Sphingomonas sp. KS 301, which was isolated from oil contaminated soil, was shown to have five different SODs (SODI, II, III, IV, V) which can be separated by DEAE-Sepharose chromatography, and SOD III was finally purified in this study by ammonium sulfate precipitation, DEAE-Sepharose chromatography, Superose 12 gel filtration and Uno-Q1 ion exchange chromatography. The molecular weight of SOD III was 23 kDa as determined by SDS-PAGE and the apparent molecular weight of the native enzyme was estimated to be approximately 71 kDa by Superose-12 gel filtration chromatography. These data suggest that the purified SOD consists of at least two subunits. The specific activity of the SOD III was higher than Mn type or Fe type SOD of Escherichia coli by 5 fold. To determine the type of SOD III, inhibitory effects of $NaN_{3},\;H_{2}O_{2},\;KCN$ were examined. 10 mM $NaN_{3}$ was able to inhibit 56% of the SOD III activity, which indicates that this SOD is Mn type. The optimum pH of the SOD III was 7.0 and the optimum temperature was $20^{\circ}C$. N-terminal amino acid sequence of purified SOD III was most similar to those of Psudomonase ovalis and Vibrio cholerae among bacteria.
A Pseudomonas strain SG3 producing biosurfactant and showing antifungal and insecticidal activities was isolated from agricultural soil severely contaminated with machine oils. The antagonistic bacterium inhibited mycelial growth of all of the tested fungal pathogens. The fermentation broth of SG3 also effectively suppressed the development of various plant diseases including rice blast, tomato gray mold, tomato late blight, wheat leaf rust, barley powdery mildew and red pepper anthracnose. An antifungal substance was isolated from the fermentation broth of SG3 by ethyl acetate partitioning, silica gel column chromatography and preparative HPLC under the guide of bioassay. The chemical structure of the antifungal substance was determined to be rhamnolipid B by mass and NMR spectral analyses. The antifungal biosurfactant showed a potent in vivo antifungal activity against gray mold and late blight on tomato plants. In addition, rhamnolipid B inhibited mycelial growth of B. cinerea causing tomato gray mold and zoospore germination and mycelial growth of P. infestans causing tomato late blight. Pseudomonas sp. SG3 producing rhamnolipid B could be used as a new biocontrol agent for the control of plant diseases occurring on tomato plants.

This research programme include investigation of the adsorption behavior of heavy metals and VOCs by Pyrolysis char for using landfill cover material. The volatile potions in the sludge gasified during the pyrolysis period and gave birth to porosity throughout the matrix. The result of the ad/desorption experiment of nitrogen to find out the formation of some pore by the gasification of the volatile matter, we can certify that the pyrolysis char($14.56\;m^2/g$) has increased twice more than the organic wasted sludge($6.68\;m^2/g$) in specific surface area. The pyrolysis char has the adsorption characteristic of medium type of Type II and V in BDDT classification, and showed a little micro pore. In the adsorption experiment of ethylbenzene and toluene, as a result of applying the Freundlich adsorption isotherms, the pyrolysis char was higher in the adsorptivity of ethylbenzene and toluene than the granite and the organic wasted sludge. The results of the heavy metal adsorption test for the char indicated that it had some ability of adsorption. It is suggest that pyrolysis char has some advantages for utilizing as landfill covers because the pyrolysis char can adsorb/absorb hazardous substances from the landfill sites and inhibit the ground water and soil contamination.

ICP-AES has been used in many laboratories due to the advantages of wide calibration range and multi-element analysis, but it may give erroneous results and suffer from spectral interference due to the large number of emission lines associated with each element. In this study, certified reference materials (CRMs) and field samples were analyzed by ICP-AES and HG-AAS according to the official Korean testing method for soil pollution to investigate analytical problems. The applicability of HG-ICP-AES was also tested as an alternative method. HG-AAS showed good accuracies (90.8~106.3%) in all CRMs, while ICP-AES deviated from the desired range in CRMs with low arsenic and high Fe/Al. The accuracy in CRM030 was estimated as below 39% at the wavelength of 193.696 nm by ICP-AES. Significant partial overlaps and sloping background interferences were observed near to 193.696 nm with the presence of 50 mg/L Fe and Al. Most CRMs were quantified with few or no interferences of Fe and Al at 188.980 nm. ICP-AES properly assessed low and high level arsenic for field samples, at 188.980 nm and 193.696 nm, respectively. The importance of the choice of measurement wavelengths corresponding to relative arsenic level should be noted. Because interferences were affected by the sample matrix, operation conditions and instrument figures, the analysts were required to consider spectral interferences and compare the analytical performance of the recommended wavelengths. HG-ICP-AES was evaluated as a suitable alternative method for ICP-AES due to improvement of the detection limit, wide calibration ranges, and reduced spectral interferences by HG.
