Effect of Corona Discharge on the Changes in Quality and Pasteurization of Ginseng Powder (코로나방전 처리 인삼분말의 살균효과 및 품질변화)

  • As a newly emerging pasteurization method for improving the shelf-life of powdered ginseng, the corona discharge generated at high voltage conditions was examined for its effects on microbial pasteurization, physicochemical characterization, and sensory evaluation. The pasteurizing effects of corona discharge on total bacteria, yeast, molds, and coliforms increased with increasing voltage. However, corona discharge treatment of 5 kv/cm did not show significant effects of pasteurization on the yeast, molds, and coliforms. Treatment with 20 kv/cm reduced about 95% of the total bacteria $(6.14{\pm}0.04{\rightarrow}\;4.59{\pm}0.04$) and coliforms ($2.43{\pm}0.05{\rightarrow}\;1.00{\pm}0.05$) and 99% of the yeast and molds ($4.65{\pm}0.06{\rightarrow}\;2.73{\pm}0.06$). The proximate analysis, minerals, free sugars, crude saponin, and colors of the control ginseng and the ginseng treated with 20 kv/cm were not significantly different at p<0.05. Corona discharge of up to 20 kv/cm did not affect the proximate analysis, minerals, free sugars, crude saponin, and colors of the powdered ginseng. Sensory evaluation scores on color, flavor, taste and overall acceptability between the control ginseng and ginseng treated with 20 kv/cm corona discharge were not significantly different at p<0.05 level. Consequently, corona discharge treatment is thought to be a good alternative pasteurization method for improving the shelf-life of powdered ginseng due to its effective pasteurization, maintenance of nutrients and good sensory characteristics.

  • Seventy two samples of feedstuff were collected from commercial channels all over Korea. As a study on the moisture contents, microflora and mycotoxin production of each sample investigated. Moisture content of the samples was $11.2{\sim}15%$. Total counts of the samples were $1.8{\times}10^{2}{\sim}2.4{\times}10^{6}$ per gram. The Coli-form group were counted from 9 to $6.3{\times}10^{5}$ per gram which composed mainly of Enterobacter and Klebsiella, whereas Escherichia coli was minor ones. The contamination of many feeds was not remarkable, and only some broiler feeds was contaminated largely with Escherchia coli. Fungi were below detectable limit in 45% of the samples and the most contaminated sample had $4.5imes}10^{5}/g fungi counts.

Effect of Chlorine Dioxide (ClO2) on the Malodor Suppression of Chicken Feces (이산화염소(ClO2) 처리가 계분의 악취 억제에 미치는 영향)

  • This study evaluated the efficacy of chlorine dioxide (ClO2) as an oxidant to reduce malodor emission from chicken feces. Two experiments were performed with the following four treatments in parallel: 1) fresh chicken feces with only distilled water added as a control, 2) a commercial germicide as a positive control, and 3) 2,000 or 4) 3,000 ppm of ClO2 supplementation. Aluminum gas bags containing chicken feces sealed with a silicone plug were used in both experiments, and each treatment was tested in triplicate. In Experiment 1, 10 mL of each additive was added on the first day of incubation, and malodor emissions were then assessed after 10 days of incubation. In Experiment 2, 1 mL of each additive was added daily during a 14-day incubation period. At the end of the incubation, gas production, malodor-causing substances (H2S and NH3 gases), dry matter, pH, volatile fatty acids (VFAs), and microbial enumeration were analyzed. Supplementing ClO2 at 2,000 and 3,000 ppm significantly reduced the pH and the ammonia-N, total VFA, H2S, and ammonia gas concentrations in chicken feces compared with the control feces (P<0.05). Additionally, microbial analysis indicated that the number of coliform bacteria was decrease after ClO2 treatment (P<0.05). In conclusion, ClO2 at 2,000 and 3,000 ppm was effective at reducing malodor emission from chicken feces. However, further studies are warranted to examine the effects of ClO2 at various concentrations and the effects on malodor emission from a poultry farm.

