Comparison for Genetic Diversity between Korean Native Commercial Chicken Brand Groups using Microsatellite Markers (Microsatellite Marker를 활용한 토종닭 브랜드 집단 간의 유전적 다양성 분석)

  • To estimate the genetic characteristics within two brands of Korean native commercial chicken, we used a total of 302 genomic DNAs from two groups (Woorichicken: 152, Hanhyup3chicken: 150). Sizes of 10 microsatellite markers were decided using GeneMapper Software (v.4.0) after analyzing ABI 3130. Genetic diversity indices including expected heterozygosity (Ex H), observed heterozygosity (Ob H) and polymorphism information content (PIC). Frequencies of microsatellites markers were used to estimate heterozygosities and genetic distances. LEI0073 showed the highest value in all genetic diversity (Ex H, Ob H and PIC). On the other hand, MCW322 showed the lowest value in all genetic diversity. The calculated genetic distance of the two brand groups is 0.199 (standard genetic distance) and 0.132 (DA distance). Genetic distances of the two groups were relatively close to each other. Each individual is ramified to two brand groups in phylogenetic dendrogram.

Comparison of Body Weight and Egg Production Ability across Nine Combinations of Korean Indigenous Chicken Breeds (토종닭 종계 9개 조합의 체중 및 산란능력 비교 연구)

  • The present study investigated production traits, including body weight, egg production, egg weight, and egg quality in Korean indigenous parent stocks. Parent stocks produced from a nine-combination association from five pure line strains (C, D, F, K, and Y) were used. The body weight analyses results showed that the CY combination had the highest body weight (1,604.9 g) and the DK combination had the lowest (1,424.4 g). The average age at first egg of the nine combinations was 118.6 days; however, that of the YD combination was 111.6 days, making it the fastest growing combination, whereas the DK combination was the slowest, at 126 days. Hen-day egg production was 74% or more for the CF, CK, and DK combinations. The DK combination hens showed excellent persistence in egg laying. Hen-housed egg production exhibited results similar to those of hen-day egg production. Egg weight was significantly higher in the DK combination than in the other combinations and was observed to increase from 20 (43.9 g) to 40 (58.1 g) weeks. The egg quality analyses results showed that the combinations based on the F and K strains had a bright eggshell color, with relatively high egg weights. Eggshell strength and thickness were the highest in the DK combination (3.8 kg/cm2, 0.38 mm). In summary, the CF and DK combinations showed excellent egg production ability and egg quality, while the YC, YD, and YK combinations with the Y strain as the paternal strain exhibited poor performance.

Quality Comparison of Chuncheon Dakgalbi Made from Korean Native Chickens and Broilers (토종닭과 육계로 만든 춘천닭갈비의 품질비교)

  • This study was conducted to evaluate a quality comparison between Chuncheon Dakgalbi made from Korean native chickens (KNC) and that made from commercial broilers. Two Korean native chickens including Woorimatdag (KNCWoori) and Hanhyup3 (KNC-Hanhyup), and two commercial broilers including grades of 18 (Broiler-18) and 13 (Broiler-13) were slaughtered at 110, 70, 38, and 31 d of ages. Chuncheon dalkalbi was prepared by mixing/dipping the meat in chili pepper sauce; it was then packed with air-packaging (Air-P) and 30% $CO_2$-MAP (0% $O_2$/30% $CO_2$/70% $N_2$), and stored at $5^{\circ}C$ for 10 d. The results showed that the KNC group had a lower pH but a higher cooking loss compared with the broiler group (p<0.05). In a texture analysis, KNC-Woori had the highest shear force value among the breeds (p<0.05). For the fatty acid composition of the thigh, the KNC-Woori contained more total saturated acids, myristic acid, palmitic acid and stearic acid, but less total unsaturated fatty acids, linoleic acid and linolenic acid than other breeds (p<0.05). Also, the n6/n3 ratios of the KNC group (19.24 and 16.77) were higher than those of the broiler group (14.02 and 14.77) (p<0.05). The total acceptability scores of Dakgalbi made from the KNC group were decreased by sensory panelists. The Dakgalbi with 30% $CO_2$-MAP delayed the protein deterioration (Volatile basic nitrogen) and lipid oxidation during storage. However, no clear evidence was observed of $CO_2$-MAP on the effect of different chicken materials. It is suggested that 30% $CO_2$-MAP instead of Air-P is used for methods for Chuncheon Dakgalbi. Furthermore, it might be unfavorable to use Korean native chickens as raw material for Chuncheon Dakgalbi from a practical quality point of view.

