Evaluation of Urease Inhibition Activity of Zerumbone in vitro (제럼본의 요소가수분해효소 활성 억제 평가)

  • A key virulence factor for urinary tract pathogens is the enzyme urease, which catalyzes the hydrolysis of urea into ammonium ions and carbonic acid. Urease activity plays an important role in the pathogenesis of urinary tract infection. In this study, the inhibitory effect of zerumbone against six urease-producing bacteria (Klebsiella oxytoca, K. pneumoniae, Morganella morganii, Proteus mirabilis, P. vulgaris, and Staphylococcus saprophyticus) and their urease activities were evaluated. The results of the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) tests showed that zerumbone had antibacterial effect against these six urease-producing bacteria. The MIC and MBC of zerumbone ranged from 0.5 to 2 mM and 1 to 4 mM, respectively. In the urease inhibitory assay, zerumbone showed better urease inhibition ($56.28{\pm}2.45-37.83{\pm}3.47%$) than the standard urease inhibitor, acetohydroxamic acid ($40.46{\pm}1.94-22.99{\pm}3.53%$). However, zerumbone did not affect the levels of the urease subunit. These results clearly indicated that zerumbone has antibacterial potential against urease-producing bacteria and possesses excellent bacterial urease inhibition properties.

Antimicrobial activity of honeybee venom against fish pathogenic bacteria (국내산 봉독의 어류병원성 세균에 대한 항균활성)

  • In this study, we examined antimicrobial activity of the bee venom isolated from honeybee (Apis mellifera L.) against fish pathogenic bacteria, Edwardsiella tarda, Vibrio ichthyoenteri and Streptococcus iniae of cultured olive flounder, Paralichthys olvaceus. The bee venom exhibited significant antimicrobial activity against the both Gram-negative bacteria, E. tarda and V. ichthyoenteri and Gram-positive bacteria, S. iniae. Minimum inhibitory concentration (MIC) and minimun bactericidal concentration (MBC) of the bee venom were 17.6 ${\mu}g$/ml 34.6 ${\mu}g$/ml against E. tarda., and 1.76 ${\mu}g$/ml, 6.8 ${\mu}g$/ml against V. ichthyoenteri, respectively. MIC and MBC of the bee venom were 3.49 ${\mu}g$/ml, 11 ${\mu}g$/ml, respectively against S. iniae. The postantibiotic effect (PAE) of the bee venom was 5 hr, 6 hr, and 7 hr against E. tarda, V ichthyoenteri, and S. iniae, respectively. In addition, its antimicrobial activity was stable under various pH conditions. According to these results, the bee venom showed the excellent antimicrobial activity against the tested pathogenic bacteria.

Antibacterial, Antioxidant Activities of Acer tegmentosum Maxim Ethanol Extract Against Staphylococcus aureus (벌나무 추출물의 황생포도상구균에 대한 항균 효과와 항산화 활성)

  • This study investigates the antibacterial and antioxidant activities of the ethanol extract of Acer tegmentosum Maxim (EATM) against Staphylococcus aureus. The total polyphenol and flavonoid content, 1,1-diphenyl-2-picryl hydrazyl (DPPH) and 2,2'-Azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging effect, and reducing power of the ferric reducing antioxidant power (FRAP) method were measured to verify the antioxidant activity of the EATM. The antibacterial activity against S. aureus using the EATM was verified by the paper disc method, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC) methods. The total polyphenol and flavonoid contents were 266.66 ㎍ GAE/mg and 6.46 ㎍ QE/mg, respectively. The DPPH and ABTS radical scavenging activity showed a concentration-dependent scavenging activity. The RC50 values of the EATM were 21.49 and 12.81 ㎍/mL, respectively. A FRAP analysis was conducted for evaluating the reducing power of the EATM and an efficacy of 0.73 ± 0.19 mM FeSO4 E/mg was observed. The antibacterial activity of EATM against S. aureus, determined using the paper disc method, showed an inhibitory ring of 3 mm at 2 mg. The MIC was confirmed at a concentration of ≥ 16 mg/mL, while the MBC was confirmed at 32 mg/mL. As the EATM shows antioxidant and antibacterial activities against S. aureus, it can be used as an effective antidote against atopic dermatitis.

  • Streptococcus mutans plays a major role in the formation of dental plaque and it is considered one of the important pathogens in the development of dental caries. Established dental plaque can be more resistant to antimicrobial agents and offers nutrient rich and relatively stable cariogenic conditions for bacteria. Thus bacteria growing in dental plaque have strong resistance to antimicrobial agents and they are not removed easily by the flow of saliva. Many researchers have been performed using natural materials, especially herbal extracts to prevent dental plaque. However, the strains of mutans streptococci used in the researches were not from Koreans. Therefore, it would be necessary to evaluate the antimicrobial activity of herbal extracts against clinical isolates of Streptococcus mutans isolated from saliva of Koreans living in Jeollabuk-do. For this study four clinical isolates were isolated from saliva samples of seventeen Korean people to investigate whether essential oils of Artemisia spp. have antimicrobial activity against them including four reference strains of Streptococcus mutans. Minimum inhibitory concentrations, Minimum bactericidal concentrations and time kill studies were performed and the results showed that Artemisia lavandulaefolia DC, Artemisia scoparia, and Artemisia capillaries have antimicrobial activity against the Streptococcus mutans clinical isolates and reference strains.