Nutrient balance during rice cultivation in the paddy of a local area under the environmental protection for drinking water supply was investigated. To compare nutrient balance in the paddy soil applied with different types of fertilization, 7 treatments were selected as followings : Recommended level of chemical fertilizers(R), Conventional fertilization(CF), Fresh cow manure(FCM), Cow manure compost(CMC), Straw compost+reduced chemical fertilizer(SCF), Fresh straw+recommended level of fertilizers(FSC), and no fertilization as control(C). Here, FCM, CMC and SCF were applied at the same level of total nitrogen as recommended in R. Rice yield was the highest in the recommendation(R) and fresh cow manure (FCM) treatments with $6,730kg\;ha^{-1}$(index 100), and followed by SCF (index 98), FSC (index 98), CMC(index 94), and CF(index 94). But statistically significant difference was not recognized among treatments except the control. Nitrogen infiltration loss was high in the simple chemical fertilizer treatments with $63kg\;ha^{-1}$ in CF and $58kg\;ha^{-1}$ in R during rice cultivation, respectively. Nitrogen infiltration loss was decreased below half level of chemical fertilizer treatments with cow manure treatments ($23kg\;ha^{-1}$ in FCM and $27kg\;ha^{-1}$ in CMC) and with reducing chemical fertilizer treatment by adding straw compost ($25kg\;ha^{-1}$). Phosphate was not leached during rice cultivation in paddy soil of a fluvial deposit type, in which oxidation horizon was developed broadly under around 15 cm depth of surface soil. Phosphate balance (A-B) was closed to 0 in all treatments except cow manure treatment (CMC), in which it was $+30kg\;ha^{-1}$ and show the possibility of over accumulation of phosphate by continuously replicated application of cow manure compost. Potassium balance was negative value in all but straw recycling treatment (FSC). It means that potassium was continuously supplied from soil minerals, uptaken by plants or eluted out of soil. In conclusion, by substituting inorganic fertilizer for organic fertilizer or reducing application rate of chemical fertilizer through mixing organic fertilizer, it would be possible to achieve the same rice yield as in the recommendation treatment and to decrease nutrient leaching below half level in rice paddy soil.
The ultimate goal of this research is to develop a botanical biofiltration system that combines a green interior, biofiltering, and automatic irrigation to purify indoor air pollutants according to indoor space and the size of biofilter. This study was performed to compare the stability of air flow characteristics and removal efficiency (RE) of fine dust within a wall-typed (vertical) botanical biofilter depending on humidifying cycle and to investigate RE of volatile organic compounds (VOCs) by the biofilter. The biofilter used in this experiment was designed as an integral form of water metering pump, water tank, blower, humidifier, and multi-level planting space in order to be suitable for indoor space utilization. As a result, relative humidity, air temperature, and soil moisture content (SMC) within the biofilter showed stable values regardless of three different humidifying cycles operated by the metering pump. In particular, SMCs were consistently maintained in the range of 27.1-29.7% during all humidifying cycles; moreover, a humidifying cycle of operating for 15 min and pausing for 45 min showed the best horizontal linear regression (y = 0.0008x + 29.09) on SMC ($29.0{\pm}0.2%$) during 120 hour. REs for number of fine dust (PM10) and ultra-fine dust (PM2.5) particles passed through the biofilter were in the range of 82.7-89.7% and 65.4-73.0%, respectively. RE for weight of PM10 passed through the biofilter was in the range of 58.1-78.9%, depending on humidifying cycle. REs of xylene, ethyl benzene, total VOCs (TVOCs), and toluene passed through the biofilter were in the range of 71.3-75.5%, while REs of benzene and formaldehyde (HCHO) passed through the biofilter were 39.7% and 44.9%, respectively. Hence, it was confirmed that the wall-typed botanical biofilter suitable for indoor plants was very effective for indoor air purification.
This study was carried out to reveal the forest land effect on water purification in mountainous watersheds. Rainfall, throughfall, stemflow, soil and stream water were monitored by pH, electrical conductivity(EC), and dissolved oxygen(DO) in Daehan-Ri and Parkdal-Ri catchments. The results were summarized as follows; 1. Rainfall pH values of Parkdal-Ri and Daehan-Ri were 7.6 and 6.4, respectively. 2. Comparing stemflow and throughfall of Pinus densiflora with Pinus rigida, the pH values of Pinus densiflora were 4.32 and 4.22 and the pH of Pinus rigida were 3.34 and 4.81, respectively. The EC values of Pinus densiflora were $119.7{\mu}S/cm$ and $96.8{\mu}S/cm$ and EC of Pinus rigida were $230.0{\mu}S/cm$ and $82.0{\mu}S/cm$. 3. All pH values were decreased as the streamflow increased except long-term runoff in Daehan-Ri. The EC values also were increased as the streamflow increased, but EC of short-term runoff in Daehan-Ri was gradually decreased as the streamflow increased due to entrance of throughfall which has high EC values at the beginning of rainfall events. The DO concentrations of all experimental plots were elevated as the streamflow increased, because reaeration occurs at the surface of the stream as the increased discharge make turbulence. 4. pH of Stemflow and throughfall in Pinus densiflora were lower than in Quercus acutissima, but EC values were higher in Pinus densiflora. 5. Water purification was mostly influenced by forest soil in forest hydrological processes. 6. Stemflow and throughfall were more influenced by dry deposition and organic acid in crown and bark than those of wet deposition. During the stemflow and throughfall passed forest soil, these acidic stemflow and throughfall were neutralized, and stream water quality was neutral or slightly alkaline.
