Vitamin C, Total Polyphenol, Flavonoid Contents and Antioxidant Capacity of Several Fruit Peels (과일 껍질의 비타민 C, 폴리페놀, 플라보노이드 함량과 항산화 활성)

  • The aim of this study was to evaluate the antioxidant activity, total phenolics, flavonoids, and vitamin C contents in peels of 11 commercial fruits commonly consumed in the Republic of Korea. The antioxidant activities were evaluated by ABTS, DPPH radical scavenging methods and reducing power. The peels of plum (Prunus spp.) and golden kiwifruit (Actinidia chinensis) exhibited the highest values of antioxidant activity and total phenolics, while the highest level of ascorbic acid was recorded in the peel of citrus fruit (p<0.05). There were positive correlations between the antioxidant activity assessed by both reducing power and ABTS methods and total polyphenol content ($R^2$=0.6356), suggesting that phenolic compounds are the major contributors to these activities. The results indicate that plum and golden kiwifruit peels can be used as natural antioxidant ingredients and in production of functional foods in the Korean diet.

Antimicrobial Activities of Botanical Antimicrobial Agent-Grapefruit Seed Extract Mixture for the Preparation of Anitimicrobial Packaging Paper (항균포장지 제조용 식물성 자몽종자추출물제재의 항균특성)

  • Botanical antimicrobial agent-grapefruit seed extract mixture(BAAG), which could be applied to the preparation of antimicrobial packaging paper, was investigated in order to prove the preservative function of fruits and vegetables. HAAG showed remarkable antimicrobial effects against Fusarium solani Botrytis cinerea, Pencillium crustosum, Erwinia carotovora, Phoma destructiva and Alternaria radicina causing the postharvest decay of fruits and vegetables. We have examined that HAAG could inhibit the growth of microorganims when treated with more than 500 $\mu$g/mL concentration. The activities of HAAG were stable in the wide spectrum of pH and temperature. Direct visualization of microbial cells by using scanning electron microscope showed the loss of microbial cell membrane function, which was destroyed by treating with the dilute solutions of HAAG. We could confirm that HAAG be an antimicrobial agent for the preparation of antimicrobial packaging paper.

Comparision of SOD Activity and Phenolic Compound Contents in Various Korean Medicinal Plants (한국산 약용식물로부터의 SOD활성 및 Phenolic Compounds 함량 비교)

  • 82 species of Korean medicinal plants were measured to their SOD (superoxide dismutase) activity, total phenol and individual phenol compounds. The SOD activity of medicinal plants was varied from 3.67% to 48.3%, and especially, 4 species including Polygala tenuifolia Willd. showed activity over 40% higher than other species. The 16 individual phenol compounds concentrations have a severe variation in medicinal plants. Codonopsis lanceolata is the highest concentration $(12,176\;{\mu}g/g)$, and 10 species including Glycyrrhiza uralensis Fisch were showed to concentrations more than $1,000\;{\mu}g/g$. And also, salicylic acid was generally found in many plants, but trans-cinnamic acid, naringenin and kampferol was little found in plants. The total phenol by Folin-Dennis methods showed a respectively variation $(3{\sim}249.731\;{\mu}g/g)$ in 82 medicinal plants, and 7 species including Astragalus membranaceus showed low total phenol concentrations less than $10\;{\mu}g/g$. In conclusion, we expect that selecting medicinal plants with high biological activity might develop to chemicals related to antioxidants, as well as these become most popular substance at many industrial materials, and suggest to more researchs in future.

Anti-proliferation Effect of Coscinoderma sp. Extract on Human Colon Cancer Cells (Coscinoderma sp.의 대장암세포 증식 억제 효과)

  • Natural products are attractive as the source of new drug development. Especially, numerous unknown marine bioresources are an object of attention because the ocean occupies three fourth of the earth. Survival of marine bioresources in extreme environment may induce the production of biological active compounds. As previous study, we examined over 40 specimens of marine sponges collected from Micronesia and screened their anti-proliferative activities in various cancer cell lines. Among them, we investigated Coscinoderma sp.'s activity and mechanism in human colon carcinoma HCT116 and RKO cells. Furthermore, we also used the p53-knockout of HCT116 cells and the p53 loss of RKO cells for elucidating the role of p53. Coscinoderma sp. inhibited cellular viability independently of the p53 status. Therefore, we compared the expression level of cell death-related proteins by Coscinoderma sp. in HCT16 and in HCT116 p53KO cells. Coscinoderma sp. increased p53 level and NOXA levels and induced apoptosis under the condition of p53 existence. On the other hand, Coscinoderma sp. increased p21 and mTOR levels in HCT116 p53KO cells. These results suggest that Coscinoderma sp. induced anti-proliferation effect through different pathway depending on p53 status.

Antioxidant Activity of the Ethanol Extract from Cooking Drips of Thunnus thynnus by Gamma Irradiation (참치 자숙액의 항산화 활성에 감마선 조사가 미치는 영향)

  • This study was conducted to examine the effect of a gamma irradiation on the antioxidant activity of ethanol extract from cooking drips of Thunnus thynnus (ECT). The electron donating ability against 1,1-diphenyl- 2-picryl-hydrazyl (DPPH), authentic hydroxyl radical scavenging activity by Ultra Weak Luminescence Analyzer, oxidative protection factors by Racimat method, ferric reducing antioxidant power (FRAP), and ${\beta}$-carotene bleaching activity were measured as indices of antioxidant activity. All of the results shown that irradiation increased the antioxidant activity of ECT. These results suggested that ECT could be used as an antioxidative component in food and gamma irradiation could enhance these antioxidative properties.

