Antibacterial effect of electrolyzed water on Streptococcus mutans (전기분해수소수의 Streptococcus mutans에 대한 항균효과)

  • Objectives: Electrolyzed water has been proven to have antibacterial effects against various microorganisms. However, there are only a few studies about effects of electrolyzed water on oral bacteria. The purpose of this study was to examine the antibacterial effect of electrolyzed water on Streptococcus mutans in vitro. Methods: S. mutans KCOM 1054 was treated with electrolyzed water for 1 or 3 minutes and plated on Mitis Salivarius agar with 15% sucrose and bacitracin. After incubation for 48 hours, colony forming units (CFU) were counted, and dental plaque was quantified by crystal violet staining. Results: The growth of S. mutans was significantly inhibited by electrolyzed water (p<0.001). In addition, the dental plaque formation by S. mutans was decreased in a time-dependent manner by exposure to electrolyzed water (p<0.001). Conclusions: Our results suggest that electrolyzed water can effectively prevent dental caries by inhibiting growth of (and the formation of dental plaque by) S. mutans.

Preventive effects of shiitake mushroom extract on candida stomatitis (칸디다성 구내염에 대한 표고버섯 추출물의 예방효과)

  • Purpose: The purpose of this study was to investigate antifungal activity of shiitake mushroom yeast and hyphal type of Candida albicans. Materials and Methods: The extract from shiitake mushroom was collected by drying the supernatant after soaking shiitake mushrooms in water or ethanol. The antifungal activity of the extracts against yeast type of C. albicans was investigated by the susceptibility assay using microplate. C. albicans biofilm was formed on 12-well plate using Ham's F-12 medium in CO2 incubator and treated with the ethanol extract. Furthermore, C. albicans biofilm was formed on denture base resin disk and treated with or without the ethanol extract in the presence of denture cleanser. Live C. albicans in biofilm was counted by cultured colony forming unit value after inoculated on agar plate. Results: Ethanol extract from shiitake mushroom showed stronger antifungal activity against yeast type of C. albicans compared to its water extract. The ethanol extract significantly reduced count of C. albicans in hyphal biofilm (P < 0.05). Also, the ethanol extract showed synergistically antifungal effect with denture cleanser on candidal biofilm on denture base resin disk (P < 0.05). Conclusion: The ethanol extract of shiitake mushroom may be a candidate for preventing candidal stomatitis as well as denture-related stomatitis.

  • The purpose of this study was to study the Relationships between Sport for All Participation and Physical and Mental Function affecting the Elderly's Self-perceived Health. For accomplishing this purpose of this study, the 308 subjects were selected by the multi-stage stratified cluster sampling method from the Seoul, in the years of 2008. presently. Out of the 360 questionnaires responded to, only 308 were used for data analysis because 52 were responded to incompletely or incorrectly. The survey questionaries were used to collect data. The questionary for physical function state were developed and standardized by kung-hee chung(1993), yang-lae Kim(2005). And mental function state were divided two parts; self-perceived scale and depress scale. The questionary for self-perceived scale were based on MMSE-K(Mini Mental State Examination-Korea). The questionary for Depression were based on Depression Scale developed by Beck(1978), adapted in Korean and standardization by Lee Young-ho & Song Jong-yong(1991), regulated by Jo Gyu-whang(2004). From the analysis of this study, the following conclusions were obtained: First, according to the elderly people's participation in leisure sport activities, it partially affected physical and mental function state. Second, according to the degree of elderly people's participation in leisure sport activities, it partially affected physical and mental function state.

Isolation and Identification of Ampicillin-resistant Bacteria in Changwon (창원근교에서의 ampicillin 내성세균의 분리 및 동정)

  • The number of antibiotic-resistant bacteria is increasing rapidly while the discovery rate of new antibiotics is in decline. A systematic study is therefore necessary to investigate which bacteria are resistant to medically important antibiotics and how high that resistance is. To that end, this study aimed to analyze which bacteria demonstrated resistance to ampicillin, one of the currently most-widely used medical antibiotics. Water samples were collected from the Changwon-Cheon that runs through Changwon City and from the pond in front of the dormitory building at Changwon University. Hundreds of ampicillin-resistant colonies were obtained and 22 morphologically distinct examples were chosen for further study. These bacteria were identified by amplifying their 16S rRNA genes and comparing those sequences with data in GenBank. The bacteria was identified as belonging to 10 families, 12 genera, and 17 species, and all were able to grow in the presence of $50{\mu}g/ml$ ampicillin while seven showed growth at ampicillin concentrations as high as 1.5 mg/ml.

