This study was conducted to investigate the effect of 20-hydroxyecdysone on diapausing pupae of Hyphantna cunea. Treatment of 20-HE at the dosage of 5 $\mu$ g or higher/g live weight terminated diapause in about 50% of Hyphantna cunea pupae treated, and some malformed adult emerged at dosages of 10 or 12.5 $\mu$g of 20-hydroxyecdysone/g live wt. The maximum concentration of 20-hydroxyecdysone in the whole body of normally developing pupae was about 3.2 $\mu$g/g on the 6th day after pupation, and in that of diapausing pupae treated with 20 -HE about 4.1 $\mu$g/g on the 6th day after the treatment. But diapausing pupae showed a low level( 1.7 $\mu$g/g) of maximum 20-HE concentration. In diapausing pupae treated with 20- HE, glycongen content was lower than in normally developing pupae, but the changing pattern was similar to that observed in normally developing pupae. Glucose(and/or sorbitol) and trehalose content of 20-HE-injected pupae reached the maximum value on the 6th day after the injection, which were higher than those of normally developing pupae. The amount of soluble haemolymph proteins was lower but the total soluble protein content of haemolymph-rernoved whole body was higher in 20-HE-injected pupae than that in normally developing pupae.
Studies were carried out to investigate effects of temperature and photoperiod on diapause induction and of juvenile hormone III and 20-hydroxyecdysone treatment on diapausing adults of the alder leaf beetle, Agelastica coerulea Baly(Chrγsomelidae: Coleoptera). Its life cycle and ovarian development in adults were also observed. The beetle had one year life cycle with egg, larval, pupal and adult periods being 7-10, 19-21, 14-15 days and about 10 months, r respectively. All adults showed a diapause syndrome when the larvae were reared at $20^{\circ}C$ or $25^{\circ}C$ in combination of photoperiods of 16L/8D, 12L/12D, or 8L/16D. Their ovarioles did not s show any development of vitellogenesis before or during diapause and even when exposed at $15^{\circ}C$ after overwintering. When diapausing adults were treated with JH III they resumed feeding and laid several eggs and broke diapause condition temporally. But diapausing adults treated with 20-hydroxyecdysone did not show any response.
곤충애 있어서 호르몬의 작용기작과 그 이용에 관하여 요야하면 다음과 같다. 곤충호르몬에는중추신경계의 신경분비세포에서 합성, 분비되는 peptide의 neuropeptide hormone (PTTH, bombyxin, diapause hormone등)이 있고, 상피계의 내분비선에서 합성, 분비되는 sesquiterpene의 유약호르몬 ( juvenile hormone)과 steroid의 탈피호르몬 (ecdysone)이 있다. 곤충호르몬은 특정한 표족세포에 있는 수용체와 높은 특이성과 높은 친화성을 가지고 결합해서 세포의 작용을 조절한다. 일반적으로 peptide hormone은 표적세포의의 세포막을 통과할 수 없으므로 표적세포의 막표면에 있는 수용체와 결합하는 것에 의해 세포내 대사제를 활성화시킴으로서 peptide hormone의 특이적인 발현이되는 것으로 알려지고 있다. 한편, ecdysone과 같은 steriod hormone이나 juvenlie hormone은 표적세포의 세포막을 용이하게 통과할수 있으므로 세포내의 세포막을 용이하게 통과할 수 있으므로 세포내로 들어가 수용체와 결합해서 hormone-receptor comlpex는 핵내로 들어가 DNA의 특이적인 영역에 결합하므로서 이들 호르몬 특이적인 기능이 발현되는 것으로 알려져 있다. 17β-estradiol (E2) is a natural hormone secreted by ovary, and continuously discharged from household and livestock wastewater into aquatic environment. Due to its strong estrogenic activity, it has adverse effects on development and reproduction in crustacean as an endocrine disrupting chemical. Although ecdysteroid signaling pathway play a key role in development in crustacean, little information on transcriptional modulation of ecdysteroid-related genes in response to E2 is available in small crustacean. Here, we investigated the acute toxicity of E2 to obtain 24-h LCx values in the brackish water flea Diaphanosoma celebensis. Time-dependent expression patterns of seven ecdysteroid pathway - related genes (CYP314a1, EcRA, EcRB, USP, ERR, Vtg, VtgR) were further examined using quantitative real time reverse transcriptase polymerase chain reaction (qRT-PCR). As results, 24-h LC50 and LC10 values were 9.581 mg/l and 4.842 mg/l, respectively. The mRNA expression of CYP314a1, EcRA, USP, VtgR was significantly up-regulated at 12 or 24 h after exposure to E2. These findings indicate that E2 can affect their molting and reproduction by modulating the expression of ecdysteroid pathway - related in D. celebensis. This study will be useful for better understanding of molecular mode of action of endocrine disrupting chemicals on molting process in small crustacean.
