Effect of Light Quality During Imbibition and Culture on Growth of Soybean Sprout (광질에 따른 콩나물의 생장)

  • Lateral roots of soybean sprout might reduce the quality. The study was done to measure the effect of light quality treated during 24 hour imbibition or 6 day culture on growth and development of soybean sprouts on the 6th day after culture. With the soybean seeds imbibed in 4 ppm benzyladenopurine (BA) solution for last 6 hours of the imbibition, blue and red lights were treated during the imbibition, but during 6 day culture, blue and red or far-red light treatments were done for 50 minutes or 5 hours a day, respectively, the periods taking for their cotyledons to turn green color, On the 6th day after culture, the soybean sprouts were classified by 4 categories on the base of hypocotyl length;>7cm, 4 to 7cm, <4cm and non-germination, and their lateral roots, hypocotyl diameters and fraction dry weights were measured. Blue and red lights treated during the imbibition completely blocked lateral root formation regardless of the lights treated during the culture, and showed nearly the same rate of hypocotyls of longer than 4cm. The period of each light treatment forced during the culture did not influence the growth of soybean sprouts. far-red light treated for 5 hours everyday, however, had the least rate of seed germination and hypocotyls of longer than 7cm of the light quality treatments. In addition, red and far-red lights almost equally having the commercial soybean sprouts of longer than 4cm hypocotyls move elongated and selenderized than blue light and dark treatment, meaning the growth and morphology of soybean sprouts was affected by light treatments during the culture.

Effects of Aeration Period and Temperature after Imbibition on Growth of Mungbean Sprouts (침종 이후의 Aeration 기간과 온도에 따른 숙주나물의 생장)

  • Lateral roots formed on mungbean sprouts should lower their quality. The study was carried out to clarify the effects of aeration periods (AP; 1, 2, 3, and 4 hours) and temperature (AT; 20, 30, and $40^{\circ}C$) after 5 hour seed imbibition into 50 ppm benzyladenopurine (BA) solution on growth and morphological characters of mungbean (cv. Keumseongnogdu, Owoolnogdu and Zhong Lu 1) sprouts. On the 6th day, the mungbean sprouts were sorted by 4 categories on the base of hypocotyl lengths; > 7cm, 4 to 7cm, < 4cm, and non-germination to calculate their composition rates, number of lateral roots, lengths of hypocotyl and root, diameters at middle and upper parts of hypocotyl, fraction fresh and dry weights were measured. AP more affected growth of the cultivars than AT showing little effect on them. In the composition rate of the above 4 categories, cv. Zhong Lu 1 had the highest rate in longer than 7cm but nearly the same rate in AP treatments. Rates of longer than 4cm hypocotyls in cv. Keumseongnogdu, Owoolnogdu were increased with longer AP but their rates of shorter than 4cm showed reverse response to the former. Formation rate and number of lateral roots per sprout were decreased with longer AP, showing more severe decrement when delayed 3 to 4 hour AP. Upper part of hypocotyls and roots were more thickened and shortened in longer AP, respectively. Total fresh weights had no significant difference between AP treatments while hypocotyl fresh weights were increased with longer AP. It was concluded that in mungbean sprout culture aeration from BA treatment to the first watering permitted at least 4 hours.

Effects of Watering Method and BA Concentration on Growth and Morphological Characteristics of Mungbean Sprouts (관수방법과 BA 처리농도가 숙주나물의 생장과 형태에 미치는 영향)

  • Utilization of synthetic benzyladenine (BA) for bean sprout production should be reduced or minimized because of elevating production cost and not ascertaining action mechanism to human body. The study was done to compare the effects of BA concentrations under different watering methods (overspraying or underwatering) on growth and morphological characteristics of mungbean sprouts. Seeds of cv. Zhong-Lu 1 were soaked for 5 hours in the solutions with different BA concentrations (0, 10, 20, 30, 40, 50 ppm) before 4 hour aeration, and then were cultured for 6 days by both watering systems. Their morphological characters, fresh and dry weights were measured. Regardless of watering methods, lateral roots were sharply dropped over 30 ppm BA concentrations, and hypocotyl, root and total lengths were shortened with increased BA concentrations although ratios of hypocotyl to root lengths and hypocotyl diameters were enlarged with their increment. Total fresh weights were increased up to 20 ppm in overspraying method but up to 30 ppm in underwatering method mainly due to increment of hypocotyl fresh weights. The sprouts were faster grown in overspraying method than in underwatering method because the former showed longer lengths of hypocotyl and root, and total fresh weights.