  • 우리나라의 닭고기는 유통주체 회사의 주도에 의한 통합생산이 일반적이며 일부 농가에서 토종닭을 이용한 자체 브랜드(PB)가 개발되어 있으나 그 수는 적다고 할 수 있다. 정예 농업인 육성과 지역 단위 농축산물의 차별화 전략, 유통회사의 브랜드 개발로 많은 닭고기 브랜드가 있으나 사료에 의한 방법으로 한약재 활용, 지방산 이용이 주류를 차지하고 있다. 앞으로 농가 및 생산자 중심의 브랜드 닭고기 개발에 있어 맛 성분에 의한 차별화에 도움이 될 수 있는 내용을 소개하고자 한다.

Crossbreeding Combination Test for the Production of New Synthetic Korean Native Commercial Chickens (신품종 토종닭 생산을 위한 교배조합 검정시험)

  • In this study, we performed a cross-combination test using offspring from two paternal and five maternal parent stock lines to produce a new synthetic breed of Korean native commercial chickens. The survival rate, body weight, feed intake, and feed efficiency were measured for 10 crosses, with 837 Hanhyup-3 chickens used as a control. Most chickens had brown-based feathers, and there was little difference in color phenotypes among combinations. The average survival rate of the combinations was 94.9%, which was significantly different from that of Hanhyup-3 at 75.3% (P<0.01). There was significant differences in body weight among the combinations at all weeks of age (P<0.01). At 10 weeks of age, the FHHY combination was the heaviest with 2,300 g, while the weight of Hanhyup-3 was 1,922 g. In addition, the weight difference between the male and female chickens was 458 g. As a result of regression analysis, the duration needed to reach 1.9 kg was about 61 days for the FHFY, FHHY, and HFFY combinations, and 70 days for Hanhyup-3, exhibiting a 9-day difference. The average feed conversion ratio of the combinations until 10 weeks was 2.42, which was significantly different from that of Hanhyup-3 at 2.97 (P<0.01). In conclusion, FHHY, FHFY, and HFFY were determined to be the best new breed combinations based on production capacity. When these combinations are used as new commercial breeds, they are expected to increase the average weight by 18% and improve feed efficiency by 19%, as compared to the existing Hanhyup-3 breed.

The Effects of Beak Trimming and Crossbreeding-Combinations on the Productive Performance and Stress Response Levels of Korean Native Chickens (한국 토종닭에 있어 부리 다듬기 및 교배조합이 생산능력과 스트레스 반응 정도에 미치는 영향)

  • This study aimed to investigate the effects of beak trimming and crossbreeding-combinations on the productive performance and stress response levels of Korean native chickens. The study divided 248 individuals from six crossbreeding-combinations into two groups: one underwent beak trimming, and the other did not. The survival rate, body weight, egg production rate, egg quality, feather damage score, HSP-70 gene expression level, H/L ratio, and intracellular DNA damage rate were measured and analyzed. The results showed that the beak-trimmed group had significantly higher survival rates and hen-housed egg production compared to the non-beak-trimmed group (P<0.05). Feather damage and DNA damage rates were significantly lower in the beak-trimmed group (P<0.05). On the other hand, there were no significant differences between the two groups in adult body weight, hen-day egg production, egg quality, HSP-70 gene expression level, and H/L ratio. Among the crossbreeding-combinations, there were significant differences in survival rate, body weight, feather damage score, egg quality, and DNA damage rate (P<0.05), while egg production rate, HSP-70 gene expression level, and H/L ratio showed no significant differences. There was an interaction between beak trimming and crossbreeding-combinations in some traits. In conclusion, beak trimming in Korean native chickens has a positive impact on productive performance, and in terms of stress response, beak trimming may not act as a stress factor or may even reduce stress after the growing period. Furthermore, there were differences in productive performance and stress response levels among crossbreeding-combinations, but the effects of beak trimming were similar across these combinations.

