Effect of Perenniporia fraxinea on Eliminating Urushiol from Rhus verniciflua Stokes Stem Bark (아까시재목버섯이 옻나무 껍질의 urushiol 제거에 미치는 영향)

  • This study evaluated the raw materials for spawning Perenniporia fraxinea, to eliminate urushiol. The growth rates of spawns on grains of millet, brown rice, and wheat were 4.92±0.05, 2.20±0.03, and 1.93±0.03 mm/day, respectively, and the laccase activity was 0.86±0.02, 0.04±0.01, and 0.01±0.00 U/mL, respectively. These observations revealed millet as the most appropriate grain for spawn production in terms of growth rate and enzyme activity. Inoculation of lacquer tree (Rhus verniciflua Stokes) stem bark with millet spawns of P. fraxinea resulted in a reduction of its urushiol contents, up to 86.6% on the third day and up to 98.5% on the seventh day. The optimal period of cultivation to eliminate urushiol was three days with 68% of residual flavonoids and 42% of phenolic components. When compared to the product cultivated from liquid spawn, the millet spawn reduced the cultivation period from 10 days to 3 days for eliminating urushiol.

  • Influence of acetic acid on xylitol production from xylose using Candida parapsilosis KFCC 10875 was investigated at the different concentrations of acetic acid. Acetic acid was totally consumed below 1.0 g/l of its concentration, whereas partially consumed above 3.0 g/l and remained in the medium during xylitol fermentation. Cell growth, xylose consumption, and xylitol production decreased when acetic acid concentration was increased. Specific growth rate of cell and specific consumption rate of xylose also decreased with increasing the concentration of acetic acid. However, the xylitol yield from xylose and specific production rate of xylitol were maximum at 1.0 g/l of acetic acid. The inhibitory effect of acetic acid on xylitol fermentation increased when pH was decreased.

Low-pathogenic Pinewood Nematode Found in Dead Trees and Resistance of Pines Induced by Its Pre-inoculation (고사목에서 발견되는 저병원성 소나무재선충 및 이의 인공접종에 의하여 유도되는 소나무의 저항성)

  • Pinewood nematode (PWN: Bursaphelenchus xylophilus) is known to kill pine tree species that are indigenous to countries where the pest was inadvertently imported, but some cultures from the extraction of dead pines do not damage trees. Experiments were conducted to examine the effect of pre-inoculation of these low-pathogenic pinewood nematode on resistance of pine trees against the pest species. The pre-inoculated pine saplings showed induced resistance which lasted for a year, and repeated inoculation of these low-pathogenic nematodes enhanced tree resistance. All nematode samples extracted from dying or dead pines that had been killed not more than three months before the extraction were pathogenic, and most of those extracted from pines that had been killed 2-3 years before were low-pathogenic. When inoculated in pine saplings, number of low-pathogenic nematodes settled, as studied two days after inoculation, was not different from that of pathogenic ones. However, as studied after 30 days of inoculation, rate of reproduction in low-pathogenic nematodes was far lower than that of pathogenic nematodes. The rate of reproduction of several nematode isolates growing on fungal mat media of Botrytis cinerea varied, but three of four low-pathogenic isolates showed same level of reproduction rates as pathogenic ones.

Composition of a Medium for Serum-free Culture of an Adipose-derived Stem Cell Line Established with a Simian Virus 40 T Antigen (Simian virus 40의 T항원 도입으로 수립한 지방유래줄기세포주의 효율적인 무혈청 배양법 및 무혈청 배지조성)

  • Adipose-derived stem cells (ADSCs) are considered promising tools for tissue regeneration. However, ADSCs have very poor proliferation capacity. Therefore, fetal bovine serum (FBS) is generally added to the culture media of ADSCs. As FBS contains many uncharacterized components that may affect cellular functions, methods for serum-free cultures of ADSCs have been widely investigated. In this study, to develop an efficient method for a serum-free culture of ADSC-T, we used an ADSC line established by introducing the simian virus 40 (SV40) T gene into primary ADSCs. We then investigated the effect of amino acids, vitamins, and other components on the growth of ADSC-T. When the ADSC-T cells were plated with DMEM/F12 serum-free medium, the cells did not proliferate, and the mixture of amino acids, vitamins, and B27 supplement did not increase the growth of the cells. However, when the ADSC-T cells were provided with serum-free DMEM/F12 after they had been cultured with serum-supplemented DMEM for 24 h, the cells proliferated, and the vitamins and B27 supplement increased the cell growth. Stem-Pro serum-free medium also appeared to be useful as a suspension culture for the ADSC-T cells. The ADSC-T cells secreted large amounts of proteins of around 70 kDa. Insulin-like growth factor (IGF) and fibroblast growth factor basic (FGF basic) were secreted by ADSC-T in larger amounts in the serum-free culture than in the serum-supplemented culture.

