Effect of Cell Size on Growth and Development of Plug Seedlings of Three Indigenous Medicinal Plants (플러그 셀 크기가 세 가지 자생 약용식물 묘 생육에 미치는 영향)

  • There have not been many studies conducted on the seedling production, especially in plug trays, of traditional medicinal plant species. In an effort to establish guide lines for seedling production, this study investigated the effect of plug cell size on the growth and development of plug seedling of three medicinal plant species. Seeds were sown in either 128, 200, or 288-cell plug trays, containing a commercial medium. Growth and development of individual seedling was generally promoted with increasing size of a plug cell in all of the three species. The greatest biomass of the seedlings gained in a plug tray was obtained in the 288-cell trays in Perilla frutescens var. acuta Kudo and Sophora tonkinensis, and the 200-cell trays in Angelica gigas Nakai. Overall growth and development of the shoot and root of a single seedling of Perilla frutescens var. acuta Kudo, except total chlorophyll and anthocyanin contents, was the greatest in the 128-cell tray. However, length of the longest root, length, width and area of the leaf, internode length, root fresh weight, and root ball formation in the 200- and 288-cell trays were not significantly different each other. In Sophora tonkinensis, although length of the longest root, stem diameter, leaf width, leaf area, shoot fresh weight, and root ball formation were not significantly different among the treatments, length of the longest root and root ball formation of a single seedling were the greatest in the 128-cell tray. Overall shoot and root growth, except total chlorophyll content, of a single seedling of Angelica gigas Nakai was the greatest in the 128-cell tray. Based on the total biomass, it is concluded that 288-cell trays are recommended for production of plug seedlings of medicinal plant species P. frutescens var. acuta Kudo and S. tonkinensis. In A. gigas Nakai, it would be more economical to use the 200-cell trays than 128-cell trays due to total biomass.

  • Inactivation in soil absorption, translocation of 2, 4-D by plants vary depending upon soil environments and herbicide formulations. Experiment was conducted in a glasshouse using rectangular pots($1350cm^2$) to evaluate the growth responses of barnyardgrass (Echinochloa crus-galli) and Indian jointvetch (Aesehyrcomene indica) to two formulations of 2, 4-D. The formulations used were 40% 2, 4-D amin salt (2, 4-D/AS) and 19.7% complex of rice husk lignin and 2, 4-D (2, 4-D/LG) which were applied at 200g ai/ha. Soil environments included fertilizer levels, soil pH, organic matter contents, and soil textures, Each treatment was replicated three times. The herbicidal activity of 2.4-D increased and lasted with increased levels of fertilizer. The activity also increased and lasted with low soil pH and decreased content of organic matter. Generally 2, 4-D/LG showed higher and longer herbicidal activity than 2. 4-D/AS for both test plants under all conditions applied. However, the herbicidal activity was influenced by the formulations more than by soil textures. It was thought that 2, 4-D/AS was released in a short time and inactivated readily while 2, 4-D/LG was slowly released and gave an opportunity of absorption by plants for a long period.

Production of Poly-3-hydroxybutyrate from Xylose by Bacillus megaterium J-65 (Bacillus megaterium J-65에 의한 xylose로부터 poly-3-hydroxybutyrate 생산)

  • A microorganism capable of producing high level of poly-3-hydoxybutyrate (PHB) from xylose was isolated from soil. The isolated strain J-65 was identified as Bacillus megaterium based on the morphological, biochemical and molecular biological characteristics. The optimum temperature and pH for the growth of B. megaterium J-65 were $37^{\circ}C$ and 8.0, respectively. The optimum medium composition for the cell growth was 2% xylose, 0.25% $(NH_4)_2SO_4$, 0.3% $Na_2HPO_4{\cdot}12H_2O$, and 0.1% $KH_2PO_4$. The optimum condition for PHB accumulation was same to the optimum condition for cell growth. Copolymer of ${\beta}$-hydroxybutyric and ${\beta}$-hydroxyvaleric acid was produced when propionic acid was added to shake flasks containing 20 g/l of xylose. Fermenter culture was carried out to produce the high concentration of PHB. In batch culture, cell mass was 9.82 g/l and PHB content was 35% of dry cell weight. PHB produced by B. megaterium J-65 was identified as homopolymer of 3-hydoxybutyric acid by GC and NMR.

Studies on the Application of Cattle Slurry in Grassland III. Grazing preference as affected by application time of cattle slurry on pasture (혼파초지에서 액상구비 시용에 관한 연구 III. 방목이용 초지에서 액상구비 시용시기에 따른 가축기호성 비교)

