Analysis of genetic relationships of Colletotrichum spp. isolated from sweet persimon with AFLP (AFLP를 이용한 단감나무 탄저병 병원균 Colletotrichum spp.의 유연관계 분석)

  • Colletotrichum species are important fungal pathogens that cause great damages on various host plant species worldwide. In Korea, Colletotrichum species cause massive economic losses on apple, peach, grape, and especially, sweet persimon productions. In the past, identification of the pathogen and the studies on the genetic relationships among the pathogenic isolates were mainly based on morphology, cultural characteristics, and the difference in pathogenicity. However, in recent years, these traditional methods have been replaced with molecular methods including AFLP. AFLP method with the merits of both RAPD and RFLP has been widely used for the genetic relationship studies of various organisms. Therefore, in this study, AFLP method was employed for the studies of genetic relationships among the different isolates of Colletotrichum species collected from various parts of sothern Korea. As a result, two specific band pattern groups were observed among different isolates of Colletotrichum species.

Exploring on the Defense Strategies against Hervivory of Broad-leaved Tree Species Growing in Taean-gun, Chuncheongnam-do (충남 태안군에서 생육중인 활엽수종의 초식에 대한 방어전략 탐색)

  • To explore on the defense strategies against hervivory of 67 broadleaved tree species, morphological characteristics of the leaf, leaf domatia structure and the number, herbivores insects and mites on the leaves, collected from the trees growing in Taean-gun, Chuncheongnam-do were investigated. 46 broadleaved tree species(68.7%) had the domatia structures, and 21 species including Quercus salicina and Magnolia grandiflora did not. 31 species including Juglans mandshurica and Carpinus laxiflora reveals tuft type, 12 species including Quercus dentata and Corylus heterophylla reveals pocket+tuft type, and 2 species, Sorbus alnifolia and Prunus yedoensis does pocket type, and Viburnum odoratissimum var. awabuki does pouch type. Domatia number per leaf proves the highest figures, 23.4/leaf for Quercus dentata. Plant defense strategies using leaf lower-surface trichomes of Magnolia grandiflora reveals dense villous, those of Populus alba and Vitis vinifera reveals dense pilose, that of Elaeagnus umbellata does dense scaly hairs, that of Pueraria lobata does dense strigose. Plant defense strategies using extrafloral nectaries were adapted 23 tree species(34.3%). Observed examples are Prunus tomentosa, Ficus carica, Viburnum dilatatum and Carpinus laxiflora. Predatory mites were observed on the leaves of 40 tree species(59.7%), and mean values of predatory mites was highest values 23.4/leaf in Quercus dentata. Minute arthropods destroying the leaf of broadleaved trees. are such as Periphyllus californiensis, P. viridis, Diaspididae sp., gall mites, thrips, and total numbers observed were odered gall mites, Diaspididae sp., aphids and thrips. Natural enemies of these hervivores arthropods are such as predatory mites, Chilocorus rubidus, Coccinella septempunctata and the nymph, Aphidius ervi. These results indicate that defense strategies including protective mutualisms may be frequent in the temperate broadleave trees.

Crown Gall of Weeping Fig Caused by Agrobacterium larrymoorei and A. tumefaciens (Agrobacterium larrymoorei와 A. tumefaciens에 의한 벤자민고무나무 뿌리혹병)

  • Crown gall on lower stem of weeping fig(Ficus benjamina Roxb.) was first observed at Daejon in 2003. Tumors were about 15 cm in size and semi-round with rough surface texture of dark brown color. Two virulent isolates among ten bacteria isolated from the tumor tissues were characterized. Their colonies were convex, glistening, circular with an entire edge, and white or tannish cream in color on potato dextrose agar supplemented with 0.5% $CaCO_3$. They were rod shape with peritrichous flagellae, gram-negative, aerobic growth, oxidase-positive, and grew on D1M agar. The isolates were identified as Agrobacterium larrymoorei and A. tumefaciens based on biochemical and physiological characteristics, fatty acid profiles and substrate utilization patterns. Seedlings of some host plants excepting grapevine produced typical galls two to three weeks after inoculation with cell suspensions of the virulent strains. This is the first report on crown gall of weeping fig in Korea.

Pathogenicity and Infection Mechanism of Pseudocercospora vitis Causing Leaf Spot Disease on Grapevine in Korea (포도나무 갈색무늬병균(Pseudocercospora vitis (Lếv.) Speg.)의 병원성과 침입기작)

  • This study was carried out to know pathogenicity of the pathogen isolated in several location of Korea and penetration mechanism of the Pseudocercospora vitis ($(L\acute{e}v)$.) Speg. Inoculation tests at room temperature were performed on both sides of leaves with different isolates of the fungus. The typical symptoms appeared on the abaxial leaf surface, but no symptom was observed on the adaxial leaf surface with all isolates. The average incubation period was nine days, since all symptoms appeared from 8 to 10 days after inoculation. In order to know the mechanism of invasion of P. vitis to grapevine, the behavior of penetration hyphae through stomata were compared in two cultivars having different level of resistance. In order to know the mechanism of resistance of grape vine to P.vitis, two cultivars having different level of resistance were compared by counting the numbers and measuring size of the stomata per leaf. In a susceptible cultivar Campbell Early, the number of stomata was more than that of a resistance cultivar 'Kyoho'. In a susceptible cultivar 'Campbell Early', the fungus entered readily into stomata after inoculation. However, in a resistant cultivar 'Kyoho', the fungus seemed to pass over or surrounded only the guard cells. In comparison of height of guard cells of stomata between the two cultivars, significant differences were observed by scanning electron microscopy and light microscopy. The height of guard cells of 'Campbell Early' showed a little higher than those of 'Kyoho' known to be resistant to the fungus.

