To estimate microbial contaminant loading discharged from diffuse sources, rainfall runoff of six rainfall events were monitored at three study watersheds of forestry and agricultural land use. Monitored indicator microorganism constituents were total coliform (TC), fecal coliform (FC), Escherichia coli (EC), and fecal streptococcus (FS). Soil loss during elevated flow rate caused higher suspended solid concentrations. Indicator microorganism concentrations were closely related with flow rate. TC event mean concentration (EMC) from unpolluted forestry was $5.3{\times}10^3CFU/100ml$, FC EMC was $1.4{\times}10^3CFU/100ml$, EC EMC was $1.1{\times}10^3CFU/100ml$, and FS EMC was $2.9{\times}10^2CFU/100ml$. From a watershed with agricultural-forestry land use, TC EMC was $1.7{\times}10^5CFU/100ml$, FC EMC was $8.5{\times}10^4CFU/100ml$, EC EMC was $8.9{\times}10^4CFU/100ml$, and FS EMC was $3.4{\times}10^4CFU/100ml$. Mixed land use of agricultural-forestry with bigger area, TC EMC was $1.9{\times}10^5CFU/100ml$, FC EMC was $9.6{\times}10^4CFU/100ml$, EC EMC was $7.0{\times}10^4CFU/100ml$, and FS EMC was $5.1{\times}10^4CFU/100ml$.
Hyporheic zone is an area where hydraulic exchanges occur between surface water and ground water. Such transient area is anticipated to facilitate diverse biogeochemical reactions by providing habitats for various microorganism. However, only a few data are available about microbial properties in hyporheic zone, which would be important in better understanding of biogeochemical reactions in whole streams. The study site is Naesung stream, located in the north Kyoung-Sang Province, of which sediment is sandy with little anthropogenic impacts. Soil samples were collected from a transect placed perpendicular to stream flow. The transect includes upland fringe area dominated by Phragmites japonica, bare soil, and soil adjacent to water. In addition, soil samples were also collected from downwelling and upwelling areas in hyporheic zone within the main channel. Soils were collected from 3 depth in each area, and water content, pH, and DOC were measured. Various microbial properties including extracellular enzyme activities ($\beta$-glucosidase, N-acetylglucosaminidase, phosphatase and arylsulfatase), and microbial community structure using T-RFLP were also determined. The results exhibited a positive correlation between water content and DOC, and between extracellular enzyme activities and DOC. Distinctive patterns were observed in soils adjacent to water and hyporheic zone compared with other soils. Overall results of study provided basic information about microbial properties of hyporheic zone, which appeared to be discernable from other locations in the stream corridor.

This study compared UV and UV/$H_2O_2$ inactivation of E.coli, a possible indicator microorganism for fecal contamination of water, and $Q{\ss}$ phage, an indicator for pathogenic viruses. UV inactivation of $Q{\ss}$, T4 and lambda phages in actual secondary effluent was investigated, too. As a result, similar inactivation efficiency between $Q{\ss}$ phage and E.coli was observed during UV treatment, while $Q{\ss}$ phage showed higher resistance to UV/$H_2O_2$ than E.coli. $Q{\ss}$ phage resistance to UV or UV/$H_2O_2$ does not reflect those of all pathogenic viruses. However, the result tells that the use of E.coli inactivation efficiency in evaluating microbiological safety of water could not always ensure the sufficient safety from pathogenic viruses. Meanwhile, $Q{\ss}$ phage showed less resistance to UV than T4 and lambda phages, indicating that the use of $Q{\ss}$ phage as an indicator virus may bring insufficient disinfection effectiveness by causing the introduction of lower UV dose than required. Consequently, it can be thought that T4 or lambda phages would be more desirable indicators in ensuring the sufficient disinfection effectiveness for various pathogenic viruses.

Biogeochemical processes in groundwater-stream water mixing zone are recently of great interest because biodegradation and natural attenuation of aquatic contaminants may occur through the processes. The objectives of this study are to investigate the hydrologic and biogeochemical processes at the groundwater-stream water mixing zone through which surface water-driven nitrate may be naturally attenuated, and to examine the effect of the vertical flow exchange flux on biogeochemical processes using correlation analysis. To examine the direction of vertical water flow in the zone, vertical hydraulic gradients were measured at several depths using mini-piezometers. Microbial populations in soil samples of the zone were also analyzed by means of the polymerase chain reaction (PCR) and Cloning methods. In addition, partial correlations among vertical flow exchange, nitrate concentration and microbial activity was investigated to examine their mutual interaction. The results showed the significant interaction among the three parameters, resulting in natural attenuation of nitrate. This study showed an example of the biogeochemical fuction of groundwater-stream water mixing zone, which can be predictable from the examination of the interaction among microbial activities, concentration of contamination and vertical flow exchange flux. temperature show a significant difference in adjacent streambed, Also, the results shows that distribution of temperature was more affected by groundwater direction than intensity of flux.

Change in microbial numbers during experimental composting has been investigated. The results show that bacteria and actinomycetes play an important role in decomposing the composting material. The number of bacteria has no relation to the efficiency of composting, though it greatly correlates to the decomposition ratio. Bacterial growth activity that shows potential of bacterial growth was originally proposed. The influence of pH and the decomposition ratio on the growth activity has been studied. It was clarified that the bacterial growth activity is useful in evaluating the efficiency of composting and the maturity of produced compost.
Hydrochemical characteristics and microbial communities of spring (Samtong), surface water, and groundwater in Cheorwon, Korea, were analyzed. Field surveys and water quality analyses were undertaken at 10 sampling points for five spring, two surface, and three groundwater samples on 15 December 2022. Hydrochemical analysis revealed that most water samples were Ca-HCO3 type and that water-rock interactions were the predominant mineral source. Radon concentrations were <1 kBq m-3 for surface water, 1~10 kBq m-3 for spring water, and 1~1,000 kq m-3 for groundwater. Microbial cluster analysis showed that the main phyla were Proteobacteria, Planctomyceta, Verrucomicrobia, Acidobacteria, and Actinomycetota.Non-metric multidimensional scaling (NMDS) analysis indicated that water temperature, pH, and Si content were closely related to microorganism content. NMDS and canonical correspondence analysis results revealed that environmental factors affecting spring water were temperature, and Mg and Si concentrations, particularly for Acidobacteria and Proteobacteria, and Pseudomonas brenneri. Both hydrochemical and microbial community analyses yielded similar results at some spring and groundwater sampling points, likely due to the effects of a basalt aquifer.

We measured the influence of antifungal antagonists Bacillus subtilis AH18 and Bacillus licheniformis K11 on soil microbial community in microcosms. Both antifungal antagonists were confirmed to suppress hot pepper phytophthora blight. Phospholipid fatty acids (PLFA) were analyzed to investigate the soil microbial community. B. subtilis AH18 changed the total PLFA composition and bio-indicators of PLFA, compared with other treatments. B. subtilis AH18 decreased the proportion of bacteria and gram negative/gram positive bacteria, and increased the fungi/bacteria and anaerobic/aerobic microorganisms. In addition cy19:0/18:$1{\omega}7c$, which means adaptation to unfavorable environmental conditions, was increased by the application of B. subtilis AH18. On the other hand the inoculation of B. licheniformis K11 or combined inoculation of both antifungal strains did not affect soil microbial community. The suppression of phytophthora blight and preservation of indigenous soil microbial community may be achieved by the combined application of B. subtilis AH18 and B. licheniformis K11.
