Saikosaponin Content in Adventitious Root Formed from Callus of Bupleurum falcatum L. (시호(Bupleurum falcatum L)의 캘러스로부터 형성된 부정근의 Saikosaponin 함량)

  • This study was carried out to analyse saikosaponin contents in adventitious root formed from callus of Bupleurum falcatum L. The induction of adventitious root from callus of 4 weeks of culture was most favorable to MS medium supplemented with 0.1 mg/L 2,4-D. The adventitious root formation ratio per callus was highest when the size of callus was 900 to 1,000 $\mu$M The content of saikosaponin a and d was 2089$\pm$124 $\mu$g and 4778 $\pm$ 214 $\mu$g at 150 days of culture respectively on the basis of g dry root wt, whereas that of saikosaponin c was 3492 $\pm$ 123 $\mu$g at 60 days of culture.

  • Plant regeneration from the stem tissue of Orostachys japonicus A. Beiger was investigated. The calli derived from shoot apex when apex when cultured on Murashige and Skoog (MS) medium supplemented with 4mg/L 2,4-dichlorophenoxyacetic acid (2,4-D)and 2 mg/L benzyl aminopurine (BAP). The calli were developed into shoot to MS medium with 0.5mg/L NAA and 2mg/L and into root with 1mg/L kinetin. The reddish pigment which might be essential for the rootregeneration was observed in the tip of regenerated root.

  • 억새(Miscanthus spp.)는 우리나라 등 동아시아가 원산이며 바이오매스량이 많아 바이오에너지 원료작물로서 잠재성이 크기 때문에 2세대 바이오에탄올 생산 원료로 주목을 받고 있고, 축산깔개 및 토양개량제 등으로도 이용되고 있다. 미국과 유럽 등에서는 생태계 교란 방지를 위해 4배체 물억새(M. sacchariflorus)와 2배체 참억새(M. sinensis)의 종간 교잡 이질 3배체인 불임성 억새(M. x giganteous)을 주로 재배하고 있으나, 단일 유전형의 품종으로 병해충과 자연재해에 취약하여 다양한 억새 품종의 개발이 시급한 실정이다. 본 연구는 억새를 재료로 하여 반수체 및 배수체 확보를 통한 배수성 별 특성 평가와 함께 기내배양을 통해 탈분화 및 재분화 시스템을 구축하여 억새의 육종 효율을 높이기 위해서 실시하였다. 억새 종자로부터 캘러스의 유도는 MS배지와 N6배지에 1mg/L 2.4-D를 첨가하였을 때 비배발생 캘러스(nonembryogenic callus)가 유도되었고, N6배지에 3~5 mg/L 2,4-D를 첨가하였을 때 배발생 캘러스(embryogenic callus)가 발생하였다. MS배지보다는 N6배지에서 캘러스 유도율이 높았으며, 식물생장조절제로 2,4-D와 BA 조합처리 보다 2,4-D 단용 처리하였을 때 캘러스 유도율이 더 높았다. 억새 종에 따른 캘러스 유도율은 물억새가 25.2~49.3%, 참억새는 30.3~52.0%였고 거대억새는 62.6~81.1%로 나타났다. 억새 신초 및 줄기로부터의 캘러스 유도율은 물억새가 4.4~17.2%, 참억새는 1.8~7.7%, 거대억새는 15.3~19.9%로 나타나 종자에 비해 매우 낮았다. 미성숙화기로부터의 캘러스 유도는 억새 종에 따른 차이가 없었으며, 3mg/L 2,4-D 첨가 배지에서 캘러스 유도율이 비교적 낮았고(60~80%), 1mg/L와 5mg/L의 2,4-D가 첨가된 배지에서 캘러스 유도율이 높게(90~95%) 나타났다.