Purification and Structure of Antioxidative Substance Derived from Tetraselmis suecica (Tetraselmis suecica유래의 항산화성 물질의 정제 및 구조)

  • Tetraselmis suecica (T. suecica) of Prasinophyta was selected because the growth rate is comparatively higher and the culturing is also easy. In order to investigate antioxidative activity, the soluble elements of T. suecica were fractionated using water and organic solvents such as methanol, hexane, chloroform, ethylacetate and butanol. The chloroform fraction of T. suecica showed strong antioxidative activity. The potential antioxidative activity was detected in hexane: ethylacetate (1:5) once used the fractions by different mixtures of organic solvents. This fraction was further purified by preparative thin layer chromatography (PTLC) and repeated reverse-phase HPLC. On the basis of chemical and spectroscopic evidences obtained by UV, FT-IR, FAB-MS and NMR, the compound purified from T. suecica was identified as pheophorbide-a. The antioxidative activity of the compound was comparable to that of $\alpha$-tocopherol and could be act as an antioxidant in foods.

Citrus platymamma inhibits the expression of pro-inflammatory cytokines, inducible nitric oxide synthase, and cyclooxygenase-2 in RAW264.7 macrophage (RAW264.7 대식세포에서 Citrus platymamma의 iNOS, COX-2, 염증성 사이토카인 발현 억제 효과)

  • Citrus platymamma hort. ex Tanaka is widely used in traditional Korean medicine because of its medicinal benefits including an anti-inflammatory effect. This study aimed to evaluate changes in the flavonoid content and anti-inflammatory activities of C. platymamma during its harvest period. Fruit peel samples were obtained between September 2015 and February 2016. The results indicate that C. platymamma peel extract (CPE) was an effective inhibitor of lipopolysaccharide (LPS)-induced NO production in RAW264.7 cells. The inhibitory effects of CPE at $100{\mu}g/mL$ concentration included dose-dependent decreases in the expression of iNOS and COX-2 proteins. In addition, CPE decreased the expression of pro-inflammatory cytokines TNF-${\alpha}$, IL-$1{\beta}$, and IL-6. The highest anti-inflammatory activity and flavonoid content were observed in CPE of C. platymamma peel harvested during the immature fruit period in September. Further, to assess the suitability of CPE for cosmetic use, we performed MTT assays using HaCaT keratinocytes and observed that CPE did not exhibit any cytotoxicity. To test the potential application of CPE as a cosmetic material, we also performed primary skin irritation tests on normal skin of 30 volunteers and no adverse reactions were observed. The results of this study indicate that CPE may be considered as an anti-inflammatory candidate for inclusion in cosmetic materials.

In vitro activities of Grape Pruning Stems for Application of Cosmetic Ingredients (포도 전정가지의 화장품 소재로서의 응용)

  • Grape pruning stems constitute a scarcely investigated class of byproducts with limited reports on their bioactive polyphenol content and/or industrial applications. Herein we present the outcome of our investigation on grape pruning stems extracts, concerning the assessment of their total polyphenolic content and the detailed evaluation of their antioxidant properties. Results obtained indicate that grape pruning stems are particularly rich in flavonoids and trans-resveratrol. The antioxidant activities was analyzed and expressed as electron donating ability, ABTS cation radical decolorization, hydrogen peroxide scavenging activity and superoxide anion radical scavenging activity, the antioxidant activity of Vitis labrusca L. pruning stems extracted (GPSE) was higher than that of BHA and L-ascrobic acid. The whitening and anti-wrinkle activities display an capability. Results herein grape pruning stems used as a valuable resource for the extraction of resveratrol, which would be added to functional cosmetics and food materials.

Lipolysis Effect of Daucosterol Isolated from Mulberry (Morus alba) Leaves (뽕잎으로부터 순수분리한 daucosterol의 lipolysis 효과)

  • Plants are reservoirs of naturally occurring chemical compounds and of structurally diverse bioactive molecules. The aim of this investigation was to screen for the presence of phytochemicals responsible for the lipolysis activity in mulberry (Morus alba) leaves, which are important in traditional Asian medicinal plants. Powdered mulberry leaves were extracted with hexane, ethyl acetate, and methanol. Daucosterol was isolated from the EtOAc extract of mulberry leaves, and its structure was elucidated by NMR spectral analyses. The NMR assignments for the compound were determined using $^1H$, $^{13}C$, DEPT, COSY, HSQC, and HMBC NMR spectral data. Daucosterol showed a concentration-dependent lipolysis activity that may impart medicinal properties that can be exploited by medical practitioners for the treatment of various diseases. However, further studies should be conducted to elucidate additional mechanisms of daucosterol.

Relievable Effect of Alpinetin on Dexamethasone-Induced Skin Aging (Alpinetin의 Dexamethasone으로 유도한 피부 노화 완화 효과)

  • Steroid hormone, glucocorticoid (GC) has strong anti-inflammatory effects by binding to glucocorticoid receptor (GR) inhibiting the expression of inflammatory genes. Therefore, agents that activate the GR have been used for the treatment of dermatitis. However, the agents have side effects such as skin barrier dysfunction and dermal atrophy, inducing skin damage as well as skin aging. It has been reported that GC is activated by 11 beta-hydroxysteroid dehydrogenase type 1 ($11{\beta}$-HSD1) to increase the activity of the GR. This study aimed to identify natural materials that can effectively inhibit dexamethasone. We found that alpinetin isolated from Alpinia katsumadai extract has a significant effect on this. Alpinetin not only inhibited $11{\beta}$-HSD1 expression, but also suppressed the increase of phosphorylated GR and cortisol concentration. Alpinetin also recovered collagen expression in dexamethasone-treated dermal fibroblasts, and the reduction of dermal thickness in dexamethasnone-treated 3D skin model. These results suggest that alpinetin prevents skin aging induced by the increase of $11{\beta}$-HSD1 expression.
