Developmental process of Natural Medicine (천연물의약품 개발프로세스)

  • 천연물의약품은 식물, 동물, 미생물 및 이의 세포내용물과 광물 등 자연계에 존재하는 물질을 의약품으로 사용한 것으로 말하며, 이들 중 대부분은 식물 유래이다. 식물추출물은 다양한 성분들이 서로 상호작용하면서 혼합되어 있는 형태로 다양한 질환에 대한 조합치료 (combination therapy)에 이용될 수 있어, 생체내에서 다중 타겟에 영향을 미치므로 천연물의 조추출물 (crude extract) 또는 다양한 성분이 함유되어 있는 용매분획물의 형태를 의약품으로 개발하거나, 이로부터 분리된 단일 성분 화합물 및 이의 합성유도체에 대하여 새로운 의약품, 즉 신약을 개발하고자 하는 노력이 시도되고 있다. 2011년 현재 전체의약품 시장에서 천연물의약품 및 천연물유래 의약품이 약 60%이상을 차지하고 있다. 그러나, 국내 천연물의약품 즉, 천연물신약의 개발은 2010년 기준으로 식약청 임상허가 신청현황에서 70%가 관절염과 관련된 의약품일 정도로 고령화에 따른 건강기능식품의 개발방향과 일치되고 있는 경향을 나타내고 있어 다양한 질환을 타겟으로 한 의약품의 개발이 미흡것으로 나타나고 있다. 이러한 경향은 천연물신약개발에 대한 프로세스에 대한 이해의 부족으로 새로운 시도가 이루어지지 않고 있음을 시사하며, 따라서 천연물신약허가에 필요한 제출자료 및 각 자료생산을 위한 방법 등 천연물신약 개발 및 허가를 위한 프로세스에 대하여 설명하였다.

Evaluation of Insecticidal Efficacy of Plant Extracts Against Major Insect Pests (주요 해충에 대한 식물추출물의 살충력 평가)

  • Insecticidal efficacies of plant extracts and environmental friendly agro-materials against green peach aphids, two spotted spider mites, and diamondback moths were evaluated in the lab condition. Oxymatrine and matrine, derivatives of Sophora flavescens, were effective to all three tested pest insect species. Selected environmental friendly agro-materials available from the market mainly contained active ingredients of nicotine, azadirachtin, and matrine were all effective to control green peach aphids and two spotted spider mites.

Antimicrobial Effect of Natural Plant Extracts against Periodontopathic Bacteria (치주염 원인균에 대한 천연 식물 추출물의 항균효과)

  • In this study, we examined the antimicroboal effect against Actinobacillus actinomycetemcomitans and Prevotella intermedia which were the bacteria causing the Periodontopathic by using 34 types of natural plant extracts. Therefore, this study measures growth inhibition activity and Minimum Inhibition Concentration (MIC) of a sample extract with the use of organic solvent extracts in order to analyze the antibacterial effect of natural plant extracts on periodontopathic bacteria. Each of the 34 types of natural plant extracts were extracted by using the ethanol, and subsequently, the size of growth inhibition zone(clear zone, ㎜) of respective extracts were measured through the disk diffusion method. As a result, it was found that the growth inhibitory activity was found for A. actinomycetemcomitans, which is the bacteria causing the Periodontitis, in 13 types of natural plant extracts such as Raphanus sativus, Akebia quinata, Paeonia lactiflora, Belamcanda chinensis, Inula britannics, Houttuynia cordata, Forsythia saxatilis, Gentiana macrophylla, Melia azedarach, Scutellaria baicalensis, Coptis chinensis, Phellodendron amurense, Kalopanax Pictus, etc. In the case of P. intermedia, the growth inhibitory activity was found in 13 types of natural plant extracts such as Raphanus sativus, Angelica acutiloba, Akebia quinata, Belamcanda chinensis, Inula britannics, Houttuynia cordata, Cinnamomum cassia, Aster tataricus, Melia azedarach, Scutellaria baicalensis, Coptis chinensis, Phellodendron amurense, Kalopanax Pictus etc. For A. actinomycetemcomitans, anti-bacterial effect was exhibited in Belamcanda chinensis, Cinnamomum cassia, Kalopanax Pictus, Phellodendron amurense, Coptis chinensis. The Coptis chinensis showed the most excellent growth inhibitory activity in all organic solvent fragment, while P. intermedia showed the growth inhibitory activity in Belamcanda chinensis, Cinnamomum cassia, Meliaazedarach, Phellodendron amurense, and Coptis chinensis.

