Quality Characteristics of Bread Made with Chungkukjang Powder (청국장 가루를 첨가한 식빵의 품질 특성)

  • This study investigated the quality characteristics of bread made with different additions of Chungkukjang powder (0, 10, 20%). In the microbiological evaluations, total aerobic bacteria increased with increases in Chungkukjang powder concentration and storage period ($20^{\circ}C$, 3 days). The pH also increased with increasing Chungkukjang powder concentration. For the color values, with increasing Chungkukjang powder content, the lightness (p<0.05) decreased in both the bread crumb and crust, while redness (p<0.05) increased. The yellowness decreased for the bread crust and increased for the bread crumb. Hardness and chewiness were higher, but springiness and cohesiveness were lower, in the breads made with 10 or 20% Chungkukjang powder as compared to the control. The control bread obtained the highest sensory scores for texture and overall acceptability during the storage period. Optimum sensory scores for color, texture, and overall acceptability were obtained for the bread with 10 % Chungkukjang powder until day 1 of storage. From the results of this study, 10% Chungkukjang powder can be added to bread without diminishing its acceptability, but further study is necessary on extending shelf-life and improving consumer acceptance.

Antimicrobial Activities of Viscous Substance from Chongkukjang Fermented with different bacillus spry. (청국장 발효 세균의 종류에 따른 청국장 정절물의 항 미생물 활성에 관한 연구)

  • To evaluate antimicrobial activities of chongkukjang slime fermented by different strains, growth characteristics were compared using various standard microorganisms with addition of chongkykjang slime. Chonghkjang slime was prepared by fermenting cooked soybean after inoculating with Bacillus circulans K-1, Baciilus spp N-1 and Bacillus subtilis CH-1, respectively. Significant antimicrobial activity was observed by chongkukjang slime on gram positive bacteria (Staphylococcus aureus, Bacillus cereus, Bacillus subtilis, Micrococcus luteus), gram negative bacteria(Escherichia coli O157:H7, Salmonella Typhimurium, Pseudomonas fluorescens), and yeast (Pichia membranaefaciens, Saccharomyces cerevisiae, Candida albicans). In case of B. cereus growth inhibition of 80% was achieved by the addition of chongkukjang slime; on the contrary, to Escherichia coli O157:H7 only 20% inhibition was observed. Slime from Bacillus subtilis CH-1, in particular, inhibition of 40% toward bacteria and yeast, whereas slime from Bacillus circulans K-1, Bacillus spp N-1 showed only 20% inhibition.

  • This study was performed to find the most desirable mixing formula for Sulgidduk added with Cheongkukjang powder for various benefits. The optimal mixing formulas were identified by evaluations of ingredients, the color, and the sensory and textual characteristics. As the amount of Cheongkukjang powder increased, the rice cake appeared redder, yellower and less luminous. The amount of crude ash was significantly smaller in the experimental groups than in the control group, and the amount of crude protein and crude fat was significantly high. The change in texture according to the amount of Cheongkukjang powder was not statistically significant, but hardness, adhesiveness, and cohesiveness showed significant differences(p<0.05). It was also found that chewiness increased while adhesiveness decreased as storage time increased. According to the results of the sensory evaluation, scores in the color and aroma decreased as the amount of Cheongkukjang powder increased. Overall acceptability score was highest in the 5% group, consisting of 68% of rice powder, 5% of Cheongkukjang powder, 10% of sugar, and 17% of water.

Characteristics of Chungkookjang Prepared by Bacillus amyloliquefaciens with Different Soybeans and Fermentation Temperatures (Bacillus amyloliquefaciens를 이용하여 콩 종류와 발효온도를 달리하여 제조한 청국장의 특성)

  • This study was carried out to investigate the characteristics of Chungkookjang depending on different soybeans and fermentation temperatures using Bacillus amyloliquefaciens C2 isolated from homemade Chungkookjang. The highest protease activity was 854 U/g in yellow soybean Chungkookjang and 847 U/g in black soybean Chungkookjang at $35^{\circ}C$. The highest amylase activity was 3.87 U/g at $40^{\circ}C$ in yellow soybean Chungkookjang and 4.96 U/g at $45^{\circ}C$ in black soybean Chungkookjang. The highest reducing sugar content was 16.11 mg/g at $40^{\circ}C$ in yellow soybean Chungkookjang and 19.08 mg/g at $45^{\circ}C$ in black soybean Chungkookjang. The highest amino type nitrogen content was 420 mg%/g in yellow soybean Chungkookjang and 194 mg%/g in black soybean Chungkookjang at $40^{\circ}C$. The highest pH was 7.92 at $40^{\circ}C$ in yellow soybean Chungkookjang and 7.59 at $45^{\circ}C$ in black soybean Chungkookjang. The highest number of viable cell was 9.3 log CFU/g at $40^{\circ}C$ in yellow soybean Chungkookjang and at $35^{\circ}C$ in black soybean Chungkookjang. On the other hand, the lowest ammonia type nitrogen content was 225 mg%/g at $45^{\circ}C$ in yellow soybean Chungkookjang and 80 mg%/g at $40^{\circ}C$ in black soybean Chungkookjang. Yellow soybean Chungkookjang showed high protease activity, pH and amino type nitrogen, whereas black soybean Chungkookjang showed high amylase activity and reducing sugar.

