Ethanol is considered as one of the most suitable substitutes for the petroleum, since it offers attractive functional features at an economical cost. Glucoamylase producing yeasts were isolated and characterized. Based on the morphological character, carbon fermentations, assimilation of carbon and nitrate, growth on vitamine-free medicine, and urease activity, five isolates of Saccharomyces diastaticus, two isolates of Saccharomycopsis fibuligera, and two of Schwanniomyces occidentalis, and each isolate of Ambrosiozyma monospora and Lipomyces kononenkoae were identified. Among 12 isolates, one of the S. diastaticus, E3 showed the highest activity of glucoamylase and identified as Saccharomyces diastaticus. The hydrolysis of starch by the E3 strain showed the release of considerable amount of reducing sugar, along with the reduction in iodine staining capacity. The product of action of glucoamylase, glucose was determined by thin-layer chromatography. The enzyme activity was found to be stable in broad pH range of $5.0{\sim}7.0$ with optimal activity at pH $5.0{\sim}6.0$. The enzyme showed optimal antivity at $50^{\circ}C{\sim}60^{\circ}C$. Soluble starch and glucose were better carbon sources for the enzyme production than xylose and glycerol. $Na^+\;and\;Mg^{2+}$ increased the glucoamylase activity, however $Hg^{2+}\;and\;Ag^{2+}$ inhibited the activity. Soluble starch was the best substrate for the enzyme activity.
A Bacillus strain from Korean soil was isolated and identified to be Bacillus licheniformis B1 through various biochemical tests, VITEK, and MIDI system analysis. The strain produced extracellular amylase and protease. Whether or not the strain can perform Chungkookjang fermentation with autoclaved soybean and Kanjang fermentation was determined in this study. In Chungkookjang fermentation, browining materials of strong anti-oxidant increased 8-fold, and 2-fold in Kanjang, compared with initiation material for fermentation. Maximal protease activity in Chungkookjang was observed one day after inoculation. Protease activities in Kanjang decreased to the half, and then maintained constant values during fermentation, probably due to the inhibitory effect of salt on protease activities. High molecular mass of nucleic acids was identified in Chungkookjang and Kanjang. Since the nucleic acids were not observed in autoclaved soybean, they seem to be originated from B. licheniformis B1. This study demonstrated successive fermentation of Chungkookjang and Kanjang by B. licheniformis B1 isolated from nature, and suggested possible development of food rich in browing and nucleic acids.
In order to isolate thermophilic compost-promoting bacteria with high activity of cellulase and xylanase, spent mushroom substrates with sawdust were collected from mushroom cultivation farm, Jinju, Gyeongnam in Korea. Among of the isolates, one strain, designated SJ21 was selected by agar diffusion method. The strain SJ21 was identified as members of the Bacillus lincheniformis by biochemical characteristics using Bacillus ID kit and VITEK 2 system. Comparative 16S rDNA gene sequence analysis showed that strain SJ21 formed a distinct phylogenetic tree within the genus Bacillus and was most closely related to Bacillus subtilis with 16S rDNA gene sequence similarity of 99%. On the basis of its physiological properties, biochemical characteristics and phylogenetic distinctiveness, strain SJ21 was classified within the genus Bacillus, for which the name Bacillus sp. SJ21 is proposed. The cellulase and xylanase activity of Bacillus sp. SJ21 was slightly increased according to bacterial population from exponential phase to stationary phase in growth curve for Bacillus sp. SJ21.
Pullulanase gene (pul) of Klebsiella pneumoniae NFB-320 which was cloned previously in Escherichia coli with plasmid pBR322. The gene was analyzed with various restriction enzymes. The cloned gene was contained within n 10 kb BamHI DNA fragment. We constructed the restriction map of the hybrid plasmid pYKL451. The optimum temperatures for pullulanases produced in E. coli (pYKL451) and K. pneumoniae NFB-320 were almost the same, 50-55 $^{\circ}C$. The optimum pHs for the reaction of the enzymes produced by E. coli (pYKL451) and K. pneumoniae NFB-320 was 6.0. Both enzyme preparations were stable under the range of pH 5.0 to 10.0 when those were kept at 40 $^{\circ}C$ for 90 min and were stable until 40 $^{\circ}C$ when allowed to stand for 1hr at various temperatures.

