Study on the Antioxidative and Physiological Activities of Saururus chinensis Extract (삼백초(Saururus chinensis) 추출물의 항산화능 및 생리활성 연구)

  • Saururus chinensis has long been widely used in oriental folk medicines to treat diseases. In the current study, organic solvent fractions obtained from the main methanolic extract of S chinensis were evaluated for their antioxidative and related physiological activities. The antioxidant activity of the fractions was measured using DPPH free radical scavenging activity, increased in a dose-dependent manner, and the $ED_{50}$ of the ethyl acetate fractions exhibited a value of 12.84 ${\mu}g/ml$ higher than 27.22 ${\mu}g/ml$ compared to the BHT. Also, the cell viability of S. chinensis on $H_2O_2$-induced HDF cell death ($IC_{50}$) showed the highest cell viability of 89.39% in 50 ${\mu}g/ml$ of ethyl acetate fraction and 67.98% of visible cell survival rate in n-butanolic fraction. Meanwhile, all fractions of the S. chinensis extract led to a slight down regulation of the mRNA expression of fibulin-5, which is related to skin elasticity, and the ethyl acetate fraction having high antioxidant activity showed a markedly inhibitory effect on chick embryonic angiogenesis using the CAM assay. These results suggest that the ethyl acetate fraction of S. chinensis extract could be a good material in therapeutic application for antioxidant and related anti-angiogenesis activities.

  • The innate immune system is important for the first line of host defence against infectious agents, which have penetrated the mechanical barriers. Mannose-binding lectin (MBL or mannan-binding protein, MBP) is a serum protein that is synthesized in the liver as a part of the acute phase response. MBL binds to carbohydrate structures presented by a wide range of pathogenic bacteria, viruses, fungi, and parasites. MBL is synthesized as a monomer that has a carboxy-terminal carbohydrate recognition domain, a neck region and a collagen region. Low MBL level was reported to be the most frequent immuno-deficiency syndrome. Although extensive studies have yielded detailed information on the structure of MBL, functions of the MBL complex are not fully understood yet. We, here, present cloning process of MBL cDNA from the rat liver and production of truncated recombinant MBL protein using a bacterial expression system in order to produce anti-MBL polyclonal antibody. Anti-MBL polyclonal antibody was raised in a New Zealand rabbit and its affinity was tested against recombinant protein using western blot technique. MBL cDNA, recombinant protein and anti-MBL antibody could be used as great arsenals to dissect cellular biochemistry of MBL.

Studies on Genetic Toxicity of Epoxidized Soy Bean Oil (에폭시화 대두유의 유전독성 연구)

  • EpoxidiBed soy bean oil (ESBO) is a plasticizer of PVC which is being widely used as a gaskets for the lid of glass jars including baby food. Using reverse mutation assay, chromosome aberration test and micronucleus test, ESBO were evaluated the mutagenicity. In the reverse mutation test, ESBO did not induced mutagenicity in Salmonella typhimurium TA98, TA100, TA1535, TA1537, TA102 with and without metabolic activation. In the chromosome aberration test using CHL cells, the results showed no increased structural and numerical aberrations in the concentration of sample producing cytotoxicity with and without metabolic activation. The in vivo induction of micronuclei was measured in polychromatic erythrocytes of bone marrow of young (3weeks old) and adult (6 weeks old) ddY mice of both sex. At 24 hours after treatment with ESBO 20, 10, 5, 2.5 g/B.W. kg/corn oil 10 ml by oral route animals were sacrificed and bone marrow cells were prepared for smear slides. The results showed no increased micronucleated polychromatic erythrocytes regardless of sex and age. It was concluded that water soluble ESBO did not show certain genotoxicity within our studies conducted.

Synthesis of Single-Crystalline InSb Nanowires Using CVD Method and Study of Growth Mechanism in Open and Close System (CVD 방법을 이용한 단결정 InSb 나노와이어의 성장과 Open/Close 시스템에서의 반응 메커니즘 연구)

  • Single-crystalline InSb nanowire was synthesized on $SiO_2$ wafer via vapor-liquid-solid (VLS) mechanism using chemical vapor deposition method. According to the source container system (open or close) which contain InSb powder and $SiO_2$ wafer, the single-crystalline InSb nanowires have different growth mechanisms. Structural characterization of the InSb nanowires was examined by scanning electron microscope (SEM). Composition of the nanowires was investigated using x-ray diffraction (XRD) and energy dispersive x-ray spectroscopy (EDS). This study demonstrates that length and diameter of the InSb nanowires are long and thick using open-boat system by VLS and additional vapor-solid (VS) mechanisms, because open-boat system can carry a large amount of vapor-phase InSb precursor than close-boat system.

