The Behavior of In-situ Top Base foundation in Granular Soil (사질토에서 현장타설 팽이기초의 거동특성)

  • Numerical analysis for the in-situ top base foundation (In-situ TBF) was carried out in order to investigate the effect of bearing capacity and the load delivering mechanisms in granular soil. The input data for the numerical model was prepared from the result obtained from the plate load test and full size in-situ TBF field tests. According to the result of numerical analysis, the behavior of in-situ TBF showed that bearing capacity of the foundation increased by $50{\sim}100%$ and settlement was reduced up to $1/2{\sim}1/3$ comparing to other types foundation. The effect of cone-shaped part of the in-situ TBF was as important as pile part for the improvement of foundation stability. The variation of the length of pile part indicated that the present length was proved satisfactory in terms of effectiveness.

The analysis of useful components in Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium (팽이버섯 자실체, 팽이버섯 균사체 및 동충하초 균사체의 유용성분 분석)

  • Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium were analyzed for their proximate composition, protein-bound polysaccharide, nucleic acid and amino acids. The content of ash and crude fiber in F. velutipes fruit body were higher than F. mycelium and C. militaris mycelium. C. militaris mycelium showed the highest crude fat content while F. velutipes fruit body had lowest. Nitrogen free extract content of the samples varied from 56.8% in F. velutipes fruit body to 61.9% in F. velutipes mycelium. The compositions of total protein and total free sugars of protein-bound polysaccharide were found to be significant differences for all samples. Nucleic acid related compounds were identified the 5'-GMP, 5'-XMP, 5'-IMP in all samples. The content of total nucleic acids were high in the orders of F. velutipes myclial (286.71 mg%), F. velutipes fruit body(187.36 mg%) and C. militaris mycelial(76.85 mg%). The highest content of 5'-GMP was found in F. velutipes fruit body. The most nucleic acid of F. velutipes mycelial and C. militaris mycelial were the 5'-XMP. As for the analysis of total amino acids, seventeen amino acids were identified by HPLC and the major amino acid was glutamic acid in all samples. The content of total amino acids were high in the orders of F. velutipes fruit body(19,919 mg%), F. velutipes mycelium(19,018 mg%) abd C. militaris mycelium(18,965 mg%). We determined the developing new food product such as amino acid drink and amino acid containing food using extracts of Flammulina velutipes fruit body, Flammulina velutipes mycelium and Cordyceps militaris mycelium.

Physical Wounding for the Enhancement of Agrobacterium-Mediated Transformation of Flammulina velutipes Mycelium (물리적 상해를 통한 Agrobacterium 이용 팽이균사체의 형질전환효율 증대)

  • In this study, Agrobacterium-mediated transformation was tested to the mycelium culture of Flamulina velutipes which is very popular as an edible mushroom in Korea. Particularly, aluminum oxide particles were used to generate wounds in F. velutipes mycelia via vigorous shaking prior to agro-infiltration. The result showed that transformants resistant to hygromycin could be obtained only from the mycelia with physical wounds. Gene transfer was verified by genomic DNA PCR. This study suggested a convenient tool to improve Agrobacterium-mediated transformation of F. velutipes.

The Bearing Capacity Characteristics of Top Base Foundations in Cohesionless Soils (비점착성 지반상 팽이기초 적용에 따른 지지특성)

  • Top Base Foundation (TBF) is widely used for the increment of bearing capacity and restraining settlement of foundations when the bearing capacity of ground is not enough. The design of bearing capacity obtained from exiting Japanese standard underestimates considerably what is compared with the observation values from the field measurement. Therefore, intensive model tests and site observation programs are carried out to establish more reasonable prediction technique and understanding of TBF in soft ground. In this study, 1/5 scale model tests are performed in order to investigate the behavior of TBF with various shapes in a laboratory. Also, about 100 sites measurement data are evaluated to investigate the field behavior of TBF in various ground conditions. Based on the results of the model tests and field measurement data, it was possible to introduce more reasonable bearing capacity equations of TBF considering N-value of soils, the effect of underground water and failure mechanisms.

  • This research performed the laboratory model tests for Top-Base Foundation developed in Japan and Floating Top-Base Foundation developed in Korea on the typical clayey soft soils, namely, clay, clayey silt and clayey sand. The performances of the two types of top-base foundation were compared with each other and evaluated by measuring load-settlement, heaving of foundation side, ground stress distribution in this model tests. The change of settlement caused by the increase of top base width was also analyzed. As a result of the model tests, Floating Top-Base Foundation showed better performance in bearing capacity improvement, settlement decrease, stress dispersion effect and lateral confinement force. And settlement caused by the increase of top base width converged to a regular value from $5{\times}5$ layout of the width.

  • ${\gamma}$-Aminobutyric acid (GABA) is basically neurotrasmitter produced by the decarboxylation of L-glutamic acid catalyzed by glutamic acid decarboxylase (GAD), which was known to convert monosodium glutamate (MSG) to GABA. To investigate enhancement of reversion rate of GABA, the yogurt fermentation with addition of nanoparticle winter mushroom and hydroponic ginseng was used. The conversion rate was revealed to nanoparticle winter mushroom and hydroponic ginseng fermenter (88%) > winter mushroom fermenter (52%) > nanoparticle winter mushroom fermenter (44%). The results showed that nanoparticle winter mushroom and hydroponic ginseng supplemented substrates for enhancement of GABA may be used more effectively as one of potential sources of functional foods.

