Big and old trees in the scenic spots with the attributes of remote villas are vulnerable to man-made damages and very sensitive to the external environment such as soil conditions, so the corresponding management plans are required. Thus this study has been conducted to survey the big and old trees in the scenic remote villas and suggest the ideal management plans. The results can be summarized as follows. First, regarding the tree heath above the ground, transformation of tree, death of branches, and death of barks are closely related to tree vigor. Particularly, the areas receiving many visitors require prompt countermeasures against the dried and dead tress above the ground and the areas in which dried and dead tress occurred and also the safety measures for the visitors and facilities. Second, regarding the soil environment, visitor traffic is closely related to the tree vigor. In 15 remote villa gardens, 64% of trees are exposed to heavy traffic and the tree vigor has declined due to an increase of visitor. Thus, there is a need to give positive consideration the installation of the complementary facilities and the plantation of herbal plants in the congested areas to form the ground surface that can tolerate the heavy visitor traffic. Third, remote gardens are in general located adjacent to ponds and mountain streams and thus the trees in the waterfront areas require the prompt countermeasures against the decline of growth due to the excess-moisture in the soil. Further the blockage of the sewage system due to the heavy rains dampens the surrounding soil, which results in lethal damages to the trees. Thus, there is a need of the maintenance of the waterfront areas and sewage system before and after the rainy season. In addition, there is a need to establish medium-long term management polices through the recognition of the importance of the main trees of remote villa gardens in scenic spots and prepare the tree management manual depending on the attributes of the corresponding areas. I strongly suggest making manuals for the systematic management as well as the extensive PR activities and education for the preservation of tress on a long-term basis; and furthermore securing the budget and manpower for the research and development of a systematic management system.
An immunochromatograhy (IC) based infectious bursal disease virus (IBDV) detection kit, which employed two anti-IBDV VP2 monoclonal antibodies, was evaluated for rapid diagnosis of infectious bursal disease virus (IBD). The detection limit of the IC kit for IBDV was $10^{3.1}$ to $10^{3.9}$$EID_{50}$/mL, indicating that the IC kit detected IBDV sensitively as same as double antigen capture ELISA but less than a RT-PCR assay. The IC kit did not detect other viral pathogens such as Newcastle disease virus, infectious bronchitis, avian influenza virus, and infectious larynotracheitis virus. When applied to tissue samples of experimental chickens died 3 or 4 days post infection after very virulent IBDV (strain Kr/D62) infection, the IC kit detected IBDV in all samples of the bursa of Fabricius, spleen, kidney, cecal tonsil and in 87.5%, 37.5% and 0% of liver, thymus and proventriculus samples. In particular, BF tissue samples showed stronger signal bands than other tissues. Positive signal was observed. All except for one thymus sample of samples having negative results by the IC kit showed the same result with DAS-ELISA but RT-PCR assay detected IBDV in some of IC kit negative samples of thymus and proventriculus. When swab samples from the bursa of Fabricius of dead chickens (n=231) on field farms were tested, the sensitivity and specificity of the IC assay relative to RT-PCR was 100% (109/109) and 97.5% (119/122), respectively and kappa value between both assay was 0.97. The kit can provide a useful aid for rapid detection of IBDV in chickens under field circumstances.

The purpose of this study is to assess doses to 18F-FDG, a radioactive drug, during PET examinations, to alleviate anxiety about radiation in patients and carers, to minimize the indiscriminate examination progress caused by medical institution personnel and space clearance problems, and health examination. The dose assessment was measured using a thermo-fluorescent dosimeter (TLD) and an electronic personal dosimeter (EPD) at the location of the cervical (hypothyroid), thorax (heart), and lower abdomen (breeding line) which are the three highest tissue areas of the radiation tissue weighting. In addition, spatial dose rates and radioactivity in urine were measured using GM counters and ion boxes. The results are as follows: First, the personal dosimeter TLD was measured 0.0425±0.0277 mSv in the cervical region, 0.0440±0.0386 mSv in the thorax and 0.0485±0.0436 mSv in the lower abdomen, with little difference in the heart dose depending on radiation sensitivity. The EPD was measured at 0.942±0.141 mSv/h immediately after the cervical position, and 0.192±0.031 mSv/h after 120 minutes. Immediately after the thorax position, 0.516±0.085 mSv/h, 120 minutes later 0.128±0.040 mSv/h. Immediately after the lower abdomen position, 0.468±0.091 mSv/h, and after 120 minutes 0.105±0.021 mSv/h were measured. The spatial dose rate at the GM counter was measured immediately at 0.041±0.005 mSv/h, 120 minutes later at 0.014±0.002 mSv/h. The radioactivity in urine using ion chamber was measured at 0.113±0.24 MBq/cc after 60 minutes and 0.063±0.13 MBq/cc after 120 minutes. As a result, 18F-FDG should be administered, dose re-evaluated two hours after the PET test is completed, and caregivers should be avoided. In addition, it is deemed necessary to provide patients and carers with sufficient explanations and expected values of exposure dose to avoid reckless testing. It is hoped that the data tested in this study will help patients and families relieve anxiety about radiation, and that the radiation workers' exposure management system and institutional improvements will contribute to the development of medical radiation.
