Analysis of Major Traits for Native Ginseng(Panax ginseng C.A. Meyer) Collected from Poonggi Area in Korea Using DNA Marker (DNA marker를 이용한 풍기 재래인삼의 주요 특성 분석)

  • In this study, a total of 25 lines consisting of five native ginseng collections from Poonggi area, five lines from Geumsam area and 15 varieties were analyzed and clustered for the selection of Poonggi native ginseng in Korea using DNA markers. The results indicated that the long cluster distance were observed between the collections of 331002, 331004, 331005, 331007 and 331026 from Poonggi area, and the collections of 332009, 332021, 332046, 332050 and 332066 from Geumsan area because of the sensible differences on the number of leaves per stem, stem color and petiole color. Thus, the collections from Poonggi area with specific characters consisting of one stem per plant, five leaves per stem and broad elliptic leaflet shape were finely classified using nine primers including OPD05, OPD20, OPG17, OPH05 and so on. In this study, the collection of 331007 from Poonggi area was considered as the respective collection with above characters. Thus, the nine primers such as OPD05 and so on, will be used to select the Poonggi ginseng in the future studies.

Growth Chracteristics, Vitamin C Content and Antioxidative Activity among Local Strains of Sedum sarmentosum (돌나물 수집종간 생육특성, 비타민 C 함량 및 항산화 활성)

  • To evaluate growth chracteristics, vitamin C content and antioxidative activity of Sedum sarmentosum, 13 local strains were cultivated in the field. Keumsan, Bucheon, and Pocheon strains had narrow and oblong leaf, long hight, and slender stem compared to the other strains. Wanju and Gunsan strains were not formed flower stalks, and did not flower during the flowering time. Vitamin C contents were ranged from 407.8 to $434.4\;mg{\cdot}kg^{-1}$, and it was high in Gwangyang, Buan, Wanju, and Gunsan strains. But vita min C contents didn't show a significant difference among strains. In antioxidative activity between fresh and freeze dried shoots, electron donating abilities (EDA) were ranged from 45.0 to 66.5% in MeOH extract of fresh shoot, and from 61.3 to 66.5% in MeOH extract of freeze dried shoot. Gwangyang, Suncheon, Wando strains showed high EDA compared to the others. Vitamine C content and antioxidant activity were highest in Gwangyang strain.

  • Agromonia pilosa Ledeb. has been used as a medicinal plant in traditional folk remedy. There are few reports on classification, physiology, ecology and morphological studies of Agromonia pilosa L. in Korea. Therefore, advanced approaches on study and development with this plant would be done urgently. Present stndy was carried out to gain basic information on genetic resources and variation with collected domestic landraces through RAPD analysis in Agromonia pilosa L. Forty two collections of Agromonia pilosa L. from nation-wide area including USA one were analyzed by RAPD test. Molecular marker size by amplified DNA band pattern ranged from 300 to 2,100bp. Among the collection, two landraces of Hadong and Cheonghak-dong showed close relation in genetic similarity. Minimum and maximum value by matrix of 1-F among 26 collected landraces were figured out as 0.365 and 0.827 showing mean value for 0.624, respectively. Those landraces were classified into two groups with cluster analysis by Nei and Li's formula from RAPD-analyzed values, and considerable genetic differences were recognized between two groups.

  • In this study, the major agronomic traits were investigated and RAPD technique was applied for the analysis of the genetic relations between the native ginsengs collected from Poonggi and Geumsan provinces in Korea. The main morphological traits were measured for a total of 54 collections of native ginseng from two areas based on UPOV standard. A total of 58 collections consisting of twenty-one native ginsengs collections from Poonggi area, twenty-nine collections from Geumsan area and four varieties of P. quinquefolium, P. japonicum, Chunpoong and Hwangsuk as controls were analyzed and clustered by RAPD. The results indicated that 01-9, 01-35 and 01-44 collections from Poonggi area were grouped into Geumsan area, while 332001, 332002 and 332003 collections from Geumsan area were grouped into Poonggi area. On comparison to the similarity of Poonggi collections (73-95%), the Geumsan collections showed 65-86% similarity in the population. Thus, the cluster should be applied according to the number of stem, number of leaves per stem and leaflet shape on the regionally native ginseng collections. The fourteen primers such as OPA02, OPA07, OPC08, OPD11, OPD20 and so on, will be used to select the native ginseng in the future studies.

  • Ears collected from 500 regions were phenotypically observed for their ear and kernel characters. Characters studied included ear shape, kernel color, kernel density, ear row number, starch quality, pop corn and waxy corn, ear length, ear diameter, kernel length, kernel width, kernel weight, ear weight, and 100 kernel weight. The Korean local corn collected were mostly flint type and were very diverse in all the characters studied. The collected local corn would be very promising for providing good germplasm for corn breeding in the future.

