The Optimal Culture Condition for the Collagenolytic Protease Production from Vibrio vulnificus CYK279H (Vibrio vulnificus CYK 279H에서 Collagenolytic Protease 생산을 위한 최적배양조건)

  • A marine bacterium for producing an collagenolytic protease was isolated from the southern sea of Korea and identified as Vibrio vulnificus and named as Vibrio vulnificus CYK279H. This strain producing an collagenolytic protease was showed high activity toward collagen and gelatin as substrate. The optimum initial pH, NaCl, and temperature for cell growth and protease production was 7.5, 2.0% and 25$^{\circ}C$, respectively. Optimization for collagenolytic protease production was composed of 0.3% D-galactose, 0.6% yeast extract, 4.0% gelatin, 0.2% (NH$_4$)$_2$SO$_4$, and 0.2 mM ferric citrate in artificial sea water. The maximum protease production was required gelatin and yeast extract. The collagenolytic protease production by Vibrio vulnificus CYK279H reached a maximum of 73 unit/l after the cultivation for 18 h under the optimized medium.

  • Microalgae has been seen all over the seawater and several species are used for human food. Specially, Nannochloropsis oculata, a photosynthetic microalgae, has been focused for a vast array of valuable nutritious compounds. In order to find high mass Nannochloropsis oculata culture conditions, some of important growth factors of pH, temperature, culture media, and $CO_2$ effect were tested. The optimal growth condition was found to be as follows : 3% artificial seawater, initial pH 8.5, and temperature $25^{\circ}C$. The alga mass and chlorophyll content were dramatically increased by applying 5% flue $CO_2$ gas (1.50 g/L algae in a continuous $CO_2$ flue; 0.76 g/L alga without $CO_2$). It was shown that the chlorophyll biosynthesis was also closely associated with alga growth.

  • Column tests were carried out to examine the effect of surfactant solution conditions on the surfactant-enhanced remediation of soil columns contaminated by toluene. The conditioned parameters of the surfactant solution for the column tests were concentration, pH, temperature and flow rate. The test results revealed that an optimum condition was achieved for 4% (v/v) of concentration, 10 of pH, $20^{\circ}C$ of temperature and 4 mL/min of flow rate respectively. The removal of 95% of toluene was obtained when optimal conditions of each surfactant solution parameter were simultaneously met. This was a marked improvement and removal efficiency increased by 6-19% compared to that with unadjusted conditions. The optimum range of these parameters may be useful for a surfactant-based remediation in the aquifer contaminated by toluene.

  • A keratinolytic protease was purified from the culture medium of Pseudomonas sp. KP-364 by use of an assay of the hydrolysis of feather keratin. Membrane ultrafiltration and DEAE-cellulose ion-exchange resin and Sephadex G-150 gel chromatographies were used to purify the enzyme. The specific activity of the purified keratinolytic protease relative to that in the original medium was approximately 72-fold high. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Sephadex G-150 chromatography indicated that the purified keratinolytic protease is monomeric and has a molecular weight of 36 kDa. The optimal pH and temperature of the keratinolytic protease activity were 6.6 and 37 C, respectively, and the keratinolytic protease was relatively stable at pH value from 3.0 to 10.0 at 37 C for 1hour. The keratinolytic protease was inhibited by EDTA and EGTA, indicating that the keratinolytic protease was a kind of metalloprotease that require Li+ for cofactor.

  • An intracellular accumulation of cadmium by the intact cell of an extremely cadmium tolerant yeast, Hansenula anomala B-7, was investigated in the presence of Triton X-100. The uptake of cadmium by the intact cell was efficiently enhanced up to approximately 40% or more by 0.1% of Triton X-100 and Aerosol OT, respectively. The Michaelis constant, Km, done by Lineweaver-Burk plot of accumulation velocity of cadmium vs. cadmium concentration was calculated to be 0.247mM. The optimal conditions of pH and the temperature for the effective cadmium uptake were from neutrality to alkali and 4$0^{\circ}C$, respectively. The accumulation of cadmium was increased approximately 3 times under the shaking incubation, with no correlation to shaking rate. By zinc the cadmium accumulation was decreased.