Awareness regarding Safe Abortion among Adolescent Girls in Rural area of Mahottari district of Nepal

  • This study aimed to assess the level of awareness on safe abortion among adolescent girls in rural area of Mahottari district of Nepal. A community based cross-sectional study was conducted in rural areas of Mahottari district of Southern Nepal between January and March 2019.A sample of 412 adolescent girls was selected using multi-stage cluster sampling. Multivariable logistic regression model was adapted to explore level of awareness among adolescent girls. This study found that 45.6% of adolescent girls had high awareness regarding safe abortion. The odds of awareness among the married adolescents was higher (AOR=2.16; 95% CI: 1.01-4.87) than unmarried adolescent whereas the odds of awareness among the adolescents who had education of secondary level and more had higher (AOR=2.21; 95%CI: 1.13-3.04) than those who had primary or lower secondary level of education. Similarly, the adolescents who had monthly family income of Nepalese Rupees (NRs.), 10,000-20,000 and more than NRs. 20,000 were respectively, 2.33 times (AOR = 2.33; 95% CI: 1.07-3.55) and 3.17 times (AOR = 3.17; 95% CI: 2.19-8.94) more likelihood to have high awareness regarding safe abortion than those their counterparts. The study showed that overall level of knowledge towards safe abortion was found low. Socio-demographic factors like marital status, level of education, and family income were the factors independently associated with level of awareness on safe abortion. Therefore, efforts should be exerted towards arising and improving the awareness of abortion care which may reduce unwanted pregnancy, abortion and other complications related to abortion.

Inhibition of growth and toxin production of ochratoxigenic Aspergillus spp. by isolated bacteria (분리세균에 의한 ochratoxin 생성 Aspergillus spp.의 생장 및 독소생성 저해)

  • Ochratoxin A (OTA) that is one of mycotoxins produced mainly by Aspergillus spp. is a common contaminant of stored grains and poses health hazards to human and livestock. The aim of this study is to explore the ability of isolated bacteria Bacillus subtilis AF13 and Streptomyces shenzhenensis YR226 to inhibit growth and OTA production of 3 ochratoxigenic Aspergillus strains. The antifungal activity against mycelial growth and sporulation of Aspergillus strains was examined by coculture with AF13 and YR226 on potato dextrose agar plate. AF13 and YR226 reduced 77.58 and 78.48% of fungal colony radius, respectively, and both strains inhibited fungal sporulation up to 99% in 10 days of incubation. YR226 also reduced more than 91% of spore germination of 3 fungal strains. When Aspergillus strains were cocultured with AF13 or YR226 in yeast extract sucrose medium, mycelial growth and OTA production decreased in all three fungal strains. In particular, AF13 completely inhibited the mycelial growth of A. alutaceus and inhibited its OTA production by 99%, and YR226 also reduced mycelial growth and toxin production up to 99%, respectively. Antimicrobial substances produced by AF13 and YR226 included siderophore, chitinase, protease, ${\beta}$-1,3-glucanase and biosurfactant. These results suggest that AF13 and YR226 can be used in a biological method to prevent valuable crops against mycotoxigenic fungi, and therefore decrease economic damage in agriculture and feed industry.

Non-hemolytic, Mucinous, Coagulase Negative MRSA Isolated from Urine (소변에서 분리된 비용혈성, 점액성, 응고효소 음성 MRSA)

  • An 84-year-old woman presented to the emergency department with a chief complaint of pressure sores of the anus. She had a urine catheter when she showed pyuria three times but had no fever. A microscopic examination revealed many grapevine-like Gram positive strains and neutrophils. After 24 hours of urine culture on blood agar, non-hemolytic mucous colonies were found and further enlarged after 48 hours of culture. The capsules were identified after India ink stain. The catalase was positive, but the tube coagulase and latex coagulase were both negative. The S. aureus was identified by Vitek-2 and mass spectrometer Vitek MS V-3 IVD. The strain was confirmed by 16S rRNA gene sequencing and multilocus sequence typing (MLST). The phenotypically atypical MRSA found in the tube coagulase and latex coagulase were both negative. MRSA often show no beta hemolysis as in this case but are rarely latex coagulase-negative. We report a woman whose urine culture showed non-hemolytic, tube coagulase-negative, and latex coagulase-negative MRSA.

