Physicochemical Characteristics of Water or Alcohol Soluble Extracts from Laver, Porphyra yezoensis (김 추출물의 이화학적 특성)

  • To confirm the possibility of seaweed extracts for functional food, water or ethyl alcohol solubles were extracted from laver, Porphyra yezoensis and evaluated those food components such as proteins, carbohydrates, nucleic acids, taurine, pigments and browning extent. The amount of proteins, polysaccharides and nucleic acids extracted decreased with increasing ethyl alcohol concentration, which was maximal when water was used as extraction solvent. The extractability of proteins, polysaccharides and nucleic acids was different between the dried and the roasted laver. Taurine was extracted about 1% from the dried and the roasted laver in the range of o~7o% ethyl alcohol concentration. The amount of carotenoids extracted by 95% ethyl alcohol from the dried and the roasted laver were 146.6 and 138.4mg%, respectively, which was 66 ~ 80% of yield extracted by methanol/acetone(1/1) solvent. The browning value of 50 ~6o% ethyl alcohol extraction group from roasted laver was highest among water/ethyl alcohol extraction group. The extraction yield was maximum when laver was extracted with water, and the value was 26.3% for the dried laver and 27.5% for the roasted layer. Organoleptic characteristics from four kinds of extraction groups containing hot water extraction showed that extracts from the roasted laver were evaluated most eminent and thought to be applicable to various preparation of food.

Utilization of the Protein Hydrolysates of Skipjack Tuna Viscera (가다랭이 내장 단백질 가수분해물의 이용)

  • For the effective utilization of skipjack tuna viscera, a study was carried out to investigate the applicability of viscera protein hydrolysate (VPH) as a protein supplement in the processing of imitation sauce and bread. The optimum extraction and hydrolyzation conditions for the production of viscera protein concentrate (VPC) and viscera protein hydrolysate (VPH) were determined. Boiled viscera could be extracted by ethyl alcohol without significant deterioration as a raw material for the further processing. High quality of VPH could be obtained by hydrolysis with 1% pepsin under its optimum condition $(pH\;1.65,\;37^{\circ}C)$. The solubilities of VPC and VPH were 40% and 90%, respectively, and the essential amino acid contents in two products were 48.7% and 63.2%, respectively. Especially, the content of taurine, a physiologically important amino acid, was 9.4% in VPH. In experimental preparations of imitation sauce and bread, panel test showed that the supplementation of 10% of VPH in imitation sauce and $3{\sim}5%$ of VPH in bread was well accepted in sensory characteristics such as color, flavor, taste and texture.

Effect of Dietary Protein and Taurine on Cysteine Catabolism in Cat Liver (식이내의 단백질과 타우린 함량이 Cysteine 대사에 미치는 영향)

  • Activieties of hepatic cysteine desulfhydration was assessed in cats fed one of the following diets for 5 weeks : 20% protein, 0% taurine diet(LPOT) ; 20% protein, 0.15% taurine diet (LPNT) ; 60% protein, 0% taurine diet(HPOT) ; and 60% protein, 0.15% taurine diet(HPNT). Cats fed LPOT and HPOT had been maintained on a taurine-free diet for 6 weeks prior to the experiment in order to deplete body taurine. Activities of cysteine desulfhydration were determined by measuring the production of H235S from 35S-cysteine in the presence and absence of $\alpha$-ketoglutarate ($\alpha$-KG) in the incubation medium. The direct pathway via cysteine desulfhydrase appears to account for the major route of cysteine desulfhydration in the cat liver since the values obtained in the absence of $\alpha$-KG were between 81 and 88% of those obtained in the presence of $\alpha$-KG. Mean$\pm$SEM of the hepatic total desulfhydration activities(umol H2S.min-1.kg body wt-1)in cats fed LPOT, LPNT, HPOT and HPNT were 117$\pm$6, 135$\pm$10, 137$\pm$10, and 190$\pm$9, respectively. The capacity of hepatic cysteine desulfhydration (UA/kg body wt) was positively cerrelated not only with the dietary concentration of taurine but also with the concentration of protein.

The Effects of Supplementation of Ascidian Tunic Shell into Laying Hen Diet on Egg Quality (산란계 사료에서 우렁쉥이껍질 첨가가 계란 품질에 미치는 영향)

  • This study was conducted to evaluate the feeding value of ascidian tunic shell the effects of its dietary supplementation on laying performance, egg-yolk pigmentation, egg-shell strength and egg taurine content. A total of 168 brown layers at the age of 29wks in commercial cage were fed for 4 wks with 7 different diets containing ascidian tunic shel1(AST) at varying levels of 0$\sim$5% Dm or 0% AST with 100ppm carophyll red. No differences were found in egg production and weight among the treatments indicating that ascidian tunic shell did not adversely affect the laying performances. Adding the ascidian tunic shell to the diets increased egg-yolk pigmentation compared to the control and resulted in simillar or better effect on egg-yolk pigmentation compared to 100ppm carophyll red. The data suggest that ascidian tunic shell may be used as feed ingredients in layer diet enrichment of egg-yolk pigmentation in the place of carophyll red(chemical pigment). Specific gravity and breaking strength of egg shell were significantly increased by the adding ascidian tunic shell to the diet, suggesting that ascidian tunic shell may be used as feed ingredients for increasing egg shell strength. Also taurine content of egg was significantly increased with increasing supplementation of ascidian tunic shell to the diet(p<0.05). Therefore, ascidian tunic shell may be used as feed ingredients in laying hen diet to improve egg quality such as egg-yolk pigmentation, egg-shell strength and egg taurine enrichment.

