The antioxidative effects of Ampelopsis brevipedunculata extracts (개머루덩굴 추출물의 항산화 효과)

  • This study was carried out to investigate the antioxidative capacity of Ampelopsis brevipedunculata 95% ethanol extracts. The concentration of A. brevipedunculata extract at which DPPH radical scavenging activity was inhibited by 50% was 0.42 mg/mL as compared to 100% by pyrogallol as a reference. Total antioxidant status was examined by total antioxidant capacity against ABTS radical reactions. Total antioxidant capacities of A. brevipedunculata extract at the concentrations of 0.1 and 1 mg/mL were 0.65 and 3.71 mM Trolox equivalents, respectively. Oxygen radical absorbance capacities of A. brevipedunculata extract at the concentrations of 5 and $100\;{\mu}g/mL$ were 22.75 and $131.25\;{\mu}M$ gallic acid equivalents, respectively. Superoxide scavenging activities of A. brevipedunculata extract at the concentrations of 0.1 and 1 mg/mL were 27.7 and 56.0%, respectively. Total phenolic contents of A. brevipedunculata extract at the concentrations of 0.5 and 2.0 mg/mL were 0.55 and 2.06 mM gallic acid equivalents, respectively. A. brevipedunculata extract at the concentration of 0.1 mg/mL inhibited 0.2 mM and 0.5 mM tert-butyl hydroperoxide induced cyototoxicity by 36.2 and 23.3%, respectively, in HepG2 cell culture system. Thus strong antioxidant and cytotoxicity-inhibiting effects of A. brevipedunculata extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in total phenolic contents.

  • 요즘 항산화 활성이 높아 인기가 많은 아로니아와 댕댕이나무의 고랭지 경사밭에 재배시 추비하는 횟수가 이들 열매의 항산화 활성에 미치는 영향을 분석하고자 본 연구를 수행하였다. 묘목은 2년생 묘를 이용하여 2016년 5월에 정식하였으며 2018년 11월 까지 재배하여 추비하였다. 추비는 매년 1회, 2회, 3회, 4회 처리를 두었으며 3년차인 2018년에 댕댕이나무는 6월 상순, 아로니아는 10월 상순에 수확하여 동결 건조하였으며 분말형태로 갈아 실험에 사용하였다. 분석결과 추비처리에 따른 아로니아 열매의 수율은 유사하였으며 총페놀 함량은 추비 1회 처리에서 76.2mg GAE/g 로 가장 높았고 그다음이 추비 4회, 2회, 3회 순이었다. 총플라보노이드 함량은 4회 추비처리에서 14.5mg QE/g로 가장 높았으며 그다음이 추비 2회, 1회, 3회 처리 순이었다. 그리고 DPPH 활성산소 제거능은 1회 추비처리에서 130.3 IC50(${\mu}g/ml$)으로 가장 효과적인 것으로 나타났으며 그다음이 추비 4회, 2회, 3회 추비처리 순이었다. 또 환원력에 있어서도 $1,000{\mu}g/m{\ell}$의 농도에서 추비 1회에서 높았고 그다음 유사하였다. 이러한 결과로 볼 때, 아로니아 열매의 항산화 활성은 추비를 1회 정도 하는 것이 좋을 것으로 생각된다. 추비처리에 따른 댕댕이나무 열매의 수율은 추비 4회 처리에서 가장 높았고 그다음이 추비 1회 처리에서 많았고 추비 2회, 3회는 유사하였다. 총페놀 함량은 추비 2회 처리에서 59.1mg GAE/g 로 가장 높았고 그다음이 추비 3회, 4회, 2회 처리 순이었다. 총플라보노이드 함량은 추비 1회 처리에서 152mg QE/g로 가장 높았으며 그다음은 모두 유사하였다. 그리고 DPPH 활성산소 제거능은 2회 추비처리에서 117.8 IC50(${\mu}g/ml$)으로 가장 효과적인 것으로 나타났으며 그다음이 1회, 4회, 3회 추비처리 순이었다. 또 환원력에 있어서도 $1,000{\mu}g/m{\ell}$의 농도에서 추비 1회와 2회 처리에서 높았고 3회 4회 처리는 유사하였다. 이러한 결과로 볼 때, 댕댕이나무 열매의 항산화 활성은 추비를 1-2회 정도 하는 것이 좋은 것으로 사료된다.

Pre-establishment of Microwave-Assisted Extraction Conditions for Chinese Ginger Oleoresins (중국산 생강의 올레오레진 제조를 위한 극초단파 추출조건 설정)

  • Some fundamental conditions were investigated in microwave-assited extraction (MAE) for oleoresin from Chinese dried ginger. The fifty watts of microwave energy(2,450 MHz), 60 mesh in particle size and 1:10 (g/mL) ratio of sample to solvent were the optimum conditions. Lower yield of soluble oleoresins was obtained at higher concentrations of ethanol. Radical scavenging ability was highest in extracts by using 25% of ethanol. Total phenolics in extracts were remarkably increased at above 50% of ethanol concentration. Overall yield of oleoresin components increased in proportional to extraction time up to 7 min. Based on the above results, it is indicated that ethanol concentration is a critical parameter in MAE.

