Expression of PTEN, Tumor Suppressor Protein, in Aggressive Fibromastosis (공격성 섬유종증에서 PTEN 종양 억제 인자의 발현)

  • Aggressive fibromatosis is a rare soft tissue tumor with locally invasive and infiltrative characteristics. The mechanism of this invasive nature was not reported until now. Mutations or reduction of PTEN, tumor suppressor gene, in cancer tissues, have been found to be associated with invasiveness and metastatic properties of cancer cells. To know the pattern of expression of PTEN in aggressive fibromatosis, we analysed the expression of PTEN with immunohistochemical stain and immunoblotting. PTEN was homogeneously expressed in the normal musculoaponeurotic tissues, but absent or very faint in tissues of patients with aggressive fibromatosis as evidenced by western blot analysis and immunohistochemical examinations. Although the meaning of decreased PTEN expression in aggressive fibromatosis is not certain, it might be involved in the growth of the aggressive fibromatosis, and associated with phenotype of aggressive fibromatosis.

Penetrating Performance of Wood-preservatives by Ultrasonic Steeping (초음파침지처리에 의한 목재방부제의 주입성)

  • In this study, we attempted to apply ultrasonic treatment for the preservative treatment of two softwood species, Korean pine and Japanese red pines to evaluate its effectiveness. Wood samples were submerged in oilborne preservative, copper naphthenate (NCU) and waterborne preservative, alkyl ammonium compound (AAC) and then treated with ultrasonic wave of 40 kHz, 400 W. After the treatment, the retention and penetration depth of these preservatives in the specimens were measured as a function of treatment time. Both the retention and penetration depth were continuously increased with increasing the treatment time up to 120 hours, where the retention reached about 95% by the application of AAC and the penetrating depth of 95% and 83% by the application of NCU, respectively for both species. The results of electronic microscopic observation showed that the improved retention capacity could be attributed to air deflation, wood extractive deflation working of ultrasonic wavelength, and destruction of wood pits which served as the pathway of preservatives. The results suggested that the use of ultrasonic treatment could be applied to thick wood veneers for the production of laminated wood products.

Effect of Cyanidin on Cell Motility and Invasion in MDA-MB-231 Human Breast Cancer Cells (Anthocyanin계 성분인 Cyanidin이 인체 유방암세포 MDA-MB-231의 이동성과 침윤성에 미치는 영향)

  • Anthocyanidins, the aglycones of anthocyanins, are natural colorants belonging to the flavonoid family. Cyanidin is one of the anthocyanidins, used for their antioxidant properties. Furthermore, previous studies have shown anthocyanidin-rich material extracts or aglycone form inhibit growth and induce apoptosis of cancer cells. But, Tumor metastasis is the most important cause of cancer death, and various treatment strategies have targeted on preventing the occurrence of metastasis. This study investigated the effects of cyanidin on metastasis processes, including motility, invasion and activity of MMP-2 and MMP-9 in MDA-MB-231 human breast cancer cell lines. We cultured MDA-MB-231 cells in presence of various concentrations 0, 5, 10 and 20 ${\mu}M$ of cyanidin. The cell motility was significantly decreased dosedependently in cells treated with cyanidin (p < 0.05) and cyanidin treatment caused the significant suppression of the invasion (p < 0.05). MMP-2 and MMP-9 activities, and MMP-9 mRNA express were not affected by anthocyanin treatment. In conclusion, cyanidin inhibits cell motility, invasion in MDA-MB-231 human breast cancer cell lines.

Wetting Agent Performance Evaluation Using Scale Model (축소모형 실험을 통한 침윤소화약제 소화성능 평가)

  • In this study, the extinguishing performance evaluation of wetting agent for wood crib was conducted by using a scale model equipment that we designed. To confirm the optimal conditions of the experiment, a test was changed amount of fire extinguishing water and the number of timber. As a result, the discrimination of the fire extinguishing performance was seen only when 20 pieces of wood and the extinguishing water of 100 mL were used. After evaluating the extinguishing performance of domestic and foreign wetting extinguishing agents under these conditions, a reignition was occurred in only when we used water. In addition, the discrimination of extinguishing performance was seen through the temperature distribution according to the time of watering. Based on the results of this experiment, this study is expected to be able to use as a basis on presenting a method of optimized performance evaluation of wetting extinguishing agent.

  • We described herein the first case of primary $Sj{\ddot{o}}gren^{\prime}s$ syndrome in Korea which presented with multiple cysts caused by only peribronchiolar lymphocytic infiltration, a rare pulmonary manifestation in $Sj{\ddot{o}}gren^{\prime}s$ syndrome, and was confirmed by surgical lung biopsy. A brief review of the literature has been included.

  • Effects of microstructure and indentation stress on fracture behavior of glass-infiltrated alumina composite for dental restorative application were investigated by the Hertzian and Vickers indentation method. Indentation stress-strain curve of glass-infiltrated alumina has showed the quasi-plastic behavior - deviation from linearity at high stress and the classical Hertzian cone crack, which could be confirmed the subsurface damage micrographs using bonded-interface specimen technique. The indentation stress-strain curves for the starting preforms are strongly dependent on porosity and microstructure of the preforms. On the other hand, the curves for the infiltrated composites are relatively insensitive to these factors. The failure of composite is originated at quasi-plastic deformation region. Damage and fracture behavior due to Hertzian stress field is theoretically examined, so that the indentation stress field plays a great role in material degradation. After Hertzian indentation annealing processing changes fracture behavior of alumina composite, so that stress field in material is healed through annealing.