Atmospheric Icing Effects on the Aerodynamic Characteristics and Performance of Wind Turbine Blade (풍력 블레이드의 결빙에 의한 공력특성 및 성능 변화)

  • A significant degradation in the aerodynamic performance of wind turbine system can occur by ice accretion on the surface of blades operated in cold climate. The ice accretion can result in performance loss, overloading due to delayed stall, excessive vibration associated with mass imbalance, ice shedding, instrumental measurement errors, and, in worst case, wind turbine system shutdown. In this study, the effects of ice accretions on the aerodynamic characteristics of wind turbine blade sections are investigated on the basis of modern CFD method. In addition, the computational results are used to predict the performance of three-dimensional wind turbine blade system through the blade element momentum method. It is shown that the thickness of ice accretion increases from the root to the tip and the effects of icing conditions such as relative wind velocity play significant role in the shape of ice accretion.

Effects of aquaculture conditions on blood chemistry property and lysozyme activity of starry flounder, Platichthys stellatus (양식 환경이 강도다리, Platichthys stellatus의 혈액 생화학적 성상 및 lysozyme 활성에 미치는 영향)

  • We have investigated lysozyme activity and protectivity of starry flounder, Platichthys stellatus against olive flounder pathogenic bacteria, Edwardsiella tarda, Vibrio ichthyoenteri and Streptococcus iniae in varying salinities, water temperatures and stocking density. Starry flounders were susceptible to E. tarda but not V. ichthyoenteri and S. iniae. Under low salinity condition, the lysozyme activity was decreased a little compared to the control but not significant. The physiological and immune activities were normal up to 26 ℃ culture temperature and 100% stocking density; they were compromised from 29℃ and 200%, respectively.

  • As a part of experiments aimed to reduce the acidic taste of a soured fruit wine, a selected yeast strain was studied on its morphological and physiological properties as well as its fermentative characteristics of apple wine. The results obtained were as follows. 1. The selected yeast strain was identified as a strain of Schizosaccharomyces pombe. 2. The tolerance of the strain to metabisulfite in apple juice was stronger than that of wine yeast. 3. The growth rate of the strain was more rapid than those of wine yeast and Sake yeast when cultured in apple juice at $25^{\circ}C$, but it was decreased below the levels of others when cultured at $15^{\circ}C$. 4. The levels of ethanol production and acid reduction by the strain were high when the apple wine brewed at $25^{\circ}C$, while these were dropped significantly at $15^{\circ}C$.

The Growth Kinetics of S. aureus Inoculated onto Potentially High Risk Foods in School Foodservice Operations (학교급식에서의 잠정적 위험식품에 접종된 Staphylococcus aureus의 증식변화)

  • The objective of this study was to model the kinetics of S. aureus survival on high risk foods in school foodservice operations. After inoculating S. aureus ATCC25923 onto the various high risk foods, the effects of competitive microorganism, storage temperatures($25^{\circ}C$, $35^{\circ}C$), and initial contamination levels ($1.0{\times}10^2\;CFU/g$, $1.0{\times}10^5\;CFU/g$) on the growth of S. aureus were investigated. Lag time decreased and specific growth rate increased with a storage temperature ($25^{\circ}C$<$35^{\circ}C$) and with a higher initial inoculation level ($1.0{\times}10^2\;CFU/g$<$1.0{\times}10^5\;CFU/g$). Previously it was shown that S. aureus is a weaker competitor than other organisms, but it proliferates aggressively in a noncompetitive environment. However, in our study, when S. aureus was used to inoculate japchae (glass noodles with sauteed vegetables) and meat ball, the growth of S. aureus was similar and more active with competitive organisms than that without competitive organisms. Regardless of other factors, the initial level of S. aureus was a more significant factor of the growth. High inoculation levels of S. aureus were reached at 6 log CFU/g within 3 hours. An incubation temperature of $35^{\circ}C$ and the animal protein component of menu items also were identified as significant factors influencing the growth of S. aureus. Therefore, the duration of time meals are stored before serving should be considered a critical control point. Food service providers must control time and temperature to insure the safety of cooked foods.

  • The stabilities of plasmid vectors in Zymomonas mobilis were tested in batch and continuous cultures. It was found that the growth of the host Zymomonas strain was greatly affected by the size of plasmids as well as the composition of nutrient media: the host cells grew taster when harboring plasmids of smaller sizes and in n non-selective medium. All the Zymomonas plasmid vectors containing antibiotics selective markers and Zymomonas replication origins could be maintained in a stable manner over 30 generations without being integrated into host chromosomes.

Maximization of cell growth and polysaccharide production from Agaricus blazei by fed-batch cultivation

  • In order to maximize the cell growth and the polysaccharide production in Agaricus blazei, two kinds of fed-batch fermentation processes were performed with varying the feeding medium compositions and the feeding process. The relationship between dissolved oxygen and polysaccharide production in batch fermentation was applied to fed-batch fermentation. The biomasss concentration was 18.2 g/L and the polysaccharide production was 10.4 g/L.