  • 최근 환경과 건강에 대한 관심이 증대되면서, 천연 섬유를 소재로 사용한 최종 제품 역시 고급화 및 고기능화되고 있는 추세이며 섬유항균가공분야에 있어서도 천연 추출물을 이용한 항균가공이 주목을 받고 있다. 현재 항균가공에 사용되고 있는 항균제는 일반적으로 합성항균제를 사용하고 있고 이는 거의 대부분 자극성 화합물로써 인체에 잠재적 유해요인이 될 수 있고 제조 및 가공과정에서 환경오염을 유발시키는 문제점도 발생되고 있다. 반면 천연추출물을 사용할 경우 합성물질에 비해 포름알데히드 검출 등의 인체 안정성의 불안이 적어 건강차원에서 그 인식이 점차 증대되고 있는 추세이다. 천연항균물질에는 chitosan이나 식물에서 추출된 천연항균물질 등이 있는데 특히 식물에서 추출된 천연항균물질은 피톤치드(phytoncide)라고 표현되고 있으며 이는 수목들이 해충이나 미생물 등으로부터 자기방어를 위해 공기 중으로 발산하는 방향성의 항생물질을 뜻하는 말이다. 본 연구에서는 식물 중에서 피톤치드 정유의 함량이 많다고 알려진 측백나무과의 편백나무 정유를 이용하였으며 편백나무 정유가 식물성 천연 오일형태이므로 가공 처리 시 물과 계면이 발생되기 때문에 HLB(Hydrophilic Lipophlic Balance) 조건에 따른 최적 유화조건을 선정하였으며 편백나무 정유를 이용한 면직물의 항균 기능 부여를 위한 시험으로 편백나무 정유 처리 농도에 따른 항균성을 평가해 보고자 하였다. 실험 결과 편백나무 정유는 HLB 15이상 계면활성제 사용 시 물에 용해성이 좋고 안정된 에멀젼 상태를 보였으며 제조된 편백나무 정유 가공액 5%이상 처리 시 Staphylococcus aureus과 Klebsiella pneumoniae의 99.9% 정균감소율을 나타내었다.

Antioxidant Activities of Extracts from Medicinal Plants (약용식물 추출물의 항산화 활성)

  • In this study, the optimal conditions for phenolic-compound extraction from medicinal plants were found to be 24 h and about 50% ethanol. The electron-donating scavenging activities (DPPH), ABTS radical-cation decolorization (ABTS), antioxidant protection factor (PF), and thiobarbituric acid reaction substance (TBAR) were measured to determine the antioxidant activities of the extracts of Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L. The total phenolic contents of the extracts of medical plants were determined to be 0.45-3.00 mg/g in the water extracts and 0.33-3.15 mg/g in the 50% ethanol extracts. The electron-donating abilities (EDA) of the water and ethanol extracts were both above 85% at the $50{\mu}g/ml$ concentration. The ABTS radical-cation decolorization was above 80% at the $100{\mu}g/ml$ concentration in all the extracts of various medicinal plants. The antioxidant protection factor (PF) of the Melia azedarach L. extracts was found to be $1.65{\pm}0.40$ PF in the water extracts at the $100{\mu}g/ml$ concentration, and was higher than those of the other medicinal-plant extracts. The TBAR inhibition rates of all the medicinal-plant extracts, except Asparagus cochinchinensis Merr., were above 85% at the $100{\mu}g/ml$ concentration. These results confirmed that the various oriental medicinal plants (Sanguisorba officinalis Linn., Citrus unshiu Markovich, Melia azedarach L., Asparagus cochinchinensis Merr., Citrus unshiu S., Polygonum aviculare L., and Leonurus sibiricus L.) that were included in this study are useful functional-food sources.