Effect of Edible Coating on Hygroscopicity and Quality Characteristics of Freeze-Dried Korean Traditional Actinidia (Actinidia arguta) Cultivars Snack (가식성 코팅처리가 토종다래(Actinidia arguta) 동결건조 스낵의 흡습과 품질에 미치는 영향)

  • The purpose of this study was to evaluate the effect of edible coating on hygroscopicity and quality characteristics of a freeze-dried Actinidia arguta snack. Freeze-dried A. arguta snacks were coated with various edible coating materials such as albumin, dextrin, and whole soy flour. There were no significant effects of coating on major quality properties such as moisture content, water activity, yield, water soluble index, water absorption index, and rehydration properties of all samples. Compared with non-coated samples, edible coated samples effectively inhibited hygroscopicity as a function of hygroscopic time. The samples coated with dextrin showed lower hygroscopicity than the other coated samples. In addition, the effects of edible coating treatment on hardness, total phenolic content, and antioxidant activity measured by 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity were investigated. Edible coated samples had higher hardness, total phenolic content, and antioxidant activity than the non-coated samples as a function of hygroscopic time. Among edible coating materials, dextrin was the most effective coating material. Dextrin as an edible coating material for freeze-dried A. arguta snack may help to prevent hygroscopicity and extend market quality and shelf-life during storage.

Antifungal and Antioxidant Activities of Extracts from Leaves and Flowers of Camellia japonica L. (동백나무 잎과 꽃 추출물의 항미생물 활성 및 항산화 효과)

  • This research was performed to investigate the possibilities of industrial usage of camellia (Camellia japonica L.) by examining the antioxidant and antimicrobial effects of methanol extract with different sections. Content of total phenolics, DPPH radical scavenging activities and antibacterial activity of young leaf, mature leaf, flower bud, flower, bark, and seed of camellia were compared in vitro experimental models. Total phenolics was contained the higher in young leaf (74.62 mg), flower bud (65.02 mg) and flower (62.42 mg) but less than 20.95 mg per 100 g of dry weight in other parts of Camellia japonica L. And effects of antioxidant measured by DPPH radical scavenger activity ($RC_{50}$, reduce concentration 50%), was shown higher $7.16{\sim}18.14\;{\mu}g/m{\ell}$ in methanol extract of young leaf, flower bud and flower than $61.23\;{\mu}g/m{\ell}$ of BHT as a chemical oxidant. Also, the antimicrobial activity of Camellia japonica L. extracts determined using a paper disc method against food-borne pathogen and food spoilage bacteria, the young leaves extracts showed the most active antimicrobial activity against 7 kinds of harmful microorganisms. Flower bud extracts showed the highest antibacterial activity against P. aeruginosa and Enterobacter spp. C1036. In addition, the minimum inhibitory concentration (MIC) of young leaf extract against B. subtillis,S. fradiae,S. aureus,E. coli,P. aeruginosa, Enterobacter spp. C1036, and S. typhimurium were revealed 1 to 15 ${\mu}g/m{\ell}$. As a result, antimicrobial activity of camellia extracts was shown higher gram positive bacteria than gram negative bacteria.

Antibacterial Activity of Vinegars on Streptococcus mutans Caused Dental Caries (충치 유발균 Streptococcus mutans에 대한 식초의 항균효과)

  • The antibacterial activity for S. mutans by using 5 kinds of vinegar was researched. As a result it was the highest at persimmon vinegar and then grape, apple, high calcium brown rice, brown rice vinegar in order. The antibacterial effect was shown at 1.5% of persimmon vinegar concentration and the growth was fully repressed at 4% of concentration. As an observation result with a transmission electron microscope(TEM), the growth was obstructed by destroying the cell wall and the cytoplasmic membrane. As a research result of the total phenol contents for each vinegar, the persimmon vinegar was the highest as 237.49 mg% and the antibacterial activity was increased when the total phenol contents were high.

Optimization of Extraction Conditions from Hericium erinaceus by Response Surface Methodology (반응표면분석법에 의한 노루궁뎅이 버섯 에탄올 추출조건의 최적화)

  • Response surface methodology (RSM) was used to monitor the characteristics of ethanol extracts from Hericium erinaceus. A central composite design was applied to investigate the effects of independent variables, ethanol concentration $(X_1)$, and sample ratio $(X_2)$ on dependent variables, soluble solid $(Y_1)$, total phenols $(Y_2)$, crude protein $(Y_3)$, electron donating ability $(Y_4)$, and browning color $(Y_5)$ of the extracts. As the sample ratio increased, the soluble solid content increased. Ethanol concentration played a minor role. Total phenols and crude protein increased with sample ratio. Sample ratio had a greater effect than alcohol concentration in the extraction of soluble solid, total phenols, crude protein, and browning color, with an exception of electron donating ability. The optimum ranges at 2 hr extraction was $3.9{\sim}5.0\;g/l00\;mL$ in sample ratio and $36{\sim}52%$ in ethanol concentration. Predicted values at the optimized conditions were acceptable when compared to experimental values.