Effects of the Grapevine Shoot Extract on Free Radical Scavenging Activity and Inhibition of Pro-inflammatory mediator Production in RAW264.7 Macrophages (포도나무가지 추출물의 프리라디칼 소거 작용 및 염증 발현 매개인자 생성 억제 효과)

  • Free radical scavengers or quenching agents for reactive oxygen species (ROS) present in consumable fruits, vegetables, and beverages have received considerable attention as potential antioxidants, and thus uses for treatment of several human diseases. In this study, grapevine shoot extract (GSE) containing high concentration of resveratrol and viniferine was evaluated for antioxidant potential and inhibition of pro-inflam-matory mediator production. Utilizing 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging activity and xanthine oxidase (XOD) inhibition assay the GSE showed inhibitory effects of DPPH radical scavenging and XOD activity with the $IC_{50}$/ values of 34.5 and 155 $\mu\textrm{g}$/ml, respectively. In addition, GSE also exhibited the inhibition of prostaglandin E$_2$ (PGE$_2$) and nitric oxide (NO) production in lipopolysaccharide (LPS)-induced mouse macrophage RAW264.7 cells with the $IC_{50}$/ value of 6.4 and 14.5 $\mu\textrm{g}$/ml, respectively. This result suggests that grapevine shoot extract has the potential activity as a natural antioxidant or antiinflammatory agent.

In the past, Identification of the pathogen and the studies on the genetic relationships among the pathogenic isolates were mainly based on morphology, cultural characteristics, and the difference in pathogenicity. However, in recent years, these traditional methods have been replaced with molecular methods to solve the difficulty of classification on pathogens. Therefore, in this study, RAPD and PCR-RFLP methods were employed for the studies of genetic relationship among the different isolates of Colletotrichum species that cause damages on sweet persimon. As a results of genetic relationship analysis, Colletotrichum species tested were divided into two big groups or five small groups.

  • The mycoflora of Korea, especially the cercosporoid taxa, have been poorly studied in the past. For this reason, a systematic revision of these taxa has been initiated, with the eventual aim of producing a monograph of the Korean cercosporoid taxa. The present study circumscribes ten species; viz., Cercospora althaeina, C. arachidicola, C. armoraciae, C. carotae, C. ipomoeae, Mycovellosiella ferruginea, Passalora miurae, Pseudocercospora araliae, P. cannabina and P. vitis. Distinguishing morphological characteristics are described and illustrated for each species.

Studies on Cultural and Morphological Characteristics of Isolate on Ganoderma species (영지속 균주의 배양적 특성 및 형태적 특성에 관한 연구)

  • Eleven fruit bodies of Ganoderma sp. were collected from eight locations throughout the forest of Kangwon province and Kyunggi province in Korea. The hosts in forest were cut trunks of Quercus dentata, Q. variabilis, Prunus peria and Alnus japonica that was newly surveyed but 5 isolates were collected at the farms of Ganoderma mushroom. Most fruit bodies were formed solitarily on the cut trunks but GS-106 isolate grown in crowds on cut trunk of Alnus japonica. Optimal temperature ranges for isolates of species studied were: G. applanatum $28^{\circ}C{\sim}30^{\circ}C$, G. lucidum $28{\sim}30^{\circ}C$, G. neo-japonicum $28^{\circ}C$, and G. tsuage $26^{\circ}C$ and all the species grew slowly at the $32^{\circ}C$. Hamada medium adjusted with pH 5.4 and 6.2 is better than other media for mycelial growth. Mycelial morphological characteristics of six species were studied: G. applanatum, G. lucidum and G. neo-japonicum produced typical type of staghoru hyphae but G. oregonens and G. valeosiacum produced staghoru hyphae with a branch of grape form. Clamp connection was observed on hypha of G. applanatum, G. lucidum, G. oregonense and G. valeosiacum except G. neo-japonicum with node type. Chlamydospore was produced by G. applanatum, G. neo-japonicum. and cuticular cells were present on hyphae of G. lucidum, G. neo-japonicum, G.oregonense and G. tsugae.

Studies on the Rooting Ability of Cutting in Elder Berry(Sambucus canadiensis) (황금(黃金)포도나무(Elder berry)의 삽목시험(揷木試驗))

  • The elder berry was known to the rich in natural food colour resources and used in as for making wine, confectionary, perfumes, natural food colour, making elder berry juice, jelly, jam and medicinal properties or oils. In the present study, wish was to find the effect of various factors on the success of the vegetative propagation of elder berry by means cutting in the exposed field and green house was carried out and those obtained results can be summarized as follows. 1. Cuttings with dormant cutting stocs in the polyethylen house with heating and water mist spray resulting 100 percent survival. And temperature and the relative humidity in which fraim during the cutting season were around $20{\sim}25^{\circ}C$ and 70-90% respectively and in case as more significant other of the 1% Level. 2. With five varieties tested, resulting 93.8 percent survival, The F. value is not significant. 3. With four organs of cutting stock tested resulting 57.5 percent survival on the cuttings with two knodes of dormant cutting stock served as better cutting stock than others. The F. value is more significantly 1% Level. 4. Dormand bud served as possible cutting stock was found to be 17.66 survival percentage. 5. Both earlier and later stage of germinated Leaves with soft wood cutting stock poor cuttings, and the degree of development of 15th June cutting stock was optimum stage on the principal factor governing the success of cutting in the soft wood cutting showing 54% survival. The F. value is more 1% Level significant.