  • Alzheimer's disease (AD), an irreversible degenerative disorder, is associated with accumulation and aggregation of amyloid-β peptides, hyperphosphorylated tau proteins, and high level of metal ions in the brain. Up to date, there is no effective therapeutic agent to stop the progress of the disease and thus early and accurate diagnosis of AD has gained increasing attention in recent years. Among several diagnostic methods, an optical imaging using fluorescent probes is one of the most promising tools to visualize AD biomarkers. In this review, we will introduce fluorescent probes that can be applied to in vivo brain imaging of AD models and also their structure. It is expected that the present review will provide useful information to many scientists in the related research fields.

Analysis of Relative Risk of Stroke by Nutrient Intake Levels - Case-Control Study in Daegu.Gyeongbuk Area, Korea - (영양소 섭취 수준에 따른 뇌졸중 위험도 분석 - 대구.경북지역 환자-대조군 연구 -)

  • The present study was performed to evaluate energy and nutrients intakes of stroke patients in Daegu Gyeongbuk region, and to analyze the relative risk of stroke related to the intake levels of energy and nutrients. The case subjects (n=100) were selected from newly diagnosed stroke patients at Kyungpook National University Hospital. The control subjects (n=150) were selected from community residents who did not have stroke history and were sex and age-matched with the case subjects. The survey was conducted by individual interviews by trained dietitians using semiquantitative food frequency questionnaires. The odds ratios were calculated by using unconditional logistic regression. In male subjects, patient group had significantly higher consumption than the control group in mean daily intakes of energy, all energy yielding nutrients, vitamin $B_1$, $B_6$, niacin, folic acid, vitamin E, phosphorus, potassium, zinc and dietary fiber, and also in the ratio of energy intake from protein and fat. In women subjects, the patient group consumed significantly lower intakes than the control group in fat, vitamin C, folic acid, vitamin E, iron, but vice versa in carbohydrate energy ratio. As for men, the increased intakes of energy, protein, carbohydrate vitamin $B_1$, E and niacin, zinc, total fatty acids, monoand poly-unsaturated, n-6 fatty acids significantly raised the relative risk of stroke. As for women, the increased intakes of fat, vitamin A, $B_2$, $B_6$, niacin, vitamin C and E, iron, sodium, potassium, selenium, mono-, poly-unsaturated, n-6 fatty acids, cholesterol, taurine and dietary fiber significantly lowered the relative risk of stroke. The results of the study demonstrated that the effect of several nutrient intake levels, such as niacin, vitamin E and fatty acids, on the relative risk of stroke was inconsistent between sex. The reason for this sex difference needs to be elucidated in a larger scale study.

Geographical Variation of the Oriental Fruit Fly, Bactrocera dorsalis, Occurring in Taiwan (오리엔탈과실파리 유전변이 - 대만 지역 집단변이)

  • This study analyzed genetic variation of the Oriental fruit fly (OFF), Bactrocera dorsalis, which is designated to be a quarantine insect pest in Korea. OFF samples endemic to Taiwan were collected at three different locations (Taipei, Taichung, and Kaohsiung) for three days from July 30 to August 1 in 2018 and assessed in their age and mitochondrial DNA sequence variations. In these places, 1,085 OFF males were collected using methyl eugenol lure while 30 males of Zeugodacus cucurbitae and one male of Bactrocera tau were collected using Cuelure. A protein diet lure attracted 6 flies including one OFF and 5 flies of Z. cucurbitae. Male heads of OFF contained pterin, which increased in contents with age from 32 to $59{\mu}g/head$. There was a local variation in pterin amounts in OFF heads, in which Kaohsiung population had lower amounts of pterin than Taipei and Taichung populations. Genetic distance among these three populations were measured by random amplified polymorphic DNA and showed that Taipei population was separated from Taichung/Kaohsiung cluster. Genetic variation was also analyzed in sequence variations in cytochrome oxidase I (CO-I) and NADH dehydrogenase I (ND-I). There was 7.8% variation in CO-I sequence (360 residues) and 6.6% variation in ND-I sequence (213 residues). These polymorphic sites are proposed to be used to develop SNP (single nucleotide polymorphism) markers characteristic to Taiwan OFF populations.
