Sugar and Amino Acid Transport in Yeast I. Glucose Transport during the Sporulation Stage with Reference to the Vegetative Stage. (효모세포의 당과 아미노산의 운반에 관한 연구 -I. 생장시기와 포자형성기의 포도당 운반-)

  • During the sporulation stage in Saccharomyces cerevisiae J170, the incorporation of $D^{14}$ C-glucose into starved cells of sporulation stage as well as the vegetative one is appeared higher at pH 6.0. Glucose transport system, in both the vegetative and sporulation stage, is associated with "energy dependent" as the result of repression by such a respiratory inhibitor as 2, 4-dinitrophenol. The Km value of glucose uptake in vegetative stage and sporulation stage was 2.1 mM and 2.5 mM respectively, indicating that the glucose is considerably reuqired for vegetative growth. Competition and countertranspoer of glucose by frutose and galactose are more distinct in vegetative stage, comparing with sporulation stage. The main sugar components of yeast cells consists of ribose, mannose, and ${\alpha}, \;{\beta}-glucose$. Amounts of mannose is lower in the aporulation stage than that in the vegetative stage.

  • Pleistophora disease is well known as microsporidiosis at eel cultural farms in Korea in recent years. Major objects of this study were to undertake the induction of Pleistophora infection experimentally and to carry out histopathological investigation from December 1984 to June 1986. Experimental infection of Pleistophora spores into young eels was carried out by oral and immersed administration. Both methods induced the same symptom successfully as that in naturally occurring diseased fish. Remarkable whitish lesions developed mainly on the body surface around the abdomen when orally administrated. On the other hand, they were scattered over the whole body when administrated through immersion. Histopathological investigation revealed that some cysts in the muscle were observed 21 days after administration. Spores were developed within the cyst. Each sporont has undergone several nuclear divisions to form a pair of multinucleate cells (Sporoblasts) enclosed within a common coat (Pansporoblast). All stages were surrounded by cyst. The cysts were destroyed and mature spores were scattered in the muscle.

Spore PCR and qPCR Methods for Rapid Detection of Five Colletotrichum Species Responsible for Pepper Anthracnose in Korea (한국의 고추 탄저병을 일으키는 Colletotrichum 5종의 신속한 검출을 위한 포자 PCR 및 qPCR 방법)

  • Pepper anthracnose, caused by Colletotrichum spp., leads to a decrease in the quantity of pepper fruit production. Molecular diagnosis is crucial for rapid identification of pathogens and determination of fungicide resistance. However, the traditional process of isolating the pathogen, extracting genomic DNA, and analyzing the gene sequence is time-consuming, which delays rapid diagnosis. In this study, we introduced a method using conidia of Colletotrichum spp. instead of genomic DNA, eliminating the need for DNA extraction or special processing for diagnosis. To elucidate this method, sensitivity was assessed through polymerase chain reaction (PCR) and quantitative real-time PCR (qPCR) tests using internal transcribed spacer-based primer pairs. Both PCR and qPCR tests showed that detection is feasible with just one conidia, with over 1,000 conidia yielding results comparable to approximately 1 pg of genomic DNA. For amplifying the cytochrome b gene for quinone-outside inhibitor fungicide susceptibility testing, detection from a single conidium is achievable, but a stable PCR product is obtained by increasing the number of cycles to 35. Additionally, the addition of 10% grinding fresh chili pepper paste to V8-Juicea gar medium, which is known for inducing conidia rapidly from the isolates, resulted in 3.2 to 6.0 times more conidia compared to the commonly used potato dextrose agar medium, enhancing the potential for swift testing. Taken together, this study presents a direct utilization of pepper anthracnose conidia through PCR or qPCR, offering a valuable technique for amplifying target genes, such as the minimum conidial amount and barcode genes, for molecular identification of anthracnose disease in pepper through PCR and qPCR analysis.

Characteristics of Spore Density and Colonization Pattern of Arbuscular Mycorrhizal Fungi on the No-tillage Soil under Greenhouse Condition (시설재배지 무경운 토양에서 녹비작물별 Arbuscular Mycorrhizal Fungi(AMF) 감염양상과 포자밀도 특성)

  • This work was studied the effects of spore density and infection of arbuscular mycorrhizal fungi (AMF) for no-tillage organic cultivation of pepper with wintering green manure crops cultivation in greenhouse field. Spore density of arbuscular mycorrhizal fungi (AMF) in green manure crops was 189 spores/30g fresh soils in control including alive spore (82 spores). Spore density of AMF in all green manure crops was totally 196~226 spores/30g fresh soil and alive spore was 84~112 spores/30g fresh soil. Spore density of AMF in soils of Pepper crop was range of 48.0~56.7 spores/30g fresh soils after cultivation of green manure crops. Infection structure of AMF was not significantly difference in soils of green manure crops and Pepper crop after cultivation of green manure crops. Infection rate of AMF in roots of green crops was low level by 2.8% in giant chickweed, 7.4% in rye, 9.3% in hairy vetch. Infection rate of AMF in roots of barley was the highest level by 20.3%. Infection rate of AMF in roots of Pepper crop was range of 5.2~7.2% after cultivation of green manure crops Also, infection rate of AMF in roots of Pepper crop was 8.1% after the harvest of barley. Infection structure of AMF in barley very well consisted of network with internal hyphae, while hairy vetch and rye tended to no network. There was not a significant relationship between spore density in soils and infection rate of AMF in rhizosphere of Pepper.