The effect of soil damage on the physicochemical characteristics and activity of the soil microbial community is not well known. This study investigates this relationship by analyzing 11 soil samples collected from various points of soil damage across Gyeonggi-do. Soil damage resulted from forest fires, landslides, and development areas, with their impacts most severe on the topsoil layer (0-30 cm). Dehydrogenase and β-glucosidase activities were notably higher at locations damaged by forest fires compared to other sites. While enzyme activities in soils influenced by landslides and development areas were relatively low, sites with a pollution history exhibited elevated dehydrogenase activity, likely due to past microbial response to the pollution. Additionally, an assessment of carbon substrate usability by soil microorganisms indicated higher substrate availability in areas impacted by forest fires, contrasting with lower availability in landslide and development sites. Statistical analysis revealed a positive correlation between organic content of sand and clay and microbial activity. These findings provide valuable insights into soil damage and associated restoration research, as well as management strategies.
A bacterium producing the neutral pretense was isolated from soil, and was identified as Bacillus sp. DS-1 by 16S rRNA sequence comparison and biochemical determinations. The production of protease from Bacillus sp. DS-1 was increased 20% and 30% by the additions of 1% glucose and 1% yeast extract, respectively. The optimum pH and temperature for the protease activity were pH 7.0 and 55$^{\circ}C$. Bacillus sp. DS-1 produced a metalloprotease as a major protease in culture medium, since the pretense activity in culture supernatant was inhibited by the presence of 1 mM EDTA significantly.
Effects of different amounts of fertilizer and manure application on changes of chemical and biological properties of soil were studied in plastic film house. Application amount of fertilizer was determined on the basis of soil test, standard application rate, and conventional rate of farmers. Lettuce in the first and second seasons and spinach in the third season were cultivated. Crop yields were highest in the plot fertilized on the basis of soil test during the three crop seasons. In the third crop season, spinach yield was lower in conventional plot applied with larger amount of pig manure compost than any other treatment. Organic matter, available phosphorus, and exchangeable potassium were accumulated in soil of the conventional plot with successive cropping. There was no difference in the abundance of soil invertebrates among the treatments, but soil enzyme activity was highest in the conventional plot.

Excessive soil organic matter (SOM) is detrimental to turfgrass quality when used intensively in sand-based root zones, thereby affecting the sustainability of turfgrass systems. As part of a major project examining the sustainable management of SOM on golf greens, microbial decomposition on soil organic matter accumulation with depth was assessed and the effect of soil air-condition improvement and Ca fertilization was investigated by soil microbial respiration (SMR). Three soil samples from three depths(0~5, 5~10, and 10~15cm) of 5 year and 30 year old green were analyzed for SOM content. In 30 year old green, SMR and dehydrogenase activity(DHA) were analyzed to assess the soil microbial decomposition with depth. It was then divided into 4 plots: untreated as a control, dolomite-treated, 0~5cm deep section-removed, and 0~5 cm deep section-removed+dolomite-treated. After treatment, three soil samples were taken at 1, 2 and 4 weeks by the above-mentioned method, and analyzed for SMR to better understand SOM decomposition. SOM accumulation in the 0~5cm depth of golf greens can be controlled by intensive cultivation such as coring, but below 5cm is more difficult as the results showed that SOM content below 5cm increased over time. Soil microbial decomposition of organic matter will be necessary to reduce SOM accumulation, but SMR below 5cm was low and wasn't significantly altered by increasing exposure to air and fertilizing with Ca. As a result, aeration treatments such as coring and Ca fertilization might not be effective at improving soil microbial decomposition below 5cm depth in aged greens.
Hyporheic zone is an area where hydraulic exchanges occur between surface water and ground water. Such transient area is anticipated to facilitate diverse biogeochemical reactions by providing habitats for various microorganism. However, only a few data are available about microbial properties in hyporheic zone, which would be important in better understanding of biogeochemical reactions in whole streams. The study site is Naesung stream, located in the north Kyoung-Sang Province, of which sediment is sandy with little anthropogenic impacts. Soil samples were collected from a transect placed perpendicular to stream flow. The transect includes upland fringe area dominated by Phragmites japonica, bare soil, and soil adjacent to water. In addition, soil samples were also collected from downwelling and upwelling areas in hyporheic zone within the main channel. Soils were collected from 3 depth in each area, and water content, pH, and DOC were measured. Various microbial properties including extracellular enzyme activities ($\beta$-glucosidase, N-acetylglucosaminidase, phosphatase and arylsulfatase), and microbial community structure using T-RFLP were also determined. The results exhibited a positive correlation between water content and DOC, and between extracellular enzyme activities and DOC. Distinctive patterns were observed in soils adjacent to water and hyporheic zone compared with other soils. Overall results of study provided basic information about microbial properties of hyporheic zone, which appeared to be discernable from other locations in the stream corridor.

In this study, for the purpose of decomposing food waste, the strain was screened from traditional fermented food and soils. The enzyme activity (protease, amylase, cellulase, lipase) experiment was carried out using the paper disc method in 212 strains isolated from 5% NaCl media. Among them, only the strains having enzyme activity of more than 2 (soil) or more than 4 (traditional fermented food) with the halozone of enzyme activity of 15 mm or more were selected first, and microorganism identification through 16S rRNA sequencing was performed. Finally, were identified such as Bacillus subtilis, Bacillus amyloliquefaciens, Bacillus siamensis, Bacillus licheniformis, Bacillus aquimaris, Bacillus megaterium, Bacillus koreensis, Bacillus stratoshericus, Bacillus aryabhattai, Bacillus safensis, Marinobacter hydrocarbonoclasticus. 11 species of mixed strains were confirmed that the culture time was 24 hours, the incubation temperature was $30^{\circ}C$ and the optimum pH was 7.0. In order to confirm the degree of decomposition of standard food wastes (100 g) by treating 11 kinds of mixed strains (25%), solid content of more than $2000{\mu}m$ was determined to be 103 g for the sterilized water group and 18 g for the mixed strains group. And the rest was decomposed to a size of less than $2000{\mu}m$.