Establishment of Korean Native Chicken Auto-Sexing Lines Using Sex-Linked Feathering Gene (한국토종닭의 깃털 발육성 반성 유전자를 이용한 자가성감별 계통 조성)

  • Although feather-sexing using sex-linked genes related to feather development is a widely used chick sexing method in the poultry industry, the feather-sexing method has yet to be used for Korean native chickens (KNCs). The purpose of this study was to construct a KNC feather-sexing line using early-feathering (EF) and late-feathering (LF) genes for industrial application. Using 557 reddish-brown KNCs as the basal flock, frequencies of the EF (k) and LF (K) genes were estimated to be 0.814 and 0.186, respectively. This indicating that it would be feasible to construct a feather-sexing line using this chicken group, and we accordingly constructed EF paternal and LF maternal lines. On the basis of test-cross for the selection of LF homozygous (KK) males in the maternal line, we confirmed that three of 40 chickens were homozygous males. The survival rate, body weight, days at first egg-laying, hen-day egg production, and egg weight were analyzed to compare the production performance of EF and LF chickens. The results revealed that EF chickens were characterized by a superior survival rate, whereas LF chickens were superior in terms of egg production rate. However, no differences between LF and EF chickens were detected with respect to other production performance parameters. In addition, assessment of the fitness of sexed chicks produced in the established KNC feather-sexing lines revealed that the accuracy of sexing was 98.6%. Collectively, these findings indicate the feasibility of constructing effective KNC feather-sexing lines with potential industrial application.

Comparison of Telomere Length and Vitality among Korean Native Chicken Breeds (토종닭 품종 간 텔로미어 길이 및 생존율 비교 분석)

  • We compared the degrees of vitality of 12 Korean native chicken breeds, such as Jaeraejong, Korean Rhode Island Red (Rhode), Korean White Leghorn, Korean Cornish (Cornish), and Korean Ogye breeds. The survival rate and telomere length were measured as vital markers. Telomere length was analyzed via quantitative fluorescence in situ hybridization method using the lymphocytes of 466 chickens. We found that the telomere length decreased linearly with increasing chronological age in all chicken breeds. Telomere length and telomere shortening rates (TSR) were significantly different among the chicken breeds after 20 weeks of age (P<0.01). Rhode had the longest telomere length and the lowest TSR, whereas Cornish had the shortest telomere length and the highest TSR. In terms of TSR, the telomere length of 50-week-old chickens was half of that of 8-week-old chickens. There was also a significant difference in survival rates among the breeds. Both Rhode and Korean Ogye had the highest survival rates, while Cornish had the lowest. There was a significant positive correlation between survival rate and telomere length, and telomere length in old age showed a higher correlation with survival rate than that in young age. Therefore, it is considered that TSR is more closely related to survival rate than the telomere length. Based on the telomere dynamics and survival rates of 12 Korean native chicken breeds, it was concluded that the Rhode breed and Cornish breed had the highest and lowest vitality, respectively.

Studies on Genetic Diversity and Phylogenetic Relationships of Korean Native Chicken using the Microsatellite Marker (Microsatellite Marker를 활용한 한국 토종닭 품종의 유전적 다양성 및 유연관계 분석)

  • In this study, genotyping was executed by using 27 microsatellite markers for genetic diversity of 469 Korean Native Chickens [20 population, each population is 24 samples but Hanhyup A line is 13 samples). in total 469 samples were collected from National Institute of Animal Science (Korean Native Chicken (NR, NY, NG, NL and NW), Ogye (NO), Leghorn F,K (NF and NK), Black and Brown cormish (NH and NS), Rhode Island Red C, D (NC and ND), Total is 12 populations] and Hanhyup [H line (HH), F line (HF), G line (HG), V line (HV), S line (HS), W line (HW), Y line (HY), A line (HA), total is 8 populations]. [The allele number were observed 5 (ADL0268) to 20 (MCW0127) each markers. Observed heterozygostiy ($H_{obs}$), expected heterozygosity ($H_{exp}$), polymorphism Information Content (PIC) were observed 0.359 to 0.677, 0.668 to 0.881 and 0.646 to 0.869, respectively. Using these markers, the calculated the heterozygote deficit within chicken line ($F_{is}$) value each population from mean 0.117. Phylogenetic tree showing the genetic relationship among 20 population using standard genetic distance calculated from 27 microsatellite markers. genetic distances revealed the closest (0.175) between NC and ND. on the other hand, Farthest genetic distances (0.710) revealed between NF and HV. STRUCTURE analysis and Principal Components Analysis (PCA) showed that results of similar phylogenetic tree. The expected probability of identity values on random individuals (Total population and only Hanhyup line) was estimated at $8.80{\times}10^{-83}$ and $3.87{\times}10^{-117}$, respectively. In conclusion, This study shows the useful data that be utilized as a basic data of Korean Native Chicken breeding and development for commercial chicken industry to meet the consumer's demand.