  • Various types of fermented soybean foods are consumed in various Asian countries, including China, Indonesia, Japan, Korea, and Vietnam. Cheonggukjang is a Korean whole soybean paste fermented by Bacillus subtilis and regarded as a healthy food. The objective of this study was to investigate the Ca-bioavailability and anti-osteoporotic effects of cheonggukjang (CGJ) containing high poly-gamma glutamate (PGA) contents in an animal model. Blood samples were collected from the jugular vein at 0, 0.5, 1, 2, 3, 4, and 5 h after a single oral dose in male rats. Ca-bioavailability in CGJ containing high PGA contents was approximately 3~5 times higher than that of natural CGJ. Female rats were either sham-operated (sham; n=5) or surgically ovariectomized (OVX; five animals per group) and then administered to OVX control, raloxifene hydrochloride (RLX) 1 mg/kg/d, or CGJ containing high contents of PGA (20 and 200 mg/kg/d) for 12 weeks. Serum osteocalcin concentration was significantly lower in the CGJ 200 mg/kg/d group compared with the OVX control group. Serum progesterone concentration was significantly higher in the CGJ 200 mg/kg/d group compared with the OVX control group. Reduction grade of the trabecular bone decreased in the RLX 1 and CGJ 200 mg/kg/d group compared with that of the OVX control group. In conclusion, CGJ 200 mg/kg/d may have inhibitory effects on osteoporosis in OVX rats, and Ca-bioavailability was improved in CGJ containing high PGA contents.

Fermentation and Quality Characteristics of Cheonggukjang prepared with Sea Mustard (미역을 첨가한 청국장의 발효 및 품질 특성)

  • Cheonggukjang that was prepared with different concentrations of sea mustard [0% (control), 5% (SM5), 10% (SM10) and 15% (SM15)] were investigated. The pH of cheonggukjangs prepared with sea mustard was higher than that of control during fermentation for 72 hrs at $37^{\circ}C$. The total aerobes of the cheonggukjang variants reached 9.40 (control), 9.45 (SM5), 9.59 (SM10) and 9.59 (SM15) log CFU/g after fermentation for 72 hrs at $37^{\circ}C$. The amino nitrogen and viscocus substance contents of the cheonggukjang variants increased with fermentation time, SM5 showed the highest amino nitrogen content ($397.13{\mu}g/mL$) and viscocus substance content (6.53%) among various cheonggukjang after 72 hrs fermentation. The total polyphenol contents of various cheonggukjangs were increased with the sea mustard increased. The DPPH radical scavenging ability of the cheonggukjang variants were 68.09% (control), 77.91% (SM5), 80.72% (SM10) and 79.40% (SM15) and their ABTS radical scavenging ability were 86.66% (control), 89.01% (SM5), 91.32% (SM10) and 92.27% (SM15) after fermentation for 72 hrs. The sensory characteristics of the cheonggukjangs prepared with sea mustard showed higher than control in taste, flavor and overall acceptability but did not significantly differ.

The Effects of Chungkukjang Powder Supplements on the Regulation of Blood Glucose and Inflammation in Diabetic Rats (청국장 분말 첨가식이가 당뇨 쥐의 혈당과 염증조절에 미치는 영향)

  • In this study, the effect of Cheonggukjang powder were investigated on the regulation of blood glucose and inflammatory in STZ-induced diabetic rats. The experimental diet used this study were three kinds of Cheonggukjang, which were soybean Cheonggukjang, Yakkong Cheonggukjang and black foods such as black rice, black sesame seeds, and sea tangle added Yakkong Cheonggukjang powder. The experimental animals were divided into 5 groups and fed experimental diets for 7 weeks; non-diabetes with normal diet group (C), diabetes with normal diet (DC), diabetes with soybean Cheonggukjang (DS), diabetes with Yakkong Cheonggukjang (DY), and diabetes with Yakkong black foods added Cheonggukjang (DYB). Blood glucose and insulin resistance of STZ-induced diabetic groups were were significantly higher than C group. But insulin levels and insulin secertory of STZ-induced diabetic groups were significantly lower than C group. However, supplementation of Yakkong or black foods added Yakkong Cheonggukjang were proven to regulation them. In diabetic group, free fatty acid level was significantly increased than C group, but this contents was significantly decreased supplementation of soybean Cheonggukjang. Leptin and adiponectin levels were significantly decreased in STZ-induced diabetic groups.

