Effects of Carbon and Nitrogen Sources in the Production of Cellulolytic Enzymes by Trametes trogii (Trametes trogii에 의한 섬유소 분해효소의 생산에 있어서 탄소원과 질소원의 영향)

  • For the purpose of utilizingcellulosesresources by cellulolytic enzymes of Trametes trogii, its cultural conditions for the production of cellulolytic enzymes in synthetic media were investigated. The optimum conditions for the production of cellulase by T. trogii in synthetic media were $30{\sim}35^{\circ}C,\;pH\;4.0{\sim}6.0,\;and\;11{\sim}15$ day's cultivation. Among the carbon sources, carboxymethyl cellulose was good for the production of avicelase and ${\beta}-glucosidase$, but cellulose was good for the production of CMCase. The optimum concentration of Na-CMC was 3% for the production of all the three cellulolytic enzymes. As the nitrogen source, $0.03{\sim}0.04%$ N as ammonium tartrate was effective for the production of the cellulases.

A Study on the Recycling of Foodwaste Leachate as External Carbon Sources Using Microbubble (마이크로버블을 이용한 음폐수의 외부탄소원으로서의 재활용 가능성 연구)

  • The purpose of this research was to examine the possibility on the recycling of foodwaste leachate as external carbon sources using microbubble. The following operating conditions were selected: pressurizing tank 3 bar, circulation flow rate 3.65 LPM, and air flow rate 0.3 LPM with batch type. Microbubble contact time of 18 hours is optimal time to satisfy the recycling of foodwaste leachate as external carbon sources with batch type. HRT 18 hours came up to standard for external carbon sources, except for T-P concentration with continuous type. Coagulants need to be used for removal of dissolved phosphorus concentration by more than 88.5% of the total phosphorus concentration. The VFA was influenced by the organic decomposition rate and the concentration in the aerobic condition. It was considered that the VFA was needed for selection the optimal HRT or the addition of acid fermentation process in order to meet recycling standard of foodwaste leachate.

  • Basic informations for direct somatic embryo formation in Rehmannja glutinosa Lib. were obtained in 500ml erlenmyer flask. The ratio of ammonium to nitrate nitrogen of 825(mg/l) : 1900(mg/l) was proper condition for somatic embryo formation from stem and petiole explants and 3% sucrose was the most effective carbon source. Full strength MS medium with 2mg/l BA was better than LS medium for somatic embryogenesis. The initial pH 5.7 of medium(full strength MS with 2.0mg/l BA and 0.1mg/l NAA) was good for embryo production. Potassium ion was taken up rapidly within 2 weeks. while $Ca^{++}$ and $Mg^{++}$ ion contents were almost constant during culture period. Sucrose hydrolysis occurred throughout the culture, while glucose and fructose were absorbed simultaneously from the third week of culture.

Medium Optimization for the Protease Production by Bacillus licheniformis Isolated from Cheongkookjang (청국장에서 분리된 Bacillus licheniformis의 Protease 생산을 위한 배지 최적화)

  • Bacillus licheniformis fermenting soybean product with highest score in consumer acceptance had been isolated from homemade Cheongkookjang. In order to develop the medium composition, effects of ingredients including nitrogen sources, carbon sources, metal ions and phosphate were examined for protease production of the isolate. Potato starch increased the protease productivity, while glucose repressed it. Yeast extract was the most effective nitrogen source for enzyme production. The calcium was found to increase protease activity slightly while cell growth and enzyme production was completely inhibited by divalent ions such as $Zn^{2+}$, $Cu^{2+}$ and $Co^{2+}$. The maximum protease productivity was reached approximately 800 unit/mL in the optimized medium consisting of potato starch (1.5%), yeast extract (1.5%), $CaCl_2$(0.7%), $K_2HPO_4$(0.03%) and $KH_2PO_4$(0.03%). The protease activity of culture filtrate was gradually decreased after incubation for 28 h.

  • An anti-inflammatory agent, serratiopeptidase, was produced from the culture of the Serratia marcescens. The effects of carbon sources, nitrogen sources and inducers on the production were investigated. Citrate was found to be inhibitory in the production of serratiopeptidase. The enzyme was synthesized in the synthetic medium without inducers, albeit low level of synthesis. But the synthesis was increased by the addition of proteinaceous substrate and leucine. Induction of extracellular proteinase by its end-product was discovered, which is not common in the proteinase synthesis in the bacteria. By the glucose fed-batch culture, we found the possible catabolite repression on the production of serratiopeptidase.