Change of Anti-Oxidative Activity and Quality Characteristics of Maejakgwa with Mugwort Powder during the Storage Period (쑥 첨가 매작과의 저장과정 중 항산화활성 및 품질특성 변화)

  • The purpose of this study was to investigate the effects of mugwort powder on the quality characteristics and antioxidant activity of Maejakgwa. Maejakgwa were prepared with mugwort powder at levels 0%, 1%, 3% and 5% ($60{\pm}1^{\circ}C$, 14 days). The lightness, redness, and yellowness values of Maejakgwa significantly reduced depending on mugwort powder. The hardness of Maejakgwa was decreased with the increase of storage period and increased with the increase of mugwort powder. In the sensory evaluations, the Maejakgwa prepared with 3% added mugwort powder received higher acceptance scores for the properties of color, taste, hardness, crispiness, adhesiveness and overall acceptability. As the mugwort powder content increased, acid value and peroxide value were decreased. With the increase of storage period, acid value and peroxide value of all sample increased but growth rate of these values decreased with the addition of the mugwort powder. DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity was improved significantly via the addition of mugwort powder and decreased as storage period increased. During storage period, Maejakgwa with mugwort powder showed a stronger antimicrobial effect in yeasts and molds than in total aerobic bacteria. Coliform bacteria were not detected in all samples. Also the antimicrobial activity was increased with the addition of the mugwort powder and decreased as storage period increased. The results show that addition of the mugwort powder to foods with fat such as Maejakgwa would be a useful way to enhance the antioxidant quality, sensory characteristics and shelf life.

Physicochemical Properties and Biological Activities of Tenebrio molitor Fermented by Several Kinds of Micro-organisms (유용 미생물을 이용한 발효갈색거저리 추출물의 이화학적 특성 및 생리활성 효과)

  • In this study, Tenebrio molitor (T. molitor) was fermented with Lactobacillus plantarum JBMI F3 (F3), Lactobacillus plantarum JBMI F5 (F5), Lactobacillus gasseri Ba9 (Ba9), Aspergillus kawachii KCCM 32819 (Ak), Saccharomyces cerevisiae KACC 93023 (Sc), and Bacillus subtilis KACC 91157 (Bs). After fermentation, the fermented products were extracted by water, ethanol, and methanol, and their physicochemical and biological properties were investigated. In a DPPH assay, the water extracts of the fermented products of T. molitor showed high antioxidant ability. Among the water extracts, the fermented product by Bs showed the highest DPPH radical scavenging activity. The total contents of phenolic compounds and flavonoids were highest in the fermented products by Ak and Bs, respectively. Reducing activity was detected the most high activity on ethanol extract of fermented product by Bs. The water extract of the fermented product by Bs exhibited strong enzymatic activity for fibrinogen and starch hydrolysis. Based on the observed physicochemical and biological properties, the fermented products of T. molitor by microorgansims can likely be applied as functional materials in various industries.

Sterilizing Effect of Electron Beam on Ginseng Powders (Electron Beam 조사에 의한 인삼분말의 살균효과)

  • The sterilizing effect of electron beam was compared with that of gamma irradiation for commercial ginseng powders. White and red ginseng powders were contaminated by about $10^5\;CFU/g$ of total bacteria and by $10^3\;CFU$ of coliforms only in white ginseng powder. Data of microbial population for the sterilizing effect of electron beam irradiation showed that no microorganisms were detected in the samples irradiated up to 7.5 kGy for total aerobic bacteria and 2.5 kGy for molds and coliforms. Such doses were effective for controlling the microbial growth in the samples during 4 months of storage at room temperature. Decimal reduction doses $(D_{10}$ value) on the initial bacterial populations were $2.85{\sim}3.75\;kGy$ in electron beam and $2.33{\sim}2.44\;kGy$ in gamma irradiation, which were influenced by the initial microbial loads and the energy applied. Compared with gamma irradiation, electron beam showed a similar result in its sterilizing effect on ginseng powders, suggesting its potential utilization in due time.

Physicochemical and Microbial Quality Characteristics of Garlic (Allium sativum L.) by Superheated Steam Treatment (과열증기 처리에 따른 마늘의 이화학적 및 미생물학적 품질 특성)

