담수산 rotifer, Brachionus calyciflorus 내구란의 상품화

  • 담수산 rotifer, Brachionus calyciflorus는 담수어류의 종묘생산시 초기 자어의 먹이생물로 많이 이용되고 있다. 특히, 이 종의 암컷은 수컷에 관계없이 암컷이 난을 생산하는 처녀생식과 수컷과의 교미에 의해 암컷이 수정란(내구란)을 형성하는 유성생식으로 나뉘어진다. 한편, 유성생식에 의해 형성된 내구란은 두터운 2차 난막으로 싸여져 있어 외부환경이 부화에 적합하지 않으면 강한 내구성을 지니고 있어 휴면 상태를 계속 유지한다. 이러한 내구란은 종의 유전적 형질을 효율적으로 보관할 수 있을 뿐만 아니라 대량배양하기 위한 종자 및 수산 생태 독성학의 실험재료로 이용할 수 있다. 또한 내구란은 Artemia cyst처럼 쉽게 부화시켜 자어에 직접 먹이로 공급할 수 있는 장점으로 인해 rotifer 대량배양에서 오는 유해 세균과 원생동물에 의한 자치어의 2차 감염을 방지 할 수 있다. 이처럼 rotifer, B. calyciflorus 내구란은 여러 가지 용도로 이용 가능하기 때문에 내구란을 대량생산할 수 있는 조건을 규명하는 것도 중요하다. 그리고 생산된 내구란을 사용 목적에 따라서 간편하게 이용하기 위해서는 내구란을 제품으로 만드는 것도 매우 중요하다. 따라서 본 내용은 담수산 rotifer 내구란을 상품화하기 위한 방법으로 내구란의 대량생산과 보존방법에 대해서 설명하고자 한다.

척추동물의 Isozyme에 관한 비교연구: V. 담수어류의 젖산 수소이탈효소 Isozyme

  • The lactate dehydrogenase isozymes in tissues of eight fresh water fishes were examined by cellulose acetate electrophoresis. The electrophoretic distribution of the isozymes showed clearly species-specific pattern. Various subbands were found more frequently in these fishes than in mammals. The isozyme pattern of muscle seems to tend to be the same with that of brain in these fishes. The fish lactic dehydrogenase suggested in many cases to be consisted of nonrandomly selected hybrids.

The Vertebrae of Some Freshwater Fishes Collected in T$\v{o}$kyusan National Park (덕유산 국립공원 일대에서 확인된 몇 종의 담수어 척추골수에 관하여)

  • The freshwater fish fauna of the Tokyusan National Park was censused during June 1993 to November 1993 at 9 sampling sites to establish baseline data for natural conservation program on the National Park. Seventeen species included 7 Korean endemic species and/or subspecies(Acheilognaths koreunus, Squazidus gracilis majimae, Microphysogobio yaluensis, Phoxinus kumganensis, Cobitis koreensis koreensis, Silurus microdorsalis and Odontobutis platycephala) belonging to 5 families were collected in this region. Sixteen species were found in Kumkang and 8 species were found in Naktongkang, however, common species were Squalidus gracilis majimae, Microphysogobio yaluensis, Puntungia herzi, Moroco oxycephalus, Zucco temminckii, Misgurnus anguillicaudatus and Odontobutis platycephala. Dominant species of this region were Moroco oxycephalus, Zacco temminckii, Odontobutis platycephala. The final aims of this study were not only accumulate baseline data on the osteology of Korean freshwater fishes which were collected at those streams during June 1993-November 1993 with soft X-ray radiography, but also evaluate the suitability of osteological analyses on the environmental damage to fish.

  • 양식산업에 있어서 어류 자어 시기의 초기 먹이생물로 rotifer가 주로 사용되고 있다. 특히, 담수산 rotifer, B. calyciflorus는 배양이 쉽고 자어의 먹이로 적당한 크기를 가지고 있기 때문에 초기 먹이생물로 적합하다. 최근 rotifer 배양수의 산소 가스 공급과 pH조절, 농축 Chlorella의 공급, rotifer 배양시 누적되는 다량의 유기물 찌꺼기 제거를 위한 filter 사용은rotifer 고밀도 배양을 가능케 했고, 현재 국내 일부 종묘 배양장에서 이러한 고밀도 배양시스템을 이용하고 있다.   • The freshwater fish fauna of Chirisan National Park, where dramatic environment changes will be expected due to develop tour and hiking, was censused to establish base line data for natural conservation program Four species belonging to 3 families included in Cyprindae, Amblycitidas and the present fishes were collected in this region. Dominant species was dark chub, therefore chareful organization of the tour development and the preservation of ichthyological fauna are strongly recommended.