  • 기후변화협약에 의해 탄소세가 부과될 경우 한국의 에너지 집약 산업에 어떠한 영향을 미치며 그에 대한 대응방안이 어떻게 마련되어야 하는지에 대한 관심이 증대되고 있다. 이에 본 연구에서는 탄소세가 부과됨으로 인하여 에너지 다소비 업종인 시멘트산업의 각 에너지원별 사용량에 미치는 파급효과를 시스템 다이내믹스 방법론에 의한 시뮬레이션 모델을 이용하여 분석하였다. 시뮬레이션 모델은 환경모듈, 경제모듈, 에너지모듈로 구성하였고 모델의 예측력 검증을 위해 의태분석을 수행하였다. BAU 시나리오와 탄소세 부과 시나리오를 설정하여 각 시나리오의 총수요의 예측치, 이산화탄소 배출량의 추이, 에너지원의 사용변화를 분석하였다. 본 연구는 향후 기후변화협약의 부담이 큰 에너지 집약산업에 있어서 에너지원의 사용 및 수요를 예측하고 이를 평가하기 위한 지침을 제시할 수 있을 것으로 기대된다.

  • Bacillus subtilis BD0310 isolated from tea leaves was used for the development of a biofungicide against Pestalotiopsis longiseta causing gray blight of tea plants. The optimum growth conditions were investigated for the mass cultivation of the microbial agent. The optimum temperature and cultivation time were determined as $12{\sim}24$ hours at $30^{\circ}C$ and the optimum initial pH was pH 7.0 in nutrient broth. Among the tested carbon sources of fructose, galactose, glucose, glycerol, inositol, lactose, maltose, sorbitol and starch, maltose and inositol were found to highly increase antifungal activity of the microbial agent against P. longiseta. Yeast extract and tryptone apparently increased antifungal activity of the microbial agent among the tested nitrogen sources of casein, tryptone, malt extract, yeast extract and $(NH_4)_2SO_4$. The results will make a contribution to mass production of the antagonistic bacterium Bacillus subtilis BD0310 for development of a microbial agent controlling gray blight of tea plants.

Effect of Complex Nitrogen Sources on Poly(3-Hydroxybutyric Acid) Synthesis by Recombinant Escherichia coli Using Sucrose as a Carbon Source (Sucrose를 탄소원으로 재조합 대장균에 의한 Poly(3-Hydroxybutyric Acid) 합성시 복합질소원이 미치는 영향)

  • When Escherichia coli W, which is able to utilize sucrose as a carbon source, harboring a high-copy-number plasmid (pSYL105) containing the Alcaligenes eultrophus polyhydroxyalkanoate(PHA) biosynthetic genes was cultured in a defined medium, the final poly(3-hydroxybutyric acid), PHB, concentration obtained was as low as $0.21g/\ell$. Ten different complex nitrogen sources were, therefore, examined for their ability to enhance PHB synthesis when supplemented to a defined medium. Addition of tryptone, casamino acids, casein hydrolysate, or soy bean hydrolysate enhanced PHB synthesis most significantly, resulting in more than 10 times higher PHB concentration compared with that obtained in a defined medium. Furthermore, PHB yield on sucrose was also increased by more than a 10 fold by the addition of these complex nitrogen sources, which suggested that PHB might be efficiently produced by the recombinant E. coli W(pSYL105) using sucrose as a carbon source.

  • The purpose of this study is to look for the optimal conditions of methane production. The conditions tested for methane production enhancement were temperature, pH, carbon source, nitrogen source, and inhibitor which can affects methane production. As a result, optimal conditions for methane production were 30$^{\circ}C$, neutral pH, methanol as a carbon source, NH$_4$Cl as a nitrogen source. 2-Bromoethanesulfonic acid was used as an inhibitor which can affects methane production. Existence in broth less than 10mM, inhibited methane production. Organic acid measurements revealed that formic acid exists in broth as majority.

  • The effects of nitrogen sources(sodium glutamate and peptone) on the production of xanthan gum were investigated. The fermentation using sodium glutamate as a nitrogen source is longer than that of peptone. In the initial nitrogen concentration of 0.4-1.0g/L, Bs was about 2.0 and ${\beta}$s was 1.2. The optimal yields were obtained when the carbon source/nitrogen concentration was 10-16. The fermentation time and product yields in the fermentation medium of mixed nitrogen source [sodium glutamate-N(0.75g/L)+peptone-N(0.25g/L)] were similar to those of peptone.