Fabrication and Properties of Reaction Bonded SiC Hot Gas Filter Using Si Melt Infiltration Method (용융 Si 침윤방법에 의한 반응소결 탄화규소 고온가스 필터의 제조 및 특성)

  • Novel fabrication technique was developed for high strength Reaction-Bonded SiC (RBSC) hot gas filter for use in IGCC (Integrated Gasification Combined Cycle) system. The room and high temperature fracture strengths for Si-melt infiltrated reaction-bonded SiC were 50-123, and 60-66 MPa, respectively. The average pore size was 60-70 $\mu\textrm{m}$ and the porosity was about 34 vol%. RBSC infiltrated with molten silicon showed improved fracture strength at high temperature, as compared to that of clay-bonded SiC, due to SiC/Si phase present within SiC phase. The thickness for SiC/Si phase was increased with increasing powder particle size of SiC from 10 to 34 $\mu\textrm{m}$. Pressure drop with dust particles showed similar response as compared to that for Schumacher type 20 filter. The filter fabricated in the present study showed good performance in that the filtered powder size was reduced drastically to below 1 $\mu\textrm{m}$ within 4 min.

In Vitro imaging of MRI and Ultrasound for Colorectal Carcinoma (직결장암 조직의 자기공명영상과 초음파 소견에 대한 비교 연구)

  • Purpose : To evaluate and compare the accuracy of magnetic resonance imaging (MRI) and ultrasound (US) for detection and estimation of invasion depth of colorectal carcinoma (CRC) by correlation with histopathologic findings in vitro, and to find out the best MR pulse sequence for accurate delineation of tumor from surrounding normal tissue. Materials and Methods: Resected specimens of CRC from 45 patients were examined about tumor detectability and invasion depth of US using high frequency (5-17 MHz) linear transducer in a tube filled with normal saline and MRI in a 8-channel quadrate head coil. The institutional review board approved this study and informed consent was waived. MRI with seven pulse sequences of in- and out-of-phases gradient echo T1 weighted images, fast spin echo T2 weighted image and its fat suppression image, fast imaging employing steady-state acquisition (FIESTA) and its fat suppression image, and diffusion weighted image (DWI) were performed. In each case, both imaging findings of MRI and US were evaluated independently for detection and estimation of invasion depth of tumor by consensus of two radiologists and were compared about diagnostic accuracy according to the histopathologic findings as reference standard. Seven MR pulse sequences were evaluated on the point of accurate delineation of tumor from surrounding normal tissue in each specimen. Results: In specimens of CRC, both imaging modalities of MRI (91.1%) and US (86.7%) showed relatively high diagnostic accuracy to detect tumor and evaluate invasion depth of tumor. In early CRC, diagnostic accuracy of US was 87.5% and that of MRI was 75.0%. There was no statistically significant difference between two imaging modalities (p > 0.05). The best pulse sequence among seven MR sequences for accurate delineation of tumor from surrounding normal tissue in each specimen of CRC was fast spin echo T2 weighted image. Conclusion: MRI and US show relatively high diagnostic accuracy to detect tumor and evaluate invasion depth of resected specimen of CRC. The most excellent pulse sequence of MRI for accurate delineation of tumor from surrounding normal tissue in CRC is fast spin echo T2 weighted image.

Bronchoscopy and Surgical Lung Biopsy for the Diagnosis and Management of Pulmonary Infiltrates in Immunocompromised Hosts (면역저하환자에서 발생한 폐침윤에서 기관지내시경과 수술적 폐생검의 유용성)

  • Background: Pulmonary infiltrate in immunocompromised hosts has many infectious and non-infectios etiologies. To evaluate the diagnostic yield and therapeutic implication of two invasive diagnostic methods, such as bronchoscopy and surgical lung biopsy, we performed retrospective analysis of these patients. Methods: All immunocompromised patients admitted to Samsung Medical Center from October 1995 to August 1998 who underwent bronchoscopy and/or surgical lung biopsy for the diagnosis of pulmonary infiltrates were included in this study. Confirmative diagnostic yield, the rate of changed therapeutic plan and patients' survival were investigated. Results: Seventy-five episodes of pulmonary infiltrates developed in 70 patients(M : F=46 : 24, median age 51). Underlying diseases of patients were hematologic malignancy(n=30), organ transplantation(n=11), solid tumor(n= 12), connective tissue disease(n=6) and others. Confirmative diagnosis was made in total 53 cases (70.7%), of which 70.2% had infectious etiology. Diagnostic yields of bronchoscopy, bronchoalveolar lavage(BAL), transbronchiallung biopsy(TBLB) and surgical lung biopsy were 35.0%(21/60), 31.4%(16/51), 25.0%(9/36) and 80.0%(20/25). Therapeutic plan was changed in 40%(24/60) of patients after bronchoscopy and in 36%(9/25) of patients after surgical lung biopsy. More patients survived (84.4% vs 60.5%, p=0.024) when therapeutic plan was changed after invasive diagnostic study. Conclusion: Bronchoscopy and surgical lung biopsy are helpful for the therapeutic implication of pulmonary infiltrates in immunocompromised hosts. Large-scale prospective case-control study may further clarify their limitation and usefulness.
