  • The Late Quaternary palynoflora from an archaeological site in Piseo-ri, Muan, Korea consists predominately of deciduous broadleaved angiosperms and subordinately of herbs and conifers. Dominant taxa are Polypodiaceae, Taxodiaceae-Cupressaceae-Taxaceae, Alnus, Carpinus, deciduous Quercus, Compositae and Gramineae. The palynoflora dated 43,000-18,000 yr BP, corresponding to the last glacial period, is similar to the modern vegetation of the cool temperate deciduous broadleaved forest in the middle part of the Korean Peninsula. Based on palynofloral composition, three palynozones are recognized. Zone I represents a cool temperate deciduous broadleaved forest, composed of Polypodiaceae, Taxodiaceae-Cupressaceae-Taxaceae, Alnus and D. Quercus, and is characterized by persistent occurrence of warm temperate taxa. Zone II shows drastic decrease in Polypodiaceae, Taxodiaceae-Cupressaceae-Taxaceae, and increase in herbs. This zone reflects a cool temperate deciduous broadleaved forest with scattered open grasslands. Zone III is characterized by dominance in herbs and increase in conifers. The palynofloral composition suggests a cool temperate climate, and variation in occurrences of main taxa indicates that climatic conditions gradually became cold and dry.

  • This study was conducted to investigate effect of temperature and NaCl on the seed germination and water absorption of Thuja orientalis. Seeds were treated with 0, 500, 1,000, 2,000 and 4,000ppm of NaCl and placed in different chambers at 15, 20, 25 and $30^{\circ}C$, respectively. And seed properties and relative water absorptions were analyzed. Germination decreased with the increase of both temperature and NaCl concentration, and especially the difference was obvious at $30^{\circ}C$. Dormancy and mortality increased with the increase of temperature in non-NaCl treatment. Two-way analysis of variance showed significant effects of temperature, NaCl concentration and interaction between temperature and NaCl concentration (p<0.001). Mean germination time increased with the increase of NaCl concentration at 15, 20 and $25^{\circ}C$ but decreased at $30^{\circ}C$ because the seeds were mortal by NaCl high concentration, Germination speed and germination performance index decreased with the increase of NaCl concentration. Those represented decreasing tendency with NaCl concentration but high positive correlation with germination. Relative water absorption decreased with the increase of NaCl concentration and represented high values at $15^{\circ}C$, and showed high positive correlations with germination, germination speed and germination performance index. It was reported that the high temperature and salinity were inhibitive factors of seed germination of Thuja orientalis.

  • In this study, the components of microwave-assisted extracts obtained from Thuja orientalis leaves were analyzed, and the cytotoxicity, antibacterial and antiviral activities were evaluated. The predominant components from microwave-assisted extraction were catechin, leucopelargonidin, arecatannin, quinolone, and kaempferol derivatives, which are classified in the flavonoid and tannin groups. We observed that the 0.11 mg/mL of extract concentration did not show cytotoxicity in HaCaT cells. The antibacterial activities were tested according to the guidelines of methods for determining the bactericidal activity of antimicrobial agents. The extracts showed 99.9% antibacterial efficiency against gram-positive S. aureus, while the antibacterial effect on gram-negative E. coli was insignificant. When the extract concentration and contact time with bacteria were increased, 99.9% antibacterial efficiency was observed for E. coli as well as S. aureus. Following the standard to assess the activity of microbicides against viruses in suspension (ASTM-E1052-20), the antiviral efficiency was more than 99.99% for influenza A (H1N1) and SARS-CoV-2. These results suggest its potential use in antiviral disinfectants, surface coatings, personal protective equipment, and textiles.

  • This study was carried out to examine the effect of soil improvement according to the shape of charcoal and the kind of carbonized tree species. As the results, all of the seedlings of Thuja occidentalis and Aesculus turbinata grew better in the charcoal-treated soil than the non-treated soil. In case of powder charcoal, Thuja occidentalis seedlings grew up best in the soil treated with powder charcoal of Pinus koraiensis and then grew well in order of Larix leptolepis > particle board > Quercus acutissima. In case of granulated charcoal, the seedlings grew well in order of the granulated charcoal of Larix leptolepis > particle board > Pinus koraiensis. It was analyzed that the soil porosity and the organic matter content were mo re in the charcoal -treated soil than the non-treated soil also. It is inferred that because the aeration property and the absorption of organic matter were increased in the root zone, the growth of seedlings was better in the char-coal-treated soil.

  • The compositions of the leaf monoterpenoids in 7 species of Thuja (Thuja orientalis, T. orientalis 'Avrea Nana', T. orientalis cv. compacts, T. occidentalis, T. occidentalis 'Boothii', T. occidentalis 'Pumila', T. occidentalis 'Tiny Tim') were analyzed by GC-MS and compared between species. These Thuja contains 30 compounds and α -pinene, camphene, sabinene, myrcene, limonene, bonyl acetate, γ-terpinene, α -terpinenyl acetatee are occurred in these all species. Compounds in these leaf extracts are remarkably different between species. The simplest monoterpenoid (11 compounds) was found in T. orientalis 'Avrea Nana' the most complex monoterpenoid was in T. occidentalis 'Pumila'(26 compounds). Based on these data, similarities are computed using presence/absence matching by PAUP (Phylogenetic Analysis Using Parsimony). It appears that four taxa are present within these Thuja plants. The minimum spanning network reveals that Thuja occidentalis and Thuja occidentalis 'Boothii' were the most similar compounds.

  • 2kg of the dried leaves of Thuja orientalis Linnaeus were ground, extracted with acetone-$H_2O$(7:3, v/v), concentrated, and fractionated with a series of hexane, $CH_2C1_2$ EtOAc and water on a separators funnel. Each fraction was freeze dried to give dark-brown powder and a EtOAc soluble portion. of the powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluents. Spectrometric analyses such as NMR and FAB-MS including TLC were performed to characterize the structures of isolated compounds. The leave of Thuja orientalis Linnaeus contained a large amount of flavononol derivatives such as quercetin-3-O-$\alpha$-L-rhamnopyranoside and myricetin-3-O-$\alpha$-L-rhamnopyranoside in addition to a small amount of flavan compounds such as (+)-catechin and (+)-gallocatechin. The antioxidative activities of each fractions and isolated compounds were done by DPPH radical scavenging test, and all of them were indicated strong antioxidative activities.

  • We have tested 41 herbal medicines to search for a natural substance with antimicrobial activity against Streptococcus mutans and five types of oral bacteria. We have also investigated antioxidative activity of these herbal medicines. Antimicrobial activity against Streptococcus mutans and five types of oral bacteria was analyzed using ethanol extracts of herbal medicines. Extracts from Illicium verum and Amomum xanthioides showed 98% inhibitory activity against Sterptococcus mutans. The effect of Thuja orientalis on S-1 and Thuja orientalis and Amomum xanthioides on S-2 were 95% and 94%, respectively. Nelumbo nucifera was 94% effective on S-5. The inhibitory activities of the herbal medicines against glucosyltransferase (GTase) were determined using purified from Streptococcus mutans and five types of oral bacteria. Extract from Illicium verum and Amomum xanthioides showed 94% effectiveness on Streptococcus mutans. Amomum xanthioides showed 95% effectiveness on S-1 and Thuja orientalis showed 96% effectiveness on S-5. In antioxidant activities of the herbal medicines, extract from Thuja orientalis showed the highest level of 81.08% DPPH radical scavenging activity and Illicium velum extract also showed high antioxidative activity of 80.45%. Thuja orientalis had a large amount of phenolic compound with $115.24\;{\mu}g/mL$ among the herbal medicines.
