  • aA series of experiments were conducted to investigate the effect of plant growth regulator treatments on the growth and lateral root formation in soybean sprouts in order to establish the effective method of producing root-less or short-rooted soybean sprouts with larger diameter in the hypocotyl. Major results can be summarized as follows. 1. Soybean sprouts showed fairly uniform elongation rate from 3 to g days after imbibition with daily increase of 3.8cm. The speed of elongation of hypocotyl was reduced whereas that of root accelerated 7 days after imbibition. Lateral roots began to emerge fairly evenly from 5 to 9 days after imbibition with a daily increase of 4.4. 2. Auxins(IAA, IBA, NAA, 2,4-D) inhibited hypocotyl elongation and formation of lateral roots and increased hypocotyl diameter without influencing root length and hook diameter at higher concentrations. The dry weight of cotyledon was increased significantly as compared to that of hypocotyl and root. Among the tested auxins, 2, 4-D was the most effective. 3. BA and 4PU-30 significantly reduced elongation of hypocotyl and root and resulted in the biggest diameter of hypocotyl when treated at higher concentrations. The lowest effective concentration of BA to prevent the formation of larval gal roots was 12.5ppm. The formation of lateral roots could be completely prevented by BA and 4PU-30 treatment but kinetin, zeatin, zeatin riboside resulted in many lateral roots and increased thickness of soybean sprouts with little influence. Cotyledon deformation was found in soybean sprouts treated by 4PU-30. 4. 2, 4-D was the most effective for increasing the hypocotyl diameter while 4PU-30 was the most effective for reducing no. of lateral roots. 5. It can be concluded that among the plant growth regulators tested, BA was effective in reducing root length and increasing hypocotyl diameter. BA 12.5 ppm or 15 ppm may thus be the more practical for production of soybean sprouts. 6. ABA showed no significant effect of growth parameter, however ABA 25 ppm inhibited only no of lateral roots with little influence on the growth of seedling. 7. Ethephon inhibited the elongation of hypocotyl and root and increased hypocotyl diameter at higher concentrations. 8. The combined effect of cytokinins and ethephon was very similar to result of BA treatment alone. As the ethephon concentration increased, hypocotyl diameter and dry weight of cotyledon tended to increase.

  • This paper has shown the experimental results about the physiological activities of water-, ethanol-, ethyl acetate-soluble fractions from ethanolic extracts of leaves, stems and roots of Cudrania tricuspidata on the skin, which has been used for a long time as a traditional herb medicine in Korea and China. The effects of these fractions on the secretion of nitric oxide and cytokines from macrophage(RAW 264.7 cell) exhibited that the ethyl acetate and water fractions from leaves inhibited the release of nitric oxide, all fractions inhibited thoses of inflammatory cytokine $IL-1{\alpha}$, and the ethyl acetate fractions of leaves, stems and roots inhibited thoses of inflammatory cytokine IL-6. The only ethylacetate fraction of leaves demonstrated significantly the reduction of melanin synthesis in melanoma cells. In order to evaluate the efficacy of collagen synthesis, the treatment with extracts on the human normal fibroblast cell(CCD-986sk cell) resulted in finding that the water fractions of leaves, stems and roots and the ethanol fractions of leaves and stems showed the increased synthesis of collagen.

Effects of $Cu^{++}-Catalyzed$ Peroxidation on Collagen Gelation ($Cu^{++}-Catalyzed$ Peroxidation이 Collagen Gelation에 미치는 영향)