Analysis of Genetic Relationship by RAPD Technique for Codonopsis lanceolata Trauty Collected from the Baekdoo Mountain and Korea (백두산지역과 국내 더덕 수집종의 RAPD에 의한 유연관계 분석)

  • Extracted genomic DNA from 16 accessions of Codonopsis lanceolata collected from South Korea and the Baekdoo Mt. areas of China were analyzed for their genetic relationships by RAPD. Twenty 10-mer-oligonucleotide primers having reproductive polymorphism were selected for the RAPD analysis. The size of amplified DNA was almost between 125 bp and 2.0 kbp. Sixteen collected Codonopsis lanceolata were analyzed with 20 primers which generated 73(49.3%) polymorphic bands among 148 PCR products. The mean number of polymorphic bands were 7.4 and varied $1{\sim}9$ per primer. It was, thus, demonstrated that RAPD was useful for detecting polymorphism in Codonopsis lanceolata. The range of 1-F value(genetic similarity) was from 0.682 to 0.959. These results indicate variable genetic similarities. By UPGMA (Unweighted Pair Group Method using an Arithmetic average) cluster analysis based on 1-F value, genetic distance among the 16 collected Codonopsis lanceolata was $0.133{\sim}0.400$. It was certainly classified into two groups between collected accessions from Korea and China, and the genetic distance was about 0.281. Both accessions collected from Korea and China showed miner differences, while the genetic relationships of Tonghua Xian and Liuhe Xian from China was farthest with other accessions collected.

  • To analyse the genetic relationship and intraspecific variations among the Eleutherococcus senticosus population, the polymerase chain reaction(PCR) was performed total genomic DNAs of 10 E. senticosus collections by random 10 primers. The genetic diversity and genetic distance among 10 collections of Eleutherococcus spp. were used to describe the dendrogram showing phylogenic relationship. Ten collections were classfied into two group(group I, II) at the similarity coefficient value of 0.50. Group I included E. senticosus of Bukhado(Japanese), youngwal(Korea), E. seoulense, and E. chiisanesis while group II included several internal and Russia collection. The range of polymorphism was from 66.7 to 90.9% in 87 amplified DNA fragments. The similarity value of all collections ranged from 0.41 to 0.92. The average of genetic distance was 0.61.

Yielding Ability and Characteristics of Collections of Local Sesame (Sesamum indicum L.) (참깨 지방수집종의 주요형질과 수량성에 관한 연구)

  • Fourteen hundred and one landraces of sesame cultivated in Korea were collected and evaluated for their major agronomic characters and yield potential under conditions of May 15 sowing, 50\timesl5cm planting density and 4-3-2 kg/l0a of N-P_2 $O_{5}$-K_2 O application in Suweon in 1977. Seventy nine percent of the collection had white seed coat color, and 29% was brown. One capsule type with two rooms-four cells was 75%, one capsule type with four rooms-eight cells was 20%, and three capsule type with two rooms-four cel1s were 5% of the varieties collected, But, two one-capsule type varieties with three rooms-six cells were also found. Days to flowering from emergence were long in the varieties collected in the central northern area, but short in the varieties collected in the southern area. Number of capsule per plant was more in the central northern varieties than in the southern varieties. Number of branch per plant was more in the one capsule type varieties with two rooms-four cells than in the three capsule type varieties with two rooms-four cells and in the one capsule type varieties with four rooms-eight cells. There were significant corelations between number of capsules per plant and grain yields in the one capsule type varieties with four cells. Grain yields had significant correlation with number of capsules per plant and number of branches per plant.

  • To evaluate the intraspecific variations among the Kasiogalpi(Eleutherococcus senticosus Max.) collections. randomly amplified DNA polymorphisms were examined. Twenty primers from 90 primers applied were selected. The range of polymorphism was $7.1{\sim}90.9%$ in 113 randomly and specifically amplified DNA fragments. Collections were divided into two major groups at the similarity coefficient value of 0.65. A considerable degree of genetic diversity was also detected among plants within the same collections. Deokyu (1, 2, 3, 4, 6), Bukhaedo (7, 8) and Odae (9, 10) col1ections showed higher degree of genetic similarity with a value of $0.65{\sim}0.86$, while Deokyu 5 showed much lower genetic similarity than other col1ections.

Morphological diversities of leaf in Korean Native Orostachys Species (자생 바위솔속(Orostachys) 엽의 형태적 다양성)

  • Korean native Orostachys species collected from 22 regions investigated morphological characters.O. japonicus species(No.3∼No.9) were similar leaf morphology and characterized by lanceolate leaves, cuspidate leaf apex.O. malacophyllus species(No.12∼No. 17) were morphologically characterized by obovate leaves and acute leaf apex. This species could be differentiate from other species by no thorn and leaf outlines formed by densely populated red dots.O. iwarenge species(No.18∼No.22) were diversified, such as obovate and elliptical leaves with acute, obtuse and round leaf apices. However, this species could be differentiate from other species bl'no thorn and grey powdery green colored leaves. Species collected from Maemuldo(No. 10) and Pocheon(No. 11) was assumed that these 2 species could be the new species which were not named classified.