Anticariogenic activity of acanthoic acid isolated from Acanthopanax koreanum $N_{AKAI}$ against oral pathogens (섬오가피로부터 분리된 acanthoic acid의 항치아우식효과)

  • This research was aimed to search for natural antimicrobial agents that are sefe for humans and specific for oral pathogens. Acanthoic was isolated from the chloroform fraction of methanol extract of Acanthopanax koreanum $N_{AKAI}$ and its structure were elucidated by 13-NMR, 1H-NNR and ESIMS. Antibacterial activity of acanthoic acid was investigated by the minimum inhibitory (MIC) and minimun bactericidal (MBC) concentration. MIC/MBC of acanthoic acid against Streptococcus mutans $N_{AKAI}$ causing dental caries was determined to be $2/4\;{\mu}g/mL$, which was much lower than these of other natural antimicrobial agents such as $8/16\;{\mu}g/mL$ of sangurinarine and $250/500\;{\mu}/mL$ of green tea extract, $500/600\;{\mu}g/mL$ of thymol and borneol. Acanthoic and significantly inhibited the growth of other cariogenic bacteria such as Streptococcus sobrinus $N_{AKAI}$ and Streptococcus sanguis $N_{AKAI}$, and Streptococcus gordonii $N_{AKAI}$ in the MIC range of $4{\sim}32\;{\mu}g/mL$. My finding suggests that acanthoic acid could be employed as a potential antibacterial agent for preventing dental caries.

  • Allyl isothiocyanate (AIT), the principle ingredient of antimicrobial ingredients from horseradish root, can be prepared from extracts of horseradish root or synthetic method. It is reported that the horseradish root extract has the antimicrobial effect against various oral microorganisms, while there is no further study about the antimicrobial effect against the oral microorganisms of synthetic AIT derived from synthetic method. The aim of the study is to compare the difference of the antimicrobial effect between horseradish root extracts and synthetic AIT. To evaluate the antimicrobial effect, we measured the minimum inhibitory concentration(MIC) and minimum bactericidal concentration (MBC), and the results are like following. 1. The MIC of horseradish root extract against 7 kinds of oral pathogenic microorganisms is about 117$\sim$1,750 ppm(0.0117$\sim$0.175%), and the MIC of the synthetic AIT is about 344$\sim$3,000 ppm(0.0344$\sim$0.3%), which have the antimicrobial effects against all kinds of microorganisms. 2. The MBC of the horseradish root extracts against the 7 kinds of oral microorganisms is about 625.2$\sim$6,000 ppm(0.06252$\sim$0.6%), and the MBC of the synthetic AIT is about 1,750$\sim$7,000 ppm(0.175$\sim$0.7%), which have the antimicrobial effects against all kinds of microorganisms.

  • This study was conducted to improve circulation system and the storage of garlic against the spoiling microorganisms during the storage. To fulfill this objective, the isolation and identification of spoiling microorganisms from horticultural products were performed at the first step. Antimicrobical activity of electrolyzed alkaline water (EAW) was tested against the spoiling microorganisms and MIC(minimun inhibitory concentration) was determined. As a result, it showed the formidable MIC value at near 2.0%(v/v) against the spoiling microorganisms.

  • To prevent contamination by Campylobacter jejuni during chicken carcass processing, the effect of emulsifiers on C. jejuni inoculated on chicken skin was investigated using confocal laser scanning microscopy. Among the 8 emulsifiers (SWA-10D, L-7D, M-7D, S-1670, L-1695, P-1670, polysorbate 20, polysorbate 80) tested for antimicrobial activity by the paper disk method, 4 emulsifiers (L-7D, L-1695, polysorbate 20, polysorbate 80) were screened further. Emulsifier L-1695 showed the largest clear zone at a concentration of 200 mg/mL. The 4 emulsifiers subjected to primary screening were screened for heat and pH stability. In the contact surface test, emulsifier L-1695 showed the lowest log CFU/㎠ value on both stainless steel and ceramic surfaces. When emulsifier L-1695 was applied via general and electrostatic spray methods, the number of C. jejuni entrapped inside chicken skin follicles was significantly reduced in both methods. In conclusion, the emulsifier L-1695 could be employed as a microbial detachment agent in the chicken carcass processing industry.

  • Purpose: This study aimed to evaluate the effectiveness of a dielectric barrier discharge (DBD) plasma module for sterilizing airborne bacteria in indoor spaces and measure the concentration of ozone generated during plasma discharge. Method: The DBD plasma module was installed in a 76m3 space, and air samples were collected under various discharge times to compare the reduction of airborne bacteria. Result: The results showed a significant decrease in airborne bacteria, ranging from 92.057% to 99.999%, with an average ozone concentration of 0.04 ppm, below the reference value. Conclusion: The study suggests that plasma discharge can be used as a means of preventing the spread of airborne bacteria and viruses, while ensuring safety for human exposure.