  • The sex ratio and gonadal histology were investigated in the Cobitis lutheri Rendahl collected from the Tamjin River of Chollanam-do Province. Korea. The ratio of males declined in proportion to increase in size, while that of females increased in proportion to increase in size. Hermaphrodites were found in the intermediate size fishes. They had degenerating testicular tissue and developing ovarian tissue simultaneously. These phenomena were discussed in relation to skewed sex ratios.

mtDNA Analysis of 5 Species of the genera Moroco and Phoxinus(Pisces, Leuciscinae) (황어아과어류 2속 5종의 mtDNA분석)

  • 한국산 담수어류의 잉어목, 황어아과(Leuciscinae) 어류 2속 5종의 계통적 유연관계를 구명하기 위하여 mtDNA분석을 실시하였다 6 base를 인지하는 10개의 제한효소를 처리하여 얻어진 강tDNA의 크기는 16.5-17.5 Kb였으며 Bcl I, Bgl I, Bgl II, Hin dIII, Pvu II, Xba I 등은 종간 차이가 뚜렷하였다. 각종의 집단간 mtDNA분화정도는 매우 낮았으나(p=1% 미만) M. oxyephalus의 무주집단과 제주집단은 예리적으로 큰 차이를 보였다(p=5.3%) Moroco속의 종간 분화정도를 비교한 결과 M. oxycephafus와 M. lagowsk서 사이가 평균 f=7.2%로 근면관계가 제일 가까웠고 M. keumkang과 M. semotilus는 타종들과 근연관계가 제일 멀었다. Moroco속과 Phoxinus속간의 평균 유전적 분화정도는 f=13.7%로 현저한 차이를 보였다. Brown 등(1979)의 공식을 이용하여 이들 황어아과 2속 5종의 분화시기를 추정한 결과 이들은 후기 선신세(Pliocene)와 흥적세(Pleistocene) 사이에 분화된 것으로 추정되었으며 이 결과는 동위효소 연구에서 얻어진 결파(Yang and Min, 1989)와 잘 일치한다.

The fish Fauna and Migration of the Fishes in the Fish Way of the Nakdong River Mouth Dam (낙동강하구의 어류상과 댐의 어도에서 어류의 이동)

  • The ichthyofauna and passing efficiency of fish ways were surveyed in the lower and upper regions of the river mouth dam in the lower reaches of the Nakdong river from June to December in 20()0. As the dam built in the river mouth, the brackish area in the estuary disappeared completely and the transitional area did not formed. The fishes collected from survey were classified into 49 species, 45 genera belonging to 29 families. Among them, the fresh water fishes composed of 10 species, peripheral freshwater fishes 11 species and sea water fishes 28 species respectively. Because of the ecological difference between the fresh and sea water, dominant species in the Myeong-ji of sea water area was Acanthogobius flavimanus (39.1%) and in the Sa-sang of fresh water area was Achanthorhodeus macropterus (44.4%). By the video camera records, fishes were identified nine species like as Coilia ectenes, Cyprinus carpio, Hemibarbus labeo, Squalidus gracilis majimae, Plecoglosus altivelis, Lateolabrax japonicus, Anguilla japonica, Mugil cephalus and Pholis nebulosa. Based on total species collected, the efficiency of the fish way for the anadromous fish was 10.1%. Therefore, efficiency evaluation of the fish way in the Nakdong River Mouth Dam was low.

Detection of Viral Hemorrhagic Septicemia Virus (VHSV) from marine fish in the South Western Coastal Area and East China Sea (남.서해안과 동중국해 자연산 어류에서 Viral Hemorrhagic Septicemia Virus(VHSV)검출)

  • Viral hemorrhagic septicemia (VHS) is one of the most serious viral disease of farmed rainbow trout and some marine fishes in Europe and North America. It has been reported in various marine fish species of Asian countries and induced cause mass mortality in Japanese flounder (Paralichthys olivaceus) culturing in Korea. The aims of this study were to monitor VHSV in wild marine fishes and to give critical information for controling the disease through prophylactic methods. Prevalence of the viral disease, geological distribution and reservoir of the virus were investigated using wild marine fishes captured in southern coast and east china sea for two years. (Reverse Transcriptase Polymerase Chain Reaction) RT-PCR results showed that VHSV were detected in 17 (10.6%) out of 160 fish. G gene sequences of viral strains isolated in this study were closely related to that of a reference strain, KVHS01-1, belonging to VHSV genotype Ⅰ. The results suggest that some of wild marine fishes are VHSV carriers and may spread the pathogen directly to fish farmed in coastal area.
