광물질 (III) -나트륨(Na), 카리(K), 염소(CI)

  • 광물질 중에서 칼슘(Ca)과 인(P) 다음으로 동물체내에 많이 함유되어 있는 광물질이 나트륨(Na), 카리(K) 및 염소(CI)이다. 칼슘과 인이 주로 골격에 많이 함유되어 있는데 비해서 이들 3가지 원소들은 주로 체액(혈액 등)과 연초지에 널리 분포되어 있다. 동물체내에서의 이들 세가지 원소들의 분포와 관련된 한가지 특징적인 사실은 카리(K)는 주로 세포내에 존재하고 나트륨(Na)과 염소(CI)는 주로 세포밖의 체액에 존재한다는 것이다. 사실상 혈청내의 염기의 93$\%$가 나트륨일 정도로 나트륨은 혈액을 구성하는 중요한 광물질이다.

  • By means of thin-layer electrophoresis in agarose gel, hemolymph protein of healthy silkworm larvae and of the nuclear polygedrosis virus infected larvae were studied. 1. In the 4th instar, 4 fractions moving toward anode were separated. Dye-binding Capacity of the fraction was increased according to the stage. 2. After 5th day in the 5th instar, 7 fractions moving toward anode were separated, and one fraction toward cathode was separated. 3. On the first day in the 5th instar, 5 fractions were separated, and on the 4th day of the same instar 5 fractions were separated. 4. As for the hemolymph protein fractions of the polyhedrosis virus infected larvae, on the 6th and 7th day, three fractions(D.E.F) were inclined to increase, whereas on the 8th day 4 fractions(A.B.D.E) were disappeared but F fraction was inclined to decrease.

Effect of Dietary Hot Red Pepper Powder on Humoral Immune Response in Rat (고춧가루 첨가 식이가 흰주의 체액성 면역기능에 미치는 영향)

  • Hot red pepper(Capsicum annum L.) has been extensively used as a spicy food additive and preservative in Korea. In this study, we investigated the effect of dietary hot red pepper powder on humoral immune response in rats. Sprague-Dawley rats were divided into 4 groups and fed experimental diets containing 0, 2, 5, 10% hot red pepper powder for 27 days. All groups were immunized with sheep red blood cells. In order to measure the immune response, plaque-forming cell number, agglutination titer, and serum antibody level were measured. Tissue ascorbic acid contents were also determined by high-performance liquid chromatogrphy. There was an increased plaque-forming cell number, agglutination titer, and serum IgG level in the groups supplemented with hot red pepper powder as compared to control. Tissure ascorbic acid contents in the hot pepper powder supplemented group were higher than those of control. The results suggest that the dietary hot red pepper powder enhances humoral immune response in rats, indicating that the hot pepper contains biological response modifier.

  • The experiment was carried out to investigate the effects of juvenile hormone analogue on changes of protein and amino acids in haemolymph of silkworm larva. Juvenile hormone analogue was topically administered to larvae at dose of 1$\mu\textrm{g}$ 10$\mu\textrm{g}$ per gm of body weight at 60hr. of the 5th instar. The results obtained were as follows ; 1. Larval duration of the fifth instar was extended about 1 day by JHA-1 and 4 days by JHA-10 as compared with the control. 2. Cocoon weight and cocoon layer weight by topical application of JHA were heavier than those of the control, but cocoon layer ratio was decreased in JHA-10 to the exclusion of JHA-1. 3. The concentration of haemolymph protein during the fifth instar was increased remarkably by the JHA application. 4. The total content of amino acids in haemolymph proteins of JHA-10 approximately doubled that of the control, with the conspicuous increase of glycine and arginine level.

  • Isolation, purification and molecular properties of adult specific protein (ASP) were investigated in the silkworm, Bombyx mori. ASP was purified from adult haemolymph by gel filtration on Sephadex G-100 and ion exchange chromatography on CM52. The preparation was shown to be homogeneous by native-PAGE and immunoelectrophoresis. The purified protein was consisted of two different subunits with molecular weights of 19.5kDa and 17.5kDa, and contained no sugars and no lipids.

