  • The control system of a direct drive servo valve is a nonlinear system, and the flow force effect on the spool motion is significant and dependent on the load pressure. To satisfy the control system design requirements, the optimal parameters of the lead-lag controller and the derivative feedback controller are searched for using a genetic algorithm and a complex constrained direct search type method. The obtained controller parameters successfully perform their role to satisfy the control system design requirements.

  • This test was performed to evaluate the skin irritation and sensitization of Creocomplex, disinfectant, Containing 10% 4-chloro-m-cresol, 10% didecyl dimethyl ammonium chloride, and 10% glutaraldehyde. In primary skin irritation test, rabbits were dermally treated with Creocomplex for 24 hrs. The disinfectant did not induce any adverse reactions such as erythema and edema on intact skin sites, but on abraded skin sites, some rabbits showed very slight erythema and edema 24hr after topical application. So, the disinfectant was classified as a practically non-irrifating material based on the score 0.13 of primary irritation index. In the skin sensitization test, guinea pigs were sensitized with intradermal injection of 0.1ml Creocomplex for 24 hr. After 1 week, Creocomplex was treated on the site of injection, and challenged 2 weeks later. Creocomplex did not induce any allergic reactions. Therefore, 10% Creocomplex was graded as a weak material from 0 in both sensitization score· and rate. From results of the present study, it is suggested that 10% Creocomplex does not cause contact irritation and sensitization.

  • Synthetic gene carriers such as poly-cationic polymers easily form complexes with plasmid DNA which contains negative charge. Chitosan is a polysaccharide that demonstrates much potential as a gene delivery system. The ability of depolymerized chitosan to condense DNA was determined using electrophoresis. Dynamic laser scattering and scanning electron microscopy were used to examine the size and the morphology of the chitosan/DNA complex. Parameters such as chitosan molecular weight and charge density influenced the complex size and the DNA amount condensed with chitosan. The cell viabilities in the presence of chitosan ranged between 84-108% of the control in all experiments. Gene expression efficacy using chitosan/DNA complex was enhanced in 293 cells relative to that using naked DNA, although it was lower than that using lipofecamine. Transfection efficacy using low molecular weight chitosan (Mw=8,517) was higher than those of the control and the other chitosan (MW=4,078). The low molecular weight chitosan (MW=8,517) with a high charge density (18.32 mV) fulfilled the requirements for a suitable model gene delivery system with respect to the condensing ability of DNA, complex formation, and transfection efficacy.
