four phases of deformation i.e. ductile shear deformation, one deformation before that, at least two deformations after that. (1) The first phase of deformation formed regional foliations and WNW-trending isoclinal folds with subhorizontal axes and steep axial planes dipping to the north. (2) The second phase of deformation occurred by dextral ductile shear deformation of top-to-the east movement, forming stretching lineations of E-W trend, S-C mylonitic structure foliations, and Z-shaped asymmetric folds. (3) The third phase deformation formed I-W trending open- or kink-type recumbent folds with subhorizontal axes and gently dipping axial planes. (4) The fourth phase deformation took place under compression of NNW-SSE direction, forming ENE-WSW trending symmetric open upright folds and asymmetric conjugate kink folds with subhorizontal axes, and conjugate faults thrusting to the both NNW and SSE with drag folds related to it. These four phases of deformation are closely connected with the orientation of regional foliation in the Yeongyang-Uljin area. 1st deformation produced regional foliation striking WNW and steeply dipping to the north, 2nd deformation locally change the strike of regional foliation into N-S direction, and 3rd and 4th deformations locally change dip-angle and dip-direction of regional foliation.

Human Neutrophil Cathepsin G: In Vivo Synthesis of Anti-HNCG Antibody, Inhibition of the Activity of HNCGs and Mechanism of the Inhibitions (사람 호중구 Cathepsin G: Anti-HNCG Ab의 In Vivo 합성, HNCG의 활성도 억제와 그 기전에 관한 연구)

  • Human neutrophil cathepsin-G, which has been known as one of the active enzymes causing inflammatory diseases, was purified by two steps procedure involving one size exclusion (Ultorogel AcA54) and one ion exchange (CM-Sephadex) chromatography. Purified HNCGs were cross-reacted with Anti-HNCathepsin-G antibodies which were radised in rabbits and purified by cathepsin-G labeled Sepharose 4B affinity chromatography. HNCGs were effectively inhibited by NSAIDs including phenylbutazone, sulindac, oxyphenbutazone, salicylic acid and salicyluric acid. $IC_{50}_s$ of these drugs for inhibition of Cathepsin G were 0.3-0.8 mM. Other NSAIDs including aspirin showed little or no inhibition effect on the activity of Cathepsin G. These results strongly indicated that NSAIDs which showed inhibition effect on the activity of HNCGs possibly be at least a part of mechanism of action which might be related to direct inhibition of cathepsin G at the tissue destruction sites beside of their known mechanism of action as an anticyclo-oxygenase in treatment of inflammatory diseases. Lipid soluble component of Korean Red Ginseng which was known as an anti-inflammatory agent inhibited HNCGs strongly, but no other fractions did inhibited HNCGs. Antibiotics including novobiosin and rifamycin showed some inhibition effect on HNCGs, i. e.., $IC_{50}$ of these drugs were 2.6 mM and 1.5 mM respectively, and other antibiotics including penicillin G showed no or negligible inhibition effect on the activity of HNCGs. However. tetracyclines inhibited HNCGs very effectively at the concentration of therapeutic range. The inhibition effect of the activity of HNCGs by tetracycline are not related to the N-dimethyl radical on the 4 position of the tetracycline molecule. Furthermore, N-dedimethylated tetracyclines may have beneficial effect for long term treatment of chronic inflammatory diseases without developing any drug resistance to microorganisms.

Characterization of Single Stranded DNA-Dependent ATPase Activities of Deinococcus radiodurans RecA Protein (Deinococcus radiodurans RecA 단백질의 외가닥 DNA-의존성 ATPase 활성 분석)

  • The RecA protein of Deinococcus radiodurans is essential for the extreme radiation resistance of this organism. The central steps involved in recombinational DNA repair require DNA-dependent ATP hydrolysis by recA protein. Key feature of RecA protein-mediated activities is the interactions with ssDNA and dsDNA. The ssDNA is the site where RecA protein filament formation nucleates and where initiation of DNA strand exchange takes place. The effect of sequence heterogeneity of ssDNA was examined in this experiment. The rate of homopolymeric synthetic ssDNA-dependent ATP hydrolysis was constant or nearly so over a broader range of pHs. For poly(dT)-dependent ATP or dATP hydrolysis, rates were generally faster, with a broader optimum between pH 7.0 and 8.0. Activities of RecA protein were affected by the ionic environment. The ATPase activity was shown to have different sensitivity to anionic species. The presence of glutamate seemed to slimulate the hydrolytic activity. Dr RecA protein was shown to require $Mg^{2+}$ ion greater than 2 mM for binding to etheno ssDNA and the binding stoichiometry of 3 nucleotide for RecA protein monomer.