Studies on the Properties of kochujang by Addition of Natural Plant Extracts (식물성 천연 추출액을 첨가한 고추장의 특성)

  • Effects of natural plant extracts on kochujang and it's sensory evaluation were investigated. Natural plant extracts was added to the general kochujang and then investigated the changes of free amino acids composition after fermentation at $30^{\circ}C$ for 60 days. The content of glutamic acid among the free amino acids was highest, and those of aspartic acid, proline, valine, histidine were also presented higher than the general kochujang, Sensory properties of natural plant extract-added kochujang were unchanged in color, hot taste, salty taste but, properties presented better in flavor, savory taste, sweet taste, overall preperence. Therefore, we may conclude that kochujang added natural plant extracts was more acceptable than general kochujang.

Activity of Nature Plants Extracts against Pseudomonas aeruginosa ATCC 27853 (Pseudomonas aeruginosa ATCC 27853에 대한 약용식물 추출물들의 활성)

  • Anti-Pseudomonas aeruginosa activities for ethanol extract of 34 medicinal plants widely used in the folk medicine were evaluated to screening of anti-Pseudomonas aeruginosa agents. The ethanol extracts of Gardenia jasminoides, Arctium lappa, Citrus unshiu, and Phellodendron amurense showed antimicrobial activities against P. aeruginosa ATCC 27853. The ethanol extracts of Gardenia jasminoides among these medicinal plants showed significant antimicrobial activity against P. aeruginosa. Therefore, we expect that these medicinal plants will be useful for nature antimicrobial agent against P. aeruginosa in future.

  • The increase in the elderly population and growing interest in skin beauty, alongside specialized scientific examination of natural substances, have led to a rise in the utilization of natural materials. This paper investigates the types of natural substances, the functionality of extracts, and extraction technologies based on the literature that utilizes natural ingredients to mitigate skin aging. To directly assess the impact of functional materials derived from natural products on skin aging mitigation, the functionality of the extracts was determined based on their ability to synthesize Procollagen and reduce MMP-1. Each natural product is categorized into plants, herbal medicines, and microalgae in the literature utilizing the above evaluation methods. This paper describes the extraction technologies for securing functional materials from each ingredient and the main outcomes, providing trends in research on extraction technologies for functional materials aimed at alleviating skin aging

The Study of Preservative System in Cosmetics using Botanical Antimicrobial (식물유래 천연항균제를 이용한 화장품에서의 방부시스템에 관한 연구)

  • Anti-microbial activities of Scutellariae Radix Extracts and Sophora flavecens Extracts was tested by formulation, such as skinsoftener, emulsion for 4 weeks. Control contained no preservative, test group contained 1.00% Scutellariae Radix extracts and 1.00% Sophora flavecens extracts and positive control contained parabens and imidazolidinyl urea. To determine the anti-microbial activity of these extract, the 4 germs such as Escherichia coil, Pseudomonas aeruginosa, Staphylococcus aures, Canida albicans were used. The test groups showed significant anti-microbial activities against the 4 germs at 2 and 3 weeks as compared with control. Anti-microbial activities of these extracts were similar to positive control. Considering that the Scutellariae Radix Extracts and the Sophora flavecens Extracts have a significant anti-microbial activities against 4 germs, it is possible as natural preservative in cosmetics.

Antimicrobial Plant Extracts as an Alternative of Chemical Preservative: Preservative Efficacy of Terminalia chebula, Rhus japonica (gallut) and Cinnmomum cassia Extract in the Cosmetic Formular (가자, 오배자, 계피 추출물을 이용한 화장품 제형에서의 방부효과)

  • This study was carried to investigate the efficiency of antimicrobial plant extracts as natural preservative in the cosmetic formulations. Ethanol extracts of different plants were tested using the disc diffusion (paper disc) method and the minimum inhibitory concentration (MIC) method for their antimicrobial activity against the common poultry pathogens. Terminalia chebula and Rhus japonica (gallut) extracts exhibited antibacterial activity against Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli. Cinnmomum cassia extract exhibited antifungal activity against Candida albicans and Aspergillus niger while the remaining plant extracts showed no activity. A study of the preservative efficacy of the cosmetic formular containing the T. chebula, R. japonica and C. cassia extracts demonstrated sufficient preservative efficacy against bacteria and eukaryotic test microbes. Also, the cosmetic formulations containing antimicrobial plant extracts more effectively inhibited the microoranisms than the mixture of traditional chemical preservatives. These results suggest that the mixture of antimicrobial plant extracts, T. chebula, R. japonica and C. cassia is incorporated as preservative in the cosmetic formulation and the mixture have considerable effect on its efficacy.
