Comparison of Cuticular Hydrocarbons of the Pine Sawyer (Monochamus saltuarius), Japanese Pine Sawyer (Monochamus alternatus) and Oak Longicorn Beetle (Moechotypa diphysis) (북방수염하늘소(Monochamus saltuarius), 솔수염하늘소(Monochamus alternatus), 털두꺼비하늘소(Moechotypa diphysis) 성충의 표피탄화수소 비교)

  • Cuticular hydrocarbons (CHCs) of the pine sawyer (Monochamus saltuarius), Japanese pine sawyer (M. alternatus) and oak longicorn beetle (Moechotypa diphysis) were analyzed by GC, GC-MS and compared. Monochamus beetles are typical vectors of pine wilt disease but Moechotypa diphysis, which belongs to the same family, is not. They possess different CHCs in carbon number: 23-25 in M. saltuarius, 25-32 in M. alternatus, and 23-29 in M. diphysis. In comparison to inter-species, these three species of adult beetles have different numbers and chains of constituents of CHCs. In comparison between male and female in intra-species, the quantities of CHCs show the difference but constituents are not. Major constituent of M. saltuarius were analyzed as n-pentacosane > n-nonacosane > n-heptacosane; those of M. alternatus were n-nonacosene > n-pentacosane > n-nonacosane; and those of M. diphysis were n-heptacosane > 13-methylheptacosane > 3-methylheptacosane. From the body surface, most saturated carbohydrates of 3 species beetles are composed of n-alkane (40.2 - 65.7%) and followed by olefines > monomethylalkanes that one or two double bonds in M. saltuarius and M. alternatus. Otherwise, M. diphysis have the difference in order of monomethylalkanes > olefins.

Differences in Ovipositional Frequency of Oak Longicorn Beetle (Moechotypa diphysis) by Oak Species Used for Lentinula edodes Cultivation Logs (표고재배용(栽培用) 참나무 원목(原木)의 수종별(樹種別) 털두꺼비하늘소의 산란빈도(産卵頻度))

  • We observed that the thick outer bark layer of Quercus variabilis hindered oak longicorn beetle (Moechotypa diphysis) from laying its eggs in inner bark (secondary phloem tissues). The outer bark thickness of Q. variabilis was average of 7.4mm, while those of Q. mongolica. Q. serrata and Q. acutissima were average of 1.1mm, 1.3mm and 2.0mm, respectively. Inner bark thickness was 4.8mm in Q. acutissima and 3.6-3.9mm in the other oak species. The outer shape of ovipositional holes on the bark by the longicorn beetle was $8-12mm{\times}6-8mm$ wide oval in Q. variabilis, whereas $5-9mm{\times}1-5mm$ narrow fusiform in the other oak species. Oak longicorn beetle drilled average of three ovipositional holes per a 1.2m-long log in a few Q. variabilis logs and its ovipositional rate was 15%. Compared to this, the longicorn beetle preferred Q. mongolica. All the 32 investigated logs of this oak species were drilled to have 20 ovipositional holes per a log and ovipositional rate was 56%. One third of the eggs laid already hatched in early June to damage the inner bark. It seems that oak longicorn beetle prefers oak species with smooth, thin and stable outer bark surface.

Isolation and Characterization of Xylanase-producing Paenibacillus sp. HY-8 from Moechotypa diphysis (털두꺼비하늘소 (Moechotypa diphysis)로부터 Xylanase를 생산하는 Paenibacillus sp. HY-8 균주의 분리 및 특성)

  • From the course of screening of useful xylanase producing microorganism from a phytophagous longicorn beetle, we isolated an extra-cellular xylanase producing strain, Paenibacillus sp. HY-8 from the intestine of Moechotypa diphysis adult. On the basis of morphological, biochemical and phylogenetic studies of the new isolate was identified as a Paenibacillus species. Production of xylanase in this strain was strongly induced by adding xylan to the growth medium and repressed by glucose or xylose. The highest xylanase production was attained in the M9 media containing 1% yeast extract and 0.5% birchwood xylan when cultured at $25^{\circ}C$ for 24 hrs. HY-8 producing xylanase showed superior hydrolytic activities against various plant source feedstuff than control xylanase produced by Tricoderma sp. at pH 6.0.

Control effects of benfuracarb and ${\lambda}$-cyhalothrin to oak longicorn beetle, Moechotypa diphysis, infesting the oak mushroom bed logs (표고원목해충인 털두꺼비하늘소에 대한 Benfuracarb와 ${\lambda}$-Cyhalothrin의 방제효과)

  • The toxicity of benfuracarb and ${\lambda}$-cyhalothrin on oak longicorn beetle, Moechotypa diphysis was investigated in terms of residual effect and control efficacy in the field. Mycelial growth inhibition of Lentinula edodes was also investigated in the laboratory. Benfuracarb and ${\lambda}$-cyhalothrin showed 100% of control values and over 90% residual activities of benfuracarb and ${\lambda}$-cyhalothrin were continued for 15 and 20 days after treatment, respectively. Benfuracarb and ${\lambda}$-cyhalothrin did not affect the mycelial growth of L. edodes Imhyup 1 variety. These results indicate that benfuracarb and ${\lambda}$-cyhalothrin might be used for the control of M. diphysis adults in the field.