Direct Polymerase Chain Reaction (PCR) method that combines biological and enzymatic amplification of PCR targets was developed for the detection of Xanthomonas axonopodis pv. glycines on soybeen seeds without DNA isolation. Primers Xag F1 and Xag R1 were designed to specifically amplify a 401 bp fragment of the glycinecin A gene of X axonopodis pv. glycines. Xag F1 and Xag R1 were used to carry out the PCR analysis with genomic DNA from 45 different bacterial strains including phylogenetically related bacteria with X axonopodis pv. glycines, and other bacterial strains of different genus and species. The PCR assay using this set of primers were able to detect X axonopodis pv. glycines with DNA concentration as low as 200 fg and $1.8{\times}10^3$ cfu/ml. The Xag was detected from the seed samples incubated for 2 hrs with shaking and the intensity of the band was increase with the incubation time of seeds. The Direct PCR assay method without DNA isolation makes detection of X. axonopodis pv. glycines on soybean seeds easier and more sensitive than other conventional methods. The developed seed assay using direct PCR method will be useful for the specific detection of X. axonopodis pv. glycines in soybean seed samples.

Bovine diarrhea is a major economical burden to the bovine industry in Korea. Since multiple infectious agents can be involved in bovine diarrhea, differential diagnosis is essential for effective treatment. Therefore, a panel of two multiplex real-time PCR assays which can simultaneously detect six major bovine enteric pathogens [i.e., bovine viral diarrhea virus (BVDV), bovine coronavirus (BCoV), group A bovine rotavirus (BRV), Salmonella spp., Escherichia coli (E. coli) $K99^+$, and Cryptosporidium parvum] was developed and applied to test 97 fecal samples collected from cattle farms in Korea. In addition, microscopic examination was also preformed on the samples to detect Coccidium oocyst. The estimated sensitivity of the multiplex PCR was 0.1 $TCID_{50}$ for BVDV, BCoV and group A BRV, 5 and 0.5 CFU for E. coli $K99^+$ and Salmonella, respectively, and 50 oocysts for Cryptosporidium. The amplification efficiency of the multiplex PCR ranged between 0.97 and 0.99 for each pathogen. Among 97 samples, 36 samples were positive for at least one of the 6 major pathogens and 6 samples were simultaneously positive for 2 pathogens by the multiplex PCR assay. Coccidium oocysts were also detected in 48 samples, which were all collected from over 1 month old calves. In conclusion, the multiplex real-time PCR panel can be a useful tool for fast and accurate diagnosis of calf diarrhea associated with BVDV, BCoV, group A BRV, E. coli $K99^+$, Salmonella, and/or Cryptosporidium and Coccidium may be an important target which needs to be included in the multiplex PCR panel in the future.
This study was performed to find differences in phototaxis response of the larva, adult emergence, sex ratio and gall formation rate of Thecodiplosis japonensis Uchida et Inouye between two areas, north and south. Larvae from these two areas, Hweng-sung in north and Moo-an in south, both considered to be the sities of severe outbreak of the insect recently, were collected from the floor and transported to the entomology larboratory, Won-Kwang University in Iri, then overwintered. These larvae were used for various experimental purposes on various dates. The following results were obtained. 1. The sizes of larvae reached 2.45 in length, 0.70 in width for north, and 2.50 in length, 0.72 in width for sourth. It seems that the differents were not significant. 2. It is known that Leaving gall of larvae to the ground for the hibernation usually occurs on rainy days. For the larvae dispersion, the phototaxis response of the larvae was hypothesized. To check this, the author manipulated some different illumination intensities adjusting the distance between the glass tube in which 100 larvae were contained and electric bulb. The glass tubes were blackened all but except a small portion. The phototaxis responses between two areas seemed to be different particularly at low light intensities, 45, 145 and 1000 lux, The larvae from north assembled to the clear portion of the glass tube were more than 30 out of 100, however less than 30 in south sample regardless of the time passage. In either cases, the saturation points came after about 8 to 10 hours. If temperature units were used, the phototaxis curve= after 1 hour illumination between two areas provided significant differences. 3. The adult emergence of south area was lagged 10 days later than that of north area. The accumulated effective day-degrees of temperature for adult emergence were $934^{\circ}C$ for north area and $1180^{\circ}C$ for south area. The emerging duration of north area appeared to be in late May through the early of July and the peak emergence occurred in middle June that was approximately 15days earlier than that of south area. The sex ratio of female to male, regardless of area, was approximately 3 : 1. 4. The rates of gall formation of the red pine(Pinus densiflora Siebold et Zuccarini), 6-year-old seedlings, were as low as 9.94% for north area and 8.87% for south area. Through the close observation, the author presumed that the population reduction was greatly affected during the prepupa stage by relative moisture content and predators, such as spiders and ants presented on the ground.

