Anti-oxidative and skin barrier effects of natural plants with a supercritical extract (초임계 추출을 적용한 식물추출물의 항산화 및 피부장벽 효과)

  • In this study, we searched for bioactive compounds from natural resources with a supercritical extract. We selected the extracts of Chrysanthemum zawadskii, Lufa cylindrica, Paeonia lactiflora, Gardenia jasminoides and Scutellaria baicalensis, as natural materials, and evaluated the effects of their skin barrier function. We found that these extracts increased the transactivation activity of the PPAR-responsive element (PPRE) and the anti-oxidation with different priorities, respectively. In addition, these extracts promoted the expression of proteins related to cornified envelope (CE) formation, such as involucrin. From these results, we suggest that natural materials from supercritical extracts will be pertinent candidates for the improvement of the epidermal permeability barrier function.

The Effects of Supercritical Carbon Dioxide on the Extraction of Perilla Oil (초임계 이산화탄소가 들기름의 추출에 미치는 영향)

  • This study was performed on the effects of extraction temperature, pressure, time on apparent solubility and extraction yield of perilla oil and tocopherol, and color and fatty acid composition of the residue in supercritical carbon dioxide $(SC-CO_2)$ extraction. Apparent solubility of perilla oil and tocopherol increased with the increase of $CO_2$ density and was found to strongly depend on extraction pressure rather than extraction temperature. The extract yield of tocopherol in $SC-CO_2$ extraction increased with an increase of temperature and decreased with an increase of pressure and extraction time. The perilla oil apparent solubility of dried perilla powder for $60\sim180$ min at $40^{\circ}C/276$ bar increased with an extraction time, on the other hands, tocopherol apparent solubility decreased. As the increase of $CO_2$ density, less redness and yellowness increased. Fatty acid composition of perilla oil showed that perilla oil extracted by $SC-CO_2$ had better unsaturated fatty acid and decreased in saturated fatty acid. $SC-CO_2$ extraction offers a safe natural method for gaining perilla oil from dried perilla seeds powder.

  • Supercritical fluid(SCF) carbon dioxide was used to extract safflower yellow pigments from Carthamus tinctorius L. In this work, supercritical fluid extractions were performed at various conditions; pressure (2000, 3000, 4000, 5000 psig), temperature $(40,\;50,\;60,\;70,\;80^{\circ}C)$ and co-solvent $(0,\;3,\;6,\;10,\;14\;wt%\;H_2O)$. Total concentrations of safflower yellow pigments extracted were determined by spectrophotometric method. A maximum yield of yellow pigments was obtained at 4000psig, $60^{\circ}C$ and 10% co-solvent. The extraction yield of pigments was also closely related to moisture content of the raw material. Extraction yield of safflower yellow pigments by SCF extraction at optimized conditions was 6% higher than that by solvent extraction. Supercritical carbon dioxide was proved to be suitable for the extraction of safflower yellow pigments from Carthamus tinctorius L.

Lipolytic Effect of Supercritical Extraction from Pine Cone (Pinus koraiensis) in Mature 3T3-L1 Adipocytes (분화된 3T3-L1 세포에서 잣송이 초임계 추출물의 지방분해 효과)

  • Seeds of Korean pine cone (Pinus koraiensis) have long been consumed as an edible food in countries located in North-East Asia, On the other hand, Korean pine cone, containing various polyphenols, is discarded as a useless garbage after removing seeds. This study investigated the lipolytic effects of pine cone extract in differentiated 3T3-L1 adipocytes. Intracellular lipid accumulation was measured by Oil red O staining, free glycerol release by colorimetric reaction, and expression of genes related to lipid metabolism by real-time PCR. Compared to control, pine cone extract reduced intracellular lipid accumulation by 8.8% and increased free glycerol release by 8.2% a concentration of $5{\mu}g/mL$ in differentiated 3T3-L1 adipocytes. mRNA levels of fatty acid synthesis were not significantly different between control and pine cone extract, but mRNA levels of lipoprotein lipase (LPL) and hormone-sensitive lipase (HSL) significantly increased by 38.7% and 94.1% at a concentration of $5{\mu}g/mL$, respectively. Thus, pine cone extract is suggested to have lipolytic effects through induction of LPL and HSL gene expression.

The Chemopreventive Effect of Heat-treated Radish Complex Extracts on Rat Colonic Aberrant Crypt Foci Induced by 1,2-Dimethylhydrazine (1,2-Dimethylhydrazine에 의해 유발된 Colonic Aberrant Foci에 대한 초임계 열처리된 무복합추출물의 대장암 효과)

  • The present study was designed to investigate the chemopreventive effects of supercritical heat radish extracts with anti-constipation acitivity in loperamide-treated rats, on 1,2-dimethylhydrazine(DMH)-induced aberrant crypt foci(ACF) in rat colonic mucosa. Supercritical heat radish extracts was administered in the drinking water(3.2 mg/ml) during the initiation (for 12 weeks) or postinitiation phase(for 9 weeks) of DMH-induced carcicogenesis. Supercritical heat radish extracts coupled with DMH for 12 weeks decreased significantly total number of aberrant crypts(pre-neoplastic lesions) and the number of ACF to 35% and 30%, respectively. The number of ACF with more than four aberrant crypts (crypt multiplicity) was significantly reduced in the colon of rats receiving Supercritical heat radish extracts for 12 weeks. In the group receiving Supercritical heat radish extracts after post-initiation for 9 weeks, only the number of ACF with ${\geq}4$ abberant crypts significantly decreased. Supercritical heat radish extracts, given alone for 12 weeks, did not induced the development of ACF. These results indicate that Supercritical heat radish extracts possess chemopreventive effects on chemically induced rat colon carcinogenesis.