Enhancement of CO2 permeance by incorporating CaCO3 in Mixed Matrix Membranes (CaCO3을 이용한 혼합매질분리막의 이산화탄소 투과도 향상)

  • With vigorous development of petroleum and chemical industry, emission of carbon dioxide has attracted tremendous attention globally due to global warming problem and abnormal climate change. To address these problems, in this study, a PEGBEM-g-POEM graft copolymer with high $CO_2$ affinity was synthesized and $CaCO_3$ was incorporated to form mixed matrix membranes (MMMs) for enhancement of $CO_2$ permeance. By varying the addition weight of $CaCO_3$ in MMMs, high separation performance of $CO_2$ over $N_2$ was obtained. At 50 wt% loading of $CaCO_3$, the greatest separation performance was obtained with an enhanced $CO_2$ permeance from 22.5 to 28.16 GPU and slightly increased $CO_2/N_2$ selectivity from 44.7 to 45.42. It resulted from the increased $CO_2$ solubility of MMMs due to specific interaction between $CaCO_3$ and $CO_2$ molecules. Upon excess loading of $CaCO_3$, MMMs exhibited loss of $CO_2$ separation performance due to the formation of interfacial defects. Based on this result, it is considered that the proper addition of $CaCO_3$ is crucial for improvement of $CO_2$ separation performance.

Estimation of Damage Degree for Mural Paintings in Maitreya Hall of Geumsan-sa Buddhist Temple, Korea (금산사 미륵전벽화의 손상도 평가 연구)

  • Since wall paintings in Maitreya Hall of Geumsan-sa temple had displayed a serious state of damage and deterioration, a detailed examination such as structural analysis of the wall, cause of damage, and the state of deterioration have been thoroughly conducted before the conservation treatment has commenced. The most seriously deteriorated part of the wall paintings was the south wall of the building in particular in its painted and surface layer. The painted layer had formed its own layer of thick, which has been separation from the surface layer. As such problem developed the whole surface layer has been separated from the wall. The problem has been caused by two reasons: 1. the heavy weight of the roof section and it caused cracks and damage on the wall; 2. the loss of function of consolidating material and it caused discolouring and the separation of surface layer from the wall. The cause of damage on the painted and surface layers can be assumed in two ways: 1. its surrounding environment such as the change of temperature and humidity level and ultraviolet rays ; 2. the loss of mechanical function of consolidating material, synthetic resin which had been applied in the past conservation treatment. The separation of layers from the wall and cracks was caused by the mistake in choosing an applicable consolidating material and dismantling technique which had ignored a different characteristic of the wall painting of Korean buddhist temples.

Automatic Anticipation Generation for 3D Facial Animation (3차원 얼굴 표정 애니메이션을 위한 기대효과의 자동 생성)

  • According to traditional 2D animation techniques, anticipation makes an animation much convincing and expressive. We present an automatic method for inserting anticipation effects to an existing facial animation. Our approach assumes that an anticipatory facial expression can be found within an existing facial animation if it is long enough. Vertices of the face model are classified into a set of components using principal components analysis directly from a given hey-framed and/or motion -captured facial animation data. The vortices in a single component will have similar directions of motion in the animation. For each component, the animation is examined to find an anticipation effect for the given facial expression. One of those anticipation effects is selected as the best anticipation effect, which preserves the topology of the face model. The best anticipation effect is automatically blended with the original facial animation while preserving the continuity and the entire duration of the animation. We show experimental results for given motion-captured and key-framed facial animations. This paper deals with a part of broad subject an application of the principles of traditional 2D animation techniques to 3D animation. We show how to incorporate anticipation into 3D facial animation. Animators can produce 3D facial animation with anticipation simply by selecting the facial expression in the animation.