Forest Green Mold Disease Caused by Trichoderma pseudokoningii in Winter Mushroom, Flammulina velutipes (Trichoderma pseudokoningii에 의한 팽이버섯 푸른곰팡이병)

  • Forest green mold incidence rate, extent of damage according to the inoculation periods, and its cultural characteristics were observed in the automatic cultural system of the winter mushroom, Flammulina velutipes. The incidence rate of the forest green mold was 7.7% in early cultivation stage and slowly increased to 14.9% in harvest stage. When the forest green mold was inoculated at cultural period, the rate was recorded at 100%, but the extent of the damage increased up to 40% (+++). There was also 100% incidence rate at early pinheading time, whereas the yield of mushroom decreased to ++ $(10{\sim}39%)$. The rate of forest green mold was greatly decreased to 34.4% at 10 days after pinheading, and its damage extent was also below 10%. A pathogen to infect the winter mushroom was identified as Trichoderma pseudokoningii. It's optimum temperature for mycelial growth is $25^{\circ}C$, and it grew 2.6 times faster than that of F. velutipes. The mycelial color of T. pseudokoningii was pale yellow or olivaceous in shades on PDA medium. Phialospore was one celled, and ellipsodal or obovoid, smooth walled, and measured $1.3{\sim}3.0{\times}1.0{\sim}2.5\;{\mu}m$. It aggregated in small heads at the tips of the phialides. The phialides were $3.2{\sim}9.2{\times}2.0{\sim}5.5\;{\mu}m$ and were of bowling pin type, solitary and alternate or more irregularly disposed at the conidiophore apex, T. pseudokoningii depressed the F. velutipes growth at the crossing cultivation when they were simultaneously. FV 4-1 (F. velutipes) cultivar was less depressed by T. pseudokoningii, but had a lower cross growth rate than the other four cultivars.

Applicability of Spent Mushroom Media as Horticultural Nursery Media (버섯재배 후 탈병배지의 원예용 상토재료 이용성 검토)

  • This study was carried out to investigate applicability of Spent Mushroom Media(SMM) as horticultural nursery media. After the mushroom has been harvested, the SMM contains a lot of organic material, different microorganism and high density of mushroom hypha. The pH, phosphate and exchangeable cation concentrations of SMM of Flammulina velutipes were higher than those of any other treatment. The CEC and $NH_4-N$ were the highest in SMM of bottle-cultivated oyster mushroom (Pleurotus ostreatus). Bacteria and fungi showed the highest density in SMM of Flammulina velutipes. Most dominant bacteria were Microbacterium sp., Rhodococcus sp. and Agrobacterium sp. in SMM of Flammulina velutipes and Bacillus sp., Pseudomonas sp., Curtobacterium sp. and Microbacterium sp. in that of Pleurotus eryngii. The SMM contained high density of mushroom hypha that inhibited germination of seed and growth of young seedlings. Therefore, composting process of the SMM is indispensible to decline of vitality of mushroom hypha. The SMM of Flammulina velutipes with 0~30% vermiculite showed high germination rate in red pepper and chinese cabbage seeds. SMM of Pleurotus eryngii with 20% vermiculite showed 100% germination rate in red pepper seeds, but chinese cabbage seeds nearly failed to germinate with 30% vermiculite. The growth of red pepper was increased according to increasing mixture ratio of vermiculite. Accordingly, we concluded that SMM of Flammulina velutipes contained 0~30% of vermiculite can be used to horticultural growth bed for red pepper.

Growth Survival of Listeria monocytogenes in Enoki Mushroom (Flammulina velutipes) at Different Temperatures and Antilisterial Effect of Organic Acids (팽이버섯에서 Listeria monocytogenes의 온도별 생존과 유기산에 의한 저감화)

  • Listeria monocytogenes (L. monocytogenes) was responsible for several recall cases owing to its incidence in mushrooms exported from the Republic of Korea. In this study, we investigated the survival of L. monocytogenes in enoki mushroom (Flammulina velutipes) at different temperatures and the antilisterial effect of its organic acids. Enoki mushrooms were innoculated with L. monocytogenes (initial concentration 4.5 log CFU/g) and stored at 1-35℃, No growth of L. monocytogenes in enoki mushrooms was observed at 1℃ for 30 days. 3.0 log CFU/g growth of L. monocytogenes was also achieved after 36 h and 24 h at 30℃ and 35℃, respectively. To evaluate the antilisterial effect of the organic acids (acetic acid, lactic acid, malic acid), enoki mushrooms were treated with 1-3% of each acid for 10-30 min. The efficacy of malic acid and lactic acid was significantly higher than that of acetic acid. Over 3.0 log reductions were observed when L. monocytogenes in enoki mushrooms was immersed in 3% lactic acid and malic acid over 10 minutes or more. Therefore, it is necessary to keep enoki mushrooms at 1℃ during the export process and treat them with 3% lactic acid and malic acid for 10 min prior to consumption.

Physicochemical Properties of Mushroom (Flammulina velutipes) Cultivated with Green Tea (녹차를 이용하여 재배한 팽이버섯의 이화학적 특성)

  • Physicochemical properties of the green tea component enriched mushroom (Flammulina velutipes) were investigated. The yield of mushroom was highest with green tea powder untreated sample and the yield was decreased by the addition of green tea powder. However, hardness was increased by the addition of green tea powder. Analysis of catechins and caffeine in mushrooms showed that catechins were not transferred into mushrooms, whereas caffeine content was increased. The content of total polyphenol in mushroom was not affected by the addition of green tea powder and crude catechins. Analyses of aroma patterns using the electronic nose based on GC with SAW sensor showed that new peaks occurred from 3 to 6 sec in green tea added mushroom. This study showed that functional components and quality of mushroom were possibly improved by incorporating green tea powder in growth medium.