Phospholipids Isolation from Squid Viscera Residues After Supercritical Carbon Dioxide Extraction (오징어 내장의 초임계 이산화탄소 추출 잔류물로부터 인지질의 분리)

  • Phospholipids were recovered from squid viscera residues by ethanol extraction after supercritical carbon dioxide($SCO_2$) extraction and from squid viscera was not processed $SCO_2$ by various organic solvent extraction. $SCO_2$ extraction were performed at $45^{\circ}C$ and 20 MPa for removal of non polar lipid molecules from freeze dried squid viscera sample. Phospholipids were extracted from freeze dried squid viscera sample by chloroform, hexane, methanol, and ethanol and from $SCO_2$extracted squid viscera sample by ethanol. The pH was fixed at 5.7 for all phospholipids extraction conditions. Phospholipid classes were analyzed by HPLC equipped with evaporative light scattering detector (ELSD). Phosphatidyl choline(PC) extracted by ethanol from $SCO_2$ extracted residues was higher than that of extracted by ethanol from squid viscera. But phosphatidyl ethanolamine(PE) and phosphatidic acid(PA) were extracted higher percentage in raw squid viscera. The fatty acid compositions in phospholipids extract by ethanol extract from $SCO_2$ extracted residues were analyzed by gas chromatography(GC). Docosahexanoic acid(DHA) was found in highest percentage in phospholipid extract.

Supercritical Extraction of Oriental Herb : Anti-aging and Anti-wrinkle Effects (한방원료의 초임계 추출을 이용한 항노화 및 주름개선 효과)

  • The reactive oxygen species generated by ultraviolet rays causes various types of cutaneous damage, such as lipid peroxidation and denaturation of the extra-cellular matrix. The accumulation of such damage contributes to skin aging, especially the formation of wrinkles. This study was carried out to develop functional cosmatic by using Oriental herb supercritical extracts (OHSE) for prevention of skin. Effects of OHSE on anti-oxidation, collagenase inhibition and collagen synthesis in normal human fibroblast were investigated. OHSE showed antioxidative activity as high as vitamin C, trolox and DL-penicillamine. Also OHSE showed promotive effect on collagen synthesis and inhibitory effect on collagenase activity. From this results, we conclude that OHSE may have the potential to be conveniently used as an additive in cosmetics for prevention and improvement of skin aging.

Supercritical $CO_2$ Extraction of Genistein from Soybean (초임계 $CO_2$를 이용한 대두 Genistein의 추출)

  • This study was carried out to examine some factors affecting the supercritical carbon dioxide extraction of genistein from soybean. The factors investigated in this study were pressure, temperature, flow rate of carbon dioxide, and concentration of modifier. It was foumd out that genistein is not extracted in the absence of modifier. Ethanol was found to be more effective modifier than methanol. 70% of genistein was extracted at 35$^{\circ}C$, 300bar and ethanol 15% (w/v) as compared with the performances of organic solvent extraction.

  • This study is about a technique for obtaining collagen by extracting fat by treating collagen-containing liposuction effluent in the presence of supercritical fluid. Using a supercritical solvent, a collagen extract could be obtained from animal-derived fat in a short time (about 6 hours), and about 2-3% of collagen by mass compared to the raw material could be obtained. The presence of collagen in the extract obtained by supercritical extraction was confirmed by SDS-PAGE, and it was confirmed that it was type 1 collagen having a relatively large molecular weight. In addition, the growth factors of IGF-1, bFGF, VEGF and NGF were analyzed to find out which growth factors were present in the collagen obtained by supercritical extraction, and it was found that these growth factors were contained in the extract. There was no significant difference in DNA content per mg of sample before and after supercritical treatment. Further in-depth studies are likely to be needed on decellularization technology using the supercritical process. In conclusion, the extracellular matrix obtained through the solvent extraction process using a supercritical fluid contains growth factors above a certain amount even after decellularization and removal of fat, so that it was found that not only biocompatibility is greatly increased, but also tissue regeneration can be rapidly induced.

Supercritical Fluid Extraction of Physiologically Active materials from Agaricus blazei Fruiting Bodies (Agaricus blazei 자실체로부터 초임계 유체를 이용한 생리활성물질 추출공정)

  • The supercritical fluid extraction(SFE) technique was applied for the isolation and purification of nonpolar physiologically acitve mateials from Agaricus blazei fruiting bodies. the qualitative analysis of extract was accomplished by gas chromatography-mass spectrometer(GC-MS) and extract was determined as linoleic acid(cis-9 cis-12-octadecadienoic acid) In order to obtain the optimum operating conditions of supercritical fluid extraction process the various temperatures and pressures were applied for process operation. From the comparison of exraction efficiencies $50^{\circ}C and 200 kg_f/cm^2$ were determined as optimum conditions.