Development of Short-span Precast Concrete Panels for Railway Bridge (철도교용 단지간 프리캐스트 콘크리트패널의 개발)

  • This paper presents experimental static test results of the precast concrete panels developed for short-span concrete bridge deck form. Different from LB-DECK, concrete rib attached to the bottom surface of concrete panel, and Top-bar is not used at the top surface of concrete panel. Number of concrete ribs and cross-section details of concrete rib are determined from the analytical results of parametric study considering the span length and the thickness of concrete bridge decks. Shear rebars are installed at the top surface of concrete panel for composite action between precast concrete panel and cast-in-place concrete. In order to evaluate the safety and the serviceability of the developed short-span concrete panel subjected to design load, static load test is conducted. Three test panels with span length of 1.6m are fabricated, and during the load test displacements, strains and cracks of test panels are measured and final failure modes are investigated. Serviceability of the test panels is evaluated based on the results of displacements, cracking load, and crack width at the design load level. Safety is also evaluated based on the comparison of the ultimate strength and the factored design load of test panels. Based on the test results, it is confirmed the short-span precast concrete panel satisfies the serviceability and safety regulated in design codes. In addition, the range of span length of concrete bridge decks for the short-span concrete panel is discussed.

Ability for Biosynthesis of C27 Brassinosteroids by an Enzyme Pool Prepared from Cultured Cells of Phaseolus vulgaris (강낭콩 현탁배양세포에서 추출된 Enzyme Pool의 C27 Brassinosteroids 합성 능력)

  • A cell-free enzyme solution prepared from suspension cultured cells of Phaseolus vulgaris successfully mediated conversions of cholesterol $\to$ cholestanol and 6-deoxo-28-norteasterone $\leftrightarrow$ 6-deoxo-28-nor-3-dehydroteasterone $\leftrightarrow$ 6-deoxo-28-nortyphasterol $\to$ 6-deoxo-28-norcastasterone $\to$ 28-norcastasterone. Al-though conversion of cholestanol to 6-deoxo-28-norteasterone intermediated by 6-deoxo-28-norcathasterone was not demonstrated, this strongly suggests that a complete set of biosynthetic enzymes catalyzing reactions from cholesterol to 28-norcastasterone via 6-deoxo-28-nor type brassinosteroids is endogenously present in the cells, which demonstrates that a $C_{27}$ brassinosteroids biosynthetic pathway, namely the late C-6 oxidation for $C_{27}$ brassinosteroids, is operative in the cells. Additionally, the enzyme solution mediated conversion of 28-norcastasterone to castasterone in the presence of S-adenosyl-methionine and NADPH, providing that the $C_{27}$ brassinosteroids biosynthesis is an important route to generate castasterone in the cells. Together with our previous finding that castasterone can be biosynthesized by the same biosynthetic pathway in tomato, this study demonstrates that the $C_{27}$ brassinosteroids biosynthesis is a common alternative process to maintain endo-genous level of castasterone, an active $C_{28}$ brassinosteroid, in plants.

Differential Activities of FOXL2 and Its Mutants on SF-1-Induced CYP19 Transcriptional Activation (SF-1을 매개한 CYP19의 전사활성에 미치는 FOXL2 야생형과 돌연변이형의 차별적 영향)

  • FOXL2 is a winged-helix/forkhead (FH) domain transcription factor, and mutations in FOXL2 gene are responsible for blepharophimosis-ptosis-epicanthus inversus syndrome (BPES). BPES is an autosomal dominant genetic disease. BPES type I patients exhibit both premature ovarian failure (POF) and eyelid malformation, while only the eyelid defect is observed in BPES type II. FOXL2-null ovaries showed a blockage of granulosa cell differentiation, suggesting that FOXL2 plays an essential role for proper ovarian folliculogenesis. Previously, we screened for FOXL2-interacting proteins and identified steroidogenic factor-1 (SF-1) which is known to be required for gonad development and transactivates steroidogenic enzymes including CYP19. In the present study, we demonstrated that FOXL2 transactivates CYP19 and stimulated the transcriptional activation of CYP19 induced by SF-1. In contrast, FOXL2 mutants found in BPES type I and II exhibited compromised abilities to enhance CYP19 induction mediated by SF-1. Thus, this study provides a functional difference between wild-type FOXL2 and its mutants which may aid to understand pathophysiology of BPES elicited by FOXL2 mutations.