  • reactive oxygen species에 의해 나타나는 reactivity에 있어서 metal ions이 관여함이 시사되고 있다. 이미 알려진 reactive oxygen species와 metal ions의 상호작용 이외에 특히 $Cu^{++}$$H_2O_2$가 강력한 peroxidative action을 나타낸다는 사실이 알려져 있으며 $Cu^{++}-H_2O_2$가 biological system 에서의 조직파괴에 관여할 가능성이 저자들에 의해서 효소 및 조직치 구조 단백질의 gradation 효과를 관찰함으로써 시사되었다. 본 연구는 $H_2O_2$ 혹은 $H_2O_2$를 생성하는 효소계(xanthine과 xanthine oxidase 및 glucose과 glucose oxidase)에 $Cu^{++}$을 첨가하여 $Cu^{++}-H_2O_2$에 의한 peroxidation의 효과를 collagen gelation을 통하여 확인코저 수행하였으며 다음과 같은 결과를 얻었다. 1) $Cu^{++}(20\;{\mu}M)$$H_2O_2$에 의하며 collagen gelation은 현저히 억제되었으며 이같은 억제효과는 양자의 농도에 비례하였다. 2) $Cu^{++}-H_2O_2$ reactivity를 확인하는 다른 방법으로 glucose oxidase system를 이용하였다. glucose oxidase$(2.5{\mu}g/ml)$ 와 glucose(0.5 mM)는 collagen gelation에 영향을 미치지 않았으나 이에 $Cu^{++}$이 존재하면 gelation이 억제되었다. 이때 억제정도는 $glucose(0.125{\sim}l.25\;mM)$$Cu^{++}$의 농도에 비례하였다. 3) 여러 reactive oxygen species 가운데 $Cu^{++}-H_2O_2$ reactivity를 xanthine oxidase system을 이용하여 확인하였다. (a) collagen gelation은 xanthine oxidase(30 munits/ml)와 xanthine$(0.25{\sim}2\;mM)$에 의하여 억제되었다. (b) 이때 나타나는 collagen gelation의 억제는 superoxide dismutase에 의하여 완전히 회복되었으나 catalase에 의해서는 더욱 촉진되었다. 그러나 catalase에 의한 억제효과의 촉진은 1,4-diazabicyclo(2,2,2)octane에 의하여 완전히 소실되었다. 따라서 이 xanthine oxidase system에서는 $O_2-,\;H_2O_2,\;^1O_2$이 관여함을 알 수 있었다. (c) 그러나 $Cu^{++}(10\;{\mu}M)$이 존재하였을 때 collagen gelation은 superoxie dismutase에 의해 더욱 억제되었고 catalase에 의해서는 완전히 회복되었다. xanthine oxidase계에서 얻어진 결과는 여러 reactive oxygen species가운데 $H_2O_2$$Cu^{++}$에 의하여 peroxidation효과를 나타냄을 알 수 있었다. 이상의 결과로 미루어 볼 때 reactive oxygen species와 metal ions과의 상호작용 가운데 $Cu^{++}-H_2O_2$는 강한 반응을 나타내는 특이한 구성요소이고 헌재 시사되고 있는 reactive oxygen species의 biological effects에 비추어 $Cu^{++}-catalyzed peroxidation$도 병적상태에서 생체에 유해한 작용을 나타내는 요소임을 시사하며 특히 염증시 조직파괴역할에 관하여 고찰하였다.

  • Hepatocyte growth factor (HGF) is a mesenchymal-derived cytokine. It exerts a motogenic effect on various target cells, which is displayed either by cell scattering, locomotion, and migration during the wound repair process of cultured cells, or invasiveness through the extracellular matrix, in vitro. Although it is known that HGF influences the motogenic effect of endothelial cells, the precise effects of HGF during migration are still poorly understood. To elucidate the role of HGF in endothelial cell migration, the effect of HGF on endothelial cell migration and MMPs and plasmin production were studied. We found that HGF induces the migration of cultured endothelial cells through increased MMPs and plasmin secretion.

Determination of Antifungal Activity on Pepper Anthracnose and Plant Growth Promoting Activity of Pleurospermum camtschaticum Root Extract (누리대 뿌리 추출물의 고추 탄저병에 대한 항균 및 생장 촉진 활성 검정)

  • This study was conducted to confirm the utilization of Pleurospermum camtschaticum root extract as an organic agricultural material. Antioxidant activity of P. camtschaticum root extract, closely related to antibacterial activity, increased in a dose-dependent manner. In mycelial growth inhibitory activity, 100% P. camtschaticum root extract supressed over 70% for Colletotrichum coccodes and over 68% for Colletotrichum dematium. In the pepper fruit anthracnose development test, the size of the lesion decreased in a dose-dependent manner, which showed the same tendency as the previous results in inhibitory activity on mycelial growth. In the pepper seed germination and red pepper growth promotion test of P. camtschaticum root extract, oposite results was confirmed. The lower the concentration, the more the seed germination and growth promotion effects were shown. The phenol content of pepper leaves was also measured after pepper growth promotion test have been completed. The phenol content related to antibacterial activity increased in all treated groups compared to the untreated group. Therefore, the results of this study showed the possibility of development as an organic material.