Plant Regeneration from Cotyledon and Hypocotyl Tissues of Chinese Cabbage (배추의 자엽과 배축 절편체로부터의 식물체 재분화)

  • The study was carried out to develop a simple and efficient system to regenerate plants from cotyledon and hypocotyl tissues of Chinese cabbage (Brassica campestris L. ssp. pekinensis cv Seoul). Among the various combinations of naphthalene acetic acid (NAA) and 6-benzyladenine (BA) tested, the best shoot induction medium for cotyledon, with 2.67 shoots per explants, contained $2.0mg{\cdot}L^{-1}$ NAA, $1.0mg{\cdot}L^{-1}$ BA and $16.7mg{\cdot}L^{-1}$ $AgNO_3$. The shoot induction medium with $1.0mg{\cdot}L^{-1}$ NAA, $5.0mg{\cdot}L^{-1}$ BA and $16.7mg{\cdot}L^{-1}$ $AgNO_3$, was best for shoot induction from hypocotyl explants, with 1.87 shoots per explants. After shoot induction, regenerated shoots were excised and rooted on rooting medium. Rooted plantlets were then hardened in the high humidity growth chamber and transplanted to pots, and then grown in the greenhouse. Regenerated plants appeared phenotypically normal and there were no changes in chromosome number.

Growth and Shape of Soybean Sprouts as Affected by Culture Method and Their Pressing (관수방법별 압착정도에 따른 콩나물의 생장과 형태 변화)

  • Shape of soybean sprouts affects their marketability. This study was carried out to understand the effects of pressure in both overspraying and underwatering cultivation methods on growth and morphological characteristics of soybean sprouts. Pressing treatments were done by continuous pressing by laying a plastic culture box over another with growing the sprouts, their alteration every day, or no pressing in overspraying method, and by continuous pressing, pressing after the 4th day cultivation with 30 kg steel case, or no pressing in underwatering method. The soybean (cv. Junjery) seeds were soaked in 2 ppm BA solution for 5 hours and then aerated for 3 hours immediately before 6 day culture. Lateral roots, hypocotyl and root lengths, hypocotyl diameters, fresh and dry weights were measured. Lateral roots per sprout were decreased with stronger pressing regardless of watering methods. In overspraying method, continuous pressing treatment resulted in shortest hypocotyl, but no and ones did the longest root. In underwatering method, however, no pressing one showed the longest hypocotyl but continuous pressing did the shortest hypocotyl. Regardless of watering methods, middle part of hypocotyl was more thickened with stronger pressing although hook diameters of the 3 treatments were not affected. In overspraying method. total fresh weight per sprout was the least in no pressing but the greatest in continuous pressing. In underwatering method, however, there was no significant difference between the treatments.

Effect of Electrolyzed Acidic Water on the Growth of Soybean Sprout. (산성 전해수가 콩나물의 생육에 미치는 영향)

  • To investigate the effect of the electrolyzed acidic water for soybean sprouts growth, the responses of characteristics of soybean sprouts were evaluated. Soybean sprouts grown by the electrolyzed acidic water showed shorter length in total body, root, and hypocotyl, etc. but they were evaluated to be increased in hypocotyl diameter and weight per sprout. Total length of soybean sprouts grown for 5 days by electrolyzed acidic water were much shorter than those by tap water. Soybean sprouts grown by tap water showed rapid growth in length even after 5 days but no more growth in length for those grown by electrolyzed acidic water. The growth of hypocotyl showed the same tendency as total length. No difference in root length among the soybean sprouts grown for 4 ~ 11 days by electrolyzed acidic water while those grown by tap water showed continuous rapid growth in length. The diameter of hypocotyl was thicker in those grown by electrolyzed acidic water than those grown by tap water and increased up 5 days. The weight of cotyledon grown by electrolyzed acidic water showed the proportional increase to the growing days but those grown by tap water showed no increase in hypocotyl weight up to 7 days, but a little bit increase after 11 days with the growth of new buds. The fresh weight per sprout was higher in those grown by electrolyzed acidic water until 7 days than tap water but it was the same weight in 11 days cultivation. The electrolyzed acidic water effected on shortening of hypocotyl and root length, thickening of hypocotyl diameter, and enlarging of cotyledon during soybean sprout cultivation.

Lateral Root Formation and Growth of Soybean Sporouts Treated with Various Solutions (다양한 물질처리에 의한 콩나물의 세근형성 및 생장)