These objectives of this study were to compute heavy metal and accumulation carbon dioxide fixing quantity from urban green space(street trees and urban parks) in Cheong-ju city and Chungju-city and thus to estimate the effect of urban green space for improving the urban environment. The results are summarized below. 1. Results of the total accumulation of the carbon dioxide fixing quantity of street trees, Cheong-ju city and Chungju-city street tree was 1, 230,000kg-C, 1,270,000 kg-C, respectively. Total accumulation carbon dioxide fixing quantity of Balssan urban park had a 25,000kg-C in Cheong-ju city, Degami sports park had a 6,400kg-C in Chungju-city. 2. Results of heavy metal for street trees, fell in the order Zn > Cu > Cr > Ni >, the highest accumulated heavy metal was Zn, and the lowest was Ni. Total heavy metal concentration according to land-use area, was observed in order, for residental areas(157.26 mg/kg) > industrial areas(141.71 mg/kg) > commercial areas(118.55 mg/kg) > and greenspace areas(61.95 mg/kg) in Cheong-ju city. 3. Total heavy metal concentration for street trees fell in the order of commercial areas(84.48 mg/kg) > residental areas(83.70 mg/kg) > and greenspace(48.23 mg/kg) according to land-use area in Chungju-city. Comparatively, Cheong-ju city had more total heavy metal concentration than Chungju-city. 4. Heavy metal for soil that planted street trees was observed in order of Zn > Cu > Pb( > Ni > Cr > As > Cd), and Zn was highest, and Cd was lowest. Total heavy metal concentration for soil fell in the order commercial area(91.82mg/kg) > industrial area(85.96mg/kg) > residental area(67.55mg/kg) > greenspace(43.13mg/kg) according to land-use area in Cheong-ju city. 5. Heavy metal for soil that planted street trees was observed in order of Zn > Pb > Cu( > Ni > Cr > As > Cd, and Zn was highest. Total heavy metal concentration for soil fell in the order commercial area(87.66mg/kg) > greenspace(72.73mg/kg) $${\geq_-}$$ residental area(70.10mg/kg) in Chungju-city.

In order to determine the horizontal and vertical distributions of metals and prospect the recent metal pollution history in Nakdong Estuary, we took surface and core sediments. Maximum value of organic matter occurs at the upstream site located 4 km from Nakdong barrage, and the concentration of trace metals (Zn, Cu, and Pb etc.) decrease seaward in the estuary. The sedimentation rates, based on $^{210}$Pb$_{ex}$ and $^{137}$Cs activities, were 0.34 cm/yr in inside of barrage (core 1) and 0.25 cm/yr in Changrim (core 4). Sediment mixing layer does not exist in core 1, where anoxic condition is known to be prevail. The topmost sediment layer of core 4 (<3.5 cm) is severely mixed. At sites 1 and 4, concentrations of Cu slowly increased during the period of 1920-1970, rapidly increased during 1970-1990, and followed by slight decrease after 1990. Zn contents increased in early 1960s and peaked in 1993, and followed by decrease after 1990s. Pb has increased continuously since early 1970s. At the downstream of the barrage, Cu and Zn have increased in the topmost layer. The trend of increase of Cu is evident after 1950 (11 cm in sediment depth). Overall trend of heavy metal concentration clearly indicates the pollution has been increasing after the construction of the barrage.
Impervious covers(IC) are artificial structures, such as driveways, sidewalks, building's roofs, and parking lots, through which water cannot infiltrate into the soil. IC is an environmental concern because the pavement materials seal the soil surface, decreasing rainwater infiltration and natural groundwater recharge, and consequently disturb the hydrological cycle in a watershed. Increase of IC in a watershed can cause more frequent flooding, higher flood peaks, groundwater drawdown, dry river, and decline of water quality and ecosystem health. There has been an increased public interest in the institutional adoption of LID(Low Impact Development) and GI(Green Infrastructure) techniques to address the adverse impact of IC. The objectives of this study were to construct the modeling site for a samll urban watershed with the Storm Water Management Model(SWMM), and to evaluate the effect of various LID techniques on the control of rainfall runoff processes and non-point pollutant load. The model was calibrated and validated using the field data collected during two flood events on July 17 and August 11, 2009, respectively, and applied to a complex area, where is consist of apartments, school, roads, park, etc. The LID techniques applied to the impervious area were decentralized rainwater management measures such as pervious cover and green roof. The results showed that the increase of perviousness land cover through LID applications decreases the runoff volume and pollutants loading during flood events. In particular, applications of pervious pavement for parking lots and sidewalk, green roof, and their combinations reduced the total volume of runoff by 15~61 % and non-point pollutant loads by TSS 22~72 %, BOD 23~71 %, COD 22~71 %, TN 15~79 %, TP 9~64 % in the study site.