  • We studied the effect of Korean medicine treatment on the clearance of vancomycin-resistant enterococcus (VRE) colonization and on improvement in symptoms in a stroke patient. The patient was continuously followed up with stool VRE cultures while being treated with herbal medicine, Chunglijagam-tang, acupuncture, and oxygen therapy. We also checked the coccygeal pressure sore condition and the oxygen levels during therapy. No VRE colonization was found after treatment and the oxygen levels and pressure sores had improved. This study proved that Korean medicine treatment could be effective for the clearance of VRE colonization and improvement of symptoms.

  • The purpose of this study was to examine the differences and the relationship between basic psychological needs and exercise addiction and to identify effects of To do this, the data were collected using the self-evaluating questionnaires on the exercise addiction, the of basic psychological needs, and the subjective were distributed to 305 participants in general exercise enthusiast for all, and the collected data. This study used the SPSS Statistics 18.0 program to process data. Exploratory factor analysis was performed for validity and Cronbach's α was performed for reliability. In addition, Frequency Analysis was carried out to analyze the demographic characteristics and Pearson's Correlation Analysis, independent t-test, one-way ANOVA, correlation analysis, multiple regression analysis Analysis were performed to verify the correlations and differences among variables. The significance level in this research was set at=.05 and the followings α are derived outcomes. The following is the comprehensive finding of the effect of general exercise enthusiast exercise addiction through basic psychological needs. First, there was no significant difference in basic psychological need and Exercise Addiction of general characteristics, but participation in 6 days was the highest in exercise amount per week. Second, Basic Psychological Need(autonomy, competition, relatedness) showed a high difference in Exercise Addiction(withdrawal symptoms, exercise need, emotional attachment). Third, correlation analysis between all variables showed statistically significant difference. Fourth, Basic Psychological Need(autonomy, competition) was found to have a high effect on Exercise Addiction(withdrawal symptoms, exercise need, emotional attachment).

Isolation and Identification of Fungi and Yeast Contaminated in Rice Cake (Garaetteok) (가래떡에 오염된 곰팡이와 효모의 분리 동정)

  • The purpose of this study was to analyze the hazard of fungi in Garaetteok (Korean rice cake) by isolating and identifying of fungi contaminated with Garaetteok and investigating the possibility of mycotoxin production. Garaetteok used in this study were the ones that were returned back to the manufacturers in Jeollanam-do due to the presence of foreign matters presumed to be fungi. The fungi foreign matter was collected and inoculated on Potato dextrose agar, Malt extract agar, and Czapek yeast extract agar, and then cultured at 25℃ for 7 days. The micro-structure was observed under an optical microscope for the colonies in which pure isolation was confirmed. The gene sequencing of the product of amplified PCR was analyzed using the ITS primer. Colony-1 and 2 maintained the same properties in each tray, confirming that they were purely isolated. Budding cells were observed from the Colony-1, thus, it was determined to be yeast. Colony-2 was determined to be a fungus that belongs to Fusarium spp. as fusiform conidia were observed. As a result of gene sequencing, a total of 76 cases of fungi of Fusarium spp. were found, among which Fusarium solani was the most observed cases (53 cases). From the morphological and genetic identification, Colony-2 was identified as Fusarium spp., specifically, Fusarium solani. The fungi found in Fusarium spp. produce mycotoxins such as nivalenol, zearalenone, and fumonisin, which may cause vomiting, diarrhea, and cancer. Conclusively, the results confirm the possibility of mycotoxin production by Fusarium spp. isolated from Garaetteok. Consequently, when an unknown fungus was found, it is necessary to isolate and identify the fungus, determine whether it is a mycotoxin producing species, and strengthen relative administrative measures, accordingly.
