Antitumor and Immuno-potentiating activity against Mouse Sarcoma 180 by Crude Polysaccharides from Fruiting Body of Lentinus giganteus (대향고(Lentinus giganteus)의 자실체에서 추출한 조다당류가 생쥐의 Sarcoma 180에 미치는 항암 및 면역 증강효과)

  • Lentinus giganteus, one of edible and medicinal mushroom belongs to Pleurotaceae of Agaricales, has been known to contain some inhibitive substances on Sarcoma 180 and curative effect on high blood pressure. Neutral saline soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting body of the mushroom. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180 and HepG2 at the concentration of $10{\sim}2,000\;{\mu}g/ml$, but crude polysaccharides from Fr. NaCl was toxic to NIH3T3 at the concentration of $10{\sim}2,000\;{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides showed life prolongation effect of $14.3{\sim}67.5%$ in mice previously inoculated with Sarcoma 180, respectively. Fr. NaCl exhibited the immuno-potentiating activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.53{\sim}1.68$ folds compared with control at the concentration of $50{\sim}200\;{\mu}g/ml$, respectively. The numbers of peritoneal exudate cells and circulating leukocytes were increased by 7.7 and 1.6 folds by injecting Fr. NaCl and Fr. MeOH into the mice at the concentration of 50 mg/ml body weight, respectively.

Soil Salinity Influencing Plant Stands on the Reclaimed Tidal Flats of Kyonggi-Bay in the Midwestern Coast of Korea (우리나라 중서부 해안 경기만 간척지에서 식생 분포에 대한 토양 염도의 영향)

  • To identify controlling factors for spatial variation of vegetation in reclaimed tidal flats, plant stands were investigated in a newly reclaimed as well as three matured tidal flats, and a natural tidal flat in the midwest coast of Korea. Electrical conductivity of saturated soil extract (ECe) was measured to assess soil salinity. Soil salinity differed significantly among plant stands. Depending on soil salinity, plant species showed different niches: glycophyte predominated low saline spots, halophyte predominated high saline spots. Soil salinity for each plant habitats was in order of as follow: bare soil or plant wilted > mixed pioneer halophyte > pioneer halophyte > mixed with pioneer halophyte and facultative halophyte > mixed facultative halophyte > facultative halophyte > mixed with facultative halophyte and glycophyte > glycophyte > mixed glycophyte stands. These results suggested that plant distribution might have been influenced by spatial edaphic gradient (soil salinity), and thus it could be utilized as an indicator for field soil salinity gradient. Relationship between soil salinity and plant distribution was not different among the aged reclaimed tidal flats, suggesting that the vegetative population might have changed into a similar direction since the reclamation.

Immuno-stimulating and Antitumor Effects of Crude Polysaccharides Extracted from Fruiting Body of Grifola frondosa (잎새버섯(Grifola frondosa)의 자실체에서 추출한 조다당류의 면역증강 및 항암효과)

  • 80% methanol and 0.9% neutral saline soluble and hot water substances (hereinafter referred to Fr. NaCl, Fr. HW and Fr. MeOH, respectively) were extracted from fruiting bodies of Grifola frondosa. In vitro cytotoxicity tests, crude polysaccharides were not cytotoxic against cancer cell lines such as Sarcoma 180 and RAW 264.7 at the concentration of 10~2000 ${\mu}g/ml$, but crude polysaccharides from Fr. NaCl was slightly toxic to HT-29 and NIH3T3 at the concentration of 2000 ${\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of 25.0~52.9% in mice previously inoculated with Sarcoma 180. Fr. HW increased the numbers of spleen cells by 1.3 fold at the concentration of 200 ${\mu}g/ml$ compared with control. Fr. NaCl improved the immuno-stimulating activity of B lymphocyte by increasing the alkaline phosphatase activity by 1.5 fold compared with control at the concentration of 200 ${\mu}g/ml$. 10~14 ${\mu}M$ of nitric acid were generated when Fr. NaCl was added to RAW 264.7 at the concentration of 50~500 ${\mu}g/ml$, while the control group produced 4.3 ${\mu}M$ of nitric oxide. The Fr. NaCl, Fr. HW and Fr. MeOH increased the production of TNF-${\alpha}$, IL-$1{\beta}$, Il-2 and IL-6 by more than 1.4 times compared with the control group. The Fr. of MeOH increased the numbers of peritoneal exudate cells and circulating leukocytes by 3.0 and 2.0 folds compared with the control at the concentration of 50 ${\mu}g/ml$, respectively. Therefore, the crude polysaccharides extracted from fruiting bodies of Grifola frondosa could improve antitumor activity of mice.

Studies on the synthesis and characteristics of calcium sulfonate complex grease (칼슘 술폰산염 컴플렉스 그리스 합성과 특성 연구)

  • In this study, we synthesized a calcium sulfonate complex grease and a lithium complex grease to investigate their physical, rheological and tribological properties. The thermal stability of the calcium sulfonate was higher than $300^{\circ}C$ and the lithium complex grease was $245^{\circ}C$ in the dropping point test. In the grease viscosity measurement, the calcium sulfonate complex grease was measured as $7.0Pa{\cdot}s$ and the lithium complex grease was as $4.5Pa{\cdot}s$. Therefore, it was confirmed that the calcium sulfonate complex grease is superior to the lithium complex grease in terms of thermal stability and cohesiveness. In the 4-ball wear test, the calcium sulfonate complex grease was measured to be 0.43 mm and the lithium complex grease to 0.85 mm. In the 4-ball extreme pressure test, calcium sulfonate complex grease was measured as 620 kgf and the lithium complex grease was as 125 kgf. Therefore, it was confirmed that the calcium sulfonate complex grease is superior to the lithium complex grease in abrasion resistance and load-bearing property. It was found that the calcium sulfonate complex grease is more effective than the lithium complex grease in the lubrication at high temperature and high load.