Growth Inhibitory Activity of Sulfur Compounds of Garlic against Pathogenic Microorganisms (마늘 황화합물의 병원성미생물 번식억제작용)

  • Efforts have been made to explore the possibility of using garlic as an antimicrobial therapeutic agent since garlic extract and its individual sulfur compounds show antimicrobial activities against all kinds of microorganisms including bacteria, molds, yeasts and protozoa. Staphylococcus aureus has been the most studied bacteria along with many other Gram positive and negative pathogenic bacteria, including species of the genera Clostridium, Mycobacterium, Escherichia, Klebsiella, Bacillus, Salmonella and Shigella. Candida albicans has been the most studied among the eukaryotic microorganisms. A pathogenic protozoa, Giardia intestinalis, was also tested. All the microorganisms tested was inhibited by garlic extract or its sulfur components. Garlic has been known to be growth inhibitory only when fresh garlic is crushed, since allicin-generating reaction is enzyme-catalyzed. Allicin is known to be growth inhibitory through a non-specific reaction with sulfhydryl groups of enzyme proteins that are crucial to the metabolism of microorganisms. Another plausible hypothesis is that allicin inhibits specific enzymes in certain biological processes, e.g. acetyl CoA synthetase in fatty acid synthesis in microorganisms. Allicin transforms into other compounds like ajoene and various sulfides which are also inhibitory to microorganisms, but not as potent as their mother compound. It is reported recently that garlic heated at cooking temperatures is growth inhibitory especially against yeasts, and that the growth inhibitory compound is allyl alcohol thermally generated from alliin in garlic.

  • A wide variety of unsaturated vinyl derivatives can be induced to undergo free-radical chain polymerization. The capability to carry out a thermodynamically feasible polymerization relies on its kinetic feasibility on whether the proceeds at a reasonable rate under a given set of reaction conditions. Initiator or promoter is often required to achieve the kinetic feasibility. Only a few unsaturated monomers including methyl methacrylate(MMA) are known to absorb light between 250 and 500 nm which is the most convenient wavelength range. Also, the polysilanes with unusual optical and electronic properties have been used as ceramic precursors, deep UV photoresists, photoconductors. The hydrosilation has been used to make many interesting types of silicon containing polymers such as copolymer, dendrimers. Bulk polymerization of monomers with different molar radio of hydrosilanes(9:1 through 1:9) were performed. A quartz test tube charged with monomer and hydrosilane was degassed and irradiated with 250 nm UV for 6 hours. The polymer was taken in toluene, precipitated in hexane, filtered off, and dried. It was found that the initiators appeared to competitively and concurrently function as both chain initiation and transfer agents in the polymerization of vinyl monomers.

Comparison of the Properties of Poly(lactic acid) Nanocomposites with Various Fillers: Organoclay, Functionalized Graphene, or Organoclay/Functionalized Graphene Complex (유기화 점토, 작용기화 그래핀 및 유기화 점토/작용기화 그래핀 복합체 등의 필러를 사용한 Poly(lactic acid) 나노 복합체의 물성 비교)

  • Poly(lactic acid)(PLA) nanocomposites containing various nanofillers were synthesized using the solution intercalation method. Organically modified bentonite clay (NSE), octadecylamine-graphene oxide (ODA-GO), and an NSE/ODA-GO complex were utilized as nanofillers in the fabrication of PLA hybrid films. PLA hybrid films with varying nanofiller contents in the range of 0-10 wt% were examined and compared in terms of their thermomechanical properties, morphologies, and oxygen permeabilities. Transmission electron microscopy (TEM) confirmed that most of the NSE and ODA-GO nanofillers were dispersed homogeneously throughout the PLA matrix on the nanoscale, although some agglomerate NSE/ODA-GO complex particles were also formed. Among the three nanofillers for PLA hybrid films, the NSE/ODA-GO complex showed the best improvement in film thermal stability. In contrast, NSE and ODA-GO exhibited the best improvement in tensile mechanical properties and oxygen barrier properties of the PLA hybrid films, respectively.

Interactions and Ionic Conductivities of Poly(epichlorohydrin) Graft Copolymer Electrolyte Membranes (Poly(epichlorohydrin) 가지형 공중합체 전해질막의 상호작용 및 이온 전도도)

  • Amphiphilic graft copolymers based on poly(epichlorohydrine) (PECH) were synthesized using atom transfer radical polymerization (ATRP). Successful graft polymerization of poly(methyl methacrylate)(PMMA) and poly(butyl methacrylate) (PBMA) from PECH was confirmed by nuclear magnetic resonance ($^1H$ NMR) and FT-IR spectroscopy. Upon the introduction of KI or LiI to the graft copolymers, the ether stretching bands were shifted to a lower wavenumber due to coordinative interactions. Ionic conductivities of PECH-g-PBMA complexes were always higher than those of PECH-g-PMMA complexes, resulting from higher mobility of rubbery PBMA chains. The maximum ionic conductivity of $2.7{\times}10^{-5}\;S/cm$ was obtained at 10 wt% of KI for PECH-g-PBMA electrolytes.