Effects of Light and Media on Pycnidial Formation of Didymella bryoniae (Auersw.) Rehm (병자곡 및 자양곡형성에 미치는 광선 및 배지의 영향)

  • The Sporulation of Didymella bryomiae were observed under diurnal cycles of light/darkness of near ultraviolet light (NUV) and artificial daylight (ADL) and continous darkness in eight isolates growing on PDA and V-8 juice agar. Light stimulated pycindial and perithecial formation of this fungus on potato dextrose agar and V-8 juice agar. Sprulation was poor in darkness, but some isolates were able to produce pycnidia and perithecia in the absence of light. Perithecial formation was much better under artificial daylight (ADL) on V-8 juice agar than those grown under near ultraviolet light (NUV). In general, cultures grown on V-8 juice agar sporulated better than cultures grown on PDA under three setsof light condition. Most of the pycnidiospores obtained from each isolates of this fungus grown on PDA were non-septate and microtype, but macrotype of non-septate and uniseptate pycnidiospores were produced on V-8 juice agar. Pycnidiospore produced on V-8 juice agar were similar to those produced on the radicle of naturally infected seeds. The appearance of perithecia were quite distinctive from pycnidia. The mature perithecia were darker than pycnidia and whitish spore masses formed on the ostiole of perithecia.

Morphological Features of Coleosporium xanthoxyli and Its Alternate Host in Korea (산초나무 잎녹병균의 중간기주 및 형태학적 특징)

  • A rust fungus has caused a serious early defoliation of Zanthoxylum schinifolium during growing seasons every year at the plantations located at Hadong and Jinju, Kyeongsangnam-Do in Korea. In order to identify the rust fungus and clarify its life cycle in Korea, aeciospores from Pinus thunbergii were artificially inoculated on the leaves of Z. schinifolium. Uredinial stage was successively formed on the leaves of Z. schinifolium. Based on the artificial inoculation test and on the morphological features of the dried specimens collected from P. thunbergii and Z. schinifolium, this rust fungus was identified as Coleosporium xanthoxyli. Morphological features of aecial and uredinial stages of the species were described. The first symptom of the infection was developed from later June to early July. And leaf infection ratio was 17.8%-58.7% during August at Hadong and Jinju regions of Kyeongsangnam-Do in Korea.

  • Nine hundred and eighty fungal strains were isolated from lesions of Trifolium repens L. and various weeds. Among them, F40362, F40496, F40497 strains were selected by the screening of herbicidal activity on Trifolium repens L. and Zoysia japonica. Selected three strains showed selective activity between Trifolium repens L. and Zoysia japonica, and were identified as Penicillium sp.

Comparison of Heat Resistance of Bacillus subtilis, Geobacillus stearothermophilus, and Bacillus atrophaeus spores (Bacillus subtilis, Geobacillus stearothermophilus 및 Bacillus atrophaeus 포자의 열 저항성 비교)

  • We analyzed the heat resistance of non-pathogenic Bacillus atrophaeus, Bacillus subtilis, and Geobacillus stearothermophilus spores which exhibit strong heat resistance and evaluated the possibility of using them to determine direct sterilization when manufacturing retort foods. The D121-values of B. subtilis, G. stearothermophilus, and B. atrophaeus spores were 2.9±0.1 min, 4.3±0.1 min, and 3.7±0.1 min, respectively. The Z-values of B. subtilis, G. stearothermophilus, and B. atrophaeus spores were 43.0±1.4℃, 25.0±1.6℃, and 35.8±1.4℃, respectively. The D121-values of B. subtilis, G. stearothermophilus, and B. atrophaeus spores were all higher than that of Clostridium botulinum spores used to confirm retort food sterilization. Considering these results, B. subtilis, G. stearothermophilus, and B. atrophaeus spores can be used instead of the pathogenic spore-forming bacteria C. botulinum when sterilizing retort food. In addition, sterilization can be confirmed in 2 to 3 days, a shorter time than the 13 days required for existing bacterial growth experiments based on the Korean food code.

Bioassay of Spores Attachment for the Weedy Green Alga Ulva fasciata on Agar and Agarose Surfaces (Agar와 Agarose 코팅필름을 이용한 갈파래의 포자 부착 검색)

  • Spore attachment of Ulva fasciata on agar and agaroses coating films showed various surfaces properties according to concentrations of agar or agaroses or via methanol. The highest number of spore attachment occurred in Sigma agarose coating film. Spore attachment on Bacto agar and other agaroses coating films showed the 12 ∼ 36 times less than Sigma agarose coated film. Comparison of spore attachment on 2.5% and 5% Seakem agarose and Bacto agar coating films differed in two concentrations while coating of 2.5% and 5% with NuSieve and Sigma agaroses did not differed. Spore attachment of coating made of 5% and 5% via 4% MeOH with Bacto agar and Nusieve agarose only differed. Overall, these results indicated agar and agarose coating films differed in spore attachment. Results of this work will be useful baseline for bioassay of antifouling activity of fouling organisms.

Sporulation and Dissemination of Pycnidiospores of Diaporthe citri in Yuzu Tree (Citrus junos Sieb) in Jeonnam Area (전남지역 유자과원의 검은점무늬병균 포자 형성과 비산)

  • Several time-course experiments were carried out to understand seasonal development of melanose on yuzu trees at koehung area, Jeonnam province, during May to October. The occurrence of dead twigs, known as a source of infection, was much more in older trees, and from June to August, mostly in July. In the experiment of pycnidia development on dead twigs seasonally collected, the number of developed pycnidia was highest on July-collected dead twigs especially with the diameter of 1.1~1.5 cm. In the collection survey of disseminated pycnidiospores, although the collected number of pycnidiospores was affected with amount of precipitation, the number of observed pycnidiospores in rainwater was relatively high from June to August, with highest in early August in 1997 and late July in 1998. In the inoculation tests on 3-year-old trees and fruits in natural condition, disease occurrences were mostly affected on twigs by inocula treatment in June, and on fruits by inocula treatment in July, respectively.