  • Dietary supplementation of folic acid to silkworm larvae in different concentration (100, 200 and 300 ${\mu}$g/ml) to the 4th and the 5th instar resulted in a significant increase in economic parameters, such as female and male cocoon weights, shell weights, egg productivity and egg hatching percentage. However, larval duration was significantly decreased, as compared with that of carrier control. The increase in the glycogen and protein contents of the fat body was not significant, whereas the increase in trehalose content of haemolymph in all the treated groups and protein content of fat body and haemolymph in 100 ${\mu}$g/ml treated groups were significant. However, the haemolymph protein was significantly decreased in 300${\mu}$g/ml treated group.

  • Insect cells were grown and infected with baculovirus for the production of recombinant protein. Later infection gave the lower expression of recombinant protein. This indicates that the expression rate is lower at higher cell concentration. This phenomena provides a well-posed optimization problem with respect to the infection time. The optimal infection time was experimentally shown to exist for the maximum productivity of recombinant protein. Also, the expression increased with the addition of 5% silkworm hemolymph. This is considered to be due to the increase of intracellular viruses and the longevity of viable cells after the infection. The production of ${\beta}$-galaclosidase increased about ten-fold with the addition of yeastolate and silkworm hemolymph for high cell density and high expression, respectively.

Detection of Mycobacterium tuberculosis by PCR from Trace Clinical Specimens and Paraffin-embedded Tissues (임상가검물과 파라핀 포매 조직에서 PCR법을 이용한 결핵균의 검출)

  • This study has been carried out to investigate the sensitivity of polymerase chain reaction (PCR) method over conventional acid-fast bacilli (AFB) staining and/culture methods for the detection of Mycobacterium tuberculosis from trace body fluid and paraffin-embedded tissues (PET) specimens. A total of 65 cases were employed for the AFB staining and culture test, and a total of 50 cases were subjected to PCR and histopathological analysis. Among the specimen showing negative reaction to AFB staining, 12.1% were positive to PCR and 3.7% of the specimen representing negative result to culture test showed positive reaction to PCR. In addition, 20.0% of the specimen with AFB negative showed positive reaction to PCR. From these results, it could be concluded that PCR method overwhelms AFB staining and culture tests in sensitivity and specificity to M tuberculosis detection.

Flavor Modification of Mideoduck (Styela clava) Drips by Maillard Reaction (Maillard 반응에 의한 미더덕 체액의 풍미개선)

  • Mideoduck drips were mixed with amino acids (Met, Tau, Gly, Ala, Thr, Cys), thiamine and sugars (Glucose, Ribose) for flavor modification and evaluation using the Maillard reaction. To mask the seafood flavor, onions, spring onions, garlic, ginger, citric orange and green tea were mixed with Mideoduck drips at $160^{\circ}C$ for 2.5 hr in a stainless still reaction bomb. The glucose/thiamine model reaction system was estimated to be lower than the ribose/thiamine model system, and an extreme case is the ribose/Met model system. Mixed system of glucose, ribose and taurine containing sulfur compounds showed fair results. Among the Mideoduck drips mixed with sugars and amino groups, only thiamine model systems were estimated to be normal. The flavor composition of Mideoduck drips/sugars model system, and long chain fatty acids were composed of 31.32~62.71% total flavor content. The 1,2-benzenedicarboxylic acid dibutylester contents made up more than 20% of the model system in groups A, B and C. From the model system in this study, drip/glucose, drip/ribose, drip/glucose/citric orange, and drip/glucose/glycine/cystine groups showed most intense good flavor.

  • Ion concentrations and osmolarities of the body fluid and oxygen consumption of the larvae of the freshwater crab, Eriocheir japonicus under the influence of various salinities and water temperatures were studied. The levels of $Na^+\;and\;Cl^-$ ions in the body fluid of the crab placed in the low salinities (freshwater and $30\%$ seawater) were higher at the low temperature $(16^{\circ}C)$ than high temperature $(30^{\circ}C)$. However, the crab in the high salinity $(90\%\; seawater)$, those ions in the body fluid were higher at the high temperature $(30^{\circ}C)$ than the low temperature $(10^{\circ}C)$ When the zoea larvae were young (first and second stages), the ion concentrations of the body fluid were more affected by salinities and temperatures of the media. The salinity levels of the media more influenced the levels of $K^+$ ions of the body fluid of the larvae than the temperature. The osmolarities of the body fluid of the larvae were almost isotonic when they were placed at $70\%$ seawater $(24.5\%o)$. Oxygen consumption of individual larva was lower in zoea 1 and 2 stages than the other stage larvae. The high oxygen consumption of the larvae was observed at $70\%$ seawater compared to the other salinity levels.