Superoxide Dismutase and Peroxidase Activity of Transformed Callus in Tomato (형질전환된 토마토 캘러스의 Superoxide Dismutase와 Peroxidase 활성)

  • This study was carried out to investigate activity difference in the superoxide dismutase (SOD) and peroxidase (POD) of tomato callus transformed with Agrobacterium containing the GUS gene. Than those of other two tomato cultivars, the hypocotyl explant of JA101 was shown to have higher POD and SOD specific activity of 23 unit/mg protein and 2,156 unit/mg protein, respectively. Relatively high frequency of callus formation was obtained from the hypocotyl explant on MS medium containing 1 mg/L 2,4-D for 30 days and its POD(47 unit/mg protein) and SOD (95,786 unit/mg protein) specific activities were higher than other 2,4-D concentration. The hypocotyl explant and callus cocultivated with Agrobacterium for 72 hours were transferred to MS medium supplemented with 1 mg/L 2,4-D, 30 mg/L kanamycin, 30 g/L sucrose and 4 g/L Gelrite. The hypocotyl explants transferred to the medium formed callus with 45.5% effeciency after 8 weeks. The transformation efficiency confirmed by GUS assay was 21.6%. POD specific activity of the transformed callus (54 unit/mg protein) were somewhat lower than the non-transformed callus (64 unit/mgg protein) and SOD specific activity of the transformed callus (30,300 unit/mg protein) were also lower than the non-transformed callus (37,077 unit/mg protein). However there was no significant difference in POD and SOD isozyme patterns between the transformed and the non-transformed calluses. From these results, it revealed that there was no difference of antioxidant enzyme activities between the transformed callus and the non-transformed callus in tomato.

  • Using mature seed-derived callus, optimal conditions for efficient callus growth and plant regeneration, and regeneration efficiency by callus type were investigated in zoysiagrass (Zoysia japonica steud.). Callus induction was highest when the seeds were cultured on MS medium containing 2 mg/L 2,4-D, 0.2 mg/L BAP, 4 mg/L thiamine-HCl and 100 mg/L $\alpha$-ketoglutaric acid. Callus growth was highest when callus were cultured on MS medium containing 0.5 mg/L 2,4-D, 0.05 mg/L BAP, 4 mg/L thiamine-HCl and 100 mg/L $\alpha$-ketoglutaric acid. Plant regeneration was highest when callus was transferred on MS medium containing 3% maltose and 1 mg/L BAP, or 1 mg/L thidiazuron (TDZ). The combinations and concentrations of 2,4-D and BAP were shown to be critical factors for both the frequency and the type of callus. And four morphologically distinct types of callus were induced from the 2,4-D and BAP treatment. Type I,II and III calli produced shoots upon subculture, while the watery callus, type IV produced roots without shoots. Of four types of callus, type I exhibited the maximum frequency (82%) of shoot regeneration and the minimum frequency (4%) of albinism.

  • To investigate the cucumber regeneration from embryogenic calli, shoot tips of aseptically-grown cucumber seedlings were used as explants for establishing tissue cultures. Growth and differentiation of callus were studied by using Murashige and Skoog's (MS) medium containing 0.5 to 2 mg/L 2,4-D. Plantlets were induced from shoot tip culture on the plant growth regulators-free MS medium. Non-embryogenic calli and viscous calli were induced on the medium supplemented with 0.5 to 2 mg/L 2,4-D, but embryogenic callus was not induced on the same medium. Segments (ca. 5∼10 mm) of aseptically-grown hypocotyl from five to seven days old seedlings after germination were placed on MS medium supplemented with 1 mg/L 2,4-D for 50 days. Embryogenic calli and embryoids were induced only from the seedlings grown in dark condition, and hypocotyl was placed on the media explanted in light condition. Foully-five point one percent of white fragile calli and 0.6% yellowish compact calli formed roots. Yellowish callus lines were investigated to have a considerably higher concentration of crude proteins than white callus lines. Plantlets derived from embryogenic calli or embryoids have been transferred to pots containing sterile vermiculite and perlite. Normal fruits were harvested from nutrient culture on aggregated hydroponics in the F-clean house.