  • The objectives of this study were to investigate the effects of superheated steam (SHS) treatment on the physicochemical and microbial properties of garlic. The garlic was treated by SHS at temperatures of 100, 150, 200, 250, 300, and $350^{\circ}C$ for 60 s. The moisture content of raw garlic was lower than that of SHS-treated garlic. The total thiosulfinate and pyruvate contents were dramatically reduced by SHS treatments. The antioxidant activities of garlic measured by ferric reducing/antioxidant power, 2,2-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt radical scavenging assay, and total phenolics content decreased by SHS. The major volatile sulfur compounds of garlic such as diallyl disulfide, allicin, allyl sulfide, diallyl sulfide, and diallyl trisulfide were significantly reduced by SHS. The antimicrobial effects of raw garlic were stronger than those of SHS-treated garlic against three strains of bacteria, including Staphylococcus aureus, Escherichia coli, and Bacillus cereus. However, total aerobic bacteria in garlic were dramatically reduced by SHS from 8.6 to 2.9 log CFU/g. The results from the sensory evaluation show that SHS treatment of garlic above $200^{\circ}C$ provides better acceptably due to reduction of off-flavor and pungency of garlic. These results suggest that superheated steam treatment can used as an efficient process for reducing garlic off-flavor and pungency.

Study of Formation Factor of Biofilm on Aluminum surface and Removal Efficiency of Biofilm by Antimicrobials (알루미늄 표면에 생물막의 형성인자 및 항균제에 의한 생물막 제거효과 분석)

  • 108 microorganism types (79 types of fungi and 29 types of bacteria) were isolated from 25 automobiles generating bad odor when the air conditioner was turned on, and 43 types of fungi and 23 types of bacteria were identified. The analysis of condensate generated by the air conditioners in the automobiles indicated pH 6.4~7.1, 12.5~34.2 mg/L carbon source, 0.9~18.6 mg/L nitrogen source, 0.5~27.8 mg/L ion contents, and 0.1~7.7 mg/L mineral contents. The biofilms were formed by the mesophiles under the summer temperature/humidity condition ($26^{\circ}C$, 70% R.H.), and they were regenerated when the environmental factors (nutritional contents and temperature/humidity) were appropriate even after they were artificially removed. Although the antimicrobials removed 99.9% of planktonic cells within 15 minutes, they were not effective in removing biofilm. Up to 1,950 ppmv of ethanol was observed in the automobile treated with the antimicrobials. Although the figure is lower than the acute toxicity level when inhaled by humans, the health safety of the chemical substances used in the antimicrobials needs to be reviewed.

Ingredients Analysis and Biological Activity of Fermented Angelica gigas Nakai by Mold (곰팡이 발효 참당귀의 유효성분 분석 및 생리활성 작용)

  • Three mold strains, Aspergillus oryzae (AOFAG), Aspergillus kawachii (AKFAG), and Monascus purpureus (MPFAG) were used for fermentation of Angelica gigas Nakai powder. The contents of polyphenolic compounds, flavonoids, minerals, decursin and decursinol angelate and the activities of DPPH (${\alpha},{\alpha}'$-diphenyl-$\beta$-picrylhydrazyl) free radical scavenging, reducing power, and tyrosinase were measured. The highest contents of phenolic compound and flavonoid were NFAG at 2.78% and MPFAG at 1.18%, respectively. Major minerals were K, Mg, Fe, Na and Ca. Decursin and decursinol angelate were the major ingredients of Angelica gigas according to HPLC analysis. Decursin area was higher in all fermented Angelica gigas than in NFAG. The activities of free radical scavenging and tyrosinase were stronger in all fermented Angelica gigas than NFAG. However, the Fe/Cu reducing powers were stronger in NFAG than all fermented Angelica gigas. Overall, these results may provide the basic data needed to understand the biological activities and chemical characteristics of Angelica gigas fermented by mold for the development of functional foods.

Effects of Storage Conditions of Rice Flour on Growth Properties of Natural Microflora (쌀가루의 저장조건에 따른 자연균총의 생육특성)

  • In order to optimize microbial safety and preservation in quality retention of rice flour, commercial hot-air dry (HT, 65/15 min) and microwave dry (MT, 700 watt/30 sec) treatments were developed, and in this study, natural microflora present in rice flour exposed to different storage temperature and periods were monitored. Changes in color (E) appeared to be less on the MT rice flour than on the rice flour. Effectiveness of the MT treatment showed reduction rates for total aerobic bacteria (2.62 log CFU/g), yeasts, and molds (0.37 log CFU/g). Total aerobic bacteria showed similar growth patterns of all the treatments during storage; however, the MT treatment inhibited the growth of this organism in rice flour. In conclusion, the MT treatment was found to be a suitable drying method to substitute the HT treatment in terms of quality of rice flour and microbial safety.