  • This experiment was carried out to determine the effects of application time of cattle slurry on grazing pasture on the grass growth, dry matter yield, nutritive value, animal preference, and efficiency of pasture utilization. Four different application times of slurry(right now after grazing, 5th day, 10th day, and 15th day after grazing), and a no-slurry(control) plot were designed, 1991∼1992. Annual application amount of slurry was 40 ㎥/ha, and slurry was applicated in spring(2 times), summer and autumn. Total NPK fertilizer applicated were same as 280, 200 and 240 kg/ha in all treatment, respectively. The pasture was grazed with growing beef cattle(initial body wt.; ca. 250 kg) at every 30∼35 cm of plant height. The grass height, dry matter yield, crude protein yield, and other nutritive value were not different among treatments. However, the animal preference and efficiency of pasture utilization showed significant difference by application time of slurry. The highest grazing preference (1: the worst∼9: the best). and utilization efficiency (0∼100%) were observed by application time of right now after grazing (7.8, 76%), which was same to control(7.8, 77%), and followed by 5th day after grazing(7.3, 74%) and l0th day after grazing(6.9, 71%). But the preference and pasture utilization were greatly decreased at application time of cattle slurry of 15th day alter grazing(5.5, 60%). From the above results, the optimum application time of cattle slurry on grazing pasture to enhance animal preference, and efficiency of pasture utilization was within 5 days after grazing (less than 13.6 cm of plant height), and within 10 days after grazing (less than 18.5 cm of plant height) at least.

  • Dynamic traffic assignment(DTA) has been a topic of substantial research during the past decade. While DTA is gradually maturing, many aspects of DTA still need improvement, especially regarding its formulation and solution algerian Recently, with its promise for In(Intelligent Transportation System) and GIS(Geographic Information System) applications, DTA have received increasing attention. This potential also implies higher requirement for DTA modeling, especially regarding its solution efficiency for real-time implementation. But DTA have many mathematical difficulties in searching process due to the complexity of spatial and temporal variables. Although many solution algorithms have been studied, conventional methods cannot iud the solution in case that objective function or constraints is not convex. In this paper, the genetic algorithm to find the solution of DTA is applied and the Merchant-Nemhauser model is used as DTA model because it has a nonconvex constraint set. To handle the nonconvex constraint set the GENOCOP III system which is a kind of the genetic algorithm is used in this study. Results for the sample network have been compared with the results of conventional method.

Inhibitory Effect of Camp Antagonist and Pka Inhibitors, and Stimulatory Effect of Adenylate Cyclase Agonist on Cathepsin K Processing in Cultured Mouse Osteoclasts (cAMP 길항제와 PKA 억제제 및 Adenylate Cyclase 촉진제의 백서 파골세포에서 Cathepsin K 생성에 대한 효과)

  • Cathepsin K (cat K) is the major cysteine protease expressed in osteoclasts and was thought to play a key role in matrix degradation during bone resorption. It was shown that the intracellular maturation of cat K was prevented by the cAMP antagonist, Rp-cAMP, and the protein kinase A (PKA) inhibitors of KT5720 and H89. In contrast, forskolin, a adenylate cyclase agonist, rather induced Cat K processing and maturation in osteoclasts. Furthermore, to determine whether cat K processing and maturation signaling involves protein kinase C (PKC), mouse total bone cells were treated with calphostin C, a specific inhibitor of PKC, however, no effect was observed, indicating that calphostin C did not affect to osteoclast-mediated cat K processing and maturation. Thus, it is indicated that the cAMP-PKA signaling pathway regulates cat K maturation in osteoclasts. Since secreted proenzymes have the potential to reenter the cell via M6P receptor, to prevent this possibility, it was tested cAMP antagonist Rp-cAMP and the PKA inhibitors KT5720 and H89 in the absence or presence of M6P. Inhibition of cat K processing by Rp-cAMP, KT5720, or H89 was observed in a dose-dependent manner. Furthermore, the addition of M6P resulted in enhanced potency of Rp-cAMP, KT5720 and H89. These dose-dependently inhibited in vitro bone resorption with a potency similar to that observed for inhibition of cat K processing.

  • The development project of Busan New Port aims to be Logistics Hub Port but there are too many things to deal with ; enlargement of harbour, interport competition, modernization of harbour loading equipment and so on. At present, 23 berths of North and South container quay are in operation and 22 berths will be constructed on west and south-side by 2020. Namely, Busan New Port will operate 45 berths in 2020. When it comes to port distripark, a large-scale of Port distripark project is underway, such as Ung-Dong district 1,2 phase, West container 1,2phase, North distripark and so on. This study is to deduce traffic system problem of Busan New Port which is caused by the development project through predicting traffic need considering the development project. According to study, there are three main problems of traffic system : 1. traffic congestion caused on main crossroad, connecting second harbour back road. 2. It has been predicted that South-North road and traffic capacity of New Port road would lack compared to traffic volume-to-be-increased. Moreover, the detour volume of traffic is caused because New Port's 1st avenue and route 2 were not connected directly. Thus, this study suggests three kinds of improvement plan for smoother traffic flow. 1st. Operate roundabout on major intersection, for example, second harbour back road, west container wharf's subway corridors(South to North), and permit only right turn on sub-intersection. 2nd. Extend New Port road(North container's port road) by utilizing side walk and median. 3rd. Install exit ramp which utilizes Route 2 connecting New Port's 1st avenue and local road 1042. The method we used to analyze the effect of improvement is Vissim of Mircro Simulation Package.