Neuropeptides produced in neurosecretory cells are the largest group of insect hormones. They regulate various physiological functions, such as fat body homeostasis, feeding, digestion, excretion, circulation, reproduction, metamorphosis, and behavior throughout all life stages. The PRXamide peptide family (X, a variable amino acid) is a well-characterized neuropeptide component with a common amino acid sequence, PRXamide (NH2), at the C-terminal end conserved across Insecta. The PRXamide peptides are classified into three subfamilies, each having diverse biological roles in insects: (1) pyrokinin (PK) includes the pheromone biosynthesis activating neuropeptide (PBAN) and the diapause hormone (DH), (2) the capability (CAPA) peptides, and (3) the ecdysis-triggering hormone (ETH). PBAN as a member of PK subfamily was first identified to stimulate pheromone biosynthesis in moths three decades ago. Since then, PBAN peptides have been extensively studied by various research groups from a broad spectrum of arthropods. In this paper, we briefly review insect PBAN molecules with emphasis on gene structure and expression, signal transduction, physiological mechanism in sex pheromone biosynthesis, and application for pest management.
Studies were carried out to elucidate the efficacy of exogeneous 20-hydroxyecdysone treatment on terminating pupal diapause in the fall webworm. Hyphantria cunea Drury. And the difference was also investigated between normal adult development and post-diapause development after 20-hydroxyecdysone treatment. In the diapause termination rate of pupae treated with 20-hydroxyecdysone after storage for various periods at 16L:8D and $25\pm$$1^{\circ}C$, at the highest rate was observed from the group stored for the longest period and the lowest rate from those stored for 1.5 months. The time needed for adult emergence was inversely proportional to the chilling (at $0\pm$$1^{\circ}C$) period, and the longer its exposure period at low temperature, the higher its sensitivity to 20-hydroxyecdysone treatment. Pupal diapause duration was almost the same, regardless of storage period in the total darkness or at the photoperiod of 16L:8D, and also they successfully emerged to adult even without any experience at low temperature. The oxygen consumption rate in normally developing pupae showed nearly a typical U-form. But, that of diapausing pupae treated with 20-hydroxyecdysone slowly increased and jumped 14 days after the treatment. Pupal diapause began before formation of adult tissues, an a timing of adult tissue formation coincided with ascending timing of the metabolic rate in both normally developing pupae and diapausing pupae treated with 20-hydroxyecdysone. The diapausing pupae treated with 20-hydroxyecdysone was similar to normally developing pupae in band patterns of proteins from haemolymph or fat body.
Juvenile hormone (JH) is an insect hormone mediating immature metamorphosis and adult reproduction. It also mediates immune responses to suppress hemocyte behavior, which is, however, activated by ecdysteroid. This study investigated an effect of a commercial pyriproxyfen (a JH agonist) formulation on a cellular immune response of the diamondback moth, Plutella xylostella, and analysed its mixture with Bacillus thuringiensis (Bt) in insecticidal potency. The commercial pyriproxyfen formulation significantly suppressed hemocyte-spreading behavior at low doses as did in pyriproxyfen technical grade. When the commercial pyriproxyfen formulation was mixed with Bt, Bt toxicity was significantly increased against P. xylostella larvae in laboratory. The mixture effect was then confirmed in field cultivating cabbage infested with P. xylostella larvae. The mixture showed a significantly enhanced mortality and reduced effective lethal time, compared to only Bt treatment.
Because of their specificity to target insects and relatively low toxicity to non-target organisms, insect growth regulators (IGRs) have been regarded as attractive alternatives to chemical insecticides. Commercially available IGRs are classified into juvenile hormone agonists (JHAs), ecdysone agonists (EAs), and chitin synthesis inhibitors (CSIs) according to their mode of action. Recently, JH-mediated interaction of methoprene-tolerant (Met), which is JH receptor, and its binding partners have been replicated in vitro using yeast cells transformed with the Met and FISC/CYC genes of A. aegypti. Using this in vitro yeast two-hybrid β-galactosidase assay, juvenile hormone antagonists (JHANs) have been identified from various sources including chemical libraries, plants, and microorganisms. As juvenile hormone (JH) is an insect specific hormone and regulates development, reproduction, diapause and other physiological processes, JHANs fatally disrupt the endocrine signals, which result in abnormal development and larval death. These results suggested that JHANs could be efficiently applied as IGR insecticides with a broad insecticidal spectrum. This review discuses JH signaling pathway mediated by Met and future prospects of JHANs as environmentally benign IGR insecticides.
A crude crystal of phytoecdysone was obtained 355 mg per Kg dry radix of A. japonica, and it was identified to $\beta$-ecdysone by TLC. Feeding silkworms on mulberry leaves sprayed with the extract, mounting time shortened and pupation ratio improved. The feeding effects of the extract was differed according to the rearing seasons, spring and summer. In spring season, only mounting time shortened without any difference in pupation ratio from the control. In summer season, not only mounting time shortened, but also pupation ratio remarkably advanced.