Influence of Hexaconazole on Biochemical Constituents of Groundnut (땅콩의 생화학성분에 미치는 Hexaconazole의 영향)

  • In this study, hexaconazole 5% SC, an ergosterol biosynthesis inhibitor, was tested on groundnut with its recommended ($500\;mL\;ha^{-1}$) and higher ($2,000\;mL\;ha^{-1}$) concentrations under greenhouse conditions in India. Its influence on biochemical constituents of groundnut plants was assessed apart from its disease management potential against late leaf spot caused by Phaeoisariopsis personata (Berk and Curt). Likewise, leaf samples were collected from hexaconazole 5% SC-sprayed plants at different time intervals. Thereafter, their analyses showed considerable differences in the plant constituents, such as chlorophyll, soluble protein, and total phenol contents and the activity of nitrate reductase enzyme. The induction activity of defense-related enzyme, peroxidase, was also analyzed. However, no difference was observed in the isozymic pattern. Moreover, the ground kernels collected from treated plants also showed no difference in the estimated carbohydrate and other constituents.

Effect of Exogenous Application of Salicylic Acid or Nitric Oxide on Chilling Tolerance and Disease Resistant in Pepper Seedlings (외생 살리실산과 일산화질소 처리가 고추묘의 저온 내성 및 병 저항성에 미치는 영향)

  • As an abiotic stress, chilling stress is one of the major factors limiting plant growth and increasing susceptibility to pathogens. Therefore, enhancing stress tolerance in plants is an important strategy for their survival under unfavorable environmental conditions. The objective of this study was to determine the effects of the exogenous application of salicylic acid (SA) or nitric oxide (NO) on chilling tolerance in pepper seedlings. Pepper (Capsicum annuum L. 'kidaemanbal') seedlings were grown under normal growing conditions ($20/25^{\circ}C$, 15 hours photoperiod, $145{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$, fluorescence lamps) for 23 days after transplanting. The solution (3 mL) of 1 mM SA and 0.3 mM NO with surfactant triton 0.1% were sprayed two times a week, respectively. Right after the completion of chemical application, seedlings were subjected to chilling condition at $4^{\circ}C$ for 6 hours under dark condition and then the seedlings were recovered at the normal growing conditions for 2 days. In order to assess plant tolerance against chilling stress, growth characteristics, chlorophyll fluorescence (Fv/Fm), and membrane permeability were determined after chilling stress imposition. Total phenolic concentration and antioxidant capacity were measured during the whole experimental period. Disease incidence for pepper bacterial spot and wilt was also analyzed. Pepper seedlings treated with SA or NO were maintained similar dry mass ratio, while the value in control increased caused by chilling stress suggesting relatively more water loss in control plants. Electrolyte leakage of pepper seedlings treated with SA or NO was lower than that of control 2 days after chilling treatment. Fv/Fm rapidly decreased after chilling stress in control while the value of SA or NO was maintained about 0.8. SA increased higher total phenolic concentration and antioxidant capacity than NO and control during chemical treatment. In addition, increase in total phenolic concentration was observed after chilling stress in control and NO treatment. SA had an effect on the reduction of bacterial wilt in pepper seedlings. The results from this study revealed that pre-treatment with SA or NO using foliar spray was effective in chilling tolerance and the reduction of disease incidence in pepper seedlings.

Antioxidant and Mixture Effects of Curry Spices Extracts Obtained by Solvent Extraction (용매에 따른 카레 향신료 추출물의 항산화 효과 및 혼합효과)

  • This study was performed to investigate the antioxidative effects of solvent extracts of eighteen curry spices. Yields of the curry spices extracted by methanol, ethylacetate and hexane were $6.4{\sim}42.9%$, $3.9{\sim}26.2%$ and $1.6{\sim}29.2%$, respectively. Methanol, ethylacetate and hexane extracts of spices were added up to 1,500 ppm in the rice bran oil and antioxidative effects of extracts were tested by Rancimat. Induction periods of spices extracted by methanol were in the order of rosemary(33.5hr)>sage(29.1hr)>ginger(28.6hr)>turmeric(26.9hr)>nutmeg(25.8hr)>oregano(25.6hr)>thyme(25.3hr)>BHT(24.5hr)>control(22.6hr). Those of ethylacetate extracts were as follows: rosemary(43.9hr)>sage(30.9hr)>oregano(29.0hr)>thyme(27.5hr)>ginger(27.1hr)>BHT(24.5hr)>control(22.6hr). Those of hexane extracts were in the order of rosemary(47.4hr)>sage(31.4hr)>oregano(27.3hr)>ginger(25.5hr)>thyme(25.1hr)>control(22.6hr). Solvent extracts of rosemary, sage, oregano, ginger and thyme showed significant antioxidative effects. Two thousands ppm hexane-extract of rosemary was the most effective. Addition of hexane-extracts of rosemary didn't show any synergism with the hexane extracts of sage, oregano, ginger, and thyme.