  • As a way of utilizing cheese whey to fortify sulfur-bearing amino acids to soybean protein, whey-soybean coagulum was made from whey-soy milk mixture and optimum conditions for coprecipitation of the two proteins were determined. Mixture of whey and soymilk in 1 : 1 volume ratio was coagulated at $0.005{\sim}0.5M$ of $CaCl_2$ concentration, pH $1.5{\sim}8.0$, and at $60{\sim}100^{\circ}C$, and absorbance at 500nm of filtrate from the coagulum was measured. Optimum conditions for the coprecipitation were 0.0125M of $CaCl_2$ concentration, pH $6.5\sim7.5$, and $70{\sim}80^{\circ}C$ which resulted in the minimum absorbance and also good physical properties of the curd.

  • The separation and purification of lysozyme enzyme from chicken egg white (CEW) solution was studied using the dead-end filtration. The optimum operation conditions of the dead-end filtration reveal that the maximum value of permselectivity of lysozyme to the other proteins of ovalbumin and conalbumin in the CEW solution was tested with change of molecular weight cut-off (MWCO) of ultrafitration membrane and pH of the CEW solution. The optimum operation conditions for the efficient permeable separation of lysozyme from the CEW solution are that the membrane MWCO is 30 kD and the pH of CEW solution is 11. At this optimum operation conditions, the maximum value of permselectivity of lysozyme to total proteins in CEW solution is about 83.

  • We investigated the growth parameters of Candida parapsilosis isolated from spoiled red ginseng extract. The optimum pH range for C. parapsilosis was 6.0, whereas the minimum and maximum pH values that permitted growth were 3.0 and 11.0, respectively. For cells grown in PG medium plus 0 and 60% sucrose, the optimum water activity(Aw) values were 0.98 and 0.97, respectively. The optimum temperature for C. parapsilosis were 3$0^{\circ}C$ at an Aw of 0.90 in 4.8% potato dextrose broth with 18% sucrose (PGS). Cations inhibiting the growth of C parapsilosis were L $i^{+}$ $Ca^{2+}$, $Mg^{+2}$, $K^{+}$ in decreasing order, while anions were S $O_4$$^{2-}$, N $O_3$$^{[-10]}$ , C $l^{[-10]}$ .TEX> .

  • Proteases are degradative enzymes which hydrolyze a peptide bond between amino acids and they are abundantly applied to commercial field. In order to investigate optimal medium compositions of carbon and nitrogen source for enzyme production, modified STY medium containing 0.15% yeast extract were used as basal medium. When galactose was used as carbon source, enzyme activity showed 1.3 higher than that of glucose. For nitrogen source addition of casamino acids to basal medium in place of tryptone showed lowest activity, whereas addition of malt extract showed maximal activity. The optimum temperature and pH of extracellular protease were found to be $35^{\circ}C$ an pH 8.5.

Hydrolysis of Egg Yolk Protein in a Packed Bed Reactor by Immobilized Enzyme (충진층 반응기에서 고정화 효소에 의한 난황 단백질의 가수분해)

  • Alkaline protease for the hydrolysis of egg yolk protein was immobilized on five carriers - Duolite A568, Celite R640, Dowex-1, Dowex 50W and Silica gel R60. Duolite A568 showed a maximum immobilization yield of 24.7%. Optimum pH for the free and immobilized enzyme was pH 8 and 9, respectively. However, no change was observed in optimum temperature ($50^{\circ}C$). Thermal stability was observed in immobilized enzymes compared to free enzymes. The immobilized enzyme retained 86% activity after 10 cycle operations in a repeated batch process. The effect of flow rate on the stability of enzyme activity in continuous packed-bed reactor was investigated. Lowering flow rate increased the stability of the immobilized enzyme. After 96 hr of continuous operation in a packed-bed reactor, the immobilized enzyme retained 83 and 61% activity when casein and egg yolk were used as a raw materials, respectively.