2-D/3-D Seismic Data Acquisition and Quality Control for Gas Hydrate Exploration in the Ulleung Basin (울릉분지 가스하이드레이트 2/3차원 탄성파 탐사자료 취득 및 품질관리)

  • To identify the potential area of gas hydrate in the Ulleung Basin, 2-D and 3-D seismic surveys using R/V Tamhae II were conducted in 2005 and 2006. Seismic survey equipment consisted of navigation system, recording system, streamer cable and air-gun source. For reliable velocity analysis in a deep sea area where water depths are mostly greater than 1,000 m and the target depth is up to about 500 msec interval below the seafloor, 3-km-long streamer and 1,035 $in^3$ tuned air-gun array were used. During the survey, a suite of quality control operations including source signature analysis, 2-D brute stack, RMS noise analysis and FK analysis were performed. The source signature was calculated to verify its conformity to quality specification and the gun dropout test was carried out to examine signature changes due to a single air gun's failure. From the online quality analysis, we could conclude that the overall data quality was very good even though some seismic data were affected by swell noise, parity error, spike noise and current rip noise. Especially, by checking the result of data quality enhancement using FK filtering and missing trace restoration technique for the 3-D seismic data inevitably contaminated with current rip noises, the acquired data were accepted and the field survey could be conducted continuously. Even in survey areas where the acquired data would be unsuitable for quality specification, the marine seismic survey efficiency could be improved by showing the possibility of noise suppression through onboard data processing.

  • Background: Gram-negative bacterial endotoxin induced septicemia is known to be a leading cause in the development of adult respiratory distress syndrome(ARDS). The mechanism of endotoxin induced lung injury is mainly due to the activated neutrophils which injure the capillary endothelial cells by releasing oxidant radical and resulted in pulmonary edema. We studied the change of antioxidant enzyme in the case of large or small, intermittant dose of endotoxin infused rat lungs. Methods: Endotoxin was given to the rat through the peritoneal cavity in the dose of 7 mg/kg body weight in the large dose group and 1 mg/kg for 10 days in the small dose group. Bronchoalveolar lavage (BAL) was done and rats were killed at 6, 12, 24 hours after single endotoxin injection in the large dose group and 3, 7, 10 days after daily endotoxin injection for 10 days in the small dose group. The lungs were perfused with normal saline through the pulmonary artery to remove the blood and were homogenized in 5 volume of 50 mM potassium phosphate buffer containing 0.1 mM EDTA. After centrifuging at 100,000 g for 60 minute, the supernatent was removed and stored at $-70^{\circ}C$ until measuring for superoxide dismutase (SOD), catalase, glutathione peroxidase (GSH-Px) and protein. Results: We observed the following results. 1) The lung wet/dry weight ratio and albumin concentration in the BAL fluids were increased to peak at 12 hours and neutrophil number in the BAL fluids were peak at 6 hours after endotoxin injection in the large dose group. 2) Cu, Zn SOD (IU/mg protein) was significantly decreased after 6, 12 hours after endotoxin injection in the large dose group. 3) There were no singnificant change in the level of Mn SOD, catalase, GSH-Px after endotoxin injection in both groups. Conclusion: Endotoxin in the large dose group produced the acute pulmonary edema and decreased the Cu, Zn SOD in the lung tissue after injecting endotoxin at 6 and 12 hours. These phenomenon may be due to the cell membrane damage by endotoxin. Further research would be necessary whther giving SOD by intratracheal route or method to increase the synthesis of SOD may lessen the acute lung injury by endotoxin.

Effect of Pimpinella Brachycarpa Ethanol Extract on Chronically Ethanol-Induced Liver Damage in Rats (참나물이 만성적으로 알코올 유도된 흰쥐의 간손상에 미치는 영향)

  • This study was performed to investigate the effect of ethanol extract of Pimpinella brachycarpa(PBE) on chronically ethanol-induced hepatotoxicity in rat liver. Sprague-Dawley rats weighing 90-130 g were divided into 5 groups; normal group(NOR), ethanol(35%, 10 ml/kg) treated group(CON), PBE 200 mg/kg treated group(P1), PBE 200 mg/kg and ethanol treated group(P2), and PBE 400 mg/kg and ethanol treated group(P3). PBE was also fractionated by the following solvent: n-hexane, chloroform, ethylacetate and n-butanol. The antioxidative capacity of the n-hexane fraction was the highest among fractions and was similar to that of butylated hydroxytoluene(BHT). The body weight gain and feed intake of the rats were decreased by ethanol administration, but were gradually increased to the similar levels of the NOR group by administering PBE. The AST activity in serum elevated by ethanol was significantly decreased by administering the high dosage of PBE, but exerted no significant change on serum ALT activity. It was also observed that the hepatic activities of xanthine oxide(XO), catalase and glutathione peroxidase(GSH-Px) increased by ethanol were markedly decreased in the combined ethanol and PBE administered groups(P2 and P3), but not in the activity of superoxide dismutase(SOD) as compared with the CON group. The glutathione(GSH) contents were decreased by ethanol adminstration, however, increased after administering PBE. These results suggest that ethanol extract of Pimpinella brachycarpa has a possible positive effect on the liver function in hepatotoxicity-induced rats by ethanol administration.