Isolation, Identification, and Fermentation Characteristics of Bacillus sp. with High Protease Activity from Traditional Cheonggukjang (전통 청국장으로부터 protease 분비능이 우수한 Bacillus sp. 균주의 분리 동정 및 발효 특성)

  • Twenty one strains strongly producing protease were isolated from Korean traditional Cheonggukjang. Eight strains selected by sensory evaluation on Cheonggukjang prepared with isolated strains were identified with based on biochemical properties a and 16S rDNA sequencing. Identified strains were Bacillus subtilis MB4, and Bacillus amyloliquefaciens A1, A2, B1, MC1, SB2, SC1, and SD1. Protease activities, important strain selection factor, were higher in Cheongjukjang prepared with B. subtilis MB4 (179.6 Unit) and B. amyloliquefaciens SB2 (201.9 Unit) than commercial traditional Cheonggukjang (97.9 Unit). Sensory evaluation revealed Cheonggukjang prepared with B. subtilis MB4 had flavor very similar to commercial traditional Cheonggukjang. Cheonggukjang prepared with B. suhtilis MB4 (0.0006 Pa s) and commercial traditional Cheonggukjang (0.0002 Pa s) revealed lower viscosities than those of Cheonggukjang prepared with B. amyloliquefaciens SB2, MC1, B1, A1, SD1, A2, and SC1 (0.006 to 0.008 Pa s at 1001/s. Results show Cheonggukjang could be prepared using single strain of B. subtilis MB4, maintaining high protease activity and very similar sensory and viscosity qualities with those of commercial traditional Cheonggukjang.

Effects of Soybean. Cheonggukjang and Doenjang on Serum Cholesterol Level and Weight Reduction in Rats Fed a High-Fat/High-Cholesterol Diet (대두, 청국장 및 된장이 고지방-고콜레스테롤식이를 급여한 흰쥐의 콜레스테롤 함량 및 체중감소에 미치는 영향)

  • This study was conducted to investigate the cholesterol lowering effect and weight reduction of autoclaved soy flour, cheonggulgang and doenjang, which are fermented using Bacillus subtilis DJI, in rats fed a high-fat/high-cholesterol diet. Male Sprague-Dawley rats weighing 250 g, were divided into five groups; a normal diet group (N), a high-fat/high-cholesterol diet group (HFC), a high-fat/high-cholesterol diet with 5% autoclaved soy flour group (HFC-S), a high-fat/high-cholesterol diet with 5% cheonggulgang group (HFC-CJ), and a high-fat/high-cholesterol diet with 5% deonjang group (HFC-DJ). The body weight gain and adipose tissues weights increased in rats fed a high-fat/high-cholesterol diet, but decreased significantly in rats fed autoclaved soy flour, cheonggulgang and doenjang, compared with the HFC group. Food intake was lower in the all HFC groups (HFC, HFC-S.HFC-CJ and HFC-DJ) than that of the N group, However, there was no differences in the food intake between HFC diet groups. Serum triglyceride level increased with a high-fat/high-cholesterol dietgroup whereas HFC-CJ and HFC-DJ groups were markedly decreased serum triglyceride levels compared with N group. Levels of serum total cholesterol and LDL-cholesterol, and level of total cholesterol in the liver were significantly lower in the HFC-S group, the HFC-CJ group and the HFC-DJ group than that of the HFC group, but exerted no significant change between the HFC groups on levels of total lipid and triglyceride in the liver. Fecal total lipid content increased in the HFC-CJ group and the HFC-DJ group than those of the HFC group and HFC-S group. The activities of heparin-releasable lipoprotein lipase (HR-LPL) and total-extractable LPL in adipose tissues decreased in the HFC-S group, the HFC-CJ group and the HFC-DJ group compared with the HFC group. These results suggest that dietary cheonggulgang and doenjang may exert cholesterol-lowering effect and prevent obesity.

The Bacterial Biological Response Modifier Enriched Chungkookjang Fermentation (면역증강물질 강화 청국장 발효)

  • The aim of this study was to produce Chungkookjang-a food produced through fermentation with Bacillus licheniformis E1-that contains an increased concentration of a bacterial biological response modifier (B-BRM). Unfortunately, sensory studies have indicated that B. licheniformis E1-fermented Chungkookjang is unacceptable for commercial use. We isolated another bacterial strain from this food product: B. subtilis S2. The optimum time and temperature for Chungkookjang fermentation with B. licheniformis E1 and B. subtilis S2 were 48 hr and $40^{\circ}C$, respectively. Sensory studies showed that Chungkookjang fermented by both B. licheniformis E1 and B. subtilis S2 was more acceptable than B. licheniformis E1 only. The amino nitrogen and crude protein content of the product were 359 mg% and 45.6% respectively. Additionally, it was confirmed that the proliferation of mouse splenic lymphocytes increased significantly, when the cells were treated with the BRM from Chungkookjang fermented using the mixture of bacterial strains in vitro. These results suggest that the enriched Chungkookjang may help patients who are medically in need of potentiation of lymphocytes proliferation.