  • Lateral roots of soybean sprouts should deeply affect their quality and marketability. The study was done to compare the effects of ocher, chitosan, corn kennel, tea (Thea sinensis L.) and hard rubber tree (Eucommia ulmoides Oliver) leaf extracts on lateral root formation, growth and morphological characters of the sprouts. Seeds of three cultivars, Pungsannamulkong, Sowonkong and Junjery, were imbibed for 5 hours into their 10% solutions and cultured for 6 days. The cultured sprouts were classified into 4 categories to calculate their composition rates on the base of hypocotyl lengths;>7 cm (A), 4 to 7 cm (B), < 4 cm (C) and not germinated (D), and their morphological characters, fresh and dry weights were measured. Composition rate of A was the lowest in Junjery of the three cultivars, while that of C showed reverse result compared to A. This results was the most distinct in hard rubber tree leaf extracts (HRTLE) of the five treatments. In HRTLE treatment, lateral root formation rate were formed in almost of Sowonkong although reduced in order of Pungsannamulkong and Junjery. However, there was no significant difference between the other treatments. Lateral roots per sprout were the lowest in HRTLE treatment of the 5 treatments. In all treatments except the chitosan treatment, the roots were most formed in Sowonkong but least in Junjery. Sprout length adding hypocotyl and root was the shortest in Junjery compared to the other two cultivars. and was the longest in tea leaf extract treatment but the shortest in HRTLE treatment. The result in total fresh weight of sprouts was similar to that of the sprout length.

Establishment of Technology for Preventing the Soybean Sprout Colletotrichum gloeosporioides Rot (열처리에 의한 콩나물 탄저병의 방제)

  • Anthracnose fungus was most pathogenic on soybean sprout, of the fungi and bacteria isolated from rotten sprout on market. Bacterial strains associated were not virulent. Dry heat (DHT) applied even as high as $65^{\circ}C$ for 30min. was not effective enough to eliminate the artificially inoculated Colletotrichum gloeosporioides propagules from seedllots. Hot water immersion treatment (HWT), at elevated temperature of $55^{\circ}C$ for 20 min, did eliminate the pathogen but reduced seed germinating and retarded sprout growth: Seed germination was practically acceptable when the seedlots were exposed to at $55^{\circ}C$ for 5 min, but about 20% anthracnose propagules survived. Accordingly, we have optimized the HWT scheme for 5 min at $60^{\circ}C$. This scheme was validated, at small to large scale production system, that surely rule out the possible carry over of the bacterial contaminant from seedlots. This result should improve the shelf-life of soybean sprout on the market.

Difference in Growth, Phenolics Content and Antioxidant Activity of Cowpea Sprouts at Different Plant Parts (동부나물의 부위별 생육, 폴리페놀 및 항산화성 차이)

  • An experiment was conducted to determine the content of phenolics and flavonoids, antioxidant activity and antioxidant enzyme status for the extracts from 5 and 7-day old sprouts (DOS) of cowpea (Vigna unguiculata L. Walp). Total phenolics [mg ferulic acid equivalents (FAE) $kg^{-1}$ DW] content was highest in cotyledon extracts (48.8 mg $kg^{-1}$), followed by roots (30.8 mg $kg^{-1}$) and hypocotyl (22.2 mg $kg^{-1}$) extracts (p < 0.05) from 5 DOS. The result of total flavonoid level [mg rutin equivalents $kg^{-1}$ DW] had same tendency to the results of total phenolics, showing lower amount ranges. The antioxidant activity of the methanol extracts from all the plant dose-dependently increased. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was higher in cotyledon extracts (82.5%) than in root (52.6%) or hypocotyl parts (35.0%) from 5 DOS. Among antioxidant enzymes, APX and CAT activities were highest in cotyledon part and POX and SOD activities in root part of 5 and 7 DOS. The results showed that total phenolics content ($r^2$ = 0.1516~0.9911) were more highly correlated with antioxidant activity than total flavonoids level ($r^2$ = 0.0113~0.9442), and that the level and activity of physiological-active substances were different depending on plant part of the sprout.

Use of Recycled PET Bottle Particles as a Medium Component for Cultivation of Tomato Plug Seedlings (재활용 PET병 입자를 배지재료로 이용한 토마토 플러그묘의 재배)

  • This Study was Conducted in 3 glasshouse to examine the Possibility for cultivation of plug seedlings using four growing media containing recycled PET-bottle-particles (PBT) as a medium component. Plug seedlings of 'Seokwang' tomato were grown in media containing $100\%$ PBT, $100\%$ peatmoss, or $50\%$ $PBT+50\%$ peatmoss. A commercial plug medium (Tosilee) was used as the control. All seeds were sown in 128 cell plug trays on July 12, 2004. Seedling growth was measured at 30 days after sowing. Percent seed germination was greater than 95% in all treatments. Plant height, shoot fresh and dry weights, leaf area, and hypocotyl length were the greatest in the control treatment. Root length and weight were the greatest in the $100\%$ PBT treatment, which had a medium pH of 7.3. Leaf area and shoot dry weight were greater in the $50\%$ $PBT+50\%$ peatmoss treatment, than in the $100\%$ PBT treatment, while they were the smallest in the $100\%$ peatmoss treatment. No toxicity symptoms caused by the use or PBT had been observed in any treatment. The results suggest a possibility of recycled PBT, a valuable reusable resource, to be used as a component for medium amendments in horticultural transplant production.