Recently, Korea has been affected by extreme weather events including extended summers and increased temperatures caused by global warming and climate change. Environmental temperature is especially important to the livestock industry because it is closely related to livestock productivity. This study was conducted to investigate the influence of different environmental temperatures on respiration rate, rectal temperature and body-surface temperature in finishing pigs. Pigs ($98.3{\pm}6.6kg$) were housed in individual cages inside an experimental chamber and exposed continuously to one of five environmental treatments ($22^{\circ}C$, $24^{\circ}C$, $26^{\circ}C$, $28^{\circ}C$, $30^{\circ}C$) for 10 days without providing additional rest time. Feed and water intake, respiration rate, rectal temperature and body-surface (head, ear, neck, back, side) temperature were measured two times daily during the experimental period. A significant increase in respiration rate from $26^{\circ}C$ and in body-surface temperature from $24^{\circ}C$ (p<0.05) was observed. At $30^{\circ}C$, the respiration rate had almost doubled and the body-surface temperature increased by about $5^{\circ}C-7^{\circ}C$. Moreover, ear skin temperature was very sensitive to environmental temperature. However, feed intake, water intake and rectal temperature did not change significantly during the experiment.
Vulnerability assessment has been required for establish climate adaptation plan to prevent damage from climate change. In this study we assessed vulnerability with 1 km resolution and determined which sectors have the highest priority in each municipality of Gangwon province based on the result of vulnerability assessment. All sectors of vulnerability assessment are composed of three criteria; sensitivity, exposure and adaptation capacity. And suitable indicators of each sector were selected and spatial data set was prepared using GIS. Priority of vulnerability was classified with the degree of vulnerability in present and variation in vulnerability between present and future. The results of vulnerability assessment were different among municipalities due to the contribution of indicators. Present and future trends in vulnerability showed similar results but high vulnerable area was predicted to expand in the future. In addition increase in temperature led whole area to be more vulnerable generally. The result of prioritizing sectors of vulnerability indicated the most considerable sectors within a municipality. Also, the municipalities which have similar geographic, climatic and social conditions tended to be classified as the same priority class. The method of vulnerability assessment and determining priorities suggested in this study could be used to support decision makers to establish adaptation plan of local area.

In order to minimize the damage on the red sea bream Pagrus major by a harmful dinoflagellate Cochlodinium polykrikoides, we investigated the effect of feeding, starvation and stocking density on the survival rate, growth, growth restoration and physiological response of P. major exposure to C. polykrikoides. The experimental groups were divided into three groups such as F-HD (feeding and high density with $6.4kg/m^3$), S-HD (starvation and high density with $6.4kg/m^3$) and S-LD (starvation and low density with $3.2kg/m^3$) according to stocking density and starvation in marine cage ($11m{\times}11m{\times}5m$). The F-HD was fed throughout the experiment for 9 weeks, whereas S-HD and S-LD were not fed for 5 weeks and then refeeding for 4 weeks. Survival rate was the lowest in F-HD (85.5%) and S-LD was the highest (97.3%). The growth rates of S-HD and S-LD were significantly lower than F-HD during starvation period for 4 weeks, but rapidly recovered after feeding. The nutritional status such as ALB, TP, TCH, TG were similar to tendency of growth data. Ht, Hb, AST, ALT and GLU levels were significantly higher in the F-HD than in the starvation groups at the same time (in 3 week) during starvation period. But starvation groups did not differ during starvation period. As a result, F-HD is more sensitive to stress than S-HD and S-LD. Thus, during C. polykrikoides bloom period, starvation and stocking density control can help survival and growth restoration of the red sea bream.
Apple (Malus pumila) is one of the most economically important fruits in Korea. but virus infection has decreased the sustainable production of apples and caused serious problems such as yield loss and poor fruit quality. Virus or viroid infection including apple chlorotic leaf spot virus (ACLSV), apple stem pitting virus (ASPV), apple stem grooving virus (ASGV), apple mosaic virus (ApMV) and apple scar skin viroid (ASSVd) have been also reported in Korea. In many cases, as apple gets infected with virus and viroid with no specific symptoms, the damage and symptoms caused by the viruses are not detected. In our research, viruses in the rootstock were eliminated for a virus-free apple dwarfing rootstock of M.9 and M.26. The virus elimination methods were apical meristem culture, thermotherapy ($37^{\circ}C$, 6 weeks) and chemotherapy($Ribavirin^{(R)}$). The detection of apple viruses was accomplished by Enzyme-linked Immuno-Sorbent Assay (ELlSA) and reverse transcription-polymerase chain reaction (RT-PCR). RT- PCR method was 10 ~ 30% more sensitive than the ELISA method. The efficiency of virus elimination was enhanced in apical meristem culture method. The acquisition rate of virus-free apple dwarfing rootstocks was 30 ~ 40% higher in apical meristem culture. After the meristem culturing of M.9, the infection ratio of ACLSV, ASPV and ASGV was 45%, 60% and 50%, respectively. In the apple dwarfing rootstock of M.26, the infection ratio of ACLSV, ASPV and ASGV was 40%, 55% and 55%, respectively. Based on this study, the best method for the production of virus-free apple dwarfing rootstocks was the apical meristem culture.