A simple mid-term preservation method (SMPM) of plant callus under low temperature conditions (저온 보존을 이용한 간편 중기 식물캘러스 저장법)

  • The repeated monthly or weekly subculture of plant callus is labor intensive and increases the risk of somaclonal variation from the parental callus line. The most effective method for preserving plant callus is cryopreservation, which involves storage in liquid nitrogen. However, this method cannot be applied to the callus of different plant species in the same manner, so it is difficult to develop a standardized cryopreservation method. In addition, the survival rate of the frozen callus after thawing and the regeneration rate after survival are uncertain. Therefore, it is necessary to develop a method to extend the subculture interval of plant callus in an active state. In this study, active plant calli of various species without freezing was incubated at 15℃ for 4 to 12 weeks without subculture. After 12 weeks, 8 lines of plant callus grew less than 2-fold when cultured at 25℃, but at least 2 times as much when cultured at 15℃. Moreover, total antioxidant activity did not differ significantly between plant callus recovered at 25℃ after culturing at 15℃ or at 25℃. These results show that the subculture interval can be extended at a temperature of 15℃ without need for modified medium composition or additional processes. In addition, positive results in all calli of several plant species are expected to reduce labor as well as somaclonal variation by increasing the subculture.

  • Perennial ryegrass의 종자유래 캘러스 및 조직 절편체로부터 식물체 재분화에 적합한 조건을 조사하였다. 종자유래 캘러스로부터 식물체 재분화 조건을 확립하기 위하여, 품종, 배지 및 생장조절물질의 조성 등에 따른 재분화율을 조사하였다. Reveille과 Modus의 품종간 비교에서 종자로부터 형성된 캘러스 생체중은 Reveille이 2,4-D 10mg/$\ell$에서 가장 좋았으며, MS배지가 SH, B5배지보다 더 좋았다.(중략)

  • Plant regeneration system from floral stem of Mymphoides indica via somatic embryogenesis was established. After four weeks of culture onto 1/2MS medium containing 2,4-D, pale-yellow globular structures and calluses were formed on the cut surface of floral stem explants. Upon transfer to 1/2MS basal medium, pale-yellow globular structures were developed into somatic embryos and normal plantlets. These results indicated that pale-yellow globular structures and calluses from floral stem were globular embryos and embryogenic calluses, respectively. The frequency of embryogenic callus formation from floral stem was reached to nearly 100% when floral stem was cultured onto 1/2Ms medium supplemented with low concentration of 2,4-D (0.1 to 0.3 mg/L). However, the higher concentration of 2,4-D resulted in decrease of the frequency of embryogenic callus formation. In this study, low concentration of 2,4-D had a stimulative role in embryogenic callus formation, whereas BA showed inhibitory role in callus formation. In comparison to floral stem, leaf explants showed low frequency of embryogenic callus formation. The highest frequency of embryogenic callus formation from leaf explants was 9.5% when leaf explants were cultured onto 1/2MS medium supplemented with 0.3 mg/L of 2,4-D. The plant regeneration system of Nymphoides indica established in this study, might be applied to mass proliferation, conservation of genetic resources and genetic transformation for molecular breeding.

Evaluation of Alcea rosea L. Callus Extract as a Natural Cosmetic Ingredient (접시꽃 캘러스 추출물의 천연화장품 원료로서의 효능 평가)

  • In this study, we investigated the biological effects of Alcea rosea L. callus extract for the development of natural cosmetics ingredients. The antioxidant activities of A. rosea L. callus extract was measured through DPPH, ABTS and FRAP assay. As a result, A. rosea L. callus extract were found to have a strong antioxidant ability in a dose dependent manner. In addition, A. rosea L. callus effectively reduced the intracellular oxidative stress induced by AAPH at a concentration of 10 mg/mL. In a tyrosinase activity assay, we found that A. rosea L. callus extract reduced tyrosinase activity by 51% at 10 mg/mL. Based on these results, A. rosea L. callus extract is considered as a promising natural ingredients for cosmetics with antioxidant and whitening functions.