  • L-Ascorbic acid 및 그 유도체는 항산화 작용과 미백작용 및 섬유아세포의 생장 촉진과 콜라젠 생합성의 증가시키는 효과가 있으므로 화장품에서 오래 전부터 사용되어 왔다. 본 연구에서는 생체에 대한 안전성과 안정성이 우수한 L-Ascorbic acid 유도체를 개발하기 위하여 인지질과 유사하게 L-Ascorbic acid 에 3-Aminopropane phosphoric acid를 결합하여 L-Ascorbic acid-3-aminopropane phosphoric acid diester를 합성하였다. ASA-APPA 는 2-Chlorotetrahydro-2H-1, 3, 2-oxazaphosphorine P-oxide 와 5, 6-isopropylidene L-Ascorbic acid 를 반응 시킨 후 산 사수분해 반응을 통하여 얻을 수 있었다. ASA-APPA는 수용액에서 안정성이 우수하였으며, 독성실험에서 무독성 물질이며, 인체 첩포 실험에서도 무자극 물질임을 확인하였다. 그리고, ASA-APPA 는 L-Ascorbic acid 와 3-APPA 의 혼합물과 거의 유사한 섬유아세포의 증식효과를 나타내었으며, melanoma 에 대한 멜라닌 생성 억제 실험에서 L-Ascorbic acid phosphate magnesium salt 와 유사한 효과를 보였다. 따라서 ASA-APPA 는 멜라닌 생성 억제 효과와 섬유아세포의 증식 효과를 갖는 새로운 화장품 원료로서 사용이 가능할 것으로 생각된다.

Effect of spent mushroom substrates on Phythopthora Blight disease and growth promotion of pepper (버섯 수확후배지의 고추 생육촉진 및 역병 억제 효과)

  • Water extracts from spent mushroom substrate (SMSE)of edible mushrooms, Pleurotus eryngii, Hericium erinaceus and Lentinula edodes promoted growth of pepper seedling. Mycellial growth rate of Phythopthora capsici and Fusarium oxysporum was dramatically inhibited by 100% and 70% on PDA added with SMSE of H. erinaceus. SMSEs from H. erinaceus, P. eryngii, and L. edodes effectively reduced the disease severity of Phytophthora blight of pepper caused by Phytophthora capsici to 75%, 10% and 35%, respectively. These results suggested that SMSE from the mushrooms have dual effects that suppress phythopthora blight disease and promote plant growth of pepper.

  • This experiments were conducted to investigate the effect of plant growth regulator treatments on the ethylene evolution in soybean sprouts. Major results can be summarized as follows. 1. IAA showed no significant effect on the ethylene evolution in soybean sprouts. 2. BA increased significantly ethylene evolution while kinetin, zeatin, zeatin riboside were less effective on the ethylene production in soybean sprouts. 3. As the BA concentration was increased, the ethylene evolution was increased. 4. BA increased ethylene production more effectively at earlier stages while 4PU-30 was more effective for ethylene production rather continuously. 5. The order of effectiveness of cytokinins and auxin on the ethylene evolution was as follows : BA 25 ppm>BA 25ppm+IAA 25ppm${\geq}$4PU-30 25ppm>IAA 25ppm>control. 6. It can be concluded that BA was the most effective in inhibiting lateral roots, among the cytokinins tested, and resulted in the biggest diameter of hypocotyl by the ethylene evolution stimulated by BA treatment.

Effects of Staurosporine and Genistein on Superoxide and HOCl Production in C5a- or PMA-activated Neutrophils (Staurosporine과 Genistein이 C5a 또는 PMA에 의하여 활성화된 호중구에서의 Superoxide와 HOCl 생성에 나타내는 영향)

  • Effects of staurosporine, genistein and pertussis toxin on superoxide and HOCl production in C5a- or PMA-activated neutrophils were investigated. A C5a-induced superoxide and $H_2O_2$ production was inhibited by staurosporine, genistein and pertussis toxin. The stimulatory effect of PMA was inhibited by staurosporine but was not affected by pertussis toxin, whereas it was further promoted by genistein. Staurosporine and genistein inhibited superoxide production by sodium fluoride, but pertussis toxin did not affect it. PMA-induced $H_2O_2$ production was inhibited by staurosporine but was not affected by pertussis toxin. Genistein did not show a stimulatory effect on PMA-induced $H_2O_2$ production. Staurosporine and pertussis toxin inhibited HOCl production by C5a- or PMA, whereas genistein stimulated it. C5a-or PMA-induced myeloperoxidase release was inhibited by genistein, in this response the effect of pertussis toxin was not detected. Staurosporine did not affect the stimulatory effect of PMA on the release. Myeloperoxidase activity was markedly increased by genistein but was not affected by staurosporine and pertussis toxin. These results indicate that the respiratory burst of neutrophils may be regulated by protein kinase C and protein tyrosine kinase. Superoxide production induced by the direct activation of protein kinase C might be affected by protein tyrosine kinase oppositely. Genistein probably pro-motes HOCl production by activating myeloperoxidase.