  • As the pest of the Letinus edoeds, five insect species and six animal species are investigated injuring the fresh mushroom. Of these, insect and animals, Achorutes armatus, Scaphidium amurense and Philomycus confusa caused severe damage to mushroom fruitbody, and a greate deal of damage was done to mushroom lamella by mites (Rhizoglyphis sp., and Histiostoma sp.). As the pest of Lentinus edodes bed logs, two insect species from immature bed logs, eight insects and one isopod species from matured bed logs were investigated. Of those, Moechotypa diphysis was found to be the most injurious species from both immature and matured bed logs. As the pest of Pleurotus ostreatus, six insects and the two mite species were investigated from sporophore. Of those, Lycoriella sp. and Mycophilla sp. are considered to be very serious. while two insects and five mite species were investigated from the mushroom bed. Of those, one insect (Lycoriella sp.) and two mites (Histiostoma feroniarum and Throglyphus bankslogior) found to be abundant and caused serious damage.

Insecticidal Activity and Ovipositional Repellency of Monoterpenoids against Moechotypa diphysis Adults (Coleoptera: Cerambycidae) (털두꺼비하늘소 성충에 대한 monoterpenoids의 살충활성과 산란기피)

  • These studies were carried out to investigate the fumigant toxicity, the contact toxicity and the ovipositional repellency of 25 monoterpenoids against oak longicorn beetle adults, Moechotypa diphysis. Monoterpenoids with 100% of fumigant toxicity were 1, 8-cinehone, fenchone, pulegone and (equation omitted)-terpinene at 20 (equation omitted)/954 ml (air) concentration, pulegone and (equation omitted)-terpinen at 10 (equation omitted)/954 ml (air) concentration, pulegone at 5 (equation omitted)/954 ml (air) concentration. Most monoterpenoids showed low or no contact toxicity but only pulegone showed about 70% mortality. Some monoterpenoids which had repellency to female adults with Y-tube olfactometer were bornylacetate, carvacrol, 1, 8-cineole and menthol at 1 (equation omitted) dose, while citronellol showed attractant response. Carveol, geraniol and perillyl alcohol of 25 monoterpenoids showed ovipoistional repellency of 82.1%, 78.3%, 87.5%, respectively, at the concentration of 1,000 ppm in the laboratory condition. In the field, the result tested with three monoterpenoids indicated that geraniol was the most effective one at the concentration of 10,000ppm and 1,000 ppm, but residual effect wasn't found.

Fumigant Toxicity of 18 Essential oils and Their Major Compounds against Adult Oak Longicorn Beetle, Moechotypa diphysis (Coleoptera: Cerambycidae) (털두꺼비하늘소(Moechotypa diphysis)로에 대한 18종 식물정유와 주요성분의 훈증효과)

  • Fumigant toxicity of 18 plant essential oils were tested against the adults of oak longicorn beetle, Moechotypa diphysis. Among them, eucalyptus, rosemary and pennyroyal oils showed 100% mortality and sage oil showed 85% mortality at 10 $\mu\ell/\ell$ (air) dose. Eucalyptus and rosemary oils showed 100% mortality within 6 hr after treatment at 10 $\mu\ell/\ell$ (air) dose. GC and GC/MS analysis of the four essential oils and bioassay of their components revealed that 1,8-cineole (a major component of eucalyptus, rosemary and sage oils), thujone (a major component of sage oil) and pulegone (a major component of pennyroyal oil) showed higher adulticidal activity than others.

Fumigant Toxicity of Essentail Oils and Agricultural Materials against Moechotypa diphysis Pascoe (Coleoptera: Cerambycidae) (털두꺼비하늘소에 대한 식물정유와 농자재의 훈증독성)

  • This study was performed to investigate the contact toxicity, fumigant toxicity, and synergistic effect using agricultural materials (AM) when mixed with selected essential oils against oak longicorn beetle, Moechotypa diphysis. Four kinds of AM, namely, chungkiller, ungsalta, jinap, purunjunsa showed 100% contact toxicity at one day after treatment. Subsequently, four AM was tested the fumigant toxicity during 24 hours. At concentration of 1 mL, jinap and chungkiller showed 100% mortality at 3 hr after treatment, prunjunsa and ungsalta showed 100% mortality at 6 hr after treatment. When applicate to the small vinyl bag (15 L), all AM (50 mL/15 L) was showed 100% mortality. Jinap showed the mortality 100% when mixed pennyroyal oil with the ratio of 5+1. It was higher than single treatment.

Screening of Thrombin Inhibitor and its DPPH Radical Scavenging Activity from Wild Insects. (자생곤충으로부터 트롬빈 저해물질의 탐색 및 DPPH radical 소거활성)

  • The in vitro thrombin inhibitory activities of 304 crude extracts from 76 kinds of korean wild insects were evaluated. Measurement of thrombin time showed that the DMSO extracts of Acrida cinerea cinerea (Thunberg), Anax parthenope julius Brauer, Eurydema rugosa Motschulsky, and Stethophyma magister (Rehn) and the water extracts of Dolycoris baccarum Linne, Lixus divaricatus Motschulsky, Metrioptera bonneti, Moechotypa diphysis (Pascoe), Nicrophorus concolor sp., and Tomapoderus ruficollis (Fabricius) had strong thrombin inhibitory activity. No prominent changes of activated partial thromboplastin time by treatment of the selected extracts suggested direct inhibition of thrombin activity by the insect extracts. DPPH scavenging activity of selected extracts showed that the extract of A. cinerea cinerea (Thunberg), D. baccarum Linne, L. divaricatus Motschulsky and N. concolor sp. has good antioxidant activity as well as antithombin activity. Our results suggested that some of korean wild insects could be developed as a natural source of antithrombosis.
